Salvia miltiorrhiza Bge (SMB) has long been used in traditional Chinese medicine to treat cardiovascular and cerebrovascular diseases. Growing clinical usage has led to a huge demand for artificial planting of SMB. Thus, continuous cropping of SMB is an important challenge that needs to be addressed. Continuous cropping can alter the metabolic profile of plants, resulting in poor growth and low yield. In this study, we tried to image the spatial location and variation of endogenous metabolites in continuously cropped SMB using matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI). Spatially resolved expressions of tanshinones, salvianolic acids, polyamines, phenolic acids, amino acids, and oligosaccharides in normal and continuously cropped SMB roots were compared. The expressions of dihydrotanshinone I, tanshinone II A, dehydromiltirone, miltirone, dehydrotanshinone IIA, spermine, salvianolic acid B/E, tetrasaccharide, and pentasaccharide in continuously cropped SMB roots were much lower than those in normal roots. There was little difference in the expressions of caffeic acid and salvianolic acid A in normal and continuously cropped SMB roots. Ferulic acid was more widely distributed in xylem of normal SMB but strongly expressed in xylem, phloem, and cambium of continuously cropped SMB. The spatially resolved metabolite information enhances our understanding of the metabolic signature of continuously cropped SMB and also provides insights into the metabolic effects of continuous cropping in other plants.

Red ginseng, a processed product of ginseng, has been shown to possess various pharmacological effects. To investigate the changes in ginsenosides during the processing of ginseng into red ginseng and to visualize their spatial distribution characteristics in tissues, ultra-high performance liquid chromatography coupled with hybrid quadrupole-orbitrap high-resolution mass spectrometry (UPLC-Q-Orbitrap HRMS) was used to detect and identify the extracts of ginseng and red ginseng. Matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI) was used to visually analyze the spatial distribution and temporal changes of prototype ginsenosides and their metabolites in ginseng. The UPLC-Q-Orbitrap HRMS results showed the identification of 42 major ginsenosides in both ginseng and red ginseng. Additionally, PCA and OPLS-DA analyses identified the processing-related markers for ginseng and red ginseng. During the processing of ginseng into red ginseng, the content of protopanaxadiol ginsenosides decreased (Rb1, Rc, Rd, etc.), while the types and content of rare ginsenosides increased (Rg3, Rg5, Rh1, etc.). The MALDI-MSI tissue distribution results demonstrated that ginsenosides in ginseng were mainly distributed in the cambium and phloem, while the processing led to the transformation of ginsenosides and an increase in distribution in the xylem and medulla. This study comprehensively reveals the chemical diversity and dynamic transformation patterns during the processing of ginseng, providing more substantial foundational information for research on red ginseng processing techniques. The transformation of these rare ginsenosides also indicates that ginseng is more beneficial for pharmacological applications such as anti-tumor, anti-allergic, and liver protection after being processed into red ginseng. Introduction Angelica sinensis is one of the most popular traditional Chinese medicines (TCM) and has been extensively used to treat various diseases. Hundreds of endogenous ingredients have been isolated and identified from this herb, but their spatial distribution within the plant root is largely unknown. Objectives In this study, we tried to investigate and map within-tissue spatial distribution of metabolites in Angelica sinensis roots. Material and Methods After optimization of experiment conditions, the 1,5-diaminonaphthalene (1,5-DAN) was chosen as the matrix and was sprayed on the surface of root sections. Then matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI) was employed to perform in situ detection and obtain detail spatial distribution information of metabolites in Angelica sinensis roots. Results The spatial distributions of a wide range of metabolites including organic acids, amino acids, oligosaccharides, and phospholipids were characterized and visualized in Angelica sinensis roots. Majority of these metabolites were located in the phloem and xylem, while ferulic acid was mainly present in the cork layer. The results revealed a dramatic metabolic heterogeneity among different regions of the roots and distinct spatial distribution patterns of different metabolites. Additionally, the metabolic pathways involved in the biosynthesis of choline were also successfully localized and visualized. Conclusion This study comprehensively characterized the spatial distribution of metabolites in Angelica sinensis roots, which would prompt the understanding of its chemical separation, biosynthesis, and pharmacological activities.

