Structural characterization and immune activity evaluation of a polysaccharide from Lyophyllum Decastes

Xiao-Jun Li,Shi-Jun Xiao,Yi Heng Xie,Jiang Chen,Hai-Rong Xu,Yuan Yin,Rui Zhang,Tong Yang,Tong-Yu Zhou,Si-Yan Zhang,Pei Hu,Li-Ming Gao,Hui-Ping Peng

An innovative acidic hydrolysate fingerprinting workflow was proposed for the characterization of Lyophyllum Decastes polysaccharide (LDP) by ultra performance liquid chromatography-mass spectrometry (UPLC-MS). The crude polysaccharides were firstly separated and purified by using DE-52 column and the BRT GPC purification system, respectively. The molecular weight and monosaccharide content of homogeneous polysaccharides were ascertained by utilizing HPGPC and ion chromatography separately. Secondly, the linkage of LDP was identified by methylation analysis and 1D/2D NMR spectra. The UPLC-MS/MS was used to scan and identify the acidic hydrolysate products of LDP using the PGC column. The oligosaccharides were collected by chromatography and identified by mass spectrometry. Thirdly, the expression of IL-1β, IL-6, iNOS, TNF-α and IFNAR-I was measured in order to assess the immunological activity of LDP. Besides, the targeted receptors identification of polysaccharides was performed by screening the expression of TLRs family protein. The results showed that oligosaccharide fragments with different molecular weights can be obtained by partial hydrolysis, which further verified that the structures of LDP polysaccharides was a 1–6-linked β-glucan. Moreover, the LDP polysaccharide can up-regulate the content of IL-1β, IL-6, iNOS, TNF-α and IFNAR-I and plays an important immunoregulation role through TLRs family.

Effects of polysaccharides from Lyophyllum decastes (Fr.) Singer on gut microbiota via in vitro-simulated digestion and fermentation

Zhang Fangfang,Xiao Ying,Pan Liang,Yu Ling,Liu Yanfang,Li Deshun,Liu Xiaojie

Introduction: Lyophyllum decastes (Fr.) Singer polysaccharides (LDSPs) have been verified to possess strong biological properties. However, the effects of LDSPs on intestinal microbes and their metabolites have rarely been addressed. Methods: The in vitro-simulated saliva-gastrointestinal digestion and human fecal fermentation were used to evaluate the effects of LDSPs on non-digestibility and intestinal microflora regulation in the present study. Results: The results showed a slight increase in the content of the reducing end of the polysaccharide chain and no obvious change in the molecular weight during in vitro digestion. After 24 h in vitro fermentation, LDSPs were degraded and utilized by human gut microbiota, and LDSPs could be transformed into short-chain fatty acids leading to significant (p < 0.05) decrease in the pH of the fermentation solution. The digestion did not remarkably affect the overall structure of LDSPs and 16S rRNA analysis revealed distinct shifts in the gut microbial composition and community diversity of the LDSPs-treated cultures, compared with the control group. Notably, the LDSPs group directed a targeted promotion of the abundance of butyrogenic bacteria, including Blautia, Roseburia, and Bacteroides, and an increase in the n-butyrate level.Discussion: These findings suggest that LDSPs might be a potential prebiotic to provide a health benefit.

Polysaccharides from Lyophyllum decastes reduce obesity by altering gut microbiota and increasing energy expenditure

Tao Wang,Junjie Han,Huanqin Dai,Jingzu Sun,Jinwei Ren,Wenzhao Wang,Shanshan Qiao,Chang Liu,Li Sun,Shuangjiang Liu,Dianpeng Li,Shenglong Wei,Hongwei Liu

Polysaccharides are known to confer protection against obesity via modulation of gut microbiota . To expand our knowledge of mushroom-derived prebiotics , we investigated the structural characteristics and anti-obesity effects of Lyophyllum decastes polysaccharides. Two heteroglycans were purified and characterized. The isolated polysaccharides effectively reduced obesity and the related disorders in the diet-induced obese (DIO) mice. An altered gut microbiota with enrichments of Bacteroides intestinalis and Lactobacillus johnsonii and an increase of secondary bile acids were detected in the polysaccharide-treated mice. Supplementation of B. intestinalis and L. johnsonii prevented the obesity and hyperlipidemia in DIO mice, demonstrating their causal linkage to the efficacy of polysaccharides. An enhancement of energy expenditure in the brown adipose tissues due to up-regulation of the secondary bile acids-activated TGR5 pathway was deduced to be one of the mechanisms underlying the effect of polysaccharides. These results confirmed Lyophyllum decastes -derived polysaccharides as new prebiotics for preventing and treating obesity.

Characterization of chitosan/zein composite film combined with tea polyphenol and its application on postharvest quality improvement of mushroom (Lyophyllum decastes Sing.)

