Litsea cubeba essential oil (LCEO) is widely used as a flavor enhancer in cosmetics and as a folk skin-care agent in southern China. The objective of this paper is to investigate whether LCEO has biological activities that might be useful in modern skin-protection formulations. The present study results revealed that citral (57.4%) was a major component, while LCEO exhibited a potent inhibitory effect on tyrosinase (IC50 values 100 μg/mL), good antioxidative activities (IC50: 17.75 mg/mL for ABTS+., 10.2 mg/mL for O2
−), and apparent protective effect against UV–TiO2–NO2
−-induced protein oxidation at 0.01 mg/mL and tyrosine nitration at 0.1 mg/mL. Additionally, the biological activities of LCEO were compared with that of its main constituents. The results demonstrated that any individual major component was not the unique contributor to the high activity of LCEO. Based on these results, we suggested that LCEO could serve as a new natural skin-whitening agent.

Essential oils have attracted increasing attentions due to their broad-spectrum antimicrobial activities and generally recognized as safe (GRAS) regulatory status. However, their low-solubility in water and reduced effectiveness in food matrices limit the commercial application in food industry. In this study, we used ultrasonic emulsification , medium-chain triacylglycerol (MCT) and phosphate-buffered saline (PBS) to formulate a highly stable and water-dispersible litsea cubeba essential oil (LCEO) nanoemulsions. After emulsification, a minimizing droplet size of LCEO nanoemulsions was achieved at approximately 100 nm in diameter, resulting in an increase in the water solubility and better stability at 4 °C and 25 °C. Furthermore, the bioactivities (antibacterial, antioxidant and antibiofilm properties) of LCEO nanoemulsions were greatly improved after emulsification, suggesting that nano-emulsification is beneficial for the value development of various essential oils and the LCEO nanoemulsions could be used as a novel food preservative for controlling bacterial growth and preventing oxidative deterioration in foods.

This study investigated the use of Litsea cubeba essential oil (LC-EO) as an anti-virulence agent targeting listeriolysin O (LLO) of Listeria monocytogenes ( L. monocytogenes ). The hemolysis assay showed a significant decrease in the hemolytic activity of LLO by 90.7% and 70.2% after exposure to 500 μg mL −1 LC-EO or 700 μg mL −1 citral (the main component of LC-EO), respectively. This suggested that citral plays a major role in the anti-virulence function against LLO. The experiments including UV–visible absorption spectroscopy , fluorescence quenching and three-dimensional fluorescence spectroscopy demonstrated that exposure to citral leads to significant changes in the conformation of LLO, potentially resulting in the unfolding of its structure and the formation of a ground-state complex. Synchronous fluorescence spectra analysis indicated that the binding site of citral could be the undecapeptide region of LLO. Additionally, the results of molecular docking and the acid-base effect experiment of different aldehydes and phenols revealed that the cysteine residue of LLO and the α, β-unsaturated aldehyde of citral are likely to form a covalent bond through Michael addition . This interaction is believed to be the primary reason for the loss of hemolytic activity. As a result, this work provides a new approach for the development of LLO-targeting anti-virulence agents and holds promise for expanding the application of LC-EO in the field of food microbiological safety.

The essential oil of Litsea cubeba demonstrates notable antibacterial properties and holds considerable promise for various applications. To explore green and effective extraction method for L. cubeba essential oil, we introduced enzyme-assisted natural deep eutectic solvents (DESs). The optimization of DESs was performed using response surface methodology with a Box-Behnken design (BBD), and the optimal parameters were determined as follows: a choline chloride to citric acid (ChCl/CA) ratio of 1:0.97, water content of 73%, and cellulase(0.5%) enzymolysis temperature of 45°C. Compared to traditional steam distillation, the DESs method significantly increased the essential oil yield from 3.20% to 4.70% and expanded the chemical profile of the essential oil, identifying a total of 79 compounds. Five new aromatic compounds, including methyl geranate, geranic acid, p-mentha-1,5-dien-8-ol, α-terpinyl acetate, and terpinyl acetate, were identified. Antifungal activity against Aspergillus flavus was confirmed by scanning electron microscopy. At a concentration of 1,000 μL/L, after 96 hours of incubation, essential oils extracted using enzyme-assisted DESs method demonstrated substantial inhibition against A. flavus , with the ChCl/EG formulation achieving a 100% inhibition rate. The content of citral, citronellal, citronellol, linalool, and new compound methyl geranate showed a positive correlation with the anti- A. flavus inhibition rate. The enzyme-assisted DESs extraction method proves to be an effective and efficient approach for extracting L. cubeba essential oil and provides a valuable reference for future research and application in essential oil extraction.

