Mechanism of Ligilactobacillus salivarius GX118 in Regulating the Growth of Rainbow Trout (Oncorhynchus mykiss) and Resistance to Aeromonas salmonicida Infection

Xiyu Cao,Bowen Wang,Wenhao Ren,Jiang Wang,Qi Liu,Yichao Ren,Xianliang Meng

Lactic acid bacteriaLigilactobacillus salivariushas been shown to be a substitute for antibiotics in the treatment of bacterial disease in high animals. However, its beneficial mechanism in fish farming is still unclear. This study evaluated the antagonistic effects of theLigilactobacillus salivariusGX118 strain onAeromonas salmonicidaand its regulation of rainbow trout growth in vivo and in vitro. The results found that GX118 produces an antibacterial substance that can directly destroy the cell wall ofA. salmonicida. Whole-genome sequencing of GX118 revealed that Enterolysin A is a type III bacteriocin with antibacterial properties. An in vivo experiment showed that the supplementation of GX118 in diet competitively inhibited the colonization ofA. salmonicidain the intestine. In addition, it was able to improve the growth performance of rainbow trout within a 21-day feeding experiment. The supplementation of GX118 increased the diversity of gut microbiota, in which the abundance ofBacteroidota,Blautia, andRhodobacteraceaeincreased. In addition, the use of GX118 activated the expression ofIFN-γandNF-κBgenes and reduced the expression level ofIL-6andIL-8, thus exhibiting a certain effect on activating the immunity of rainbow trout. This study provides a scientific basis for the development of antibacterial probiotics in the healthy farming of rainbow trout.

Improved functionality of Ligilactobacillus salivarius Li01 in alleviating colonic inflammation by layer-by-layer microencapsulation

Yao Mingfei,Lu Yanmeng,Zhang Ting,Xie Jiaojiao,Han Shengyi,Zhang Shuobo,Fei Yiqiu,Ling Zongxin,Wu Jingjing,Hu Yue,Ji Shouling,Chen Hao,Berglund Björn,Li Lanjuan

The low viability during gastrointestinal transit and poor mucoadhesion considerably limits the effectiveness of Ligilactobacillus salivarius Li01 (Li01) in regulating gut microbiota and alleviating inflammatory bowel disease (IBD). In this study, a delivery system was designed through layer-by-layer (LbL) encapsulating a single Li01cell with chitosan and alginate. The layers were strengthened by cross-linking to form a firm and mucoadhesive shell (~10 nm thickness) covering the bacterial cell. The LbL Li01 displayed improved viability under simulated gastrointestinal conditions and mucoadhesive function. Almost no cells could be detected among the free Li01 after 2 h incubation in digestive fluids, while for LbL Li01, the total reduction was around 3 log CFU/mL and the viable number of cells remained above 6 log CFU/mL. Besides, a 5-fold increase in the value of rupture length and a two-fold increase in the number of peaks were found in the (bacteria-mucin) adhesion curves of LbL Li01, compared to those of free Li01. Oral administration with LbL Li01 on colitis mice facilitated intestinal barrier recovery and restoration of the gut microbiota. The improved functionality of Li01 by LbL encapsulation could increase the potential for the probiotic to be used in clinical applications to treat IBD; this should be explored in future studies.

Probiotic characterization of Ligilactobacillus murinus ZQL11 isolated from healthy dog feces

Gong Duqiang,Zhang Luyao,Hong Lianhong,Mo Shengjun,Hu Ruihong,Wang Xiaonan,Zhang Jiantao

Currently, canine-derived probiotics, especially those specifically developed for dogs, remain relatively scarce. Our study aimed to isolate bacterial strains from the feces of healthy dogs and to screen for those with excellent probiotic properties. A total of 12 strains were isolated and identified through morphological observation and 16 S rRNA gene sequencing. These strains were systematically evaluated for key probiotic characteristics, including tolerance to simulated gastrointestinal conditions, antimicrobial activity, adhesion-related properties, and overall safety. Among them, Ligilactobacillus murinus ZQL11 (ZQL11) demonstrated the most promising probiotic profile. This strain exhibited strong acid tolerance, with a survival rate of 75.23% after 3 h at pH 2.0, and notable bile salt resistance, maintaining 78.86% survival after 4 h in 0.3% bile salts. Antibacterial assays revealed significant inhibitory effects against Escherichia coli , Salmonella enterica , and Staphylococcus aureus . Adhesion-related evaluations showed high cell surface hydrophobicity together with strong auto-aggregation and co-aggregation abilities. In vivo safety assessment in mice showed that ZQL11 did not induce detectable toxicity or adverse effects; no significant differences were observed in routine blood parameters between the ZQL11-treated and control groups, and no obvious histopathological lesions were found in the major organs. In summary, ZQL11 exhibited favorable tolerance, adhesion-related properties, safety, and antibacterial activity, suggesting its potential for further application in animal microecological preparation.

