Based on indigenous maps collected in British Burma by James George Scott (1851–1935) and kept at the Cambridge University Library, this paper offers an alternative history of the first years of British colonisation in Upper Burma in the 1880s and 1890s. The entangled scripts – Chinese characters, Shan, Burmese and English – as well as the materiality and the visual codes of these documents bring into view forms of contact that did not last long. Contextualising these maps with other kinds of sources – including Scott’s diaries and administrative reports – allows us to reconstruct their production as part of processes of intelligence gathering and frontier settlement. By tracing the more or less willing role of Burmese clerks, notables, guides and interpreters in the cartographic processes implemented by the English on the ground, we can reintroduce these actors into a history of cartography that has long been Eurocentric. Doing so reveals how the British had to rely on indigenous knowledge to control a territory quite unknown to them during the early years of colonisation.

Elucidating ligand–protein interactions is important in understanding the biochemical machinery for given proteins. Previously, formaldehyde (FH)-based labeling has been employed to obtain such structural knowledge, since reactive residues that participate in ligand–target interactions display reduced accessibility to FH-labeling reagents, and thus can be identified by quantitative proteomics. Although being rapid and efficient for probing proteinaceous lysine accessibility, here, we report an acetaldehyde (AcH)-labeling approach that complements with FH for probing ligand–target interactions. AcH labeling examines lysine accessibility at a more moderate reaction speed and hence delivers a cleaner reaction when compared to that of FH. The subsequent application of AcH to label RNase A without and with ligands has assisted to assign lysines involved in ligand–RNase A binding by detecting the time-dependent changes in accessibility profiles. We further employed multiple reaction monitoring (MRM) to quantify these ligand-binding-responsive sites when a variety of potential ligands were queried. We noted that the time-resolved abundance changes of these peptides can sensitively determine the ligand-binding sites and differentiate binding affinities among these ligands, which was confirmed by native mass spectrometry (MS) and molecular docking. Lastly, we demonstrated that the binding sites can be recognized by monitoring the chemical accessibility of these responsive peptides in cell lysates. Together, we believe that the proposed combined use of AcH-based lysine accessibility profiling, native MS, and MRM screening is a powerful toolbox in characterizing ligand–target interactions, mapping topography, and interrogating affinities and holds promise for future applications in a complex cellular environment.

Monolithic ZrB 2 , ZrB 2 -SiC, ZrB 2 -TiC and ZrB 2 -SiC-TiC were fabricated by hot pressing. Contour maps depicting grain size and mechanical properties were generated on the ZrB 2 -SiC-TiC ternary diagram through experimental data and software fitting. These visual representations effectively establish the relationships between chemical composition and material properties, despite the similar high hardness of three main compounds. Notably, the average grain size of ZrB 2 -SiC-TiC is significantly reduced by an order of magnitude compared to monolithic ZrB 2 . Both strength and hardness exhibit a strong correlation with grain size, while the elastic modulus demonstrates insensitivity to microstructure changes. The evolution of toughness proves to be complex.

Summary Drosophila has long been a successful model organism in multiple biomedical fields. Spatial gene expression patterns are critical for the understanding of complex pathways and interactions, whereas temporal gene expression changes are vital for studying highly dynamic physiological activities. Systematic studies in Drosophila are still impeded by the lack of spatiotemporal transcriptomic information. Here, utilizing spatial enhanced resolution omics-sequencing (Stereo-seq), we dissected the spatiotemporal transcriptomic changes of developing Drosophila with high resolution and sensitivity. We demonstrated that Stereo-seq data can be used for the 3D reconstruction of the spatial transcriptomes of Drosophila embryos and larvae. With these 3D models, we identified functional subregions in embryonic and larval midguts, uncovered spatial cell state dynamics of larval testis, and revealed known and potential regulons of transcription factors within their topographic background. Our data provide the Drosophila research community with useful resources of organism-wide spatiotemporally resolved transcriptomic information across developmental stages.

