Phenolamides from Rootstocks of Lycium ruthenicum Murr by Ligand Fishing and Their Neuroprotective Effects

Yi-Kao Hu,Ayzohra Ablat,Xiao-Lin Bai,Xiang Yin,Chao Ma,Xun Liao

Lycium ruthenicum Murr is an important edible and medicinal plant widely cultivated in Northwest China. Our previous study found that its fruit possessed rich polyphenols exhibiting significant neuroprotective effects, inspiring us to investigate the active constituents of its rootstocks, which have never been reported. In this study, we rapidly identified four phenolamides (1–4) possessing monoamine oxidase B (MAO-B) inhibitory effect by using MAO-B-functionalized cellulose filter paper-based ligand fishing, and their structures were elucidated with NMR and MS. Compounds 1, 2, and 4 exhibited moderate inhibitory effect on MAO-B with IC50 values of 71.44 ± 1.81, 28.97 ± 1.20, and 25.58 ± 0.74 μM, respectively, and enzyme kinetic analysis, molecular docking, and molecular dynamics further revealed the inhibition mechanism. All four phenolamides displayed a neuroprotective effect against 1-methyl-4-phenylpyridinium -induced SH-SY5Y cells model, among which 2 and 4 significantly reduced intracellular MAO-B activity and improved the ATP content. Western blot revealed that compounds 2 and 4 alleviated the oxidative stress through up-regulation of the expression of Nrf2 and HO-1 protein, promoting mitophagy via activating the PINK1/Parkin pathway. Moreover, compounds 2 and 4 extended the lifespan, dopamine and ATP levels, locomotor behavior, and olfactory ability of the PINK1B9 flies, a drosophila model of Parkinson’s disease. Meanwhile, compounds 1–4 showed potent antioxidative activities on DPPH, ABTS+, and PTIO radical scavenging assays. These results revealed the health-promoting effects of the rootstocks in preventing and treating neurodegenerative diseases, laying the scientific foundation for the exploitation of this plant byproduct for the development of functional food formulations and pharmaceutical products.

Extract from Lycium ruthenicum Murr. Incorporating κ-carrageenan colorimetric film with a wide pH–sensing range for food freshness monitoring

Jingrong Liu,Hualin Wang,Min Guo,Linlin Li,Minmin Chen,Suwei Jiang,Xingjiang Li,Shaotong Jiang

This study was to design a novel wide pH–sensing colorimetric film of κ-carrageenan incorporated with extract from Lycium ruthenicum Murr. (Car-LRM) for food freshness monitoring. The structure, basic properties (thermal stability, mechanical, barrier and antioxidant activity), and release kinetics of LRM from the films were investigated. Attentions were centered on colorimetric response of films and their applications for freshness monitoring of milk and shrimps. A low incorporation of LRM (no more than 2.5%) led to an enhancement in thermal stability and water vapor barrier of films to a certain extent. At above level of incorporation, LRM could be well-distributed in Car matrix by the interactions of hydrogen bonds. In addition to good antioxidant activity , the film displayed colorimetric response at a wide range of pH (2−10) and the color change was reversible. The release of LRM from films followed Fickian's law which was controlled by diffusion mechanism rather than swelling mechanism. The successful applications of Car-LRM2.5 for the freshness monitoring of milk and shrimps indicate that Car-LRM2.5 may have great potential for monitoring the freshness of milk and aquatic products.

