Interaction between Bacteroidetes species in the fermentation of Lycium barbarum arabinogalactan

Cui Cao,Lilong Wang,Xueqian Zhang,Chunqing Ai,Zhongfu Wang,Linjuan Huang,Shuang Song,Beiwei Zhu

The present study investigated the utilization of an arabinogalactan from Lycium barbarum (LBP-3) by intestinal Bacteroidetes species. The mixed-culture assay showed 58.4 % LBP-3 was utilized, and Bacteroides caccae and Phocaeicola vulgatus utilized more LBP-3 in single-culture compared to others. During in vitro fermentation of LBP-3, P. vulgatus favored arabinose while B. caccae preferred galactose. Moreover, 9 and 25 oligosaccharides were identified by HPLC-MS n in conditioned media (CM) derived from B. caccae and P. vulgatus , respectively. All of 3 tested Parabacteroides species ( P. distasonis , P. goldsteinii , and P. johnsonii ) markedly proliferated in CM of B. caccae and P. vulgatus , and proliferations of B. uniformis , B. finegoldii , B. ovatus and B. thetaiotaomicron also increased significantly in CM of B. caccae. The study suggests that the ability of Bacteroidetes species to degrade LBP-3 and sheds light on cooperative interactions of Bacteroides, Phocaeicola , and Parabacteroides species in the presence of LBP-3.

Lycium barbarum pulp addition improves the dough properties and gluten protein structure

Fei Hu,Yu-Zhu Song,Jin-Yu Li,Kiran Thakur,Jian-Guo Zhang,Zhao-Jun Wei

This study investigated the effects of Lycium barbarum pulp (LBP) on the properties of mixed dough and gluten protein. The results showed that appropriate addition of LBP (5 %) significantly improved the performance of the dough, promoted the aggregation of gluten protein, enhanced the water binding ability, and delayed the gelatinization of starch during cooking. Compared with the control group, the peak temperature (T p ) of the LBP sample gradually increased from 63.23 °C to 65.56 °C, the expansion force reduced by about 21.56 %, the absolute Zeta potential lowered by about 18.4 %, and the α -helix content and β -folding increased by 32.36 % and 10.23 %, respectively, indicating the more orderly and stable overall structure. However, LBP did not change the crystal configuration of starch and still showed typical type A line diffraction. Moreover, the addition of LBP increased the polyphenol content, which further improved the antioxidant properties and provided the possibility to improve the health potential of the flour.

Lycium barbarum polysaccharides grafted with doxorubicin: An efficient pH-responsive anticancer drug delivery system

Ying Wang,Feng Bai,Qiang Luo,Mingliang Wu,Gang Song,Hongmei Zhang,Jian Cao,Yanqing Wang

Lycium barbarum polysaccharide-doxorubicin (LBP-DOX) nanoparticles were synthesized via a one-step Schiff base reaction between oxidized Lycium barbarum polysaccharide (oxidized LBP) and doxorubicin (DOX). The structure of LBP-DOX nanoparticles was characterized by multi-spectral methods, 1 H nuclear magnetic resonance ( 1 H NMR), transmission electron microscopy (TEM), and thermo gravimetric analysis (TGA). The rheological properties of LBP-DOX nanoparticles were studied. With the increase of shear rate, the viscosity of LBP and LBP-DOX nanoparticles gradually decreases at 25 °C. Under the same strain, the storage modulus (G′) and loss modulus (G″) of LBP-DOX nanoparticles were inferior to LBP. The DOX release from LBP-DOX nanoparticles was pH-dependent and was accelerated by decreasing pH. Cytotoxicity study showed that the LBP-DOX nanoparticles have significantly enhanced cytotoxicity in vitro, especially for human Hepatic cancer cell line HepG2.

Effects of Lycium barbarum on gluten structure, in vitro starch digestibility, and compound noodle quality

Fei Hu,Jin-Yu Li,Peng-Ren Zou,Kiran Thakur,Jian-Guo Zhang,Mohammad Rizwan Khan,Zhao-Jun Wei

