The recently characterized Limosilactobacillus reuteri N1 GtfB (LrN1 GtfB) from glycoside hydrolase family 70 is a novel 4,6-α-glucanotransferase acting on starch/maltooligosaccharides with high enzyme activity and soluble protein yield (in heterogenous system). In this study, the influence of the treatment by LrN1 GtfB on the fine structure and functional characteristics of three maize starches were furtherly investigated and elucidated. Due to the treatment of LrN1 GtfB, the starch molecules were transformed into reuterans containing linear and branched (α1 → 6) linkages with notably smaller molecular weight and shorter chain length. Moreover, the (α1 → 6) linkage ratios in the GtfB-modified high-amylose maize starch (GHMS)/normal maize starch (GNMS)/waxy maize starch (GWMS) increased by 18.3 %/12.6 %/9.0 % as compared to their corresponding controls. In vitro digestibility experiment revealed that the resistant starch content of GHMS, GNMS and GWMS increased by 16 %, 18 % and 25 % as compared to the starch substrates. Furthermore, the butyric acid yielded from GHMS, GNMS and GWMS in the in vitro fermentation experiments were 1.4, 1.5 and 1.4 times higher than those of commercial galactose oligosaccharides . These results indicated that the highly-branched short-clustered reuteran synthesized by LrN1 GtfB might serve as novel potential prebiotics , and provide insights for the synthesis of promising prebiotic dietary fiber from starch.

Limosilactobacillus reuteri DSM 17938 ( L. reuteri DSM 17938) was one of the most widely used probiotics in humans for gastrointestinal disorders, but few studies have investigated its role in drug-induced liver injury (DILI). Here, we evaluated the efficacy of L. reuteri DSM 17938 using a mouse model of DILI induced by triptolide. Pregavage of L. reuteri DSM 17938 for 1 week remarkably lowered hepatic inflammatory cytokines level and oxidative stress, with diminished serum alanine transaminase and aspartate aminotransferase levels. Metabolomics and RT-qPCR analysis confirmed its ability in ameliorating TP-disrupted hepatic fatty acid β oxidation. Genome annotation of L. reuteri showed its ability to modulate energy metabolism. Targeted metabolomics demonstrated that L. reuteri DSM 17938 modified the short fatty acid profiles in cecum, especially enhancing propionate levels. Further experiments found that L. reuteri DSM 17938 can activate AMPK signaling by upregulating gut microbiota–derived propionate level, thus restoring impaired mitochondrial biogenesis and energy supply processes to recover energy homeostasis, which leads to diminished ROS production and oxidative stress injury in hepatocytes. Besides, AMPK inhibitor dorsomorphin abolished all the effects on propionate protecting mitochondria and energy metabolism. This study established probiotic therapy of L. reuteri DSM 17938 as a preventive intervention for DILI in clinical. We also revealed that L. reuteri DSM 17938 can activate AMPK signaling by propionate, facilitating a deeper understanding of the action mechanism of L. reuteri DSM 17938 against acute liver injury and contributing to the development of its postbiotics and wider applications. Graphical Mechanism illustration of Limosilactobacillus reuteri DSM 17938 alleviating TP-induced liver injury by enhancing AMPK signaling pathway via propionate (Created with Biorender. com).

Limosilactobacillus reuteri 121 4,6-α-glucanotransferase GtfB (Lr 121 GtfB), belonging to glycoside hydrolase family 70 (GH70), synthesizes linear isomalto/malto polysaccharides having (α1→6) linkages attached to the nonreducing ends of (α1→4) linked maltose oligosaccharide segments using starch or maltodextrin as a substrate. Since Lr 121 GtfB has low catalytic activity and efficiency, it leads to substrate regeneration and reduced substrate utilization. In this study, we superimposed the crystal structure of Lr 121 GtfB-ΔNΔV with that of L. reuteri NCC 2613 GtfB-ΔNΔV (Lr 2613 GtfB-ΔNΔV) to identify the acceptor binding subsites +1 to +3 and constructed five single-residue mutants and a random mutagenesis of N1019. Compared with the wild-type, N1019D Lr 121 GtfB-ΔN did not alter the product specificity, increased the catalytic activity and efficiency by 420 and 590%, respectively, and maintained >80% relative activity in the pH 3.5–6.5 interval. The findings will contribute to the industrial application of Lr 121 GtfB and provide new solutions for starch synthesis of higher value derivatives.

