Cloning and functional verification of Geraniol-10-Hydroxylase gene in Lonicera japonica

Shuping Zhang,Zhenhua Liu,Jia Li,Qian Liu,Yongqing Zhang,Gaobin Pu

Background Geraniol 10-hydroxylase (G10H) is a cytochrome P450 monooxygenase involved in regulation, which is involved in the biosynthesis of monoterpene. However, G10H is not characterized at the enzymatic mechanism and regulatory function in Lonicera japonica . Methods and Results A gene related to the biosynthesis of monoterpenoid, geraniol 10-hydroxylase, has been cloned from the medicinal plant Lonicera japonica . The gene, LjG10H , encodes a peptide of 498 amino acids with a predicted molecular weight of 54.45 kDa. LjG10H shares a homology of 72.93–83.90% with G10H from other plants. Phylogenetic analysis suggests that the protein encoded by this gene belongs to the cytochrome P450 monooxygenase family. Tissue-specific expression analysis revealed that LjG10H is most highly expressed in flowers. Through heterologous expression in E. coli , the LjG10H protein was purified and its catalytic activity was studied. The results show that the enzyme can catalyze the hydroxylation of geraniol to 10-hydroxygeraniol. Additionally, analysis of Lonicera japonica seedlings with silenced LjG10H revealed a reduction in monoterpenoid content. Conclusions This study demonstrates that LjG10H plays an important role in the biosynthetic pathway of iridoids. This is the first article that ascribes G10H to be associated with the biosynthetic pathway of iridoid. This study provides a theoretical basis for the functional mechanism of LjG10H in regulating iridoid synthesis and provides a valuable resource for molecular breeding studies.

Lonicera japonica Thunb. as a promising antibacterial agent for Bacillus cereus ATCC14579 based on network pharmacology, metabolomics, and in vitro experiments

Nan Xu,Li-hua Du,Yan-chao Chen,Jin-hao Zhang,Qian-feng Zhu,Rong Chen,Guo-ping Peng,Qi-ming Wang,Hua-zhong Yu,Li-qun Rao

Lonicera japonica Thunb. has attracted much attention for its treatment of bacterial and viral infectious diseases, while its active ingredients and potential mechanisms of action have not been fully elucidated. Here, we combined metabolomics, and network pharmacology to explore the molecular mechanism of Bacillus cereus ATCC14579 inhibition by Lonicera japonica Thunb. In vitro inhibition experiments showed that the Lonicera japonica Thunb.'s water extracts, ethanolic extract, luteolin, quercetin, and kaempferol strongly inhibited Bacillus cereus ATCC14579. In contrast, chlorogenic acid and macranthoidin B had no inhibitory effect on Bacillus cereus ATCC14579. Meanwhile, the minimum inhibitory concentrations of luteolin, quercetin, and kaempferol against Bacillus cereus ATCC14579 were 15.625 μg mL−1, 31.25 μg mL−1, and 15.625 μg mL−1. Based on the previous experimental basis, the metabolomic analysis showed the presence of 16 active ingredients in Lonicera japonica Thunb.'s water extracts and ethanol extracts, with differences in the luteolin, quercetin, and kaempferol contents between the water extracts and ethanol extracts. Network pharmacology studies indicated that fabZ, tig, glmU, secA, deoD, nagB, pgi, rpmB, recA, and upp were potential key targets. Active ingredients of Lonicera japonica Thunb. may exert their inhibitory effects by inhibiting ribosome assembly, the peptidoglycan biosynthesis process, and the phospholipid biosynthesis process of Bacillus cereus ATCC14579. An alkaline phosphatase activity assay, peptidoglycan concentration assay, and protein concentration assay showed that luteolin, quercetin, and kaempferol disrupted the Bacillus cereus ATCC14579 cell wall and cell membrane integrity. Transmission electron microscopy results showed significant changes in the morphology and ultrastructure of the cell wall and cell membrane of Bacillus cereus ATCC14579, further confirming the disruption of the cell wall and cell membrane integrity of Bacillus cereus ATCC14579 by luteolin, quercetin, and kaempferol. In conclusion, Lonicera japonica Thunb. can be used as a potential antibacterial agent for Bacillus cereus ATCC14579, which may exert its antibacterial activity by destroying the integrity of the cell wall and membrane.