Lycium ruthenicum Murray exosome-like nanovesicles alleviated Alzheimer's disease–like symptoms induced by Aβ protein in transgenic Caenorhabditis elegans through the DAF-16 pathway

Yadan Zhang,Xiaoyu Zhang,Jie Zhou,Yuting Li,Tianhan Kai,Lin Zhang

Alzheimer's disease (AD) is predominantly characterized by cholinergic dysfunction, mitochondrial impairment, oxidative stress, and inflammation, primarily driven by amyloid-beta (Aβ) peptides. This study investigates the protective effects of Lycium ruthenicum Murray–derived exosome-like nanoparticles (LELN) in AD models using transgenic Caenorhabditis elegans ( C. elegans ). Findings showed that C. elegans effectively internalized LELN, which remained stable in vivo. Compared with untreated controls, treatment with 600 μg/mL LELN significantly extended the lifespan of CL4176 [ myo-3p ::Aβ 1–42 ] and CL2006 [ unc-54 /Aβ 1–42 ] worms by 34.78 % and 34.85 %, respectively, and delayed Aβ-induced paralysis by 52.42 % and 42.72 %, respectively. Furthermore, LELN increased the chemotaxis index of CL2355 [ snb-1 ::Aβ 1–42 ] worms from 11.11 % to 55.56 %. Mechanistically, LELN reduced the levels of Aβ oligomers and monomers via the DAF-16 pathway, consequently alleviating AD-like symptoms in transgenic C. elegans . The effects of LELN include inhibiting acetylcholinesterase activity to mitigate cholinergic dysfunction, restoring mitochondrial membrane potential and adenosine triphosphate production to ameliorate mitochondrial dysfunction, and reducing oxidative stress and inflammation. Collectively, these results highlight the protective role of LELN against Aβ-induced AD pathology and underscore their potential as a therapeutic candidate for AD treatment.

Extraction optimization and purification of anthocyanins from Lycium ruthenicum Murr. and evaluation of tyrosinase inhibitory activity of the anthocyanins

Meilun Shen,Kang Liu,Yefei Liang,Guangxin Liu,Jie Sang,Cuiqin Li

The aim of this study was to extract and purify anthocyanins from Lycium ruthenicum Murr. and evaluate their tyrosinase inhibitory activity. Response surface methodology was devoted to optimize enzyme-assisted extraction of anthocyanins from L. ruthenicum dried fruits. Extraction at 38 °C for 37 min using water-containing pectinase (52.04 mg/100 g dried fruit) rendered an anthocyanin extraction yield of 19.51 ± 0.21 mg/g. The purified anthocyanins were separated from the extract by macroporous resin XDA-6. Antioxidant tests in vitro suggested that the extract and the purified anthocyanins exhibited a potent 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging capacity, 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging capacity, hydroxyl radical scavenging capacity, superoxide radical scavenging capacity, and total reducing power. Thirteen anthocyanins from L. ruthenicum dried fruits were analyzed by HPLC-MS. Moreover, the purified anthocyanins had inhibitory effect on tyrosinase monophenolase (IC 50 = 1.483 ± 0.058 mg/mL), and the type of inhibition was competitive inhibition ( K i = 39.83 ± 1.4 mg/mL). The maximum inhibitory activity of the purified anthocyanins (3.00 mg/mL) on tyrosinase diphenolase was 42.16 ± 0.77%, and the type of inhibition was anticompetitive inhibition ( K is = 2.387 ± 0.10 mg/mL). Practical Application The anthocyanins from L. ruthenicum dried fruits can be used as tyrosinase inhibitors in medicine, cosmetics, and food preservation industries.

Polyphenolic Glycosides from the Fruits Extract of Lycium ruthenicum Murr and Their Monoamine Oxidase B Inhibitory and Neuroprotective Activities