Elaidic acid suppresses hepatocellular carcinoma growth through modulating the production of intestinal Ligilactobacillus murinus-derived spermidine

Yini Li,Tongtong Tian,Qian Yu,Huiqin Jiang,Te Liu,Hao Wang,Ran Huo,Chenzheng Gu,Yu Liu,Ying Zhao,Chunyan Zhang,Yan Zhou,Jiyan Wang,Lin Ding,Chuyu Wang,Xinyi He,Wei Guo,Wenjing Yang,Beili Wang

Dietary intervention provides a novel approach for cancer therapy. Elaidic acid (EA), which accounts for 80-90% of total trans fatty acids in foods, has recently been found to exert anti-tumor effects. However, the biological functions and underlying mechanisms of EA remain elusive in hepatocellular carcinoma (HCC). In this study, targeted fatty acid metabolomics demonstrated that among 44 types of fatty acids, the concentration of EA decreased most significantly when comparing plasma from HCC patients with plasma from healthy people. Through in vivo assays using HCC orthotopic and xenograft mouse models, we further revealed that dietary EA attenuates HCC growth. Notably, when gut microbiota was depleted using a cocktail of antibiotics, the anti-tumor effect of EA was diminished, confirming that EA suppresses HCC tumor growth by modulating gut microbiota. Mechanistically, analysis of 16S ribosomal RNA sequencing showed that dietary EA markedly increases the abundance of intestinal Ligilactobacillus murinus (L. murinus) . Subsequent untargeted metabolomic sequencing analysis further demonstrated that dietary EA drives the production of L. murinus -derived spermidine (SPD), which attenuates HCC growth in vitro as well as in vivo . The observed impact correlated with the phosphorylation of p38 MAPK and the upregulation of biomarkers pertinent to apoptosis and proliferation, including tumor protein 53, bcl-2-associated X protein, and cysteine-requiring aspartate protease 3. Taken together, our findings highlight the important role of intestinal L. murinus -derived SPD in EA-mediated HCC suppression, thereby offering a promising dietary strategy for HCC treatment.

Probiotic extracellular vesicles from Ligilactobacillus murinus alleviate colonic injury and inflammation via miR-146a-5p-driven macrophage polarization

Donglin Sui,Shugang Li,Chunqing Ai,Shuang Song,Yixuan Li,Shouhao Zhao,Huan Li,Hong Chang,Xiaomeng Ren

Our prior studies have indicated that the isinglass polysaccharide derived from Larimichthys crocea (CIP) can enhance the growth of Ligilactobacillus murinus , which may play a significant role in mitigating colitis. This study investigated the potential of Ligilactobacillus murinus and its derived extracellular vesicles (LM-EVs) as therapeutic and prophylactic agents. LM-EVs demonstrated remarkable ameliorative efficacy in colitis models, substantially mitigating colonic injury, atrophy, and hematochezia, coupled with a significant dampening of inflammatory markers. These beneficial effects are underpinned by significant in vitro antioxidant capacity and a favorable initial safety profile, alongside marked immunomodulatory functions. Mechanistically, we propose a significant role for the miR-146a-5p/TRAF6 signaling axis in LM-EV-mediated immunomodulation, specifically its influence on macrophage polarization. Our findings indicate that LM-EVs modulate the expression level of miR-146a-5p within macrophages. miR-146a-5p subsequently targets and suppresses the TRAF6/NF-κB signaling axis, driving macrophage polarization from the M1 to the M2 phenotype, thereby effectively ameliorating colitis. These insights reveal a promising avenue, potentially utilizing probiotic extracellular vesicles for alleviating colitis and paving the way for new frontiers in gut health modulation.

