Objectives The study investigated the solid reactants of amorphous calcium phosphate (ACP) nanoparticles with 10-methacryloyloxydecyl dihydrogen phosphate (MDP) in ethanol-aqueous solution, and the effect of ACP-MDP suspension as a novel primer on dentin micro-tesnisl bond strengths (MTBS) prior to application of self-etch adhesives. Materials and methods The ACP nanoparticles were synthesized. The solid reactants of ACP nanoparticles with MDP ethanol-aqueous solution were analyzed by TEM , SEM, XRD and AFM . After the ACP-MDP complexes were attacked by alkaline, acidic and self-etch adhesive challenges, they were analyzed with TEM. The polished mid-coronal dentin surfaces of six third molars were treated with ACP-MDP suspension, 5% MDP solution or not (served as control), and thoroughly water-sprayed. The pretreated dentin surfaces were applied with a self-etch adhesive (Clearfil S3 Bond) and placed with a composite resin (Clearfil Majesty). After 24 h, the resin-bonded samples were cut into multiple beams before the MTBS tests were performed. The failure modes were determined by a stereo-microscopy. The pretreated dentin surfaces were analyzed by SEM. The resin-dentin interfaces were analyzed with TEM. Results The ACP nanoparticles in the MDP ethanol-aqueous solution at certain molar ratio of Ca:MDP (0.12:1) could self-assemble into ACP-MDP complexes consisting of nanolayering structures. The ACP-MDP self-assembly went from ACP nanoparticles, beaded structures, and twig-like structures to networks. The ACP-MDP complexes can be resistant to adhesive challenge, but not to alkaline and acidic challenges. ACP-MDP complexes possessed a similar modulus of elasticity to dentin, and were stable at least for 3 months. Furthermore, the dentin surfaces treated with ACP-MDP suspension could significantly increase the dentin MTBS when compared with control and those treated with 5% MDP solution ( P < 0.05). Conclusion The ACP-MDP suspension at certain molar ratio of Ca:MDP (0.12:1) could be self-assembled into ACP-MDP complexes consisting of nanolayering structures, and could be used as a novel primer to greatly improve the dentin bond strengths.

Objective This study aimed to evaluate the potential interaction of chlorhexidine (CHX) and 10-methacryloyloxydecyl dihydrogen phosphate (MDP) and its effects on the durability of dentin bonding. Methods Two commercial adhesives were tested: a MDP-free adhesive (Single Bond 2, SB2) and a MDP-containing adhesive (Single Bond Universal, SBU). Teeth were randomly assigned to six groups and tested for micro-tensile bond strength (μTBS): Ctr, direct bonding with SB2; CHX, CHX conditioning and SB2; MDP, MDP conditioning and SB2; CHX + MDP, combined CHX and MDP conditioning and SB2; SBU, direct bonding with SBU; CHX + SBU, CHX conditioning and SBU. The potential interaction of CHX and MDP was assessed by measuring nanoleakage, in situ zymography, and chemoanalytic characterization via Fourier transform infrared spectroscopy (FTIR), X-ray photoelectron spectroscopy (XPS) and nuclear magnetic resonance (NMR). Specimens for μTBS and nanoleakage tests were first subjected to water storage for 24 h or 6 months. Results The initial μTBS values of the Ctr and CHX groups were significantly lower than those of the other four groups (P < 0.05). Water storage for 6 months significantly weakened all groups (P < 0.05), with the Ctr group showing the lowest μTBS. This group also showed more obvious nanoleakage than the other five groups. In situ zymography revealed that the Ctr group showed the strongest fluorescence and that the CHX + MDP group showed greater fluorescence than either CHX or MDP group. FTIR, XPS, and NMR indicated that MDP can interact with hydroxyapatite. NMR detected no Ca 2+ salt peak for MDP when it was combined with CHX. Significance The application of either CHX or MDP alone can improve dentin bond durability. However, CHX may interfere with the formation of MDP–Ca salts.