Anthocyanins from buds of Lonicera japonica Thunb. var. chinensis (Wats.) Bak.

Yuan Yuan,Jian Yang,Xiaodan Yu,Luqi Huang,Shufang Lin

Recently, a new kind of Lonicera japonica (FLJ) tea well known for its red color, flavor, and taste has become popular in the market. This tea comes from the buds of Lonicera japonica Thunb. var. chinensis (Wats.) Bak. (rFLJ), a wild mutant of the Lonicerae plant. The present study aimed to identify and quantify, for the first time and in detail, the anthocyanin compounds in the buds of rFLJ. Anthocyanins were extracted with methanol–water–formic acid–trifluoroacetic acid (70:27:2:1, v/v/v/v) and quantified as 108.06   ±   4.32   mg/100   g of fresh weight (cyanidin-3-glucoside equivalent) by using the pH-differential method. Eight different anthocyanins were identified by HPLC–DAD–ESI-MS/MS analysis and were quantitated by HPLC–DAD analysis. The main anthocyanins in the buds of rFLJ were two non-acylated anthocyanins: cyanidin-3,5-diglucoside and cyanidin-3-glucoside. Moreover, orthologous genes in the biosynthesis of anthocyanins were cloned and their structure and transcript levels in the buds of FLJ and rFLJ were further compared. The results revealed a 12-residue insertion (IKPTIEGVLGII) in LjcDFR compared with LjDFR. Furthermore, transcript levels of DFR, LDOX, BZ1 and GT1 were decreased in the buds of FLJ compared with rFLJ, as constitute with the content of cyanidin-3-glucoside. Here, a rapid and effective method to identify and quantify all major anthocyanins in the buds of rFLJ is established. Furthermore, the results of this study may explain which anthocyanin produces the red color of rFJL, an important characteristic that distinguishes it from FLJ.

Ultrasound-assisted extraction of polyphenols from Lonicera japonica leaves and their α-glucosidase inhibition

Gang Li,Chunming Bao,Hao Zhang,Lu Bai

Introduction: The search for natural bioactive compounds that act as α-glucosidase inhibitors is a central focus in diabetes treatment research. Methods: This study utilized ultrasonic-assisted extraction to optimize the extraction of polyphenols from honeysuckle leaves through response surface methodology (RSM). Enzyme inhibition and fluorescence quenching experiments were conducted to examine the inhibitory activity and binding behavior of the extracted polyphenols. Results: The results indicated that the optimal conditions for polyphenol extraction were as follows: ethanol concentration, 64%, liquid–solid ratio, 45 mL/g, ultrasonic power, 700 W, ultrasonic time, 6 min. Under these conditions, the yield of polyphenols from honeysuckle leaves was 9.16 ± 0.19%, which closely aligns with the predicted value of 9.18%. The total phenolic content of the polyphenol extracts was 20.6 ± 0.67%, with chlorogenic acid and luteoloside contents measuring 5.65 ± 0.40% and 2.51 ± 0.14%, respectively. The inhibitory effect of polyphenol extracts (IC50, 0.14 ± 0.01 mg/mL) on α-glucosidase was better than that of chlorogenic acid (IC50, 0.55 ± 0.02 mg/mL). Fluorescence quenching experiments indicated that the polyphenol extracts interact with α-glucosidase, resulting in alterations to the microenvironment of amino acid residues.Discussion: This interaction can reduce the binding affinity between the substrate and α-glucosidase, thereby contributing to the objective of lowering postprandial hyperglycemia. Our research results can provide reference for the development and utilization of honeysuckle leaves.

