Integrated cell metabolomics and network pharmacology approach deciphers the mechanisms of Astragali Radix MIX in repairing podocyte injury

Aiping Li,Xiaoyu Zhang,Zheng Ju,Tingting Luo,Ting Cui,Xuemei Qin,Guangzhen Liu

To elucidate the molecular mechanisms of the MIX (combined in proportion to the content in Astragali Radix (AR), named the MIX) repairing podocyte damage to ameliorate nephropathy. MTT assay and western blot analysis were used to evaluate the protective effects of MIX on MPC5 cells induced by Adriamycin (ADR). Screening of potential pharmacodynamic markers, relevant drugs and disease targets were conducted by using metabolomics combined with bioinformatics, and the most relevant metabolic pathways were identified by analyzing shared target and KEGG pathways. The key mechanism was subsequently validated by cell adhesion assays, western blot assays, and immunofluorescence staining. The results showed that the MIX has the capacity to repair adriamycin-induced damage in MPC5 cells, as evidenced by enhanced cell viability and synaptopodin expression. Additionally, the MIX shows promise in potentially reinstating the podocyte adhesion through the modulation of the expression of podocyte adhesion-related proteins linked to nucleotide metabolites. The MIX has the potential to beneficially affect podocyte injury by modulating the cell adhesion pathway, contributing to one of the pharmacodynamic mechanisms of AR treatment of nephrotic syndrome. This has implications for the development and utilization of AR resources and achievement the social benefit of empowering rural revitalization.

Engineering bioluminescent immunoassay for mix-to-measure parathion detection in foodstuffs

Qiu-Yue Pan,Yi-Fan Liang,Yu Wang,Yi-Hong He,Yan-Hong Chen,Jin-Yi Yang,Yu-Dong Shen,Zhen-Lin Xu,Bruce Hammock,Surat Hongsibsong,Hong Wang

The growing demand for simple and efficient methods to detect pesticides has highlighted the importance of homogeneous immunoassays. In this study, a competitive bioluminescent homogeneous immunoassay was developed for detecting the highly toxic pesticide parathion, using nanoluciferase binary technology (NanoBiT). The assay design involved fusing the large subunit (11S) of nanoluciferase (NanoLuc) to the anti-parathion nanobody VHH9, while conjugating the small subunit (86) to the coating antigen H1-OVA.The interaction between VHH9 and H1-OVA brought 11S and 86 into close proximity, enabling NanoLuc reassembly and luminescence emission upon the addition of coelenterazine h (CTZ-h). In the presence of parathion, luminescence intensity decreased proportionally to parathion concentration. This one-step, 10-min assay required no washing steps and demonstrated a half-maximum inhibition concentration (IC 50 ) of 6.67 ng/mL with a linear range of 1.44–30.9 ng/mL. The NanoBiT immunoassay achieved recovery rates of 88.30 % to 103.85 % and correlated well with results from standard gas chromatography-tandem mass spectrometry (GC–MS/MS). These results indicated that the homogeneous NanoBiT immunoassay is a rapid, convenient, and reliable tool for detecting parathion in food.

Mix-charged polyamide membranes via molecular hybridization for selective ionic nanofiltration

Junhui Zhao,Xinda You,Guangzhe Wang,Jinqiu Yuan,Yafei Li,Chao Yang,Shiyu Zhang,Xiaoyao Wang,Runnan Zhang,Hong Wu,Zhongyi Jiang

Nanofiltration membranes that can selectively remove multivalent ions from salty water hold great promise in water treatment and salt utilization. However, developing nanofiltration membranes with high rejection to both divalent anions and cations remains a grand challenge. Herein, we design a kind of mix-charged polyamide membranes (MCPMs) via molecular hybridization to achieve the removal of versatile divalent ions. Amino-functionalized polyhedral oligomeric silsesquioxane (NH 2 -POSS) was employed as molecular filler added into piperazine and reacted with trimesoyl chloride at the aqueous-organic interface to generate cross-linked polyamide-NH 2 -POSS mixed network. The protonated amino groups from NH 2 -POSS offered positively charged sites for enhanced divalent cation repulsion while the deprotonated carboxyl groups from polyamide network can afford negatively charged sites for divalent anion repulsion. The optimized membrane displayed high rejections to both divalent anionic and cationic ions (Na 2 SO 4 rejection of 98.2%, MgCl 2 rejection of 98.0%), together with high mono-/divalent salt selectivity (NaCl/Na 2 SO 4 of 48.2, LiCl/MgCl 2 of 43.9) that outperformed the benchmark nanofiltration membranes. This molecularly-hybridized charge modulation method may enlighten the structural design of membranes for efficient ionic separations.