Yi-Kao Hu,Xiao-Lin Bai,Hao Yuan,Yi Zhang,Emmanuel Ayodeji Ayeni,Xun Liao

The fruits ofLycium ruthenicum Murr have long been consumed as health food and used in folk medicine in China. Apart from the well-known polysaccharides, the active small molecular constituents in this fruit have not been fully studied. In this work, a systematic phytochemical study was carried out to investigate the small molecules and their potential health benefits. Nine new polyphenolic glycosides, lyciumserin A–I (1–9), together with 16 known compounds (10–25), were isolated and elucidated by high-resolution electrospray ionization mass spectrometry and comprehensive NMR analyses in combination with chemical hydrolysis. Compounds 1, 2, and 16 exhibited moderate inhibitory activity of monoamine oxidase B (MAO-B), while compounds 1 (50 μM) and 2 (100 μM) displayed significant neuroprotective effects (69.22 and 72.38% of cell viability, respectively) in the 6-hydroxydopamine-induced injury of the PC12 cell model (54.41%), comparable to the positive drug rasagiline (70.45%). The neuroprotective effect of 1 and 2 was further evidenced by the observation of the morphological change and fluorescein diacetate/propidium iodide staining. In addition, the levels of the major active compounds (1, 3, 5/6, and 16–18) vary from 21.5 to 892.3 μg/g. This is the first report on phenolic glycosides from the fruits ofL. ruthenicum Murr that possess both significant MAO-B inhibitory and neuroprotective effects, indicating the promising potential of the fruits for the development of health care products and even therapeutic agents for the treatment of Parkinson’s disease and other neurodegenerative diseases.

Lycium ruthenicum Murray derived exosome-like nanovesicles inhibit Aβ-induced apoptosis in PC12 cells via MAPK and PI3K/AKT signaling pathways

Yadan Zhang,Xiaoyu Zhang,Tianhan Kai,Lin Zhang,Anping Li

Plant-derived exosome-like nanovesicles (ELNs) are nano-sized vesicles extracted from edible plants. Lycium ruthenicum Murray (LRM) has been gaining increasing attention due to its nutritional and medicinal value, but the ELNs in LRM has not been reported. In this study, LRM-ELNs were obtained, and the proteins, lipids, microRNAs (miRNAs) and active components in LRM tissues and LRM-ELNs was analyzed by LC-MS/MS, LC-MS, high-throughput sequencing techniques, and physical and chemical analysis. LRM-ELNs can be uptaken by PC12 cells through macropinocytosis and caveolin-mediated endocytosis primarily. Transcriptomic and western blot experiments indicate that LRM-ELNs can inhibit Aβ-induced apoptosis in PC12 cells through the MAPK and PI3K/AKT signaling pathways, with miRNAs playing a crucial role. These results indicate that LRM-ELNs have the protection effect on PC12 cells and can be considered as dietary supplements for alleviating neurodegenerative diseases.

Study on anthocyanins from Lycium ruthenicum Murr via ultrasonic microwave synergistic extraction and its antioxidant properties

Dong Yuhe,Zhong Wenting,Yang Chunmiao,Zhang Yongze,Yang Dongsheng

The objectives of this study are to optimize the ultrasonic microwave synergistic extraction (UMSE) of anthocyanins from Lycium ruthenicum Murr (ALRM) by response surface methodology and also to investigate its antioxidant activity in vitro. Based on the single-factor experiments, we optimized the ALRM extraction process by response surface methodology assuming anthocyanin extraction rate as the response point and microwave power, ultrasonic power, extraction time, and liquid-to-material ratio as the investigating parameters as well as performed correlation analysis between DPPH·, ·OH, ABTS·, O2-· radical scavenging potential, and ferric-reducing antioxidant power. The results showed that the influence on anthocyanin yield was in the order of liquid to material ratio > microwave power > ultrasonic power > extraction time; the elicited extraction conditions were: ultrasonic power 216.253 W, microwave power 89.311 W, time 26.141 min, liquid to material ratio 17.294 mL/g, the forecast content was 10.157 mg/g, while the actual value was consistent with the preview value. Meanwhile, ALRM obtainable by UMSE was identified as yielding favorable antioxidant properties firstly, when the concentration of purified ALRM (ALRM-1) was increased, the antioxidant capacity was strengthened. Theoretically, we provide a basis for the extraction procedure of ALRM and its antioxidant activity, which serves as a promising antioxidant and free radical scavenger.