Background The continuous mutations in the haemagglutinin gene in A/H1N1 influenza viruses drives antigenic drift, necessitating frequent vaccine updates. Pseudovirus systems, with their flexible HA protein presentation and high-throughput luciferase-based reporter assays, provide a versatile approach for mapping the antigenic landscape of A/H1N1 viruses. Methods This study constructed a pseudovirus library comprising 123 representative strains and 21 vaccine strains to analyse the antigenic evolution of human A/H1N1 from 1918 to 2023. Additionally, a machine learning-based antigenicity evaluation model, PN-AgEvaH1, was developed using pseudovirus neutralisation assay data. Findings The pseudovirus neutralisation experiments identified eight antigenic clusters, each associated with distinct epidemiological characteristics. All vaccine strains were found to align closely with the prevalent circulating strains of their respective years. Key amino acid residues contributing to pdm09 antigenic clusters were also identified. Furthermore, the PN-AgEvaH1 model achieved a high antigenicity evaluation accuracy with a receiver operating characteristic area under the curve (ROC AUC) score of 0.990, providing a reliable tool for characterising A/H1N1 antigenic clusters and guiding vaccine selection. Interpretation The pseudovirus neutralisation (PN) assay demonstrated high accuracy in identifying antigenic matches between vaccine and circulating strains, highlighting its value in vaccine strain selection. This study represents the large-scale antigenicity evaluation of human A/H1N1 viruses using the PN assay and underscores its potential as a complementary or alternative approach to the HI assay in both experimental and computational applications. With growing attention being devoted to autonomous vehicle (AV) safety, people have recently attached importance to high-definition (HD) maps. HD maps are not limited by environmental factors and can limit AVs driving in certain lanes. HD maps provide accurate auxiliary information on factors such as road geometry, traffic sign placement, and traffic topology. Nowadays, most HD maps are made from point clouds data, and this data contains accurate 3D position information. However, the production costs associated with HD maps are substantial. This article proposes an algorithm that reduces a great amount of time and human resource. The algorithm is divided into three phases, lane lines’ extraction from point clouds, modelling lane lines with attributes, and building OpenDRIVE file. The algorithm extracts lane lines resting on intensity value within the range of roads. Next, it models lane lines by cubic spline interpolation with the result of first phase, and build the OpenDRIVE file following the announcement of OpenDRIVE. The final result is compared with the verified HD map from the mapping company to analyze the accuracy. The root mean square (RMSE) obtained were 0.069 and 0.079 m for 2D and 3D maps, respectively.

The current zwitterionic antifouling (AF) coatings generally have limited broad-spectrum AF ability, poor compatibility with coating matrixes and weak mechanical properties due to high water adsorption. In this work, we tried to incorporate zwitterionic component, namely, ( N -methoxyacylethyl)-3-aminopropyltriethoxysilane (MAPS) and ZnO nanoparticles into a moisture-curable polyacrylate (MCP) to fabricate marine AF coatings. AF behaviors were examined by laboratory and field assays towards different target foulants. Laboratory anti-protein ability towards bovine serum albumin promoted with the increase of MAPS and/or ZnO contents, verifying their synergistic AF effect towards the earliest pollutant in biofouling process. Laboratory AF test towards bacterium Pseudoalteromonas marina and diatom Cylindrotheca closterium also exhibited the synergistic AF effect between zwitterions and ZnO nanoparticles, however, depending on their concentrations. Under the highest contents of MAPS ( i.e., 30 %) and ZnO nanoparticles ( i.e., 50 %), the coatings showed the best laboratory AF properties, with the antibacterial rate, sterilization rate and anti-diatom rate of 86.7 %, 71.8 % and 87.6 %, respectively. As for the marine field AF performance, the ZnO nanoparticles content had a significant effect on improving the field antibacterial ability, while the effect of MAPS was not obvious. But for the field anti-diatom assay, MAPS and ZnO nanoparticles depicted synergistic effect on diatom resistance. After field exposure for 52 d, the coatings with 30 % MAPS and 50 % ZnO nanoparticles revealed the best comprehensive AF performance, i.e., 82.4 %, 95.0 % and 95.5 % reductions towards bacteria, diatoms and mussels, respectively, and moreover, maintained pencil hardness of 2 H after immersed in ASW for 9 months. Besides, the coatings were capable of restoring their original oleophobicity and AF capability after mechanical abrasion.

Determination of condensate oil saturation in condensate gas reservoirs is the key parameter for estimating condensate oil blockage and gas well productivity. In PVT experiments, the constant volume depletion (CVD) method is usually adopted to obtain condensate oil saturation. However, in this method, the effect of porous media is not considered. This study applied a low field nuclear magnetic resonance (NMR) test instrument in which 1D T2 and 2D T1–T2 NMR techniques are carried out to investigate the condensate oil saturation of “condensate gas-saturated” core plugs in the depletion process of 29 to 5 MPa at 25 °C. It was observed here that the T1/T2 ratios of condensate oil and gas at the signal peak are approximately 20 and 60, respectively, and the condensate oil and gas could be distinguished by positions in 2D NMR T1–T2 maps. Additionally, a standard equation (the relationship between the signal intensity and the condensate oil saturation) with an excellent linear relationship was established for the calculation of the condensate oil saturation in core plugs. The results indicated that the condensate oil first appears in micropores, and the retrograde condensation phenomenon of the core mainly comes from the contribution of mesopores and macropores; furthermore, the maximum condensate oil saturation in the porous medium is 5.36% higher than that in the PVT cylinder, and the dew point pressure in the porous medium is 11.83% higher than that in the PVT cylinder. The T1–T2 NMR technique may become an efficient approach for assessing fluid distributions and saturations, simultaneously possessing potential applications for characterizing fluid occurrence states in the core.