The aim of this study was to explore the alterations in physicochemical characteristics, bioactivity, flavor and metabolic profiles of mango juice fermented by Limosilactobacillus reuteri FJG2526 ( L. reuteri FJG2526). The results exhibited that L. reuteri FJG2526 had strong adaptability in mango juice, and reduced the total sugar, polyphenolics and flavonoids content of mango juice. L. reuteri FJG2526 fermentation ameliorated the flavor profiles of mango juice, particularly promoted the production of acids, alcohols, and esters. Moreover, 107 metabolites in the mango juice were drastically altered after 48 h L. reuteri FJG2526 fermentation by metabolomic analysis, including 73 remarkably upregulated metabolites and 34 remarkably downregulated metabolites, primarily involving amino acid metabolism. In addition, L. reuteri FJG2526 fermentation also enhanced the ability to scavenge DPPH and OH free radicals of mango juice, and inhibited lipase and α-glucosidase activities. This study offers new insights into the mango juice fermentation and will contribute to the application of L. reuteri in functional juices.

Recent studies have shown that lactic acid bacteria (LAB) fermentation has great potential to relieve hypertension. In this study, we verified the antihypertensive effects of Limosilactobacillus reuteri Z09, Lactobacillus helveticus Z11 and their mixed fermented milk (MIX-FM) on spontaneously hypertensive rats (SHRs). Four weeks of administration showed that all three LAB fermentations had an antihypertensive effect on SHRs, while MIX-FM showed the best effect. MIX-FM had the highest angiotensin-converting enzyme inhibitor peptide contents, and it can improve intestinal dysbiosis better than the other two groups. The above results suggest that these two strains have the potential to develop antihypertensive dairy products.

Limosilactobacillus reuteri 121 4,6-α-glucanotransferase (GtfBΔN) modifies starch by cleaving (α1 → 4) linkages and introducing non-branched (α1 → 6) linkages to produce functional starch derivatives. Research has mainly focused on GtfBΔN converting amylose (linear substrate), whereas the conversion of amylopectin (branched substrate) has not been studied in detail. In this study, we used GtfBΔN to understand amylopectin modification and performed a set of experiments to analyze this modification pattern. The donor substrates were segments from the non-reducing ends to the nearest branch point in amylopectin as shown from the results of the chain length distribution of GtfBΔN-modified starches. Decreased and increased contents of β-limit dextrin and reducing sugars, respectively, during the incubation of β-limit dextrin with GtfBΔN indicated that the segments from the reducing end to the nearest branch point in amylopectin act as donor substrates. Dextranase was involved in the hydrolysis of the GtfBΔN conversion products of three different substrates groups, maltohexaose (G6), amylopectin, and G6 plus amylopectin. No reducing sugars were detected, therefore, amylopectin was not used as an acceptor substrate, and no non-branched (α1 → 6) linkages were introduced into it. Thus, these methods provide a reasonable and effective approach to studying GtfB-like 4,6-α-glucanotransferase in analyzing the roles and contribution of branched substrates.

