Analysis of the CPZ/Wnt4 osteogenic pathway for high-bonding-strength composite-coated magnesium scaffolds through transcriptomics

Zewen Shi,Fang Yang,Tianyu Du,Qian Pang,Chen Liu,Yiwei Hu,Weilai Zhu,Xianjun Chen,Zeming Chen,Baiyang Song,Xueqiang Yu,Zhewei Ye,Lin Shi,Yabin Zhu,Qingjiang Pang

Magnesium (Mg)-based scaffolds are garnering increasing attention as bone repair materials owing to their biodegradability and mechanical resemblance to natural bone. Their effectiveness can be augmented by incorporating surface coatings to meet clinical needs. However, the limited bonding strength and unclear mechanisms of these coatings have impeded the clinical utility of scaffolds. To address these issues, this study introduces a composite coating of high-bonding-strength polydopamine-microarc oxidation (PDA-MHA) on Mg-based scaffolds. The results showed that the PDA-MHA coating achieved a bonding strength of 40.56 ± 1.426 MPa with the Mg scaffold surface, effectively enhancing hydrophilicity and controlling degradation rates. Furthermore, the scaffold facilitated bone regeneration by influencing osteogenic markers such as RUNX-2, OPN, OCN, and VEGF. Transcriptomic analyses further demonstrated that the PDA-MHA/Mg scaffold upregulated carboxypeptidase Z expression and activated the Wnt-4/β-catenin signaling pathway, thereby promoting bone regeneration. Overall, this study demonstrated that PDA can synergistically enhance bone repair with Mg scaffold, broadening the application scenarios of Mg and PDA in the field of biomaterials. Moreover, this study provides a theoretical underpinning for the application and clinical translation of Mg-based scaffolds in bone tissue engineering endeavors.

Interface ligand engineering of Au nanoclusters with near-infrared boosted peroxidase-like activity for total antioxidant capacity analysis

Chen Sun,Yutong Ye,Xueting Luo,Wentao Xiang,Cao Li,Bingbing Jiang,Xiaoying Wang,Ziqiang Xu

Total antioxidant capacity (TAC) is a significant indicator for evaluating nutritional value of fruit and beverage industries, in the aspects of their reactive oxygen species (ROS) scavenging activity. The sensitivity and selectivity of the TAC assay based on the peroxidase (POD) assay is restricted by their poor catalytic activity. Herein, a simple and sensitive strategy for assessing TAC in fruits based on light (808 nm) boosted POD like activity of Au 25 nanoclusters (NCs) have been reported. The absorption bands of mercaptohexanoic acid (MHA)-protected Au 25 NCs could be extended into the near-infrared region (∼770 nm) by introducing neutrally charged ligands or 2,2’(ethylene dioxy) diethyl thiol (EDT)) as bi-thiol ligands, which may contribute to more obvious light enhanced POD activity. The most intense absorption at 770 nm can be obtained by the ratio of MHA/EDT=1.7:0.3 (denoted as MEA-17). Since we have confirmed that ROS is involved in the light enhanced POD like activity, the concentration of various antioxidants AA, GSH and L-Cys, can be analyzed by their inhibitory effect to the oxidation of TMB. More importantly, light enhanced POD like activity of MEA-17 have been successfully utilized to analysis TAC value in various fresh fruits. This work not only provide novel direction for the rational development of near-infrared light-enhanced POD-like nanozyme, but also further expands the prospect of the application of Au 25 NCs protected with thiols for TAC analysis.

Highly efficient separation of organic substances from high salinity brine by nanofiltration

Yahui Li,Daoguang Wang,Junfeng Wang,Yi Nie

Efficient separation of organic compounds from the high salinity brines during the treatment of reverse osmosis concentrate (ROC) is highly desirable and also very challengeable for the purpose of zero liquid discharge (ZLD). For the general treatment process, the efficient removal of organic compounds from the NaCl rich solution before the evaporation-crystallization stage is one of the key challenges. In this work, separation of N-(2-hydroxypropyl)morpholine (NMH), 4-morpholineacetate (MHA), the major organic components of ROC in refinery, and monovalent/divalent ions from the high salinity brines by three commercial nanofiltration (NF) membranes, i.e. Desal-5DK, Desal-5DL and NF270 were systematically investigated. The effects of various parameters on the rejection performance of these three NF membranes were experimentally investigated. The results showed that the retentions of NMH and MHA were dominated by flux and pH. Single stage filtration experiments showed that the rejection to NMH and MHA with various operating parameters was high and stable for DK. In comparison, DL and NF270 had higher separation efficiency for monovalent and divalent ions . The experiments determined that the rejections of NMH and MHA were 99.16% and 98.42% under the optimized conditions, respectively, indicating that NF membranes were highly efficient for separating organic compounds from the high salinity brine.