Lonicera japonica Attenuates Carbon Tetrachloride-Induced Liver Fibrosis in Mice: Molecular Mechanisms of Action

Hui Miao,Yi Zhang,Zhenlin Huang,Bin Lu,Lili Ji

Liver fibrosis is a worldwide clinical issue that generally causes hepatic cirrhosis. Lonicerae Japonicae Flos (dried flower buds of Lonicera japonica Thunb) is a traditional heat-clearing and detoxifying herbal medicine in China. This study aims to observe the protection of the water extract of Lonicerae Japonicae Flos (FL) from carbon tetrachloride (CCl 4 )-induced liver fibrosis in mice. Liver fibrosis was induced in mice by intraperitoneal injection of 2 ml/kg CCl 4 twice a week for 4 weeks. FL’s attenuation of CCl 4 -induced liver fibrosis in mice was evidenced by the results of Masson’s trichrome and Sirius red staining, liver hydroxyproline content and serum amount of collagen IV. FL reduced hepatic stellate cells (HSCs) activation and reversed the epithelial-mesenchymal transition (EMT) process in mice treated with CCl 4 . FL also alleviated liver oxidative stress injury and enhanced the activation of nuclear factor erythroid 2-related factor 2 (Nrf2) anti-oxidant signaling pathway in mice treated with CCl 4 . Additionally, the main phenolic acids in FL including chlorogenic acid (CGA) and caffeic acid (CA) both reduced HSCs activation in vitro . In summary, FL attenuates CCl 4 -induced liver fibrosis in mice by inhibiting HSCs activation, reversing EMT and reducing liver oxidative stress injury via inducing Nrf2 activation. CGA may be the main active compound contributing to the antifibrotic activity of FL.

Lonicera japonica-Derived Nanovesicles Ameliorate Acute Lung Injury by Suppressing Neutrophil Extracellular Traps Formation in Mice

Xi Lin,Jian Lin,Yezi Zhang,Xinyan Zhang,Xinghua Liu,Hongying Cao,Huan Shen,Jianming Liang,Zhenhua Dai,Siyuan Xu,Xingdong Lin

Acute lung injury (ALI) is a lethal respiratory disease. The inflammatory storm triggered by neutrophil extracellular traps (NETs) is the critical pathogenesis of pulmonary injury. But the effective clinical treatments remain limited for ALI. Hence, developing a pulmonary-targeting nano-drug is urgent in the clinic. Lonicera japonica -derived nanovesicles (LDNVs) was isolated and characterized. Distribution of LDNVs were enriched in the pulmonary neutrophils. LDNVs restored the body weight, physical activity and attenuated the pulmonary injury of LPS-induced ALI mice. The inflammatory cytokines and neutrophil-associated chemokines were decreased with LDNVs treatment. LDNVs also decreased pulmonary accumulation of neutrophils and NETs. The LC3B II was suppressed and p62 was enhanced by LDNVs treatment. Finally, LDNVs suppressed the translocation of LC3 in nuclear and lysosomal damage. But these therapeutic effects of LDNVs in ALI and NETs were both blocked by a neutrophils and NETs activator, phorbol myristate acetate (PMA). LDNVs attenuate LPS-induced ALI by inhibiting NETs formation. These effects may be associated with LDNVs suppressing the autophagic flux.

Anti-hyperuricemic and anti-gouty arthritis activities of polysaccharide purified from Lonicera japonica in model rats

Qiuxuan Yang,Qilong Wang,Wenwen Deng,Congyong Sun,Qiuyu Wei,Michael Adu-Frimpong,Jixiang Shi,Jiangnan Yu,Ximing Xu

In this present study, we investigated the anti-hyperuricemic and anti-gouty arthritis effect of a puried water-soluble polysaccharide (LJP-1) obtained from Lonicera japonica . A series of characterization of the purified polysaccharide were carried out in this paper. Monosaccharide analysis showed that LJP-1 composed of glucuronic acid, glucose, galactose, arabinose, and xylose at the ratio of 2.43:1:2.09:1.95:1.96, respectively. The estimated molecular weight of LJP-1 was 17.5 kDa. LJP-1 belonged to pyranose and possessed α- and β -glycosidic configurations. Congo red test showed that LJP-1 had a spatial triple helix structure. In pharmacodynamic experiments, the anti-hyperuricemic activity of LJP-1 was studied using hyperuricemic SD rat model induced via potassium oxonate and hypoxanthine. The result showed that LJP-1 could obviously decrease the serum uric acid level and suppress xanthine oxidase (XOD) activity. Moreover, in the gouty arthritis model established by sodium urate crystals, the degree of swelling of the ankle joint, IL-1β, IL-6, TNF-α and COX-2-related inflammatory factors levels in murine serum all declined. Taken together, these results demonstrated that LJP-1 has anti-gouty arthritis effect. Therefore, LJP-1 could serve as a promising candidate for developing novel natural anti-gouty agent.