ObjectiveHigh intake of caffeoylquinic acid (CQA)-rich dietary supplements, such as green coffee bean extracts, offers health-promoting effects on maintaining metabolic homeostasis. Similar to many active herbal ingredients with high pharmacological activities but low bioavailability, CQA has been reported as a promising thermogenic agent with anti-obesity properties, which contrasts with its poor oral absorption. Intestinal tract is the first site of CQA exposure and gut microbes might react quickly to CQA. Thus, it is of interest to explore the role of gut microbiome and microbial metabolites in the beneficial effects of CQA on obesity-related disorders. Results Oral CQA supplementation effectively enhanced energy expenditure by activating browning of adipose and thus ameliorated obesity-related metabolic dysfunctions in high fat diet-induced obese (DIO) mice. Here, 16S rRNA gene amplicon sequencing revealed that CQA treatment remodeled the gut microbiota to promote its anti-obesity actions, as confirmed by antibiotic treatment and fecal microbiota transplantation. CQA enriched the gut commensal species Limosilactobacillus reuteri (L. reuteri) and stimulated the production of short-chain fatty acids, especially propionate. Mono-colonization of L. reuteri or low-dose CQA treatment did not reduce adiposity in DIO mice, while their combination elicited an enhanced thermogenic response, indicating the synergistic effects of CQA and L. reuteri on obesity. Exogenous propionate supplementation mimicked the anti-obesity effects of CQA alone or when combined with L. reuteri, which was ablated by the monocarboxylate transporter (MCT) inhibitor 7ACC1 or MCT1 disruption in inguinal white adipose tissues to block propionate transport. Conclusion sOur data demonstrate a functional axis among L. reuteri, propionate, and beige fat tissue in the anti-obesity action of CQA through the regulation of thermogenesis. These findings provide mechanistic insights into the therapeutic use of herbal ingredients with poor bioavailability via their interaction with the gut microbiota.

Chronic diarrhea in pets features high incidence, often causing vitamin B12 deficiency. Probiotics, which can alleviate or even treat chronic diarrhea by improving intestinal microbial balance, have shown promise. Previously, our laboratory isolated a strain of Limosilactobacillus reuteri (CLAB) with vitamin B12-producing capability from canine feces. However, it had suboptimal palatability and insufficient efficacy against chronic diarrhea and vitamin B12 deficiency. In this study, the CLAB was combined with Enterococcus faecalis and Saccharomyces cerevisiae to formulate a compound probiotic paste. The properties of the paste were comprehensively evaluated via a series of experiments including sensory evaluation, pH testing, microbial colony counting, palatability tests, disease activity index scoring, histopathological analysis, biochemical indicator analysis, and RNA quantification. The results demonstrated that the paste exhibited good stability, safety, and favorable palatability in both dogs and cats. Meanwhile, it significantly alleviated chronic diarrhea and vitamin B12 deficiency in mice. Specifically, the high-dose compound probiotic paste decreased myeloperoxidase and malondialdehyde levels in diarrheic mice from approximately 15.9 and 2.5 nmol/mgprot to about 4.2 and 1.7 nmol/mgprot; increased the total antioxidant capacity content and superoxide dismutase activity from approximately 0.5 nmol/mgprot and 1.0 U/mgprot to about 0.6 nmol/mgprot and 1.6 U/mgprot; and downregulated the relative mRNA levels of interleukin-6, interleukin-1β and tumor necrosis factor-α from approximately 2.1, 2.1 and 2.7 to about 1.2, 1.1 and 1.3, respectively. In conclusion, this study supports the potential use of the compound probiotic paste for preventing and treating chronic diarrhea and improving vitamin B12 deficiency in pets.

This study aims to investigate changes in the microflora and metabolites during in vitro fermentation of galactooligosaccharides (GOS), Limosilactobacillus reuteri M94 ( L. reuteri M94), and healthy human fecal microbiota, and to explore the anti-inflammatory and barrier-protective effects of the in vitro fermentation products on the Caco-2/RAW264.7 cell co-culture model. In vitro fermentation of GOS, L. reuteri M94, and healthy human fecal microbiota significantly enhanced the abundance of beneficial bacteria ( e.g. , Bifidobacterium ). It induced a substantial shift in the short-chain fatty acid (SCFA) profile, characterized by a significant increase in acetate and total SCFAs. In contrast, the concentrations of other major SCFAs (propionate, butyrate, valerate) were decreased. The in vitro fermentation products from GOS, L. reuteri M94, and healthy human fecal microbiota significantly alleviated LPS-induced inflammatory response in RAW 264.7 cells by suppressing the nuclear factor kappa-B (NF-κB) inflammatory signaling pathway, markedly downregulating NO release and pro-inflammatory cytokines (TNF-α, IL-6, IL-1β) expression, while upregulating the anti-inflammatory cytokine IL-10 and activating the Nrf2 pathway, reducing reactive oxygen species (ROS) levels. Additionally, these products upregulate tight junction protein expression (ZO-1, Claudin-1, Occludin) in the Caco-2/RAW 264.7 cell co-culture model, suggesting enhanced intestinal epithelial cell barrier function. Overall, the in vitro fermentation products of L. reuteri M94, GOS, and healthy human fecal microbiota demonstrate significant anti-inflammatory and barrier-protective potential to ameliorate gut barrier dysfunction.

