Structural characterizations and α-glucosidase inhibitory activities of four Lepidium meyenii polysaccharides with different molecular weights

Wen Luan,Wu Zhou-Wei,Lin Li-Wu,Al-Romaima Abdulbaset,Peng Xing-Rong,Qiu Ming-Hua

Four polysaccharides (MCPa, MCPb, MCPc, MCPd) were obtained from Lepidium meyenii Walp. Their structures were characterized by chemical and instrumental methods including total sugar, uronic acid and protein content determination, UV, IR and NMR spectroscopy, as well as monosaccharide composition determination and methylation analyses. Four polysaccharides were a group of glucans with different molecular weights ranging from 3.12 to 14.4 kDa, and shared a similar backbone chain consisting of (1→4)-glucose linkages with branches attached to C-3 and C-6. Furthermore, bioactivity assay showed that MCPs had concentration-dependent inhibitory activity on α -glucosidase. MCPb (Mw = 10.1 kDa) and MCPc (Mw = 5.62 kDa) with moderate molecular weights exhibited higher inhibitory activity compared with MCPa and MCPd. Graphical Abstract

The structures of two polysaccharides from Lepidium meyenii and their immunomodulatory effects via activating NF-κB signaling pathway

Zhou-Wei Wu,Xing-Rong Peng,Xiao-Cui Liu,Luan Wen,Xin-Yu Tao,Abdulbaset Al-Romaima,Ming-Yi Wu,Ming-Hua Qiu

Lepidium meyenii Walp., also known as the “Peruvian national treasure”, is a popular functional food in the daily lives of Peruvian people due to its bioactive with main polysaccharides. However, studies on polysaccharides isolated from Lepidium meyenii were few. Two new highly heterogeneous polysaccharides, MCP-1a and MCP-2b, were isolated and purified from the tuber of Lepidium meyenii . The structure characterization revealed that MCP-1a primarily consisted of D-Glc and had a molecular weight of 6.6 kDa. Its backbone was composed of 1,4,6- α -D-Glc, while branches feature T- α -L-Ara, 1,5- α -L-Ara, and T- α -D-Glc attached to the O -6 positions. MCP-2b was a rare arabinogalactan with a molecular weight of 49.4 kDa. Interestingly, the backbone of MCP-2b was composed of 1,6- β -D-Gal, 1,3,6- β -D-Gal with a few 1,3- β -D-Glc p A-4-OMe units inserted. Side chains of MCP-2b were mainly composed of 1,3- β -D-Gal, T- β -D-Gal, T- α -L-Ara, 1,5- α -L-Ara, with trace amounts of 1,4- β -D-Glc and T- β -D-Glc. The bioactivity assay results revealed that MCP-1a and MCP-2b increased the release of NO, IL-1 β , TNF- α , and IL-6 from RAW 264.7 cells at concentrations ranging from 50 μg/mL to 400 μg/mL. Furthermore, MCP-1a and MCP-2b could promote the expression of key transcription factors (IκB- α , p-IκB- α , p65, and p-p65) in the NF-κB pathway, indicating that MCP-1a and MCP-2b had potential immunomodulatory activities.

Micelle nanovehicles for co-delivery of Lepidium meyenii Walp. (maca) polysaccharide and chloroquine to tumor-associated macrophages for synergistic cancer immunotherapy

Ye Yang,Tingting Guo,Junwei Xu,Yin Xiong,Xiuming Cui,Yang Ke,Chengxiao Wang

Here, we fabricated amphiphilic polysaccharide micelles for synergistic cancer immunotherapy targeting tumor-associated macrophages (TAMs). Lepidium meyenii Walp. (maca) polysaccharide (MP), a naturally derived macromolecule with a strong TAM-remodeling effect, was grafted on a hydrophobic poly(lactic- co -glycolic acid) (PLGA) segment, with a disulfide bond for redox-sensitive linkage. The amphiphilic polysaccharide derivatives could self-assemble into core (PLGA)–shell (MP)-structured micelles and encapsulate chloroquine (CQ) into the hydrophobic core. By using a 4T1-M2 macrophage co-culture model and a 4T1 tumor xenograft mouse model, we showed that the prepared micelles could co-deliver MP and CQ to the tumor sites and selectively accumulate at TAMs because of the specific properties of MP. Furthermore, the nanoparticles exerted synergistic tumor immunotherapeutic and antimetastatic effects, which might be attributable to the enhanced cell internalization of the micelles and the multiple regulatory mechanisms of MP and CQ. Thus, immunomodulatory MP may be a promising biomaterial for cancer immunotherapy.