An effective approach for synthesizing green and sustainable biodiesel fuel from low-value vegetable oils has been explored. Herein, a series of catalysts of zinc supported on zirconia ( x Zn/ZrO 2 ) were successfully synthesized, characterized, and applied for the production of biodiesel from Litsea cubeba kernel oil (LCKO) through the transesterification process. The results showed that the monoclinic phase structure of the prepared catalysts transformed to the more stable tetragonal phase structure after supporting different Zn loadings (≥5%). Compared with the pure ZrO 2 , the highly dispersed Zn species on the surface of x Zn/ZrO 2 could be divided into Zn oxide and ZnOH + , which could promote the transesterification of LCKO. Thus, the 7%Zn/ZrO 2 catalyst exhibited the best catalytic activity of the LCKO conversion 82.5%, which was attributed to the positive effects of both the ZrO 2 support and Zn species. Furthermore, the various reaction parameters (such as reaction time, stirring rate, n CH3OH :n LCKO , catalyst amount, and reaction temperature) were investigated systematically and optimized carefully. And a possible mechanism of the transesterification process catalyzed by Zn/ZrO 2 catalysts was also proposed. Finally, the physical properties of the as-synthesized biodiesel accorded with the current international biodiesel standards.

Early sex identification of the dioecious and medicinal spice plant, Litsea cubeba (Lour.) Pers (LC), is necessary for expanding the production and application of LC. We describe a Raman spectroscopy (RS) and surface-enhanced Raman spectroscopy (SERS) method combined with machine learning techniques. Based on the RS and SERS features, we combined principal component analysis and linear discriminant analysis (PCA-LDA) to dimension reduction for the data. And we combined seven machine learning algorithms, including logistic regression (LR), Naive Bayes (NB), decision tree (DT), random forest (RF), k-nearest neighbor (KNN), support vector machines (SVM), and extreme gradient boosting (XGBoost) algorithms to construct a gender prediction model for LC. The results demonstrated that the surface reinforcement treatment with drops of silver sol combined with LR, SVM, and XGBoost models achieved an accuracy of 84.62% in distinguishing male and female leaves. Furthermore, compared to the discrimination effect of scanning the leaf surface only, the gender recognition accuracy of the surface reinforcement treatment increased by 17.31%, 19.24% and 19.24%, respectively. Portable Raman spectroscopy combined with machine learning algorithms can be promoted for use as a tool for early sex identification in most plants, which can be applied to large-scale plant cultivation and breeding programmes.

Litsea cubeba (LC) is an indigenous industrial crop in China with fruits possessing interesting essential oil content of 3-5%, which production leads to a large number of kernels as wastes. In this study, the performance of four alternative solvents on the extraction of LC kernel oils was firstly evaluated in comparison with n-hexane as the reference solvent. Regardless of the solvent used, all extracted oils were quantitatively and qualitatively analyzed, where green solvents (dimethyl carbonate-DMC and cyclopentyl methyl ether-CPME) were preferred to alcoholic solvents (ethanol and isopropanol) considering their higher extraction yields and similar acylglycerol and fatty acid compositions to oils extracted by n-hexane. Interestingly, micronutrients in oils extracted by green solvents were quantified much higher than that extracted by n-hexane. Apart from common physicochemical properties measured, Hansen solubility parameter and antioxidant activity in vitro were also studied to make a more comprehensive profile for solvent effects and oil functionality. The results indicated that CPME could be the optimal alternative solvent to n-hexane with comparable extractability and selectivity in the green extraction of LC kernel oils as a new promising natural plant oil resource.