This study aimed to evaluate the effects of the combined application of dimethylaminohexadecyl methacrylate (DMAHDM) and 10-methacryloyloxydecyl dihydrogen phosphate (MDP) on dentin bonding and its antimicrobial properties. DMAHDM was incorporated into the adhesives at 5% by mass, where two commercial adhesives were used: (1) MDP-free adhesive (single bond 2); (2) MDP-containing adhesive (single bond universal, SBU). The microtensile bond strength (μTBS) and nanoleakage were analyzed on the combination. The effects of DMAHDM on the interaction between MDP and hydroxyapatite and the degree of conversion were assessed by Fourier transform infrared spectroscopy (FTIR). Antimicrobial properties were examined by live/dead staining assay, colony-forming units (CFU), and metabolic activity, and cytotoxicity was measured using cell counting kit-8 (CCK-8) assay. 5% DMAHDM or MDP-containing groups showed increased immediate μTBS compared with the control group. Nanoleakage showed that 5% DMAHDM or MDP-containing groups presented little amount of silver penetration, while the control group exhibited extensive silver uptake along the bonding interface after water storage aging. FTIR showed that the interaction between MDP and hydroxyapatite was not affected by DMAHDM, besides DMAHDM did not hamper the polymerization of the bonding agents. The adhesives added with DMAHDM inhibited biofilm formation and reduced the biofilm CFU and metabolic activity of Candida albicans and Streptococcus mutans ( P < 0.05). No cytotoxic or slight cytotoxic effect was detected for the experimental materials used (MDP and 5% DMAHDM). The addition of DMAHDM along with MDP can obtain improved dentin bonding with increased antimicrobial performance.

Objectives The chemical affinity between 10-methacryloyloxydecyl dihydrogen phosphate (MDP) and hydroxyapatite (HAp) is an important factor in the enamel bonding provided by MDP-based self-etch (SE) adhesives, besides microinterlocking mechanisms. This study aimed to investigate how phosphoric acid pre-etching affects MDP–Ca salt formation in the application of MDP-based SE adhesives. Methods Single Bond Universal (SBU), All Bond Universal (ABU), Clearfil Universal Bond Quick (CBQ), and a MDP-based all-in-one adhesive (EXP) were used in both SE and etch-and-rinse (ER) modes, along with Clearfil SE Bond and untreated enamel (UE) as controls. The MDP–Ca salts produced with or without etching were examined by nuclear magnetic resonance, Raman spectroscopy, X-ray photoelectron spectroscopy, X-ray diffraction, and Fourier-transform infrared spectroscopy. Zeta potential , contact angle, and scanning electron microscopy measurements were employed to elucidate the mechanism behind the changes in MDP/HAp chemical affinity upon pre-etching. Results The percentage of MDP–Ca salt in EXP_ER (73.13%) was higher than that in EXP_SE (43.27%). Characteristic CH 2 (1130, 1441, 2853, and 2909 cm −1 ), C C (1641 cm −1 ), and C O (1718 cm −1 ) bands were observed in the Raman spectra of EXP_ER. Pre-etching increased the negative zeta potential of the enamel surface compared to that of UE ( P < 0.001). The contact angles of MDP-based adhesives applied to pre-etched enamel were significantly lower than those of the self-etched surface ( P < 0.05). Significance The increased MDP–Ca salt formation is a significant advantage of phosphoric acid pre-etching, improving the MDP/HAp chemical affinity in addition to increasing surface wettability.

Objectives This study introduced the prime-and-rinse approach with an methacryloyloxydecyl dihydrogen phosphate (MDP)-containing primer to augment the performance of modified self-etch adhesives loaded with amorphous fluorinated calcium phosphate (AFCP) nanoparticles. Methods The modified self-etch adhesive was acquired by adding 20 wt% AFCP nanoparticles into Clearfil S3 Bond. An experimental primer containing 15 wt% MDP was prepared. The bond performance was assessed through micro-tensile bond strength (MTBS) tests. Additionally, a single-layer collagen model and demineralized dentin were used to investigate mineralization characteristics. Cytotoxicity was evaluated using a modified transwell insert model on human dental pulp stem cells. Results The incorporation of AFCP nanoparticles into the Clearfil S3 Bond led to a significant reduction in MTBS. While the application of MDP primer could improve the bond performance of modified Clearfil S3 Bond. The MDP primer further augmented the mineralization of single-layer collagen model and demineralized dentin induced by modified self-etched adhesive. The relative cell viability was around 100 % in all groups in the study. Clinical significance These results indicated that the prime-and-rinse approach with MDP primer not only compensated for the reduced bond strength caused by the incorporation of AFCP nanoparticles but also enhanced mineralization capabilities and maintained biocompatibility, offering a promising strategy for improving the longevity of dental restorations.

