Engineered Escherichia coli Nissle 1917 targeted delivery of extracellular PD-L1-mFc fragment for treating inflammatory bowel disease

Yuhong Wang,Lin Hu,Lei Wang,Chonghai Zhang,Wenhao Shen,Hongli Yang,Min Li,Xin Zhang,Mengmeng Xu,Muxing Zhang,Kai Yang,Xiaopeng Tian

Inflammatory bowel disease (IBD) is an autoimmune disorder involving complex immune regulation, where balancing localized and systemic immunosuppression is a key challenge. This study aimed to enhance the therapeutic efficacy by engineering the probiotic Escherichia coli Nissle 1917 (EcN). We removed endogenous plasmids pMUT1 and pMUT2 from wild-type EcN and expressed the mPD-L1 (19‒238 aa)-mFc fusion protein on the bacterial surface using a cytolysin A (ClyA) fragment. This modification stabilized mPD-L1 (19‒238 aa) protein expression and promoted its recruitment to outer membrane vesicles (OMVs). The engineered strain, EcNΔ pMUT1/2 -ClyA-mPD-L1-mFc (EcN-ePD-L1-mFc), features conditional ePD-L1-mFc expression under the araBAD promoter, enhancing gut-targeted release and reducing systemic side effects. This strain improved treatment targeting and efficiency by enabling direct ePD-L1-mFc interaction with immune cells at inflammation sites. OMVs from this strain induced Treg proliferation, inhibited effector T cell proliferation in vitro , and significantly improved intestinal inflammation and colonic epithelial barrier repair in vivo . Additionally, the bacterium restored intestinal microbiota balance, increasing Lactobacillaceae and reducing Bacteroides . This study highlights the engineered bacterium's potential for targeted intestinal immune modulation and offers a novel local IBD treatment approach with promising clinical prospects.

Impact of bioelectricity on DNRA process and microbial community composition within cathodic biofilms in dual-chambered bioelectrode microbial fuel cell (MFC)

Luhan Cai,Yubiao Lu,Haiguang Zhu,Binxin Liu,Xinyi Li,Tianbo Jia,Jianxin Wang,Xueting Wang,Peng Li

The synchronous bioelectricity generation and dissimilatory nitrate reduction to ammonium (DNRA) pathway in Klebsiella variicola C1 was investigated. The presence of bioelectricity facilitated cell growth on the anodic biofilms, consequently enhancing the nitrate removal efficiency decreasing total nitrogen levels and causing a negligible accumulation of NO 2 – in the supernatant. Genomic analysis revealed that K. variicola C1 possessed a complete DNRA pathway and largely annotated electron shuttles. The up-regulated expression of genes narG and nirB , encoding nitrite oxidoreductase and nitrite reductase respectively, was closely associated with increased extracellular electron transfer (EET). High-throughput sequencing analysis was employed to investigate the impact of bioelectricity on microbial community composition within cathodic biofilms. Results indicated that Halomonas , Marinobacter and Prolixibacteraceae were enriched at the cathode electrodes. In conclusion, the integration of a DNRA strain with MFC facilitated the efficient removal of wastewater containing high concentrations of NO 3 – and enabled the environmentally friendly recovery of NH 4 + .

Bioelectricity facilitates carbon dioxide fixation by Alcaligenes faecalis ZS-1 in a biocathodic microbial fuel cell (MFC)

Xinyi Li,Tianbo Jia,Haiguang Zhu,Luhan Cai,Yubiao Lu,Jianxin Wang,Hengcong Tao,Peng Li

The CO 2 fixation mechanism by Alcaligenes faecalis ZS-1 in a biocathode microbial fuel cell (MFC) was investigated. The closed-circuit MFC (CM) exhibited a significantly higher CO 2 fixation rate (10.7%) compared to the open-circuit MFC (OC) (2.0%), indicating that bioelectricity enhances CO 2 capture efficiency. During the inward extracellular electron transfer (EET) process, riboflavin concentration increased in the supernatant while cytochrome levels decreased. Genome sequencing revealed diverse metabolic pathways for CO 2 fixation in strain ZS-1, with potential dominance of rTCA and C4 pathways under electrotrophic conditions as evidenced by significant upregulation of the ppc gene. Differential metabolite analysis using LC-MS demonstrated that CM promoted upregulation of various lipid metabolites. These findings collectively highlight that ZS-1 simultaneously generated electricity and fixed CO 2 and that the ppc associated with bioelectricity played a critical role in CO 2 capture. In conclusion, bioelectricity resulted in a significant enhancement in the efficiency of CO 2 fixation and lipid production.

