Enhanced cellular compatibility of chitosan/collagen multilayers LBL modified nanofibrous mats

Fangfang Dai,Jia Yu,Mengqin Yuan,Zhimin Deng,Yanqing Wang,Yaqi Fan,Hongbing Deng,Yanxiang Cheng

Developing novel, lightweight and biocompatible mesh for pelvic organ prolapse, a gynecological disease, has attracted the global interest of biomedical researchers. In our study, polycaprolactone and nylon 6 mats with excellent mechanical properties were fabricated via co-electrospinning. Positively charged chitosan with antimicrobial properties and negatively charged collagen with good biocompatibility were then assembled on the nanofiber mats via layer by layer (LBL) technology. SEM and mechanical property tests showed that LBL deposition increased the nanofibers’ diameter and elastic modulus. To evaluate the successful deposition process, the zeta potential, The Fourier-transform infrared, X-ray diffraction, and X-ray photoelectron spectroscopy analyses were conducted to systematically study the mats’ chemical characterization. The results indicated that the mats’ composition and structure significantly changed after LBL modification. Cell biology experiments demonstrated that the LBL-structured mats can facilitate cell growth, proliferation, and adhesion. Antibacterial tests confirmed their excellent antibacterial activity. Chitosan/Collagen-coated Polycaprolactone/Nylon 6 mats demonstrate considerable potential as implant materials for pelvic reestablishment in pelvic organ prolapse patients.

LBL deposition of chitosan/heparin bilayers for improving biological ability and reducing infection of nanofibers

Long Li,Xianguo Wang,Dan Li,Jinfa Qin,Ming Zhang,Kaijie Wang,Jinping Zhao,Li Zhang

The complexity of cardiovascular disease requires that the materials for preparing vascular grafts possess good biocompatibility, high mechanical property, and even some excellent additional properties. In this study, polycaprolactone (PCL) with good mechanical property and natural source silk fibroin (SF) were electrospun into PCL/SF nanofibers to obtain the nanofibrous substrate. With the addition of SF, the mechanical property of PCL/SF nanofibrous mats was maintained to a certain extent. While, the hydrophilicity of PCL/SF nanofibrous mats was greatly improved which is more suitable for immersive layer-by-layer assembly (LBL). The oppositely charged heparin (Hep) and chitosan (CS) were alternatively deposited on the surface of PCL/SF nanofibers via LBL. After implanting human umbilical vein endothelial cells (HUVEC) on the LBL-structured nanofibrous mats for 48 h, it was confirmed that the CS/Hep bilayers enhanced the biocompatibility of the nanofibers. Furthermore, the results of the antibacterial test showed that the antibacterial effects of the LBL-structured nanofibrous mats for Escherichia coli and Staphylococcus aureus were both achieved 95% when the number of Hep/CS bilayer was 10. It can be demonstrated that the LBL-structured nanofibrous mats with improved biocompatibility and reduced infectivity had been prepared successfully, and can be potentially used in vascular grafts.

Long-term antibacterial activity by synergistic release of biosafe lysozyme and chitosan from LBL-structured nanofibers

Jun Wu,Fangtian Liu,Chaoji Chen,Ze Zhao,Yumin Du,Xiaowen Shi,Yang Wu,Hongbing Deng

Biosafe antibacterial agents are urgently demanded in treating infection especially chronic infection. However, efficient and controlled release of those agents remains great challenging. Two nature-derived agents, lysozyme (LY) and chitosan (CS), are selected to establish a facile method for long-term bacterial inhibition. We incorporated LY into the nanofibrous mats, then deposited CS and polydopamine (PDA) on the surface by layer-by-layer (LBL) self-assembly. In this vein, LY is gradually released with the degradation of nanofibers , and CS is rapidly disassociated from the nanofibrous mats to synergistically result in a potent inhibition against Staphylococcus aureus ( S. aureus) and Escherichia coli ( E. coli) over a period of 14 days. Besides long-term antibacterial capacity, LBL-structured mats could readily achieve a strong tensile stress of 6.7 MPa with an increase percentage of up to 103%. The enhanced proliferation of L929 cells arrives at 94% with help of CS and PDA on the surface of nanofibers. In this vein, our nanofiber has a variety of advantages including biocompatibility, strong long-term antibacterial effect , and skin adaptability, revealing the significant potential to be used as highly safe biomaterial for wound dressings.