en This link goes to a English section pt This link goes to a English section Maca ( Lepidium meyenii Walp) polysaccharides (MP) with purity of 99.2% were obtained to investigate their structural characteristics and antifatigue effect in vivo . The physicochemical properties of MP were analyzed through high-performance gel filtration chromatography, IR, monosaccharide composition, methylation, GC-MS, and NMR analyses. The antifatigue effect of MP was evaluated by using a mouse weight-loaded swimming model. MP is an acidic heteropolysaccharide with an average molecular weight ( M w ) of 793.5 kDa. It is composed of D-GalA: D-Glc: L-Ara: D-Man: D-Gal: L-Rha = 35.07:29.98:16.98:13.01:4.21:0.75 (mol, %). The findings revealed that MP contained β-1,3-Gal p (A), β-1,3-Glc p , and α-1, 3-Man p linked alternatingly to form a backbone (5:4:1). MP (above mid-dosage 50 mg/kg bw/d) could effectively elongate swimming durations and accelerate average swimming speeds (within the 1st 5 min) of mice ( P < 0.05) and improve the serous biochemical parameters of mice. Compared with the control model, high-dosage (100 mg/kg bw/d) MP treatment could significantly enhance glutathione peroxidase and creatine kinase activities ( P < 0.05) and decreased lactate dehydrogenase activity ( P < 0.01). High-dosage MP could significantly reduce the levels of blood urea nitrogen, lactic acid, and malondialdehyde ( P < 0.05). MP is an acidic polysaccharide with a high D-GalA content, which could be responsible for the antifatigue effect of maca. Practical Application en This link goes to a English section pt This link goes to a English section The antifatigue activity of polysaccharides isolated from maca ( Lepidium meyenii Walp) was investigated in this study. Polysaccharides, as a major component of maca (above 10% in dry weight), could be explored as a natural antifatigue functional food in the future.

Evaluation of Biological Activity of a Lepidium meyenii (Maca) Polysaccharide-Zinc Complex

Xiufeng Yang,Rui Chen

Zinc, an important metal element in the human body, fulfills multiple biological roles, such as influence on cell division and differentiation. In this paper, a Maca polysaccharide-zinc (MPS-Zn) complex was prepared by water extraction and alcohol precipitation process. A granular MPS-Zn complex with 0.2360 μg/mg Zn was obtained at 60°C and pH = 3.0 for 60 min. The antioxidant activities of this MPS-Zn complex were evaluated by the free radical scavenging activity toward DPPH and ABTS. The rate of ABTS free radical scavenging of 4.0 mg/mL MPS-Zn complex is equal to that of control group (vitamin C). The abilities of this complex against three cancer cell lines including breast cancer MCF-7, colon cancer SW480, and liver cancer SMMC-7721 were used to evaluate its antiproliferative activity by MTS assay. The results demonstrate that this complex has an inhibitory effect on the above cancer cell lines. Especially, the inhibitory effect of it on colon cancer SW480 is more effective than that of cisplatin. The toxicity of this complex on human normal pulmonary epithelial cell BEAS-2B is lower than that of taxol and cisplatin. This complex has the potential use as the zinc-containing nutritional food and/or anticancer drug.

Discovering anti-osteoporosis constituents of maca (Lepidium meyenii) by combined virtual screening and activity verification

Hao Liu,Wenwen Jin,Chunhua Fu,Penfei Dai,Yuantao Yu,Qin Huo,Longjiang Yu

Maca ( Lepidium meyenii Walp.) is a traditional medicine and nutritional supplement from South America with many pharmacological effects, such as enhancing female and male fertility, improving sexual dysfunction, preventing osteoporosis and relieving menopausal syndrome. Until now, the active principles behind most of these effects have not been clarified, severely hindering the exploitation and application of maca products. In this study, the effective anti-osteoporotic components of maca are uncovered. Through virtual screening against estrogen receptor and verification of pharmacological activity on osteoblasts, we found that N-benzyl-palmitamide is an active constituent of maca in preventing osteoporosis. As well, N-benzyl-palmitamide promotes osteoblast proliferation, differentiation and mineralization, plus it benefits bone formation through enhanced expression of osteogenesis-related genes, such as bone morphogenetic protein-2, core binding factor alpha 1, type 1 collagen and alkaline phosphatase. The effects of N-benzyl-palmitamide on enhanced bone formation are very likely through the estrogen receptor pathway, as N-benzyl-palmitamide also increases the expression of ERα and ERβ genes. Clarifying single-component biological activity will greatly enable exploitation and application of maca as a medication and health supplement to prevent osteoporosis.