Efficient separation of nanoemulsion and simultaneous membrane cleaning by electroflotation coupled membrane filtration process

Hongwei Ge,Dafan Chen,Mutai Bao,Dan Zhang,Zhenzong Lu,Xiuping Chen,Yiming Li

The unique stability and complexity of oil-in-water (O/W) nanoemulsions originating from various fields of life and production pose significant challenges for their separation. In this work, a novel oil/water separation membrane (MFC/PPy/Ni–Co–P) is constructed by loading polypyrrole (PPy) particles on cellulose (MFC) membranes to provide growth sites for Ni–Co–P alloys, so as to endow the membrane with in situ bubble-generating capability. Under the external electric field, the MFC/PPy/Ni–Co–P membrane is integrated into the electroflotation filtration system as a cathode, and the effective membrane cleaning and demulsification of nanoemulsions can be realized through the micro-aeration environment constructed in situ . The MFC/PPy/Ni–Co–P membrane demonstrates outstanding performance in gravity-driven separation of various oil/water mixtures, with a rejection rate of >99.99 % and a flux of >3000 L∙m −2 ∙h −1 . The interaction energy between oils with different viscosities and membranes is quantified by the extended Derjaguin-Landau-Verwey-Overbeek (XDLVO) theory, revealing the dominant role of acid-base interactions in the anti-fouling process. Notably, the integration of MFC/PPy/Ni–Co–P membrane into an electroflotation separation system enables the coalescence and separation of small-sized, highly stable nanoemulsions with separation efficiencies of >99.86 %. This study provides a novel approach for simultaneous and efficient nanoemulsion separation and membrane fouling remediation, showing a great potential in the development of efficient and sustainable oil/water separation technologies.

An MFC-IIP functionalized SNMNS optical fiber sensor for Cd2+ detection

Shaojun Zhang,Ziyu Hu,Shaowa Lü,Xueyan Bi,Yanyang Lei

This study presents a novel optical fiber sensor based on a MoS 2 @Fe 3 O 4 @CTS ion-imprinted polymer (MFC-IIP) functionalized single-mode fiber-no-core fiber-multi-mode fiber-no-core fiber-single-mode fiber (SNMNS) structure for the quantitative detection of trace cadmium ions (Cd 2+ ) in solution. The MFC-IIP nanomaterial was coated onto the surface of the SNMNS fiber structure as a selective sensing membrane to enable highly sensitive and specific detection of Cd 2+ . The experimental results demonstrate that the sensor achieves a maximum detection sensitivity of 119.06 nm/μM for cadmium ions, with a limit of detection (LOD) as low as 0.77 nM. This study introduces a novel detection approach applicable to real-time online water quality monitoring, early diagnosis of chronic cadmium poisoning, and the rapid analysis of heavy metal cadmium in complex traditional Chinese medicine matrices.

A novel long-wavelength fluorescent probe for discrimination of different palladium species based on Pd-catalyzed reactions

Ting Yu,Guoxing Yin,Peng Yin,Ying Zeng,Haitao Li,Youyu Zhang,Shouzhuo Yao

We have synthesized a novel long-wavelength fluorescent probe MFC for detection of palladium (Pd0). This probe not only shows high selectivity for palladium, but also could quickly discriminate different palladium species (Pd0, Pd2+, Pd4+) in phosphate buffered solution. After the reaction with Pd(PPh3)4, the UV absorption of the probe MFC shifts from 334 nm to 594 nm within a few minutes. The probe MFC conjugated with alkyl carbamate, which destroys the principle of intramolecular charge transfer (ICT) and shows weak fluorescence. Based on the Pd0-catalyzed Tsuji–Trost deallylation reaction, the fluorogen was released and the fluorescence intensity at 612 nm was observed with 13-fold enhancement along with an obvious color change from yellow to purple with low detection limit.