Isolation, antioxidant property and protective effect on PC12 cell of the main anthocyanin in fruit of Lycium ruthenicum Murray

Jilong Tang,Yamei Yan,Linwu Ran,Jia Mi,Yi Sun,Lu Lu,Yuxiao Gao,Xiaoxiong Zeng,Youlong Cao

The fruit of Lycium ruthenicum Murray has been widely used as a precious Chinese herbal medicine. In this study, the major anthocyanin in L. ruthenicum fruit was obtained via an AKTA purifier system with YMC-Pack ODS-A column and identified by mass and nuclear magnetic resonance spectroscopy to be petunidin 3- O -[6- O -(4- O -( trans - p -coumaroyl)-α- l -rhamnopyranosyl)-β- d -glucopyranoside]-5- O -[β- d -glucopyranoside] (compound 1 ). Compound 1 exhibited higher scavenging activities against 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid), 1,1-diphenyl-2-picrylhydra-zyl and superoxide radicals than the crude extract of anthocyanins. Moreover, compound 1 was demonstrated to have potential cytoprotective effect on hydrogen peroxide-induced intracellular reactive oxygen damage in neuronlike rat pheochromocytoma line cells by promoting cell proliferation, mitigating lipid peroxidation and regulating endogenous antioxidant enzymes (glutathione peroxidase, superoxide dismutase and catalase).

Modification of Lycium ruthenicum pomace dietary fibers: Enhanced physicochemical, structural, and functional properties

Huiling Li,Keyu Tian,Ruiqi Wang,Ruyu Shangguan,Jingna Yang,Nana Yang,Xintao Meng,Xiang Li,Ting Zhang,Hongjie Lei

Physical and functional properties of LPDFs were improved by modification methods. • Modification methods effectively increased soluble dietary fiber (SDF) content. • High-pressure homogenization (HPH) could loosen the dense structure of LPDFs. • Alkaline hydrogen peroxide (AHP) effectively enhanced thermal stability of LPDFs. • Complex enzymatic hydrolysis (CE) exhibited superior hydration capacity of LPDFs. Lycium ruthenicum pomace, a juice-processing by-product, is characterized by low soluble dietary fiber (SDF) content and limited functionality, which restricts its further application. In this study, the pomace dietary fibers (LPDFs) were modified using high-pressure homogenization (HPH), complex enzymatic hydrolysis (CE), and alkaline hydrogen peroxide (AHP), and the resulting changes in physicochemical, structural, and functional properties of both SDF and insoluble dietary fiber (IDF) fractions were evaluated. Modification treatments effectively increased SDF content (from 6.89% to 15.44%) and enhanced functional attributes. AHP produced the highest SDF yield (15.44%) and crystallinity (16.54%), and improved thermal stability. In contrast, CE and HPH treatments loosened the compact structure, reduced particle size, and increased antioxidant activity and viscosity. Notably, CE treatment yielded a substantial SDF increase (12.61%) and exhibited superior overall functional performance, including higher viscosity, rehydration capacities, and antioxidant activities. Our findings suggest that CE represents a more appropriate modification approach for LPDFs. Download: Download high-res image (264KB) Download: Download full-size image

Interspecies Developmental Differences in Metabonomic Phenotypes of Lycium ruthenicum and L. barbarum Fruits

