The present study focuses on the synthesis and evaluation of the gelatin/nanohydroxyapatite (Gel/n-HA) composite microspheres fabricated by the water-in-oil (W/O) emulsification process as potential substrates for bone repair applications. Herein, 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide (EDC)-crosslinked Gel/n-HA (10 wt%) composite microspheres were synthesized. The morphology, physicochemical properties, and cellular behavior of the EDC-crosslinked Gel-HA microspheres (ECM) were characterized. In vitro cell culture assays demonstrated that the ECM could effectively promote the adhesion and growth of MG63 cells and in vivo bone repair experiments suggested that ECM possessed sustainable regenerative effects. This study provided a simple strategy to prepare functionalized microspheres that showed great potential in promoting bone regeneration as injectable microscaffolds.

Osteosarcoma is among the most common malignant bone tumors in human skeletal system. The conventional treatment of osteosarcoma mainly consists of combining neoadjuvant chemotherapy with surgical approach. However, it is crucial to design an artificial implant that possesses excellent biomechanical properties and is capable of sustaining local release of chemotherapeutics. In this study, we envision that the highly efficient combination of gemcitabine (GEM) hydrochloride loaded liposomes with gelatin methacryloyl (GelMA) of in situ photocrosslinkable hydrogel will lead to a multifunctional implant with unique antitumor, mechanical, and biodegradable properties. A sustained controlled release was observed; more specifically, the release of GEM in vitro lasted for 4 days long. Furthermore, its capability in killing MG63 cells was further explored by using the lixivium of GEM-Lip@Gel and GEM-GelMA hydrogel in vitro (composite hydrogel by GEM loaded liposomes blending with GelMA, short for GEM-Lip@Gel), which agreed with the drug release outcome. In addition, these hydrogel showed excellent ability in inhibiting osteosarcoma in vivo by Balb/c mice bearing MG63 cells. Therefore, GEM-loaded lipo-hydrogel certainly has presented itself as a promising strategy for the development of implant in the field of osteosarcoma treatment.

The implant-associated infection remains a serious problem in clinic. A better compromise between the anti-infection property and biocompatibility of bone filling materials is still required. In this work, carboxylated chitosan/silver-hydroxyapatite (CMCS/Ag-HA) hybrid microspheres were fabricated via a facile gas diffusion method. CMCS and Ag ion were evenly distributed into hybrid microspheres. CMCS regulated the morphological characteristics of the as-synthesized hybrid microspheres. As the content of CMCS increased, the size of the sample increased and dispersion exacerbated. In addition, antimicrobial studies demonstrated that CMCS/Ag-HA hybrid microspheres exhibited an excellent antimicrobial activity against Staphylococcus aureus because of the synergistic effect of Ag + and CMCS. In vitro cell tests indicated that the as-prepared CMCS/Ag-HA hybrid microspheres promoted the proliferation and adhesion of MG63 cells. CMCS/Ag-HA hybrid microspheres showed excellent bactericidal property and biocompatibility; thus, they could be used in biomedical applications, such as infection-resistant bone replacement materials.

Treatments for malignant bone tumors are urgently needed to be developed due to the dilemma of precise resection of tumor tissue and subsequent bone defects. Although polyether-ether-ketone (PEEK) has widely attracted attention in the orthopedic field, its bioinertness and poor osteogenic properties significantly restrict its applications in bone tumor treatment. To tackle the daunting issue, we use a hydrothermal technique to fabricate novel PEEK scaffolds modified with molybdenum disulfide (MoS 2 ) nanosheets and hydroxyapatite (HA) nanoparticles . Our dual-effect synergistic PEEK scaffolds exhibit perfect photothermal therapeutic (PTT) property dependent on molybdous ion (Mo 2+ ) concentration and laser power density, superior to conventional PEEK scaffolds. Under near-infrared (NIR) irradiation, the viability of MG63 osteosarcoma cells is significantly reduced by modified PEEK scaffolds, indicating a tumor-killing potential in vitro . Furthermore, the incorporation of HA nanoparticles on the surface of PEEK bolsters proliferation and adherence of MC3T3-E1 cells, boosting mineralization for further bone defect repair. The results of micro-computed tomography (micro-CT) and histological analysis of 4-week treated rat femora demonstrate the preeminent photothermal and osteogenesis capacity of 3D-printed modified scaffolds in vivo . In conclusion, the dual-effect synergistic orthopedic implant with photothermal anticancer property and osteogenic induction activity strikes a balance between tumor treatment and bone development promotion, offering a promising future therapeutic option.

Alginate (Alg) hydrogels possess desirable advantages for application in tissue engineering; however, they are limited by their weak mechanical properties , poor chronical stability in phosphate buffered saline , and absence of mammalian cell recognition sites, severely restricting their biomedical applications. To overcome these limitations, we integrated Alg hydrogels with nano-silica (SiO 2 ) to produce nano-SiO 2 reinforced Alg–chitosan–gelatin nanocomposite hydrogels (Alg/SiO 2 –CHI–GA NCH) for biomedical purposes, utilizing Chitosan (CHI) and gelatin (GA) in an alternate electrostatic adsorption. Specifically, we investigated the regulatory and promotional effects of the nano-SiO 2 on the morphological structure, mechanical properties, thermal stability , rheological properties, swelling, biodegradability , biomineralization and cytocompatibility of the resultant Alg/SiO 2 -CHI-GA NCH. The experimental findings demonstrate that the constructed Alg/SiO 2 -CHI-GA NCH exhibited uniform morphology and a regular structure. Upon freeze–drying, the internal cross-sections of the NCH exhibited a honeycomb porous structure. Furthermore, the physicochemical properties and biological activities of the prepared Alg/SiO 2 –CHI–GA NCH were regulated to some extent by nano-SiO 2 content. Notably, nano-SiO 2 inclusion enhanced the attachment and viability of MG63 and MC3T3-E1 cells and induced three-dimensional cell growth in ALG/SiO 2 –CHI–GA NCH. Among the fabricated NCH, Alg/SiO 2 –CHI–GA NCH with 0.5% and 1.0% (w/v) nano-SiO 2 exhibited significant proliferative activity, which is attributable to their high porosity and uniform cell adhesion . Furthermore, the alkaline phosphatase activity in the cells gradually increased with increasing of nano-SiO 2 amount, indicating the favorable effect of nano-SiO 2 on the osteogenic differentiation of MG63 and MC3T3-E1 cells. Our study findings provide a comprehensive foundation for the structural- and property-related limitations of Alg hydrogels in biomedicine , thereby expanding their potential applications in tissue engineering.