Lycium barbarum , a plant of the Solanaceae family with medicinal and food origins, is rich in many bioactive substances. This study examined the effects of Lycium barbarum pulp (LBP) on the gluten structure, starch digestibility, and compound noodle quality. The addition of LBP (typically less than 5%) could improve the performance and structure of gluten proteins. Compared with the control group, when LBP addition was 5%, T p and ΔH increased significantly ( p  <  0.05 ), and the stability of gluten protein thermal increased. At this time, LBP also had a certain reduction effect on the surface hydrophobicity (from 0% to 5%), and the concentration of free thiol group increased from 1.93 μmol/g to 4.19 μmol/g. In addition, the LBP addition improved the gluten structure resulting in favorable texture, stretchability, cooking properties, and flavor of noodles. The hardness and recovery significantly increased by about 25.97% and 11% ( p  <  0.05 ), and the cooking yield and cooking loss also changed significantly ( p  <  0.05 ). More than 5% of an addition was reported to exhibit detrimental effects. The addition of LBP greatly reduced the in vitro starch digestibility and the expected glycemic index ( p  <  0.05 ). The scanning electron microscope (SEM) and atomic force microscope (AFM) analysis showed that the bioactive components of LBP can mix with gluten protein, bringing the structure of noodles closer together and intensifying the cross-linking of the gluten structure. Our results indicated that gluten protein and the starch-gluten composite system were overall improved by LBP addition, while the starch digestibility was lowered.

Characterization and Evaluation of Antioxidant and Anti-Inflammatory Activities of Flavonoids from the Fruits of Lycium barbarum

Tingting Yang,Yuhang Hu,Yamei Yan,Wangting Zhou,Guijie Chen,Xiaoxiong Zeng,Youlong Cao

The fruits ofLycium barbarumare rich in flavonoids, which may contribute to the health-promoting function ofLycium barbarum. However, the composition of flavonoids in the fruits ofLycium barbarum(LBFs) has received little attention. Thus, the goal of this work was to identify more kinds of flavonoids from fruits ofLycium barbarumby liquid chromatography–mass spectrometry. The potential antioxidant and anti-inflammatory activities of LBFs in vitro were also investigated. Thirteen flavonoid compounds were identified in LBFs, of which daphnetin, 6,7-dihydroxycoumarin, astragalin, taxifolin, eriodictyol, naringenin, and chrysoeriol were identified for the first time in the fruits ofLycium barbarum, which greatly enriched the variety of flavonoids in the fruits ofLycium barbarum. LBFs showed a similar superior antioxidant activity to vitamin C. Furthermore, LBFs exhibited an anti-inflammatory activity by suppressing the production of nitric oxide and pro-inflammatory cytokines, including tumor necrosis factor-alpha, interleukin-1β, and interleukin-6, in lipopolysaccharide-treated RAW264.7 macrophage cells. This study demonstrated the potential development of LBFs as functional foods. Keywords:Lycium barbarum;flavonoids;liquid chromatography mass spectrometer (LC-MS);antioxidant;anti-inflammatory

Lycium-Barbarum Polysaccharide-Loaded Dual-Crosslinked Rigid Hydrogel Enhances Bone Healing in Diabetic Bone Defects by Scavenging Reactive Oxygen Species

Wenjie Zhong,Wenao Liao,Lingcong Xu,Niezhenghao He,Ke Xu,Caiyuan Liu,Fei Wang,Wei Zhang,Jiang Hu,Haowen Cui

Diabetes-induced oxidative stress can lead to poor bone defect healing, severely affecting the quality of life for patients. Studies show that improving the microenvironment and promoting bone formation can effectively accelerate the healing of bone defects. However, traditional local drug delivery methods face various challenges during the treatment process. Therefore, this study develops a novel hydrogel (HLBP) loaded with natural protein polysaccharides (LBP) extracted from goji berries, aiming to enhance the healing of diabetic bone defects. The hydrogel is composed of freeze-dried polyvinyl alcohol (PVA) and photocrosslinked poly (ethylene glycol) diacrylate (PEGDA). This hydrogel exhibits excellent biocompatibility. Additionally, it demonstrates effective loading capacity for the LBP. LBP's bioactivity enables ROS scavenging and promotes bone regeneration at defect sites. In vitro, experimental results show that HLBP significantly reduces ROS levels and enhances osteogenic differentiation ability and cell viability of human bone marrow mesenchymal stem cells. In vivo studies using BKS-db diabetic mice show that HLBP implantation at bone defects achieves over 80% healing, highlighting its strong healing potential. This method effectively avoids potential toxicity from systemic drug administration and significantly promotes regeneration at the bone defect site, providing a new strategy for treating diabetic bone defects.