Ovotransferrin Alleviates Ulcerative Colitis by Modulating Ligilactobacillus murinus-Mediated Bile Acid Metabolism via the Ptgs2/JAK-STAT Signaling Axis

Donglei Zheng,Xiaojuan Xin,Hui Ouyang,Lulu Ma,Hao Wang,Xin Liu,Xiangxin Liao,Xinzi Chu,Ying Gao,Jiaying Huo,Shugang Li

Ovotransferrin (OVT) is a functionally active protein demonstrated to alleviate ulcerative colitis (UC), although its mechanism remains incompletely understood. Notably, UC is closely associated with dysbiosis of the gut microbiota. This study investigates the potential of OVT to alleviate colitis by reshaped gut microbiota composition in a DSS-induced UC mouse model. Specifically, Ligilactobacillus murinus (L. murinus) was isolated from the gut microbiota of OVT-treated UC mice and stimulated by OVT in vitro. Integrated whole-genome sequencing and targeted metabolomics revealed that L. murinus encodes bile salt hydrolase, thereby driving secondary bile acid biosynthesis. Mechanistically, fermentation products (FP) derived from coculture of L. murinus with bile exerts potent anti-inflammatory in both Caco-2 cells and a colitis model by Ptgs2 (COX-2) downregulation and the JAK-STAT signaling pathway inhibition. These results established the “OVT-L. murinus-bile acids-Ptgs2/JAK-STAT” axis as a novel mechanism for UC treatment, providing a foundation for high-value applications of OVT in functional foods.

Synergistic Addition of Ligilactobacillus acidipiscis YJ5 to a Probiotic Consortium for Enhanced Alleviation of Loperamide-Induced Constipation in Mice

Fei Shen,Congguo Wei,Zhiguo Zhang,Yujie Zhou,Yunjing Han,Juan Du,Fengqin Feng,Guanghua He,Hao Zhong

Probiotics have emerged as a promising therapeutic strategy for relieving constipation. The strategy of employing multi-strain probiotic combinations to achieve synergistic therapeutic effects is widely adopted in commercial applications. This study demonstrates that incorporating Ligilactobacillus acidipiscis YJ5 as a synergistic keystone strain into a base three-strain probiotic consortium (CP3-L) creates a potent low-dose formulation (CP4-L) with enhanced efficacy against loperamide-induced constipation in mice. Although the CP3-L consortium ( Lactiplantibacillus plantarum SYS1 and T34, Limosilactobacillus fermentum W1) itself outperformed its constituent single strains, the CP4-L consortium provided improvement in defecation parameters, intestinal barrier integrity, and inflammatory markers. It effectively restored interstitial cells of Cajal (ICC) markers (c‑Kit/SCF) to enhance intestinal motility, downregulated colonic aquaporins to increase fecal water content, and selectively modulated the gut microbiota to raise beneficial Bifidobacterium and fecal butyrate levels. These findings validate a mechanism-guided approach to probiotic consortium design, where functional complementarity between strains yields synergistic, multi-target anti-constipation activity at a reduced dosage.

The Role of Dihydroresveratrol in Enhancing the Synergistic Effect of Ligilactobacillus salivarius Li01 and Resveratrol in Ameliorating Colitis in Mice.

Yiqiu Fei,Shuobo Zhang,Shengyi Han,Bo Qiu,Yanmeng Lu,Weixing Huang,Fang Li,Deying Chen,Björn Berglund,Hang Xiao,Lanjuan Li,Mingfei Yao

Currently approved therapeutical strategies for inflammatory bowel diseases (IBD) suffer from variable efficacy and association with risk of serious side effects. Therefore, efforts have been made in searching for alternative therapeutics strategies utilizing gut microbiota manipulation. In this study, we show that the probiotic strain Ligilactobacillus salivarius Li01 (Li01) and the phytochemical prebiotic resveratrol (RSV) have synergistic effect in ameliorating colitis in mice. Oral coadministration of Li01 (10 9  CFU/d) and RSV (1.5 g/kg/d) promoted restoration of various inflammatory injuries and gut microbiota composition, exhibiting a favorable anti-inflammatory effect in DSS-induced colitis mice. The combination treatment was associated with reductions in the levels of proinflammatory cytokines IL-1 β and IL-6 and increases in the levels of the anti-inflammatory cytokine IL-17A in mouse serum. Moreover, the combination treatment was found to alter the composition and metabolism of the gut microbiota, especially influencing the production of short chain fatty acids and anti-inflammatory related molecules. The mechanism underlying the improved anti-inflammatory effect from the RSV and Li01 combination treatment was found to be associated with the environmental sensor mammalian aryl hydrocarbon receptor (AHR) and tryptophan metabolism pathway. Administration of RSV in combination with Li01 in different mouse model led to enhanced conversion of RSV into metabolites, including dihydroresveratrol (DHR), resveratrol-sulfate, and resveratrol-glucuronide. DHR was found to be the dominant metabolite of RSV in conventional and colitis mice. An increased DHR/RSV ratio was confirmed to activate AHR and contribute to an enhanced anti-inflammatory effect. DHR is considered as a potential AHR ligand. The DHR/RSV ratio also affected the serotonin pathway by controlling the expression of Tph1, SERT, and 5-HT 7 R leading to amelioration of colitis in mice. Our data suggest that treatment with a combination of Li01 and RSV has potential as a therapeutic strategy for IBD; further investigation of this combination in clinical settings is warranted.