Lonicera japonica Thunb. leaves were used to biosynthesize silver nanoparticles (AgNPs), while its antibacterial and antifungal properties were investigated. Reaction conditions of AgNPs which characterized were optimized. The maximum absorption wavelength of the AgNPs was located about 400 nm, with a spherical shape and a particle size of 14.87 ± 4.07 nm. Produced AgNPs showed effective antibacterial activities against tested bacterial strains. Minimal inhibitory concentrations and minimum bactericidal concentrations of the AgNPs were 0.25 and 0.5 mg/ml for Escherichia coli ( E. coli ), and 0.5 and 1.0 mg/ml for Staphylococcus aureus , respectively. According to the mechanism study, AgNPs generated morphological changes in E. coli and disrupted membrane integrity. Furthermore, AgNPs which showed effective dose-dependent inhibitory effects against Penicillium citrinum could suppress the growth of fungal mycelium on citrus peels. Conclusively, AgNPs generated by this facile and green method showed effective antimicrobial activity, which is promising in food processing and preservation. Practical applications Silver nanoparticles have attracted enormous attention because of their intrinsic properties which are advantageous in many applications, especially in agriculture and food industry. Silver nanoparticles were synthesized by medicine food homology plants Lonicera japonica Thunb. leaves for the first time, which was a green synthesis method. Synthesized AgNPs showed good antibacterial and antifungal properties, which has the potential use in citrus preservations.

Luteolin in Lonicera japonica inhibits the proliferation of white spot syndrome virus in the crayfish Procambarus clarkii

Hai-Feng Jiang,Cheng Chen,Xin-Yuan Jiang,Jing-Lei Shen,Fei Ling,Peng-Fei Li,Gao-Xue Wang

White spot disease caused by the white spot syndrome virus (WSSV) is a highly lethal infectious disease of crustacean aquaculture, with no effective means of control. Active compounds from herbal medicines are promising candidates for the development of antiviral agents for WSSV. We evaluated the efficacy of 12 herbal medicines in inhibiting WSSV proliferation in a WSSV infection model ( Procambarus clarkii ), Lonicera japonica extract had the strongest inhibitory effect on WSSV replication. Luteolin (LUT), the major bioactive compound of L. japonica extract, was further investigated and revealed to have a strong inhibitory effect on WSSV (96.04%, 50 mg/kg). LUT treatment decreased the WSSV load and the expression of important viral genes (immediate-early gene ie1 , DNA polymerase DNApol and envelope protein Vp28 ) in a dose-dependent manner and increased the survival of WSSV-infected crayfish by 60.00% (50 mg/kg). Co-incubation and time-of-addition assays revealed that LUT did not affect the infectivity of viral particles or viral absorption and entry. Expression of immune-related genes such as barrier-to-autointegration ( BAF ), heat shock protein 70 ( Hsp70 ) and signal transducer and activator of transcription ( STAT ), which may be beneficial for viral replication, were significantly decreased ( P  < 0.01) in LUT-treated crayfish. LUT also modified the expression of antioxidants ( SOD , CAT and GST ), pro-inflammatory factor ( COX-1 and COX-2 ), and apoptosis-related ( BAX and BI-1 ) genes ( P  < 0.01) to enhance antioxidant defenses, mitigate inflammation and induce apoptosis, respectively. Overall, LUT could be used as a prophylactic or therapeutic agent against WSSV infection in crustacean aquaculture.