Litsea cubeba (Lour.) Pers. is an important industrial tree in southern China and Southeast Asia. We surveyed and sampled wild L. cubeba in 32 provenances spread across nine provinces of China. We used nested variance analysis, multiple comparisons analysis, hierarchical clustering analysis, Pearson’s correlation analysis, path analysis with multiple stepwise regression, detrended correspondence analysis, redundancy analysis, and variance partitioning analysis to evaluate the phenotypic diversity of fresh fruit yield (FFY) and essential oil (EO) characteristics of L. cubeba , and to explore the relationship between the main habitat factors and phenotypic diversity. We detected wide phenotypic diversity in FFY, EO content (EOC), citral yield (CitrY), and the proportions of the seven main components of EOs (citral, D -limonene, 3,7-dimethyl-3,6-octadienal, 4-methyl-3-pentenal, linalool , citronellal , and 1-(cyclopanecarbonyl)piperidin-4-one). The variation was mainly derived from among provenances ( P ≤ 0.01) rather than within provenances, and the variation in FFY and CitrY was greater than that of EOC and the seven dominant components among provenances. We also detected several region-specific components of EOs in fresh fruit, including α-terpineol, geraniol , eucalyptol , and 7-methyl-3-methyleneoct-6-enal. Most of the provenances with higher FFY were distributed in southwestern China (lower longitude and latitude, and higher elevation), while most of those with higher EOC were distributed in southeastern China (higher longitude and latitude, and lower elevation). The three provenances with excellent FFY were YN-XC, GZ-ZY, and SC-KJ; the two with higher EOC were SC-KJ and HN-DX; and CitrP was higher in GZ-ZF than in other provenances. Longitude, total potassium content (TotalKC), and bulk density (BD) were the most important factors that influenced FFY diversity, while latitude, elevation, annual average relative humidity (AARH), and organic carbon content (OCC) most strongly influenced EOC diversity, and annual average temperature and annual average maximum temperature (AAMaxT) influenced CitrP diversity. The six main habitat factors together explaining the largest proportions of phenotypic diversity were longitude, AARH, AAMaxT, elevation, latitude, and soil total phosphorus content (TotalPC). Higher soil TotalKC, BD, and TotalPC were suitable conditions for cultivation of L. cubeba to achieve higher FFY, while lower AARH and OCC were favorable for accumulation of EOs, and only lower temperature was more likely conducive to citral synthesis. The wide diversity of FFY and EO characteristics should be taken into account when selecting germplasms for protection and utilization, and the identification of the most important environmental factors affecting phenotypic diversity provides a theoretical basis for the optimization of L. cubeba cultivation.

Although Litsea cubeba essential oil (LCEO) is widely used for against fungi, antifungal mechanism of it is not fully understood. In the study, antifungal potential and mechanism of LCEO on Colletotrichum scovillei , the main pathogen fungi caused pepper anthracnose , were studied. Results of gas chromatograph-mass spectrometry (GC-MS) showed that citral (67.37%) and d -Limonene (11.72%) were the major components of LCEO. LCEO was effective in inhibiting germination of C. scovillei spores , and the minimum inhibitory concentration (MIC) was 0.60 mg mL −1 . To determine influence of LCEO on cell membrane integrity and permeability of C . scovillei , SEM observation, OD 260 nm (nucleic acids), OD 280 nm (proteins), extracellular electric conductivity , cell membrane permeability (propidium iodide, PI), cell wall integrity (calcofluor white, CFW), malondialdehyde (MDA) content and ergosterol content were analyzed. Spores exposed to LCEO were partly deformed, collapsed, shrunken and shriveled. The OD 260 nm, OD 280 nm and the extracellular electric conductivity of LCEO treatment group increased by 2.0, 1.5 and 1.2 times, respectively, at 12 h after treatment of LCEO (0.6 mg mL −1 ). Besides, high accumulation of MDA content indicated that LCEO significantly enhanced membrane lipid peroxidation . Moreover, LCEO induced a decrease in ergosterol content of C. scovillei . These findings suggest that LCEO exhibited potent inhibitory activities against C. scovillei , and the cell membrane permeability and cell wall integrity of C. scovillei treated by LCEO were significantly destroyed.