Biofunctionalization of electrospun fiber membranes by LbL-collagen/chondroitin sulfate nanocoating followed by mineralization for bone regeneration

Jing Zheng,Naureen Rahman,Longfei Li,Jingshuang Zhang,Haozhe Tan,Yun Xue,Yu Zhao,Jiliang Zhai,Nana Zhao,Fujian Xu,Liqun Zhang,Rui Shi,Yuri Lvov,Jiajia Xue

It is of great significance to develop osteoinductive artificial scaffold for bone repair and regeneration. We constructed a biomimetic apatite interface on electrospun polycaprolactone fibers by combining layer-by-layer (LbL) nanocoating with mineralization to fabricate an osteoinductive artificial scaffold. After polydopamine modification, cationic type-І collagen and anionic chondroitin sulfate were sequentially adsorbed on the fiber surface. The fibers coated with the multilayer components served as the precursor matrix to induce apatite deposition. By adjusting the number of the layers and duration of mineralization, the nanoscale morphology of composite fibers was optimized. When ten bilayers of the collagen and chondroitin sulfate were deposited onto the fibers followed by one day-mineralization, the obtained polycaprolactone–apatite composite scaffolds significantly promoted the adhesion, proliferation, and osteogenic differentiation of MC3T3-E1 cells. In a subcutaneous implantation in mice, this composite fiber membrane enhanced in vivo ectopic osteogenesis. Our nano-architectural scaffolds were able to mimic the composition and structure of the bone matrix to a certain extent, holding great potential for bone repair and regeneration.

Glucosamine/collagen assembled biomimetic nanofibrous mats via LBL deposition for cartilage engineering

Guomin Wu,Xiao Ma,Yiding Wang,Le Fan,Yining Wang,Hongbing Deng

In order to highly simulate the chondrocyte extracellular matrix, electrospinning and layer-by-layer (LBL) self-assembly techniques were applied in this study. Polycaprolactone (PCL) with good mechanical property was selected as the substrate, while D-glucosamine sulfate (GAS) and collagen type I (COL) with chondroprotective properties were coated on the surfaces of PCL nanofibers. The morphology, chemical and physical investigations suggested the successful deposition of GAS/COL, improved mechanical properties and significantly ameliorated surfaces hydrophilicity after LBL medication. The release of GAS/COL was proved to be slow and sustainable. Besides, CCK-8 assay and cell morphology observation manifested that LBL structured mats were more suitable for rat articular chondrocytes (rACs) adhesion and proliferation, while quantitative Real-Time polymerase chain reaction analysis and glycosaminoglycans (GAG) production measurement indicated the introduction of GAS/COL upregulated glucuronosyltransferase I (GlcAT-I) gene expression and GAG content in rACs. Additionally, LBL structured mats induced GlcAT-I expression and reduced interleukin‐1β (IL‐1β) and Matrix metalloproteinases 13 (MMP13) expression in IL‐1β-induced rACs. Moreover, subcutaneous model in SCID mice further confirmed that GAS/COL modified PCL mat could facilitate the GAG production and collagen type II expression of rACs.

A highly elastic conductive film prepared by bidirectional AS-LBL method

Pingwei Zhu,Zhichao He,Siqi Liu,Li Liu,Yudong Huang,Jun Li

Development of elastic conductive film is critical to prepare flexible electronic devices. Herein, bidirectional elastic conductive films based on polyurethane (PU) and different dimensional fillers were prepared by using an novel alternating stretching layer-by-layer self-assembly (AS-LBL) technology. It is shown that wrinkle structures were constructed in the conductive films, which allows them to cope with the deformation under bidirectional stretching to a certain extent, while maintaining stable conductivity. Basically, the one-dimensional MWCNTs exhibit the better construction speed of the conductive path than zero-dimensional Ag NPs . In addition, under the synergy of Ag NPs and MWCNTs, the conductivity of PU/Ag-MWCNTs films reaches the best. After adsorbing 6 layers of Ag-MWCNTs, the conductivity of the film increased to 0.24 S cm −1 . More importantly, during the stretching process, the PU/MWCNTs and PU/Ag-MWCNTs films show a more stable conductivity than PU/Ag. As the strain increases from 0 to 500%, the conductivity of PU/Ag-MWCNTs films decreases by only 9.6%. However, during the stretch-release cycle, the zero-dimensional small-sized Ag NPs display a more superior ability of conductive path reconstruction than MWCNTs. When the elongation is 100%, after 100 stretching cycles, the conductivity of PU/Ag film is only reduced by 8%.