The activation of matrix metalloproteinases (MMPs) is responsible for degradation of the hybrid layer . The effect of 10-methacryloyloxydecyl dihydrogen phosphate (10-MDP) on biodegradation of the hybrid layer remains unknown. This study aimed to evaluate the effects of 10-MDP and its Ca salt on MMP-9 activity and dentin-bonding durability. 10-MDP-Ca salt was synthesized and characterized by X-ray diffraction (XRD) and nuclear magnetic resonance (NMR). The effects of chlorhexidine (CHX), 10-MDP, and 10-MDP-Ca salt on MMP activity were evaluated by in-situ zymography, MMP-9 inhibition assay, and hydroxyproline (HYP) assay. Dentin-resin bonded specimens were fabricated with using CHX or 10-MDP, including Single Bond 2 (Ctr), CHX + Single Bond 2 (CHX), 10-MDP + Single Bond 2 (10-MDP), and SBU (SBU). Micro-tensile bond strength (μTBS) and nanoleakage tests were performed after 24-h or two-years water storage. XRD and NMR confirmed the presence of 10-MDP-Ca salt. The relative inhibition percentages of recombinant MMP-9 for 10-MDP and 10-MDP-Ca salt were 8.04 ± 0.83% and 55.96 ± 1.82% respectively. HYP assay showed that 10-MDP, CHX, and 10-MDP-Ca salt conditioning decreased collagen degradation. Water storage weakened μTBS of all the groups, with the Ctr and CHX groups showing more significant decreases ( P < 0.05). After aging, the Ctr and CHX groups also presented more nanoleakage, and the CHX group showed more in-situ zymography fluorescence than the 10-MDP and SBU groups. The capacity to inhibit MMP-9 activity, decrease HYP release, and reduce nanoleakage of 10-MDP and its salt contribute to improvement of dentin-bonding durability.

Objectives The study investigated the effects of novel prime-&-rinse mode using MDP (10-methacryloyloxydecyl dihydrogenphosphate) and matrix metalloproteinase (MMPs) inhibitors on dentin microtensile bond strengths (MTBS) of self-etch adhesive, resin-dentin interface degradations, and activity of recombinant human (rh) MMP-8, -9. Materials and methods Eight experimental primers were prepared using 5% and 15% of MDP ethanol-aqueous (1:1) solution in combination with/without MMPs inhibitors (1%benzalkonium chloride (BAC), 1000 μm/mL polyvinylphosphonic acid (PVPA) and 15%proanthocyanidin (PA)). Ninety human mid-coronal dentin surfaces were applied with the experimental primers, water-sprayed and gently air-dried (prime-&-rinse mode), or not (control, self-etch mode). The specimens were bonded with self-etch adhesive (Clearfil S3 Bond) and composite resin (Clearfil Majesty). The resin-bonded specimens were prepared into multiple micro-beams for MTBS tests after 24 h and 1 yr of water storage. The resin-dentin interfaces were analyzed with SEM/TEM. The inhibitory effects of eight primers on rhMMP-8, 9 were determined. The data were analyzed using two-way ANOVA and LSD multiple comparisons tests. Results Compared with control, all the primers used in prime-&-rinse mode could significantly improve long-term dentin MTBS ( P < 0.05), while 5%MDP-BAC, 15%MDP and 15%MDP+MMPs inhibitors could significantly increase the short-term dentin MTBS ( P < 0.05). The SEM/TEM findings revealed that the resin-dentin interfaces were stable over time when the prime-&-rinse mode used. Eight primers possessed the high inhibitory ratio of rh MMP-8, 9. Conclusions The novel prime-&-rinse mode using 5%MDP-BAC, 15%MDP and 15%MDP+MMPs inhibitors could significantly increase the short- and long-term dentin MTBS of self-etch adhesive. This might be a supplement to contemporary dentin bonding strategies.