In this study, we systematically investigated the mechanisms of the antioxidation and anti-lipid accumulation effects of antioxidant peptides fromLitsea cubebaon a free fatty acid (FFA)-induced NAFLD model of HepG2 cells. The NAFLD cell model was constructed by inducing the HepG2 hepatocellular carcinoma cell line with 0.5 mmol/L FFAs, and AQRDAGLL, QEGPFVR, and DVPPPRGPL were given to the culture to study their lipid-lowering and antioxidant activities on NAFLD cells. The lipid-lowering activities of the three antioxidant peptides were evaluated by Oil Red O staining and TG and TC content assays, and the results showed that all three peptides had strong ameliorating effects on FFA-induced lipid accumulation in NAFLD cells. The intracellular antioxidant protease (CAT, GSH, and SOD) activity levels and lipid peroxidation (MDA) content were measured and intracellular ROS levels were detected. The results showed that after intervention with the antioxidant peptides, the intracellular ROS levels in the NAFLD model cells were significantly reduced, the SOD and CAT activities were increased, the GSH content was elevated, and the MDA content was reduced, which indicated that AQRDAGLL, QEGPFVR, and DVPPPRGPL were able to inhibit the oxidative stress of the cells effectively and to achieve the effect of intervening in NAFLD. JC-1 fluorescence staining experiments showed that the mitochondrial membrane potential function of NAFLD cells was restored under the effect of the antioxidant peptides. Molecular dynamics simulations revealed that the main driving force between QEGPFVR and Keap1 protein was van der Waals forces, ΔG = −62.11 kcal/mol, which indicated that QEGPFVR was capable of spontaneously binding to Keap1 protein.

As an important natural product, the sufficient separation of plant essential oil (EO) is helpful to improve its utilization value. In this work, deep eutectic solvent-homogenate based microwave-assisted hydrodistillation (DES-HMAHD) was developed and applied to isolate EO from the fruits of Litsea cubeba (Lour.) Pers. Different types of DES were investigated in terms of the EO kinetics and composition, among which oxalic acid/choline chloride (OA/ChCl) had obvious advantages. Following, molar ratio of OA and ChCl (1:1), water content (50%), liquid-solid ratio (12.5:1 mL/g), homogenate time (2 min), and microwave power (700 W) were found to be the optimum conditions. Gas chromatography-mass spectrometer (GC-MS) analysis showed that the EO isolated from DES-HMAHD contained a large proportion of m -cymene and trans -linalool oxide, which were quite different from the conventionally reported L. cubeba EO. In addition, the proposed DES-HMAHD resulted in higher separation efficiency and economic value, as well as lower environmental impact, as compared with other techniques. Afterwards, the EO isolated by different methods was evaluated from the perspective of biological activity. The EO obtained by DES-HMAHD showed higher antioxidant activity (DPPH and ABTS) but lower antifungal activity, which was related to its chemical composition. In general, DES-HMAHD produced a kind of L. cubeba EO with different components, which provided a scientific foundation for the sufficient isolation of plant EO and its application in the natural products.

With increasing demands for environmental protection and food safety, biodegradable active food packaging is gaining attention. Hereby, konjac glucomannan/carrageenan blend films (KCNTO) incorporating Litsea cubeba essential oil (LCEO) Pickering emulsion stabilized by cellulose nanocrystals/tannic acid were developed and evaluated for their preservation effect on pork. The formation, structure, physical properties, antioxidant and antimicrobial activities of KCNTO, as well as the release of LCEO were investigated. The results showed that the viscosity and pseudoplasticity of KCNTO film-forming solutions decreased due to LCEO Pickering emulsion interfering with the molecular entanglement network of konjac glucomannan/carrageenan. LCEO droplets were homogeneously distributed in KCNTO to make its structure loose. Hydrogen bonding interactions occurred between LCEO Pickering emulsion and the film matrix. The addition of LCEO Pickering emulsion improved KCNTO's elongation at break, water contact angle, water vapor barrier and UV shielding, while giving it excellent antioxidant and antimicrobial activities. LCEO was slowly and sustainably released from KCNTO at 25 °C in air. KCNTO-8 maintained the quality of pork by slowing down the increase in pH, aerobic plate count and thiobarbituric acid reactive substances during storage at 4 °C. The findings suggested that KCNTO was a promising active packaging material for pork preservation.