The Tourism Demarketing Mix: Working to Reduce Revisits to Egyptian Nature Reserves

Ahmed M. Elsetouhi,Mohamed E. Elshourbagi,Ahmed A. Hammad,Alaa A. Elnazer

This study tests the impact of the tourism demarketing mix (product, price, place, promotion) on tourists’ intention to reduce revisits to nature reserves in Egypt. In particular, this work focuses on the moderating role of switching cost. Data were collected from 401 tourists of the Wadi Degla and Petrified Forest nature reserves in Egypt. Path analysis is employed to test the research hypotheses using WarpPLS 6. Findings confirm that the tourism demarketing mix significantly affects tourists’ perception and intention to reduce revisits to the nature reserves. Lower switching cost partially enhanced the relationship between the tourism demarketing mix and tourists’ intention to reduce revisits. Regarding implications, managers can use findings from this study to design marketing plans that contribute to sustainable stewardship of destinations and protect nature reserves. Limitations and conclusions were also provided.

Multi-Objective Mix Design Framework for Solid-Waste-Based Self-Compacting Concrete

Xiaolong Jia,Feng Jin,Guangqi Xiong,Tao Ma,Xiwen Zou,Guangxiang Ji,Xudong Ma,Pengjun Li

The growing demand for sustainable construction has highlighted the need to effectively utilize solid waste materials in concrete production, yet achieving satisfactory workability, strength, and durability simultaneously remains challenging. A multi-parameter mix-design methodology is proposed for solid-waste-based self-compacting concrete (SCC). This method couples minimum water demand, control of paste film thickness, and multi-performance balancing. The ternary solid-waste powder system (silica fume, fly ash, and supersulfated solid-waste-based cement) was first optimized through minimizing water demand to achieve maximum packing density. The resulting composition was then blended with varying dosages of ordinary Portland cement (OPC) to form the final cementitious binder. Aggregate gradation was proportioned to minimize voids, and paste volume was determined using an equivalent-paste-film-thickness model. Under comparable mixture conditions, SCC with OPC contents of 70–40 wt.% and paste film thicknesses of 2.0–2.6 mm was evaluated for fresh performance, compressive strength, freeze–thaw resistance, and material cost. Mixtures with a paste film thickness of 2.4 or 2.6 mm satisfied the self-compactability criterion—the mix with 50 wt.% OPC and a paste film thickness of 2.4 mm showed the best overall performance balance, achieving higher 28 d strength than higher-OPC mixtures while improving freeze–thaw resistance and reducing cost. Results from TGA, XRD, ATR–FTIR, and SEM–EDS analyses indicated enhanced calcium hydroxide (CH) consumption, increased formation of C-(A)-S-H and ettringite, and a denser interfacial transition zone (ITZ), supporting the proposed multi-objective design approach. While the framework was validated for a specific ternary binder system, it provides a reproducible proportioning strategy applicable to a broader range of solid-waste-based concrete systems, with potential for extension to other waste streams and exposure conditions, thus supporting the development of more resource-efficient and environmentally sustainable concrete.

Mesoporous dendritic fibrous nanosilica (DFNS) stimulating mix matrix membranes towards superior CO2 capture

Songwei Li,Xu Jiang,Hongguang Sun,Shanshan He,Liling Zhang,Lu Shao

Herein, for the first time, mesoporous dendritic fibrous nanosilica (DFNS) is utilized to fabricate mixed matrix membranes (MMMs) to increase the membrane properties and push gas separation performance of low-molecular-weight polyethylene glycol (PEG) impregnated MMMs toward superior CO 2 capture. The basic physicochemical and gas transport properties of various membranes have been examined in detail for clarifying the mechanism of the significant enhancement in membrane performance. The polymer chains around the DFNS became rigidly according to the enhanced T g of the MMMs, gas permeability of the gas improved mainly ascribe to the increase of gas diffusivity . Because of the high porosity of DFNS with the mainly specific pore size range of 2–4 nm, the incorporation of novel mesoporous DFNS in cross-linked CO 2 -philic poly (ethylene oxide) (PEO) matrix can not only promote gas transportation, but also increase the loading rate of low-molecular-weight PEG in membranes. Meanwhile, the introduction of PEGDEM-500 into the DFNS reduces the influence of non-selective interface on the selectivity. We identified the optimized performance of low-molecular-weight PEG impregnated PEO-based MMMs demonstrates the superior CO 2 permeability (up to 2281.1 Barrer, a 478.5% increment than that of original UV cross-linked PEO membrane) and high selectivity (up to 48.1 for CO 2 /N 2 ), which can easily surpass the (2008) Robeson's Upper Bound. The long-term stability of our membrane is also excellent, which makes great promise for CO 2 capture in CCS towards sustainably environmental remediation.