Antibiotics have important medical value, but they need to be monitored when used as veterinary drugs. We report α-cyano-4-hydroxycinnamic acid/ p -phenylenediamine (CHCA/PPD) hybrid as a novel matrix for enhanced matrix-assisted laser desorption/ionization-mass spectrometry imaging (MALDI-MSI) in situ the spatial distribution of antibiotic drugs in grass carp tissues. Method We have used MALDI-TOF-MSI in positive ion mode for the analysis of quinolones and sulfonamides in grass carps. A novel CHCA/PPD matrix was prepared and applied using a simple method to improve the analysis. Results Compared with the traditional matrix, CHCA/PPD significantly improved the detection intensity of quinolones and sulfonamides with better sensitivity (17.20%–94.30%) and reproducibility. For demonstration, this novel matrix was successfully applied to visualize enrofloxacin (ENR) in grass carp tissues, with the entire abundance differences clearly observed based on MALDI-MSI. The concentration levels in different tissues were determined, with a calibration curve of 10–2000 μg/ml (R 2 > 0.993). Conclusion This study was the first to introduce CHCA/PPD as a novel matrix, and the classical acid–base mixing was used to improve the ionization effect of the traditional matrix CHCA in MALDI. Based on CHCA/PPD, MALDI-MSI detected ENR in different grass carp tissues for the first time and realized the spatial distribution and concentration detection.

In recent years, the development and optimization of biodegradable coronary stents have become the research focus of many medical device manufacturers and scientific research institutions since they can be completely degraded and absorbed, and they restore vascular function. However, there is a lack of in situ quantification of these stents spatially in tissue in vivo . In this study, matrix-assisted laser desorption/ionization (MALDI) Fourier transform ion cyclotron resonance (FT ICR) and time-of-flight (TOF) mass spectrometric imaging (MSI) were used to analyze the time-dependent distributions of a biodegradable vascular scaffold, which consisted of copolymers of lactic acid and glycolic acid (PLGA) and its degradation products in cross-sections and longitudinal sections of blood vessels. The MALDI-MSI methods for analyzing the distribution of PLGA and its derivatives in vivo were established by optimizing the conditions of sample pretreatment and mass spectrometry (MS). In order to semi-quantify the contents of PLGA degradation products in blood vessels, self-made stainless-steel and indium tin oxide (ITO) target plates were developed to compare and establish the standard curves for semi-quantitative analysis. The target plate can be placed on the target carrier of MS simultaneously with the conductive slide, which can simultaneously carry out vapor deposition or spray on the substrate, to ensure the parallelism of the pretreatment experiments between the standards and the actual vascular samples. The proposed method provided a powerful tool for evaluating the distributions and degradation process of biological stent materials in the coronary artery, as well as provided technical support for the research and development of degradable biological stents and product optimization.

This study focuses on the potential hazards of borneol (BO) to aquatic organisms and human health. BO has antibacterial, anti-inflammatory and antioxidant activities, and is widely used in medicine, cosmetics, and detergents. In this study, zebrafish was used as a model organism to systematically evaluate the effects of BO on the heart, liver, kidney, and nervous system. The effects of BO on metabolites of zebrafish were studied using MALDI-MSI. The results showed that a high concentration of BO (500 μM) could induce morphological abnormalities (swim-bladder loss, spinal curvature, body-length shortening), cardiotoxicity (decreased heart rate, increased SV-BA distance), hepatotoxicity (reduced liver area index), and neurotoxicity (impaired behavioral ability, and dopamine neuron development deficits), but there was no renal toxicity observed in zebrafish. Additionally, MALDI-MSI analysis showed that BO exposure significantly altered the levels of metabolites, including phospholipids, fatty acids, choline, and amino acids. The contents of PC-34:1, PC-34:2, PI-36:4, PE-36:1, LysoPE-22:5, LysoPC-18:1, FA-18:2, phenylalanine, lysine and glutathione were significantly increased, while the contents of PC-38:6 and PC-40:6 were significantly decreased. Notably, BO elicited a significant alteration in the mRNA expression levels of genes associated with phospholipid metabolism, fatty acid metabolism, choline metabolism, and amino acid metabolism (such as elovl5 , chpt1 , chka , setd7 , hgd ). This study revealed that BO exerted toxicity on multiple organs and demonstrated that BO causes metabolic dysregulation in zebrafish. These findings provide a novel insight into the toxicity of BO.