Ligand fishing of anti-neurodegenerative components from Lonicera japonica using magnetic nanoparticles immobilised with monoamine oxidase B

Gui-Fang Wu,Xiao-Lan Jiang,Yu-Zhou Gong,Ya-Dong Hu,Xiao-Lin Bai,Xun Liao

In this work, monoamine oxidase B was immobilised onto magnetic nanoparticles to prepare a new type of affinity solid-phase extraction adsorbent, which was used to extract the possible anti-neurodegenerative components from the Lonicera japonica flower extracts. Coupled with high-performance liquid chromatography with mass spectrometry, two monoamine oxidase B ligands were fished-out and identified as isochlorogenic acid A and isochlorogenic acid C, which were found to be inhibitors of the enzyme for the first time, with similar half maximal inhibitory concentration values of 29.05 ± 0.49 and 29.77 ± 1.03 μM, respectively. Furthermore, equilibrium-dialysis dissociation assay of enzyme-inhibitor complex showed that both compounds have reversible binding patterns to monoamine oxidase B, and kinetic analysis demonstrated that they were mixed-type inhibitors for monoamine oxidase B, with K i and K is values of 9.55 and 37.24 μM for isochlorogenic acid A, 9.53 and 35.50 μM for isochlorogenic acid C, respectively. The results indicated that isochlorogenic acid A and isochlorogenic acid C were the major active components responsible for the anti-degenerative activity of the flowers of L. japonica , while magnetic nanoparticles immobilised monoamine oxidase B could serve as an efficient solid-phase extraction adsorbent to specifically extract monoamine oxidase B inhibitors from complex herbal extracts.

Global lysine succinylation analysis unveils post-translational regulation effect on phenylpropanoid metabolism remodeling during Lonicera japonica flower development

Yao Chen,Luhuizi Shan,Wenxi Zheng,Jie Chen,Linfang Deng,Xu Tian,Ruili Xie,Yunhong Yang,Lin Zhang,Bingxian Yang

Lonicera japonica plays a significant role in traditional Chinese medicine and as a food source, making it a focus of studies on protein succinylation and its potential role in regulating secondary metabolism during flower development. This study aimed to clarify the regulatory mechanism of protein succinylation on phenylpropanoid-related phenotypic changes by conducting a global lysine succinylation proteomic analysis across different flowering stages. A total of 586 lysine succinylated peptides in 303 proteins were identified during early and late floral stages. Functional enrichment analysis revealed that succinylated proteins primarily participated in the tricarboxylic acid (TCA) cycle, amino acid metabolism, and secondary metabolism. The abundance of succinylated aspartate transaminase (AT), 4-coumarate-CoA ligase (4CL), and phenylalanine N-hydroxylase (CYP79A2) in phenylpropanoid metabolism varied during flower development. In vitro experiments demonstrated that succinylation increased AT activity while inhibited 4CL activity. Decreased levels of total flavonoids and phenolic acids indicated significant alterations in phenylpropanoid metabolism during later floral stages. These results suggest that succinylation of TCA cycle proteins not only influences flower development but also, together with AT-4CL-CYP79A2 co-succinylation, redirects phenylpropanoid metabolism during flower development in L. japonica .

Effect of Lacticaseibacillus rhamnosus L08 fermentation on xanthine oxidase inhibitory activity and flavour profile of honeysuckle (Lonicera japonica Thunb.)

Yan Song,Qing Liu,Maliha Munir,Yunfeng Gao,Jian He,Libo Liu,Guofang Zhang,Xing Li,Chun Li,Xiaoxi Qi

Xanthine oxidase (XOD) catalyses the oxidation of hypoxanthine and xanthine to uric acid, and inhibition of its activity has become the main strategy to reduce uric acid production. Honeysuckle (HON) which is rich in polyphenols potentially has the capacity to inhibit XOD activity. However, it carries the bitter and astringent flavor as well as many traditional Chinese medicines, along with that most of the polyphenols in HON exist in bound form, the exploitation and application of HON in food is limited. In this study, we investigated the effects of Lacticaseibacillus rhamnosus L08 fermentation on the bioactive components, XOD inhibitory activity and flavour characteristics of HON. The results showed that the XOD inhibitory activity was increased by 2.08 times after 96 h of fermentation. It was demonstrated that the increase in the total flavonoid content and total phenol content was the main reason for the enhancement of the fermentation solution's XOD inhibitory activity and antioxidant activity. Moreover, the fermentation also enriched the composition of volatile components of HON and reduced levels of undesirable flavour substances, which made the usage of fermented HON as functional food or in pharmaceutical use feasible and applicable.