Anodic-potential-tuned bioanode for efficient gaseous toluene removal in an MFC

Juping You,Han Chen,Liangliang Xu,Jingkai Zhao,Jiexu Ye,Shihan Zhang,Jianmeng Chen,Zhuowei Cheng

Microbial fuel cell (MFC) is an innovative technology to stimulate the anaerobic degradation of hydrocarbons and recover their chemical energy. Typically, for the hydrocarbon oxidation, its performance depends upon the activity of the bioanode. In this work, four anodic potentials pertained to the redox of four different membrane proteins of the microbes, namely -0.52, -0.35, -0.16, and 0.17 V vs. Ag/AgCl, were applied to tune the bioanode. Experimental results revealed that, the more negative the bioanode potential was, the more obvious the enhancement became. The biofilm tuned at -0.35 V exhibited a highest toluene removal efficiency of 99% (800 mg/m 3 ), and a maximum power density of 8.97 mW/m 2 with the Coulomb efficiency of 2.06%. The microbial community analysis indicated that the dominant microbes in the bioanode were Comamonas sp., Petrimonas sp., Sedimentibacter sp., Ignavibacterium sp. and Parachlamydia sp., which was almost unaffected by the applied anodic potentials. The cyclic voltammetry analysis showed that the bioanode tuned at 0.17, -0.16 and -0.35 V exhibited three completely different electron transfer pathways. Furthermore, three metabolic pathways with o -cresol as an intermediate were proposed under the four applied anodic potentials. These results demonstrated that the anodic potential acclimation offers an effective enhancement method for the gaseous toluene removal combined with power generation.

Polyaniline-modified iron oxide nanoparticles to improve the overall performance of MFC-BOD sensors

Jianlei Yang,Min Du,Wanqing Wu,Qing Chen,Jiaqi Niu,Ziyang Zhang

Biochemical oxygen demand (BOD) has attracted considerable attention as an important indicator of water pollution. Microbial fuel cells (MFC) are considered a promising method for BOD detection. In this work, the MFC-based BOD sensor was proposed considering the performance and cost factors, and the anode material was modified by iron oxide nanoparticles (IONPs) to improve the detection performance of the sensor and enable targeted selection of microorganisms, resulting in the dominant bacterial group accounting for 92.39 % of the total bacterial group. Polyaniline (PANI) was used as an adhesive to enhance the electrical performance of the anode material. The composite anode (CE) has superior electrical properties and biocompatibility compared to traditional carbon felt materials. The modified MFC sensor ensures the accuracy of detection and can detect BOD within 20 min with a detection limit of 73.3–440 mg/L. In addition, a good correlation (R 2  = 0.9969) was obtained between the BOD concentration and the actual performance, which proved the suitability of the system for real wastewater. The whole BOD detection system has the advantages of fast sensing speed, large detection range and easy operation while ensuring accuracy, which has high potential for engineering applications.

CW-MFC for Efficient Degradation of Ofloxacin-Containing Wastewater and Dynamic Analysis of Microbial Communities

Li Yanhong,Zhao Zikang,Zhao Yao,Tang Shen,Zhang Lihao,Zhang Jie,Peng Yuqing

The overuse of antibiotics poses critical threats to environmental and human health. This study developed a constructed wetland-microbial fuel cell (CW-MFC) system integrated with pre-acclimated Shewanella oneidensis MR-1 to enhance ofloxacin (OFL) removal from wastewater. The CW-MFC demonstrated exceptional removal efficiency (>96%) for OFL across tested concentration gradients, significantly outperforming conventional constructed wetlands (CW) in both antibiotic elimination and conventional pollutant reduction. Microbial community analysis revealed that S. oneidensis MR-1 inoculation enhanced microbial diversity under OFL stress while promoting enrichment of electroactive Firmicutes phylum members compared to non-inoculated systems. The synergistic integration of bioelectrochemical processes and microbial consortia modification in CW-MFCs effectively addressed antibiotic contamination challenges. These findings establish CW-MFC technology combined with electroactive bacteria acclimation as a sustainable solution for pharmaceutical wastewater treatment, providing critical insights for optimizing antibiotic removal strategies in engineered ecosystems.

Spontaneously producing syngas from MFC-MEC coupling system based on biocompatible bifunctional metal-free electrocatalyst