Layer-by-Layer (LBL) Self-Assembled Biohybrid Nanomaterials for Efficient Antibacterial Applications

Yuanhao Wu,Yubo Long,Qing-Lan Li,Shuying Han,Jianbiao Ma,Ying-Wei Yang,Hui Gao

Although antibiotics have been widely used in clinical applications to treat pathogenic infections at present, the problem of drug-resistance associated with abuse of antibiotics is becoming a potential threat to human beings. We report a biohybrid nanomaterial consisting of antibiotics, enzyme, polymers, hyaluronic acid (HA), and mesoporous silica nanoparticles (MSNs), which exhibits efficient in vitro and in vivo antibacterial activity with good biocompatibility and negligible hemolytic side effect. Herein, biocompatible layer-by-layer (LBL) coated MSNs are designed and crafted to release encapsulated antibiotics, e.g., amoxicillin (AMO), upon triggering with hyaluronidase, produced by various pathogenic Staphylococcus aureus (S. aureus). The LBL coating process comprises lysozyme (Lys), HA, and 1,2-ethanediamine (EDA)-modified polyglycerol methacrylate (PGMA). The Lys and cationic polymers provided multivalent interactions between MSN-Lys-HA-PGMA and bacterial membrane and accordingly immobilized the nanoparticles to facilitate the synergistic effect of these antibacterial agents. Loading process was characterized by dynamic light scattering (DLS), transmission electron microscopy (TEM), thermogravimetric analysis (TGA), and X-ray diffraction spectroscopy (XRD). The minimal inhibition concentration (MIC) of MSN-Lys-HA-PGMA treated to antibiotic resistant bacteria is much lower than that of isodose Lys and AMO. Especially, MSN-Lys-HA-PGMA exhibited good inhibition for pathogens in bacteria-infected wounds in vivo. Therefore, this type of new biohybrid nanomaterials showed great potential as novel antibacterial agents.

Zero-order release of polyphenolic drugs from dynamic, hydrogen-bonded LBL films

Ya-Nan Zhao,Jianjun Gu,Siyu Jia,Ying Guan,Yongjun Zhang

Drug carriers capable of releasing drugs at a constant rate, or following zero-order kinetics, can lead to the best control of plasma drug concentration. Here we demonstrated that zero-order release of polyphenolic drugs, including tannic acid, epigallocatechin gallate, proanthocyanidins, and theaflavin-3′-gallate, could be achieved using hydrogen-bonded layer-by-layer films as the drug carrier. The films were fabricated using the polyphenolic drugs as hydrogen donors and polyethylene glycol (PEG) as the hydrogen acceptor. Because the drugs and PEG are bonded with reversible, dynamic hydrogen bonds, the films disintegrate gradually in aqueous solutions, and thus release the drugs into the media. Furthermore, because the PEG polymers have a narrow molecular weight distribution, the films disintegrate and release the polyphenolic drugs at a constant rate. Besides allowing for zero-order release, the drug carrier developed here also provides various ways to tune the drug release rate. The drug release rate increases with decreasing molecular weight of PEG. More importantly, the release rate could be tuned using external stimuli. Increasing the pH or temperature results in accelerated drug release, while the addition of salt retards the drug release.