Osteonecrosis of the femoral head (ONFH) is a common orthopedic disease characterized by disability and deformity. To better understand ONFH at molecular level and to explore the possibility of early diagnosis, instead of diagnosis based on macroscopic spatial characteristics, a matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI MSI) method was developed for ONFH disease for the first time. The most challenging step for ONFH MSI is to deal with human bone tissues which are much harder than the other biological samples studied by the reported MSI studies. In this work, the MSI sectioning method of hard bone tissues was established using tender acids and a series of test criteria. Small-molecule metabolites, such as lipids and amino acids, were detected in bone sections, realizing the in situ detection of spatial distribution of biometabolites. By comparing the distribution of metabolites from different regions of normal femoral head, ONFH bone tissue (ONBT), and adjacent ONFH bone tissue (ANBT), the whole process of femoral head from normal stage to necrosis was monitored and visualized at molecular level. Moreover, this developed MSI method was used for metabolomics study of ONFH. 72 differential metabolites were identified, suggesting that disturbances in energy metabolism and lipid metabolism affected the normal life activities of osteoblasts and osteoclasts. This study provides new perspectives for future pathological studies of ONFH.

Acquisition of tissue spatial distribution information is essential for identifying sites of action, assessing pharmacological effects and elucidating mechanisms, thereby providing evidence for drug development. Anthraquinones, including rhein, emodin, aloe-emodin, chrysophanol and physcion, are the main pharmacologically active constituents of rhubarb. However, as exogenous compounds, most anthraquinones are difficult to detect by conventional matrix-assisted laser desorption/ionization time-of-flight mass spectrometry imaging (MALDI-TOF-MSI) because of low tissue abundance, poor ionization efficiency and strong ion suppression from endogenous matrices. To overcome these limitations, we established a highly sensitive on-tissue chemical derivatization (OTCD) workflow for MALDI-TOF-MSI analysis of anthraquinones in mouse tissues. By optimizing matrix selection and OTCD labeling conditions, we first visualized the spatial distributions of rhein and chrysophanol in mouse small intestine and kidney tissues. The derivatization reagent 2-fluoro-1-methylpyridinium p-toluenesulfonate (FMPTS) selectively labels phenolic hydroxyl groups, enabling rapid and efficient in situ derivatization of anthraquinones on tissue sections. In on-tissue spiking experiments (1 μL/spot), FMPTS-OTCD method achieved a practical lowest tested concentration producing a signal of 100 pg/spot, with diagnostic ions clearly distinguishable above the local tissue background. In contrast, the underivatized workflow required a higher amount (≥10 ng/spot) for detectable signal, demonstrating that FMPTS-OTCD effectively overcomes the low-response bottleneck of traditional MALDI-TOF-MSI method for anthraquinones. The resulting derivatization increased signal to noise ratio by 20–1154 times and enabled reliable in situ visualization of rhein and chrysophanol in mouse tissues.

Epimedii Folium (EF) is a renowned traditional herbal medicine known for its kidney-tonifying effects. However, the spatial distribution of its components in renal tissue and the resulting metabolic regulations remain poorly understood. To facilitate quality control and efficacy evaluation of EF, MALDI-MSI integrated with UPLC-MS/MS was employed to comprehensively analyze the compositional and metabolic profiles. 97 compounds including 30 flavonoids identified, MALDI-MSI analysis revealed origin- and species-specific accumulation patterns of these bioactive compounds with samples from Longnan in Gansu Province exhibiting superior phytochemical quality. Subsequently, spatial metabolomics analysis was conducted on mouse kidney tissues to investigate the tissue-specific distribution and metabolism of EF components. It demonstrated that icariin was rapidly metabolized into bioactive derivatives in the mouse kidney. To elucidate the renal mechanism of EF in greater detail, region-of-interest (ROI) analysis was implemented, enabling precise localization of metabolic changes within microstructural regions of the kidney. Results indicated that EF modulated several key renal metabolic pathways: it enhanced antioxidant capacity and urea cycling through amino acid metabolism; reduced levels of xanthine and uric acid in purine metabolism; and increased phosphatidic acid and sphingosine in lipid metabolism, thereby improving membrane antioxidant defenses.