Xiaomin Wang,Xue Huang,Fengjuan Zhang,Fanyun Hou,Fangxuan Yi,Xia Sun,Qingqing Yang,Xiangbo Han,Zhanli Liu

In this study, chitosan/zein composite film combined with tea polyphenol was fabricated, and the effect of tea polyphenol on the structure and physical properties of chitosan/zein film was investigated. Low-field nuclear magnetic resonance spectroscopy indicated that the distribution and content of water molecules in chitosan/zein film were changed with the addition of tea polyphenol. The results of fluorescence measurement and secondary structure analysis proved the enhanced physical and structural properties of tea polyphenol composite film. And the film showed favorable stretching resistance and flexibility. Furthermore, the effect of composite film on quality and physiological indexes of mushroom ( Lyophyllum decastes Sing.) stored at 4 °C for 15 d were systematically evaluated. The results indicated that the tea polyphenol composite film showed the effectiveness of maintaining color, texture as well as integrity of cell membrane evidenced by relatively lower membrane permeability and malondialdehyde content of mushrooms. Besides, weight loss and respiration rate were efficiently retarded. In comparison with control and chitosan/zein film, the tea polyphenol composite film inhibited the activity of polyphenol oxidase and peroxidase, while enhanced the activity of antioxidant enzymes (superoxide dismutase and catalase). In a word, the chitosan/zein composite film incorporated with tea polyphenol showed the potential as an effective technology for maintaining the postharvest quality of mushroom ( Lyophyllum decastes Sing.).

Optimization of Protoplast Preparation Conditions in Lyophyllum decastes and Transcriptomic Analysis Throughout the Process

Xiaobin Li,Ying Qin,Yufei Kong,Samantha Chandranath Karunarathna,Yunjiang Liang,Jize Xu

Protoplasts are essential tools for genetic manipulation and functional genomics research in fungi. This study systematically optimized protoplast preparation conditions and examined transcriptional changes throughout the preparation and regeneration processes to elucidate the molecular mechanisms underlying the formation and regeneration of protoplasts inLyophyllum decastes. The results indicated an optimal protoplast yield of 5.475 × 106cells/mL under conditions of fungal age at 10 days, digestion time of 2.25 h, enzyme concentration of 2%, and digestion temperature of 28 °C. The Z5 medium supplemented withL. decastesmycelial extract achieved a high regeneration rate of 2.86. RNA-seq analysis revealed 2432 differentially expressed genes (DEGs) during protoplast formation and 5825 DEGs during regeneration. Casein kinase I, cytochrome P450 (CYP52), and redox-regulated input receptor (PEX5) were significantly upregulated during the protoplast stage, while β-1,3-glucan synthase (SKN1), chitin synthase (CHS2), hydrophobin-1, and hydrophobin-2 showed significant upregulation during the protoplast regeneration phase. These findings provide a reference for the efficient preparation and regeneration of protoplasts and offer new insights into the molecular mechanisms of protoplast formation and cell wall regeneration in fungi.

Exploring the Prebiotic Effects of Composite Fungal Polysaccharides From Hericium erinaceus and Lyophyllum decastes on Gut Microbiota and Metabolism

Xueying Yang,Yijun Zhou,Haining Zhuang,Lingyun Yao,Min Sun,Huatian Wang,Shiqing Song,Heng Yue,Qian Liu,Wencui Kang,Xueping Zheng,Chuang Yu,Tao Feng

Fungal polysaccharides have attracted considerable scholarly interest due to their potential as prebiotics, with the ability to modulate gut microbiota and enhance host health. Despite increasing recognition, the precise effects of composite polysaccharides from various fungal species on gut microbial composition and metabolic activity remain inadequately understood. This study sought to investigate the digestibility and fermentation characteristics of composite polysaccharides derived from Lyophyllum decastes (LDUP) and Hericium erinaceus (HEUP), combined at ratios of 1:1, 1:2, and 2:1 (LDHE1_1, LDHE1_2, and LDHE2_1). In vitro simulated gastrointestinal digestion and fermentation assays were conducted to assess pH variations, short-chain fatty acid (SCFA) production, and shifts in microbial communities. The findings revealed that the LDHE1_1 formulation exhibited the most significant capacity to reduce pH and produce SCFAs, particularly propionic and butyric acids. Furthermore, the LDUP and LDHE2_1 groups favored increased acetic acid production. Importantly, these composite polysaccharides substantially enhanced gut microbial diversity, with distinct formulations promoting specific bacterial taxa. The LDUP and LDHE1_1 groups increased Firmicutes and decreased Bacteroidota , while HEUP, LDHE1_2, and LDHE2_1 groups exhibited the reverse. At the genus level, the LDHE1_1 boosted Parabacteroides , and LDHE2_1 increased Phascolarctobacterium , Akkermansia , and Oscillibacter . Both the LDUP and HEUP notably raised Alistipes levels, and all polysaccharide groups suppressed potential pathogens such as Klebsiella and Escherichia-Shigella . This study provides new insights into how specific fungal polysaccharide combinations can modulate gut microbiota composition and metabolism, contributing to the design of targeted prebiotic formulations and functional foods.