Ligilactobacillus murinus Confers a Dual Benefit: Counteracting Crotonis Fructus-induced Intestinal Toxicity and Synergizing with Its Processed Form against Ulcerative Colitis

Weiying Liu,Zijian Zhang,Wanxing Wu,Xiaoyu Yan,Yijia Huang,Han Feng,Qiaoxing Mou,Jie Wan,Mengfan Yan,Huayang Tang,Jie Liang,Ying Zhang,Cheng Peng,Xiaoqi Pan

Ethnopharmacological relevance Ulcerative colitis (UC) poses a major clinical challenge. Classical Chinese medical texts record the use of Crotonis Fructus (CF), the seeds of Croton tiglium L. , for treating conditions like “chronic dysentery” presenting symptoms similar to UC. However, the clinical application of both raw and processed CF is limited due to intestinal toxicity. Aim of the study This study investigates the role of gut microbiota in mitigating the intestinal toxicity induced by CF and in enhancing the anti-UC efficacy of its processed form. Methods Metagenomic analysis investigated CF-induced intestinal toxicity. The benefits of probiotics combined with CF or processed CF cream were evaluated in Caenorhabditis elegans ( C. elegans ) and a dextran sulfate sodium (DSS)-induced mouse model. Their combined effect was further assessed in DSS-exposed C. elegans , with qRT-PCR measuring intestinal barrier integrity. Results Metagenomic analysis revealed that CF-induced intestinal toxicity was associated with gut microbiota dysbiosis characterized by a pronounced reduction in Ligilactobacillus murinus ( L. murinus) . Supplementation with L. murinus alleviated CF-induced damage in C. elegans . In DSS-induced UC mice, both L. murinus and processed CF cream ameliorated colitis and suppressed TNF-α, IL-6, and IL-1β. When co-administered in DSS-exposed C. elegans , two agents acted synergistically, leading to greater restoration of intestinal barrier integrity and more pronounced upregulation of barrier-function genes. Conclusion This study demonstrates that L. murinus plays a dual role: it mitigates CF-induced intestinal toxicity and acts synergistically with processed CF cream to enhance UC treatment, providing a microbiome-based strategy for safer clinical application.

Insights into the roles of Ligilactobacillus, Tetragenococcus and Levilactobacillus in flavor metabolite formation during low-salt Doubanjiang fermentation: Pathways and mechanisms

Yaoyao Tian,Yufeng Du,Qian Yang,Chi Zhao,Fengju Zhang,Suyi Zhang,Zhihua Li,Qi Li

To address health risks of traditional high-salt Doubanjiang , reduced-salt fermentation has been adopted, often accompanied by enrichment of specific lactic acid bacteria (LAB) including Ligilactobacillus , Tetragenococcus and Levilactobacillus . Nevertheless, their key roles and potential molecular mechanisms in flavor formation remain incompletely elucidated. This study aimed to clarify the flavor formation mechanisms driven by the three LAB strains in low-salt Doubanjiang . First, sterilized low-salt Doubanjiang (10 %, w /w) was individually inoculated with our laboratory-isolated Ligilactobacillus acidipiscis YQ11, Tetragenococcus halophilus YQ19 and Levilactobacillus brevis YQ20 for axenic fermentation. Then, physicochemical analyses showed all strains significantly reduced pH and increased total acid by 12.3 %–21.5 %. Next, metabolomic profiling revealed the inoculation raised volatile flavor compounds from 9 to 34–40 types, with 23 key differential metabolites identified (VIP ≥ 1, p  < 0.05). Each strain had distinct traits: YQ11 accumulated lactic acid (7.80 mg/g) and acetic acid (15.38 mg/g); YQ19 produced smoky 4-ethylphenol and the highest benzyl alcohol (18.06 mg/kg); YQ20 excelled in ester synthesis and specifically accumulated umami L-aspartic acid. Furthermore, whole-genome sequencing and metabolic pathway analysis of the strains were conducted. Finally, integrated genomic and metabolomic analysis uncovered core flavor-related genes, revealing potential molecular mechanisms. This study clarifies the distinct roles of enriched LABs in low-salt Doubanjiang flavor development, providing core microbial resources for rational strain selection and theoretical support for precisely modulating low-salt fermented food flavor.