Abstract Oxidative stress following spinal cord injury (SCI) triggers diverse metabolic alterations, including those in the acrolein (ACR)-associated pathway, which involves reactive carbonyl species (RCSs), polyamines, amino acids, fatty acids, and lipids. Therefore, comprehensive mapping of these metabolites is critical for understanding disease progression of SCI. Although matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI MSI) is widely used to map endogenous compounds, the simultaneous broad-spectrum detection of multiple analyte classes within the oxidative stress network remains challenging. Thus, we developed a cooperative matrix (co-matrix) composed of 2-hydrazinoquinoline (2-HQ) and α-cyano-4-hydroxycinnamic acid (CHCA) for the MALDI MSI of rat spinal cord tissue after SCI. In this system, ion-pair formed between 2-HQ and CHCA achieved crystallization uniformity and spot-to-spot reproducibility, while 2-HQ selectively derivatizes RCSs in situ. Density functional theory calculations further revealed that the ionicity of the co-matrix enhances analyte ionization. Using this co-matrix, we imaged 41 distinct metabolites in rat SCI tissue sections with high signal-to-noise ratios, including two carbonyl compounds associated with ACR-related oxidative stress pathways. In comparison, CHCA alone allowed imaging of only 15 metabolites (predominantly lipids), while 2-HQ alone detected one. Temporal heatmap analysis of nine metabolites within the ACR-associated pathway revealed coordinated dynamics between ACR and glutathione, both rebounding at day 7 (one-way ANOVA, p < 0.01). Overall, this study provides an efficient strategy for the comprehensive imaging analysis of multiple metabolite classes in complex injured biological systems.

Neurotransmitters (NTs) and neuromodulators (NMs) are two of the most important neurochemicals in the brain, and their imbalances in specific brain regions are thought to underlie certain neurological disorders. We present an on-tissue chemoselective derivatization mass spectrometry imaging (OTCD-MSI) method for the simultaneous mapping of NTs and NMs. Our derivatization system consists of a pyridiniumyl-benzylboronic acid based derivatization reagent and pyrylium salt, which facilitate covalent charge labeling of molecules containing cis-diol and primary amino, respectively. These derivatization systems improved the detection sensitivity of matrix-assisted laser desorption/ionization (MALDI)-MSI and simplified the identification of amino NTs and nucleoside NMs by the innate chemoselectivity of derivatization reagents and the unique isotopic pattern of boron-derivative reagents. We demonstrated the ability of the developed method on brain sections from a hypoxia mouse model and control. The simultaneous imaging of NTs and NMs provided a method for exploring how hypoxic stress and drugs affect specific brain regions through neurotransmitter modulation.

Abnormal glucose metabolism has been identified as a key characteristic of tumorigenesis. Visualizing the levels of monosaccharides in the lesion tissues and bodies of cancer patients is conducive to uncovering patterns of glucose metabolic reprogramming and assisting in the early diagnosis of cancer. Consequently, based on 7-(diethylamino)coumarin-3-carbohydrazide (DCCH), which has mass spectrometry/optical dual-signal enhancement functionality, both an on-tissue derivatization strategy suitable for matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI) analysis and an in-solution derivatization strategy for capillary electrophoresis/laser-induced fluorescence (CE-LIF) analysis have been developed. During the on-tissue derivatization reaction, the nonvolatile MALDI matrix, 2,5-dihydroxybenzoic acid, was innovatively adopted as a proton donor. This approach not only efficiently catalyzes the derivatization process but also significantly minimizes analyte redistribution, and a low-cost airbrush application can achieve satisfying in situ derivatization. For further addressing the issue of the coexistence of multiple conformational isomers in biosamples, a rapid and efficient CE-LIF method was proposed for the separation of five DCCH-labeled monosaccharides (including three conformational isomers) within 8 min. By focusing on endometrial cancer (EC), the synergy of DCCH-mediated MALDI-MSI and CE-LIF analysis successfully achieved high-resolution in situ visualization of an obvious depletion difference in monosaccharides across the focal areas (tumor tissues, peritumoral tissues, and paracancerous tissues), and a comparative analysis was conducted on the serum monosaccharide profiles between patients with EC and those with benign uterine diseases. These analyses yielded novel molecular-level evidence, thereby facilitating a deeper understanding of the monosaccharide metabolic abnormalities associated with EC.