Wang Wei,Borse Rahul Anil,Xie Jiafang,Wang Yaobing

The preparation of high-value fuels and chemicals through the electrochemical carbon dioxide reduction reaction (CDRR) is of great significance to the virtuous cycle of carbon dioxide. However, due to the high overpotential involved in this reaction, high power consumption and high-cost noble-metal-based catalysts are required for driving this process. Herein, the electrochemical CDRR was achieved on biocompatible metal-free nitrogen, phosphorus co-doped carbon-based materials (NP-C) in the microbial fuel cell-microbial electrolysis cell (MFC-MEC) coupling system. As the bioelectrochemistry in MFC supplied power to drive the electrocatalysis in MEC, syngas was spontaneously produced from this coupling system without external energy input. With the NP-C materials as the excellent bifunctional electrocatalyst for the CDRR and oxygen reduction reaction (ORR), the current density of the MEC reached −0.52 mA cm −2 , and the Faradaic efficiencies (FEs) of CO and H 2 were 60% and 40%, respectively, at a load resistance of 10 Ω. Moreover, the CO/H 2 product ratio can be changed by adjusting the load resistance, which will widely meet various demand of syngas usage in further reactions. This study provides a spontaneous and tunable production of syngas in biogas digesters via a electrochemical strategy.

Bioinspired anisotropic architectures in AgNWs/MFC/SA/PDMS composite aerogels for enhanced wearable piezoresistive sensors

Fuping Bian,Yuliang Zhou,Yuhang Shao,Mengna Peng,Yan Lu,Zhenhao Lian,Xiang Li

A bioinspired honeycomb composite aerogel was fabricated by freeze-drying. • AgNWs-based conductive networks enabled stable piezoresistive sensing behavior. • PDMS impregnation improved mechanical stability and environmental resistance. • The composite elastomer exhibited antibacterial activity (∼95%). • Wearable sensors demonstrated high sensitivity, fast response and stable cycling. Wearable piezoresistive sensors show great potential for motion analysis, health monitoring, and human–computer interaction. However, conventional sensing materials often suffer from limited sensitivity, insufficient flexibility, poor environmental adaptability, and inadequate antibacterial performance for skin-contact applications. Herein, a bioinspired anisotropic AgNWs/MFC/SA composite aerogel (30 A-55MS) was fabricated by directional freeze-drying, using silver nanowires (AgNWs) as the conductive and antibacterial network, microfibrillated cellulose/sodium alginate (MFC/SA) as the biomass-derived polymer scaffold, and CaCl 2 as the ionic-gelation agent. The directional porous architecture was inspired by the transport-channel structure of taro stalks. Subsequently, polydimethylsiloxane (PDMS) was introduced by vacuum-assisted impregnation to obtain the 30 A-55MS/PDMS composite aerogel elastomer. PDMS encapsulation enhanced the mechanical stability and environmental resistance of the elastomer, which also exhibited bacterial reduction efficiencies of 95.57% against E. coli and 95.21% against S. aureus . The wearable piezoresistive sensor assembled from the 30 A-55MS/PDMS elastomer exhibited a high gauge factor (GF = −20.792 in the 0–2% strain range), rapid response/recovery times of 65/40 ms, and stable cycling performance over 1000 compression cycles. Moreover, real-time finger, neck, elbow, wrist, and knee movements were successfully monitored. This work provides a promising strategy for developing multifunctional aerogel-based wearable sensors for health monitoring, motion analysis, and human–computer interaction.

MFC based in situ electrocatalytic persulfate activation for degradation of 2,4-dichlorophenol: Process and mechanism

Minjie Zhu,Xiaomei Liu,Linan Liu,Qinglong Liu,Fengxiang Li,Jingchun Tang

Electrocatalytic persulfate (PS) generation of sulfate radicals has been extensively studied for the removal of organic pollutants , while continuous electricity output causes high energy consumption. Microbial fuel cell (MFC) is an environmentally friendly approach that can catalyze PS for degradation of pollutants through bioelectricity and achieve electricity recovery and utilization at the same time. In this study, a MFC based on electrocatalytic PS activation in cathode (PS-MFC) was constructed to remove 2,4-dichlorophenol (2,4-DCP) with simultaneous electricity recovery. Results showed that 2,4-DCP was successfully removed by PS-MFC with a maximum removal rate of 68.7% after 220 mins of reaction, and the removal process followed pseudo-second-order kinetic. In addition to the wide pH reaction range (pH 3–11), PS-MFC exhibited excellent electrical performance with a higher maximum power density (300.28 mW/m 2 ) than reported studies of the similar type of MFC. Active species for the degradation of 2,4-DCP by PS-MFC system are O 2 • − , • O H , S O 4 • − and O 2 1 , with O 2 • − and • O H accounting for major part. The degradation pathway of 2,4-DCP in PS-MFC was proposed by free radical quenching experiments and intermediate product analysis. This work provided a green and sustainable solution for the removal of refractory organic pollutants and promoted research for the further practical application of MFCs.