Lycium barbarum glycopeptide reduces inflammation and fibrosis in corneal injury by modulating the NF-κB/NLRP3/IL-1 β signaling pathway and microRNA-21a-5p/SMAD7

Yarong Yan,Shulei Xing,Jinghua Liu,Xinlin Yan,Yi Guan,Zhixin Jiang,Wei Zhang,Xuan Li

Lycium barbarum glycopeptide (LbGp), derived from the Chinese medicinal plant Lycium barbarum, has demonstrated anti-inflammatory properties; however, its precise role and mechanism in corneal repair following injury remain elusive. The present research investigated the mechanisms and effects of LbGp on corneal repair following alkali burn injury using in vivo mouse models of corneal alkali burn and in vitro human keratocyte fibrosis models. Corneal inflammation, opacity, and epithelial defects were assessed via a slit lamp microscope. Results showed that LbGp-treated mice exhibited reduced edema, accelerated re-epithelialization, and decreased corneal opacity compared to the phosphate-buffered saline (PBS)-treated controls. Proteomic analysis revealed altered proteins enriched in the extracellular matrix among the control, injury, and LbGp treatment groups. Moreover, LbGp significantly attenuated TGFβ-1-induced myofibroblasts transdifferentiation from keratocytes. Consistently, LbGp treatment inhibited the upregulation of fibrosis markers (αSMA, fibronectin, and collagen III) at both the protein and mRNA levels after corneal alkali burns. LbGp also effectively suppressed the activation of the NF-κB/NLRP3/IL-1β signaling pathway and neutrophil infiltration following corneal alkali burn injury. Additionally, miR-21 was upregulated in TGFβ-1-stimulated keratocytes and in the alkali-burned mouse cornea. LbGp decreased miR-21 expression, while increasing expression of its target, Smad7, thereby dampening the TGFβ/Smad2/3 signaling pathway. This research demonstrates that LbGp promotes corneal healing by inhibiting inflammation and fibrosis after alkali burns, suggesting its potential as a supplementary therapy for corneal injury repair.

In vitro digestion under simulated saliva, gastric and small intestinal conditions and fermentation by human gut microbiota of polysaccharides from the fruits of Lycium barbarum

Yu Ding,Yamei Yan,Yujia Peng,Dan Chen,Jia Mi,Lu Lu,Qing Luo,Xiaoying Li,Xiaoxiong Zeng,Youlong Cao

In the present study, the digestion under simulated saliva, gastric and small intestinal conditions and in vitro fermentation by human gut microbiota of Lycium barbarum polysaccharides (LBPS) were investigated. As a result, LBPS contained carbohydrates, uronic acids, protein and polyphenols of 60.96 ± 5.58, 20.98 ± 2.49, 5.30 ± 0.12 and 1.04 ± 0.06%, respectively. Furthermore, LBPS was mainly consisted of glucuronic acid (5.12 mol%), galacturonic acid (12.40 mol%), glucose (2.15 mol%), galactose (39.67 mol%) and arabinose (40.66 mol%). Under simulated saliva, gastric and small intestinal conditions, LBPS was not affected. Whereas, LBPS could be utilized by gut microbiota based on the consumption of monosaccharides, decreased molecular weight and lower content of total carbohydrates after fermentation in vitro . With the proceeding of fermentation, LBPS significantly promoted the production of short-chain fatty acids. Meanwhile, LBPS could alter the microbial community after 24 h fermentation, especially enhancing the relative abundances of genera Bacteroides , Bifidobacterium , Phascolarctobacterium , Clostridium XlVb , Prevotella and Collinsella. All the results suggested that LBPS could be developed as special ingredients for modulating gut microbiota composition and promoting the growth of beneficial bacteria like Bifidobacterium .

The polysaccharides from the fruits of Lycium barbarum L. confer anti-diabetic effect by regulating gut microbiota and intestinal barrier

Wangting Zhou,Tingting Yang,Weiqi Xu,Yujie Huang,Linwu Ran,Yamei Yan,Jia Mi,Lu Lu,Yi Sun,Xiaoxiong Zeng,Youlong Cao

The antidiabetic effect and potential mechanisms of the polysaccharides from the fruits of Lycium barbarum L. (LBPs) by the mouse model of high-fat diet/streptozotocin-induced diabetes were investigated. Six-week oral administration of LBPs (200 mg/kg/day) resulted in improvement in the levels of fasting blood glucose (13.51% decrease) and glycated hemoglobin and β-cell function in diabetic mice, and simultaneously induced a 3.3-fold increment in one taxon belonging to genus Allobaculum in gut bacterial community. The experiments of fecal microbiota transplantation and antibiotics treatment confirmed that the LBPs-mediated gut microbiota participated in the glycemic control of the diabetes management. Moreover, LBPs intervention guarded the intestinal barrier function via upregulating the expression of zonula occludens 1 both in vivo (analyzing the gut permeability in diabetic mice) and in vitro (using intestinal-like Caco-2/RAW264.7 cells co-culture inflammation model). Collectively, our study showed that LBPs could confer anti-diabetic effect through modifying gut microbiota and intestinal barrier.