Limosilactobacillus reuteri 121 4,6-α-glucanotransferase (Lr121 4,6-α-GTase), belonging to the glycosyl hydrolase (GH) 70 GtfB subfamily, converts starch and maltodextrins into linear isomalto/malto polysaccharides (IMMPs) with consecutive (α1 → 6) linkages. The recent elucidation of its crystal structure allowed identification and analysis of further structural features that determine its reaction and product specificity. Herein, sequence alignments between GtfB enzymes with different product linkage specificities (4,6-α-GTase and 4,3-α-GTase) identified amino acid residues in GH70 homology motifs, which may be critical for reaction and product specificity. Based on these alignments, four Lr121 GtfB-ΔN mutants (I1020M, S1057P, H1056G, and Q1126I) were constructed. Compared to wild-type Lr121 GtfB-ΔN, mutants S1057P and Q1126I had considerably improved catalytic efficiencies. Mutants H1056G and Q1126I showed a 9% decrease and an 11% increase, respectively, in the ratio of (α1 → 6) over (α1 → 4) linkages in maltodextrin-derived products. A change in linkage type (e.g., (α1 → 6) linkages to (α1 → 3) linkages) was not observed. The possible functional roles of these Lr121 GtfB-ΔN residues located around the acceptor substrate-binding subsites are discussed. The results provide new insights into structural determinants of the reaction and product specificity of Lr121 GtfB 4,6-α-GTase.

Background/Objectives: Raffinose is a prebiotic trisaccharide fermented by human gut microbiota, yet the strain-level determinants of its degradation and associated metabolic outputs remain poorly characterized. Methods: In the present study, anaerobic fermentation, 16S rRNA gene amplicon high-throughput sequencing, culturomics, and metabolomics were conducted to address this question. Results: In vitro fermentation of raffinose was performed using fecal samples from 16 healthy donors, and substantial interindividual variation was observed in substrate consumption, microbiota composition, and short-chain fatty acid production. Through culturomics, 204 bacterial strains spanning 18 species were isolated, among whichLimosilactobacillus reuterisubsp.reuteriexhibited the highest raffinose-degrading efficiency. Untargeted metabolomics comparingL. reuterisubsp.reutericultured on raffinose versus glucose revealed a distinct metabolic shift, with 290 metabolites significantly upregulated, including the putatively identified fructooligosaccharide 1-kestose—a compound with documented prebiotic and anti-inflammatory properties. KEGG enrichment further highlighted coordinated remodeling of nucleotide and amino acid metabolism. Conclusions: Collectively, this study provides a strain-level framework for understanding how raffinose shapes the gut microbiota and identifiesL. reuterisubsp.reuterias a key metabolic hub linking prebiotic consumption to the production of bioactive metabolites.

In this study, we investigated the protein structures, powder characteristics, as well as rehydration and emulsifying properties of spray-dried egg yolk powder after short-time lactic acid fermentation (3.5 h). Results indicate that fermentation improved the rehydration and emulsifying properties of yolk powder. Limosilactobacillus reuteri -fermented yolk powder exhibited better wettability due to the porous structure of particles and higher hydrophilicity. Lacticaseibacillus rhamnosus -fermented yolk powder had an enhanced coefficient of stability due to its smaller particles and higher surface charge. The higher water solubility of fermented yolk powder samples is mainly attributed to their lower hydrophobicity and higher zeta potential. The enhanced emulsifying activity of fermented yolk powder samples is primarily related to their increased β-turn structure and better solubility. Furthermore, fermentation treatment altered powder moisture content and bulk densities, while not affecting its flow behavior and thermal stability. This study provides an effective approach to improving the quality of yolk powder.