Objectives To investigate the effect of neutral 10-methacryloyloxydecyl dihydrogen phosphate salt (MDP-Na) on the dentin bond strength and remineralization potential of etch-&-rinse adhesive. Methods Two experimental etch-&-rinse adhesives were formulated by incorporating 0 wt% (E0) or 20 wt% (E20) neutral MDP-Na into a basic primer. A commercial adhesive, Adper Single Bond 2 (SB, 3 M ESPE), served as the control. Sixty prepared teeth were randomly allocated into three groups ( n = 20) and bonded using either one of the experimental adhesives or SB. Following 24 h of water storage, the bonded specimens were sectioned into resin-dentin sticks, with four resin-dentin sticks obtained from each tooth for microtensile bond strength (MTBS) test. Half of the sticks from each group were immediately subjected to tensile loading using a microtensile tester at a crosshead speed of 1 mm/min, while the other half underwent tensile loading after 6-month incubation in artificial saliva (AS). The degree of conversion (DC) of both the control and experimental adhesives ( n = 6 in each group) and the adsorption properties of MDP-Na on the dentin organic matrix ( n = 5 in each group) were determined using Fourier-transform infrared spectrometry. Furthermore, the effectiveness of neutral MDP-Na in promoting the mineralization of two-dimensional collagen fibrils and the adhesive-dentin interface was explored using transmission electron microscopy and selected-area electron diffraction. Two- and one-way ANOVA was employed to assess the impact of adhesive type and water storage on dentin bond strength and the DC (α = 0.05). Results The addition of MDP-Na into the primer increased both the short- and long-term MTBS of the experimental adhesives ( p = 0.00). No difference was noted in the DC between the control, E0 and E20 groups ( p = 0.366). The MDP-Na remained absorbed on the demineralized dentin even after thorough rinsing. The intra- and extra-fibrillar mineralization of the two-dimensional collagen fibril and dentin bond hybrid layer was confirmed by transmission electron microscopy and selected-area electron diffraction when the primer was added with MDP-Na. Conclusions The use of neutral MDP-Na results in high-quality hybrid layer that increase the dentin bond strength of etch-&-rinse adhesive and provides the adhesive with remineralizing capability. This approach may represent a suitable bonding strategy for improving the dentin bond strength and durability of etch-&-rinse adhesive.

This study investigated effects of polyacrylic acid (PAA) on nanolayering self-assembly, and effects of salt ions on the formation of nanolayering after their chemical interaction with 10-methacryloyloxydecyl dihydrogen phosphate (MDP). ACP–MDP suspension was prepared by dispersing 0.01 g ACP nanoparticles in 4 mL of a 5 wt% MDP solution. Then a PAA solution (100 or 500 μg/mL) was mixed into ACP–MDP suspension. The reaction products were analyzed by transmission electron microscope (TEM). After one of the following metal chlorides (each 0.076 mM of CaCl2, BaCl2, AlCl3, FeCl2, FeCl3, MnCl2, CuCl2, ZnCl2, or MgCl2) was dispersed in a 4 mL MDP solution, they were further analyzed by X-ray diffraction (XRD) and TEM. ACP nanoparticles in the MDP solution could self-assemble into ACP–MDP nanolayering composites. The TEM images show that an amount of ACP nanoparticles were aggregated and fused together after the addition of PAA into ACP–MDP suspensions, while no nanolayering was found. The XRD data and TEM images reveal that nanolayering was self-assembled after the chemical interaction of MDP with some metal chlorides (CaCl2, BaCl2, AlCl3, FeCl2, FeCl3, MnCl2), but not with the others (ZnCl2, CuCl2, MgCl2). PAA could inhibit the self-assembly of nanolayering. Some salt ions play an important role in highly ordered, self-assembled nanostructure after their chemical interaction with MDP. The finding is helpful for functional materials fabrication.

Objectives To examine whether solvents and changing the molecular structure of 10-Methacryloyloxydecyl dihydrogen phosphate (10-MDP) affect its chemical affinity to Yttria-stabilized tetragonal zirconia polycrystals (Y-TZP). Methods The present work investigated the chemical affinity between Y-TZP and 10-MDP dissolved in different solvents (acetone/ethanol/water or mixture) using X-ray photoelectron spectroscopy, Fourier-transform infrared spectroscopy, and thermodynamic calculations. Shear bond strength (SBS) tests were used to evaluate the influence of different solvents on 10-MDP bonding. In addition, several phosphate ester monomer variants were created by changing the 10-MDP molecular structure. Changes included extending/shortening the spacer chain-length, and installing hydroxyl or carboxyl groups as side chains at different positions along the spacer chain. The thermodynamic parameters of the complexes formed between the 10-MDP variants and tetragonal zirconia were evaluated. Results The acquired data indicated that solvent is necessary for the formation of Zr–O–P bonds between 10-MDP and Y-TZP. Solvents affected the chemical affinity of 10-MDP to Y-TZP; acetone facilitated the best bonding, followed by ethanol. Changing the molecular structure of 10-MDP affected its chemical affinity to Y-TZP. The variants 15-MPDP, 12-MDDP, 6-hydroxyl-10-MDP and 6-carboxy-10-MDP all exhibited higher thermodynamic stability than 10-MDP when coordinated with tetragonal zirconia. In contrast, 2-MEP, 5-MPP, 10-hydroxyl-MDP, 10-carboxy-MDP, 5,6-dihydroxyl-10-MDP and 5,6-dicarboxy-10-MDP exhibited lower thermodynamic stability. Significance 10-MDP coordinates with zirconia through dissociating in solvents. Changing the molecular structure of 10-MDP theoretically affects its chemical affinity to Y-TZP.