Lycium barbarum lipid-based edible nanoparticles protect against experimental colitis

Menghang Zu,Heliang Song,Jiabin Zhang,Qiubing Chen,Shiqin Deng,Brandon S.B. Canup,Yi Yuan,Bo Xiao

Edible plant-derived nanoparticles (NPs) have attracted increasing attention in the treatment of ulcerative colitis (UC). Lycium barbarum (LB), a popular functional fruit, possesses various biological functions. Here, fat-soluble contents were extracted from LB and further processed into LB lipid-derived NPs (LBLNs). The resultant NPs had an average hydrodynamic diameter around 189.2 nm, narrow size distribution (polydispersity index = 0.2), and negative surface charge (-34.9 mV). Moreover, they could be efficiently taken up by UC therapy-related target cells (macrophages), and over 69.0 % of macrophages internalized LBLNs after 4 h co-incubation. We further found from the in vitro results that LBLNs had strong capacities to inhibit the secretion of the main pro-inflammatory cytokines (TNF-α and IL-12) and up-regulate the expression of the typical anti-inflammatory factor (IL-10). Finally, mice experiments confirmed that LBLNs after oral administration could specifically accumulate into inflamed colon tissues, and further attenuate UC-relevant symptoms ( e.g. , bodyweight loss, colon shortening, increase of spleen weight, and histopathological appearance, as well as ulceration). Collectively, this study demonstrates the excellent therapeutic outcomes of LBLNs against UC and provides a promising edible nanotherapeutic for UC treatment.

Properties of Novel Components of Polysaccharides Isolated From Lycium barbarum Fruits

Yuling Chen,Xueluan Liu,Genglin Lu,Xuan He,Dandan Li,Yunong Tian,Xiaolin Feng,Yong Ye

This study has established structure-function relationships of Lycium barbarum polysaccharides (LBPs) relevant to antioxidant and tyrosinase-inhibitory functions in food systems by integrating optimized extraction, stepwise purification, comprehensive structural analysis, and functional verification, offering a novel multimethod approach to explore structure-based mechanisms of bioactivity. Firstly, hot-water extraction optimized at 30 mL/g, 90°C, and 5 h yielded 7.44 % ± 0.17 % crude LBPs. Secondly, ion-exchange followed by gel-filtration chromatography separated two purified fractions: LBP-1 (115.1 kDa) and LBP-2 (73.5 kDa). LBP-1 exhibited a compact morphology and featured spectral characteristics indicative of a triple-helix structure, along with both α - and β -glycosidic linkages. In contrast, LBP-2 showed a more amorphous configuration without triple-helix features and predominantly β -linkages. Their monosaccharide profiles also differed, with LBP-1 enriched in glucose, mannuronic acid, galacturonic acid, and glucuronic acid, while LBP-2 contained glucose, galacturonic acid, mannuronic acid, and arabinose. Both fractions exhibited in vitro antioxidant and tyrosinase-inhibitory activities, with LBP-2 demonstrating relatively stronger effects. The enhanced performance of LBP-2 may be associated with its dispersed structure and uronic acid–rich β -linked motifs, which increase the accessibility of functional sites. These insights support the potential application of both LBP-1 and LBP-2 in functional foods, with LBP-2 showing greater promise for use in beverages and fresh-cut fruit products requiring natural antibrowning and antioxidant solutions.

Optimization of ultrasonic extraction of Lycium barbarum polysaccharides using response surface methodology

Jitian Song,Shi Dongqi,Su Hang,Feng Yongxia,Tian Wei

Ultrasonic extraction was a new development method to achieve high-efficiency extraction of Lycium barbarum polysaccharides instead of hot water extraction. In this paper, the single factor method combined with the box Behnken design of response surface method was used to study the influence of extraction time, extraction temperature, material liquid ratio and ultrasonic power on the extraction rate of L . barbarum polysaccharide. The results indicated that the best extraction rate of L . barbarum polysaccharide was 12.54 ± 0.12% under the conditions of 80 min for extraction time, 73 °C for extraction temperature, 1 g:38 mL for material to liquid ratio, and 185 W for ultrasonic power. Under the same operating conditions, the yield of L . barbarum polysaccharide using ultrasonic extraction was 83.3%, which was higher than that hot water extraction. Moreover, the extraction time of ultrasonic extraction was only 47% of that using hot water extraction. This suggested that there was great potentials of using ultrasonic extraction in the realization of high-efficiency extraction of L. barbarum polysaccharide. The results of this study could also provide a theoretical basis for the coupling of ultrasonic extraction and ultrasonic concentration process to develop the integrated equipment of both ultrasonic extraction and ultrasonic concentration.