Exploring the anti-Listeria monocytogenes mechanisms of the postbiotics derived from Ligilactobacillus salivarius LSA-6 and its biopreservative effects on Inner Mongolian cheese

Ce Shi,Li Jia,Jinlong Xu,Na Guo,Haiying Cui,Tariq Aziz,Abdulhakeem S. Alamri,Walaa F. Alsanie,Majid Alhomrani,Changzhu Li,Lin Lin

Food contamination caused by Listeria monocytogenes has been a major concern worldwide, and postbiotics derived from lactic acid bacteria are promising in controlling foodborne pathogens, while the antibacterial effects and mechanisms are still largely unclear. This study aimed to investigate the anti - L. monocytogenes activity and mechanisms of the postbiotics derived from Ligilactobacillus salivarius LSA-6 (Postbio-LSA-6), and its potential application in Inner Mongolian cheese preservation. Results showed Postbio-LSA-6 exhibited efficient anti - L. monocytogenes activity, the minimal inhibitory concentration (MIC) was 4 mg/mL. Postbio-LSA-6 at 2MIC could significantly disrupt the bacterial membrane integrity, the membrane-damaged cells percentage increased to 49.6 %, leading to the intercellular materials leakage and cell death. Besides, Postbio-LSA-6 at 2MIC could induce the oxidative stress in bacterial cells, the membrane-fluorescence cells percentage increased to 46 %, resulting in lipid peroxidation. Untargeted metabolomics analysis showed organic acids and fatty acids such as erythronic acid, citric acid, 2-hydroxybutyric acid, and oleic acid were identified as the main differentially expressed metabolites in Postbio-LSA-6. Furthermore, Postbio-LSA-6 could effectively maintain the moisture content and pH stability, reduce the color and texture alterations, and extend the shelf life of cheese samples by suppressing microbial proliferation and lipid oxidation. These results offered new insights on the antibacterial activities of postbiotics against foodborne pathogens, and provided a low-cost, label-friendly biopreservation strategy for local dairy products preservation.

Lactobacillus rhamnosus RL-H3-005 and Pediococcus acidilactici RP-H3-006 ameliorate Atopic Dermatitis in offspring mice by modulating the gut microbiota

Mingxin Li,Shugang Li,Chunqing Ai,Wenxiu Zhu,Donglin Sui,Yixuan Li,Binbin Hou,Xiaomeng Ren

The increasing incidence of atopic dermatitis (AD) presents a public health issue, and thus there is an urgent need for safe and effective preventive strategies. In this study, a strategy to reduce the risk of AD in offspring by administering probiotics to the mother was explored. Female mice were given Lactobacillus rhamnosus RL-H3-005 (RL) and Pediococcus acidilactici RP-H3-006 (RP) during pregnancy and lactation period, and their inhibition effects on AD symptoms in offspring mice were evaluated. The results showed that supplementation with RL and RP during pregnancy and lactation period altered not only the gut microbiota community but also microbial community in milk in pregnant mice. Further study showed that pups from RL- and RP-treated pregnant mice had mild symptoms of AD, characterized by reduced epidermal thickness in the ear tissues and numbers of mast cells, which was associated with the reduction in secretory immunoglobulin A (sIgA), immunoglobulin E (IgE), IL-4, IL-5, IL-13, thymic stromal lymphopoietin (TSLP), and eosinophil cationic protein (Ecp). The underlying mechanism of RL and RP on the inhibition of AD symptom in offspring was closely associated with the alterations of the gut microbiota community in offspring, including an increase in Ligilactobacillus and Bacteroides . This study implies that RL and RP hold the potential to reduce the risk of AD in offspring via gut-mammary axis during the pregnancy and lactation period.