Objectives This study investigated the effects of the surface conditioning of nano-zirconia fillers with 10-methacryloyloxydecyl dihydrogen phosphate (MDP) and dipentaerythritol penta-acrylate phosphate (PENTA) on the degree of conversion and mechanical properties of Bis-GMA- and UDMA-based resin composites containing the fillers. Methods MDP or PENTA conditioned nano-zirconia fillers were characterized by Fourier transform infrared spectroscopy (FTIR) and X-ray photoelectron spectroscopy (XPS). The conditioned nano-zirconia fillers were then used to prepare Bis-GMA- and UDMA-based resin composites. The bending strength and elastic modulus of the prepared composites were evaluated, and Weibull analysis was adopted to evaluate the reliability of the bending tests. Degree of conversion (DC) was measured by FTIR. Results FTIR and XPS analysis confirmed formation of Zr O P bonds in the MDP-conditioned and PENTA-conditioned zirconia fillers. MDP-conditioning and PENTA-conditioning increased bending strength of Bis-GMA-based composites (100.70 ± 7.15 MPa, 98.65 ± 4.76 Mpa) and UDMA-based composites (99.02 ± 4.45 MPa, 99.92 ± 5.78 MPa) compared to the groups with untreated nano-zirconia fillers (Bis-GMA-based, 84.98 ± 5.13 MPa; UDMA-based, 88.24 ± 4.61 MPa). The same trend was observed for the Weibull modulus ( m ) and elastic modulus. The MDP/PENTA conditioning did not affect the DC of the composites. Conclusion MDP or PENTA conditioning of nano-zirconia fillers improved the mechanical properties of the composite compared to the composites containing untreated nano-zirconia fillers; however, it did not affect DC. Clinical significance Mixing MDP- or PENTA-conditioned nano-zirconia fillers directly with gamma-MPS-conditioned silica to the resin composite has the potential to improve the composite’s mechanical properties.

Purpose To investigate the effects of nanozirconia fillers conditioned with 10-methacryloyloxydecyl dihydrogen phosphate (MDP) with or without zirconium hydroxide precoating on bending strength, Vickers hardness, and translucence of dental resin composites. Methods We obtained nanozirconia fillers coated with different concentrations of Zr(OH)4 using wet-chemical synthesis. We analyzed coating quality by observing electron-diffraction patterns using transmission electron microscopy. We conditioned zirconia fillers, with or without prior Zr(OH)4-coating, using MDP-containing primers and evaluated the formation of chemical bonds using nuclear magnetic resonance (NMR) and X-ray photoelectron spectroscopy (XPS). We then performed three-point bending-strength tests, Weibull analysis, Vickers hardness, and translucence-parameter analysis with or without addition of different concentrations of zirconia using untreated zirconia fillers as controls. Results We achieved desirable Zr(OH)4 coating using 5 mmol/L zirconium chloride. NMR and XPS analysis detected stronger Zr–O–P peaks on MDP-conditioned zirconia fillers with prior Zr(OH)4-coating compared with MDP-conditioned fillers alone, suggesting that MDP bonding with zirconia was enhanced by zirconium hydroxide. Our three-point bending-strength tests revealed that increasing levels of untreated zirconia fillers decreased the three-point bending strength of the resin composites, while MDP-conditioned zirconia fillers with or without prior Zr(OH)4 coating improved three-point bending strengths. Adding 5 wt% and 7.5 wt% MDP-conditioned zirconia fillers with prior Zr(OH)4 coating achieved the highest three-point bending strength. Furthermore, addition of zirconia fillers decreased the translucence of silica-based resin composites. Conclusion MDP conditioning with prior Zr(OH)4 coating is recommended for treating nanozirconia fillers of resin composites.