Qi Wang,Shaohua Zeng,Xiangyu Wu,Hehua Lei,Ying Wang,Huiru Tang

Fruits of Lycium ruthenicum (LR) and L. barbarum (LB) in Solanaceae family contain abundant bioactive metabolites used widely as functional food and natural medicine. To characterize the fruit developmental molecular phenotypes, we comprehensively analyzed metabolite composition of both Lycium fruits at three developmental stages using the combined NMR, liquid chromatography–tandem mass spectrometry, and gas chromatography-flame ionization detector/mass spectrometry methods. The metabonomes of these fruits were dominated by over 90 metabolites including sugars, amino acids, tricarboxylic acid (TCA) cycle intermediates, fatty acids, choline metabolites, and shikimate-mediated plant secondary metabolites. Metabolic phenotypes of two species differed significantly at all three developmental stages; LB fruits contained significantly more sugars and amino acids but less TCA cycle intermediates, fatty acids, and secondary metabolites than LR. Interspecies differences for fatty acid levels were much greater after color-breaking than precolor-breaking. Furthermore, LR fruits contained more osmolytes than LB fruits indicating different osmoregulation requirements for these fruits during development. Significant differences were also present in biosynthesis of shikimate-mediated plant secondary metabolites in LR and LB. These findings provided essential metabolic information for plant physiology of these Lycium species and their utilizations demonstrating the usefulness of this metabonomic phenotyping approach for studying fundamental biochemistry of the plant development.

Anthocyanin and spermidine derivative hexoses coordinately increase in the ripening fruit of Lycium ruthenicum

Xiaoman Yang,Shuang Lin,Yongxia Jia,Fazal Rehman,Shaohua Zeng,Ying Wang

The Lycium ruthenicum (Lr) fruit is a widely used nutritional food that contains various bioactive components such as anthocyanin and spermidine derivatives. In the present study, ultra-high-performance liquid chromatography with tandem mass spectrometry was utilized to profile the metabolic dynamics of four developmental stages of Lr fruit. A total of 49 compounds, including anthocyanin, alkaloids, hydroxycinnamic acid derivatives, flavonoids, and amino acids, were tentatively identified. Principal component analysis distinguished the fruit at four developmental stages using 15 (9 were tentatively identified) potential marker compounds. Pearson correlation analysis suggested that anthocyanin and spermidine derivative hexoses had a strong positive correlation coefficient. A glucosyltransferase (HG27071) was confirmed to glucosylate both anthocyanidin and spermidine derivative in vitro . Our results provide insight into the metabolic linkages among bioactive components in Lr fruits. The glucosyltransferase identified in this study will promote its potential use in functional foods and natural pigment resources.

Fatty Acid and Phytosterol Composition, and Biological Activities of Lycium ruthenicum Murr. Seed Oil

Xiaohui Zhao,Banmacailang Dong,Pi Li,Wei Wei,Jun Dang,Zenggeng Liu,Yanduo Tao,Hongping Han,Yun Shao,Huilan Yue

Lycium ruthenicum Murr. (LRM, Solanaceae), also called “black wolfberry” in China, is extremely tolerant drought. Although black wolfberry has seeds that are rich in oil, being considered as a potential alternative for edible oil production, little information has been found regarding its characterization. The present study evaluated the fatty acid profile of the seed oil and its phytosterol contents. Moreover, its antioxidant potential and its inhibition activities on pancreatic lipase, and cholesterol esterase (CEase) were also determined. The key fatty acids of studied oil were linoleic (74.58%), oleic (11.82%), γ -linolenic (6.60%), palmitic (4.88%), and stearic (1.40%) acids, which together comprised 99.28% of the total fatty acids. Phytosterols including 24-methylenecholesterol, campesterol, stigmasterol, dihydrolanosterol, 24-methyldesmosterol, β -sitosterol, Δ 5 -avenasterol, cycloartenol, and Δ 7− avenasterol were identified for the 1st time. The inhibition values (IC 50 ) for the DPPH and ABTS radical scavenging capacities of the oil were 7.86 and 15.49 mg/mL, respectively. Additionally, LRM seed oil was also demonstrated to possess strong pancreatic lipase, and CEase inhibition activities with IC 50 values of 12.38 and 2.63 mg/mL, respectively. The mechanism study indicated that the inhibitory manner of the oil on CEase belonged to uncompetitive inhibition of reversible inhibition. The results derived from above studies suggest that LRM seed oil could be a potential source of high value phytochemicals for developing novel functional food.