Hyaluronan derivative decorated calcium carbonate nanoparticle as a potential platform for breast cancer synergistic therapy via blood coagulation and drug delivery

Fei Zhou,Huiru Li,Yuying Liu,Haotian Deng,Jianhua Rong,Jianhao Zhao

Malignant tumor is seriously threatening human lives, and synergistic therapy has recently become a promising strategy. In this study, we developed a methacrylated hyaluronan-polyethylene glycol (mHA-PEG) decorated and DOX-loaded CaCO 3 NPs ( [email protected] 3 /mHA-PEG NPs) for the potential synergistic therapy of breast tumors via Ca 2+ -induced blood coagulation and drug delivery. This core/shell nanoparticle , with ∼300  nm in size and −8.6 mV in charge, was composed of 27% of DOX , 65% of CaCO 3 , and 8% of mHA-PEG in weight percentage. [email protected] 3 /mHA-PEG NPs were stable in the environment mimicking the bloodstream due to the crosslinked shell with almost no premature delivery of DOX and Ca 2+ , both of which however were fast released by pH/Hyals stimuli in the mimicking microenvironment of breast cancer cells. The released Ca 2+ caused fast in vitro blood clotting within 12 min. These nanoparticles were highly internalized by MCF-7 and MDA-MB-231 breast cancer cells, followed by the intracellular release of DOX and Ca 2+ . The released DOX migrated into the cell nucleus for killing cancer cells, while the released Ca 2+ moved outside the cells so as to induce tumor vessel coagulation. Herein, [email protected] 3 /mHA-PEG NPs is a potential candidate for breast cancer synergistic therapy by the combination of starvation therapy and chemotherapy.

Ligand Engineering of Gold Nanoclusters for NIR-II Imaging

Meili Guo,Gang Zhang,Ruoli Zhao,Huizhen Ma,Yuxing Yan,Shuyu Yang,Jian Meng,You Huang,Xiao-Dong Zhang,Hao Wang,Ruiping Zhang

Near-infrared-II (NIR-II) imaging has shown great potential in medical diagnosis and surgical navigation, but developing safe, photostable, high-brightness molecular probes remains a great challenge. Due to their ultrasmall size resulting in highly efficient renal clearance, gold nanoclusters have shown great clinical potential. In this work, we systematically explored the effect of different ligands on the luminescence and bioactivity of gold nanoclusters. Our results show that gold nanoclusters protected by thioglycolic acid (TGA) and 6-mercaptohexanoic acid (MHA) exhibit the strongest fluorescence, while 3-mercaptopropionic acid (MPA)- and sodium sulfide (Na2S)-protected gold nanoclusters show the weakest NIR-II signal. Further doping showed that the Cd-doped MPA and MHA-protected gold nanoclusters exhibited enhanced fluorescence, but the cysteine (Cys)-, glutathione (GSH)-, and Na2S-protected gold nanoclusters showed fluorescence quenching after doping, indicating significant ligand selectivity. Because of the unique multi-energy structure and the large number of electronic states at the highest occupied molecular orbital energy level, MPA- and MHA-protected gold nanoclusters exhibit high stability and photostability. In addition, gold nanoclusters with different ligands exhibited different selective enzyme-mimicking activities of peroxidase (POD), superoxide dismutase (SOD), and catalase (CAT). Imaging in vivo showed that gold nanoclusters could accomplish reliable imaging of cerebral vasculature, hindlimb vessels, and spine as well as monitor renal clearance. The stable nanoclusters allow the time window of imaging to reach 125 min for hindlimb imaging and 270 min for spinal imaging. The gold nanoclusters exhibit a high signal-to-noise ratio of up to 11 for whole-body imaging and show efficient renal clearance and low toxicity at an injected dose of 50 mg/kg.