Antibacterial multilayer films fabricated by LBL immobilizing lysozyme and HTCC on nanofibrous mats

Weijuan Huang,Xueyong Li,Yue Xue,Rong Huang,Hongbing Deng,Zhaocheng Ma

Cellulose acetate nanofibrous mats were prepared by electrospunning technique. Positively charged and negatively charged composites were alternately deposited on negatively charged CA mats via layer-by-layer self-assembly technique. The morphology and inhibition rate of samples were investigated by regulating number of deposition bilayers and composition of outermost layer. Field emission scanning electron microscopy images indicated that the average diameter of fibers was increased with increasing the number of coating bilayers. Additionally, the catalytic activity of immobilized LY was measured and results showed “layer-by-layer alternative oscillation” phenomenon. The results of antibacterial assay indicated that (LY–HTCC/ALG) 10.5 films coating had the best inhibitory effect.

LBL fabricated biopolymer-layered silicate based nanofibrous mats and their cell compatibility studies

Rong Huang,Yuejun Li,Xue Zhou,Qi Zhang,Huanguang Jin,Jiemin Zhao,Siyi Pan,Hongbing Deng

N-(2-hydroxyl) propyl-3-trimethyl ammonium chitosan chloride (HTCC) was synthesized from chitosan (CS). Organic rectorite (OREC) added into cellulose acetate (CA) was used to fabricate electrospun nanofibrous mats with improved thermal properties, as a result of depositing multilayers of the positively charged HTCC-OREC composites and the negatively charged sodium alginate (ALG) via layer-by-layer (LBL) technique. The morphology was affected by the number of deposition bilayers and the component of the outmost layer. Observed from the field emission scanning electron microscopy (FE-SEM) images, the LBL structured nanofibrous mats had much larger fiber sizes than CA-OREC nanofibrous mats. X-ray photoelectron spectroscopy (XPS) and X-ray diffraction (XRD) results further confirmed that HTCC-OREC was assembled on nanofibrous mats. Additionally, cell experiments and MTT results demonstrated that OREC had little effect on the cytotoxicity of LBL template, but obviously affected both the cytotoxicity and the cell compatibility of LBL structured mats when OREC was in the deposition films.

LBL structured chitosan-layered silicate intercalated composites based fibrous mats for protein delivery

Wei Li,Ruifen Xu,Liangqun Zheng,Jing Du,Yuli Zhu,Rong Huang,Hongbing Deng

Organic rectorite (OREC) was used to prepare intercalated composites with chitosan. The negatively charged cellulose acetate (CA) fibrous mats were modified with multilayers of the positively charged chitosan or chitosan–OREC intercalated composites and the negatively charged bovine serum albumin (BSA) via electrostatic layer-by-layer (LBL) self-assembly technique. The morphology and protein delivery properties of the resultant samples were investigated by regulating the number of deposition bilayers, the outermost layer and the composition of coating bilayers. The thickness of LBL films coated CA mats increased as the number of bilayers was increased and OREC was added. X-ray photoelectron spectroscopy indicated that chitosan and OREC were deposited on CA fibers. Small angle X-ray diffraction patterns showed that OREC was intercalated by chitosan. The in vitro BSA encapsulation and release experiments demonstrated that OREC could affect the degree of protein loading capacity and release efficiency of the LBL films coating.

The effect of polysaccharide types on adsorption properties of LbL assembled multilayer films

Jie Xu,Lixing Yang,Xiaoxia Hu,Shimei Xu,Jide Wang,Shun Feng

Three types of biocompatible films were fabricated via electrostatic layer-by-layer (LbL) adsorption of oppositely charged cationic polyurethane and anionic polysaccharides with different primary structures, including sodium hyaluronate, sodium carboxymethyl cellulose and sodium alginate. The adsorption behaviors of films were investigated by using the cationic dye methylene blue (MB) as a model drug at various pH values and salt concentrations. The relationship between the type of polysaccharide and the adsorption behavior of LbL films was comparatively studied. It was found that the adsorption capacity increased with an increase of the initial concentration of MB in the concentration range of the experiment to all of the films, and the pH of environment ranged from 3.0 to 9.0. The Langmuir equation fit perfectly to the experiment data. In addition, a pseudo second-order adsorption model can well describe the adsorption behaviors of MB for three films. The results showed that the type of side chains and the charge density of the polysaccharides played key roles in the adsorption properties of the PU/polysaccharide multilayer films.