Preparation and properties of silane coupling agent modified zeolite as warm mix additive

Xiaobin Han,Jianying Yu,Zhilong Cao,Ruiyang Wang,Wei Du,Peng He,Yangyang Ge

Zeolite is a common additive for warm mix asphalt. In order to improve the dispersion and water loss rate of zeolite in asphalt, it was modified by γ-ammoniapropyltriethoxysilane (KH550, namely KH550-Zeolite) and γ-(2,3-epoxypropoxy)propytrimethoxysilane (KH560, namely KH560-Zeolite). FTIR demonstrated KH550 and KH560 were grafted to the surface of zeolite through chemical reaction, and SEM showed that the agglomeration of the modified zeolites was weaker than that of zeolite. Laser particle size analyzer revealed the particle size of zeolite, KH550-Zeolite and KH560-Zeolite mainly distributed in 7–20 μm, 4–10 μm and 3–8 μm, respectively. The KH550-Zeolite and KH560-Zeolite were more compatible with n-heptane than zeolite and the water contact angles of zeolite, KH550-Zeolite and KH560-Zeolite were 0°, 36.75° and 63.5°. In hot asphalt, the water loss of zeolite was non-uniform (first fast, then slow), while that of KH550-Zeolite and KH560-Zeolite was uniform, which results in that the viscosity reduction effect of KH550-Zeolite and KH560-Zeolite on asphalt was better than that of zeolite. The residual Marshall stability, flexural breaking strain and dynamical stability of warm mix asphalt with KH550-Zeolite or KH560-Zeolite were higher than those of warm mix asphalt with zeolite. When the mixing and compaction temperature of warm mix asphalt was 30 °C lower than that of hot mix asphalt, the comprehensive road performance of warm mix asphalt with KH550-zeolite was better than that of hot mix asphalt.

Chemical inhibition of wax precipitation to improve cracking resistance of warm mix asphalts

Haopeng Zhang,Yanjun Qiu

In recent years, the wax-based warm mix additives are always doped into the asphalt to lower the construction temperatures, which can lead to the substantial energy savings and promote energy sustainability. However, few studies have considered the methods to inhibit the premature precipitation of wax-based warm mix additives in warm mix asphalt, which will deteriorate the cracking resistances of asphalt pavement. This paper introduced a new method from the essence of the phase separation between the wax-based warm mix additives and asphalt leading to the reduction of cracking resistance, which chemically inhibited the premature precipitation of common additives (linear aliphatic hydrocarbon (LAH), polyethylene (PE), fatty acid amides (FAA), and Fischer-Tropsch (F-T) waxes) by wax inhibitors (WIs) to improve the compatibility of additives, and the cracking resistances of warm mix asphalts with addition of WIs were also investigated through the multi-stress creep recovery (MSCR), linear amplitude sweep (LAS), dynamic mechanical analysis (DMA), and thermal fatigue tests. The results show that the wax precipitation temperature (WPT) of warm mix asphalt reduces with the addition of WIs to inhibit the precipitation of additives to improve the compatibility, and the wax inhibition efficiencies of WIs are better with the asphalt doped with short-chain additives at higher content. The WIs can provide the warm mix asphalt with the additional elasticity and decrease the dependency on the applied strain, which increases the high-temperature rutting and intermediate-temperature fatigue resistances. The cracking and thermal fatigue resistances of warm mix asphalt at the low temperature can also be improved after doping with WIs, which is leaded by the precipitation inhibition of additives. Therefore, it is promising to improve the cracking resistances of warm mix asphalts and develop a new energy-saving pavement by chemically inhibiting the precipitation of additives.

A Novel Lysosome Targeting Chimera for Targeted Protein Degradation via Split-and-Mix Strategy

Jinpeng Wang,Yuechen Wang,Fenfang Yang,Qinhong Luo,Zhanfeng Hou,Yun Xing,Fei Lu,Zigang Li,Feng Yin

Targeted protein degradation is becoming more and more important in the field of drug development. Compared with proteasomal-based degraders, lysosomal-based degraders have a broader target spectrum of targets, which have been demonstrated to have great potential, especially in degrading undruggable proteins. Recently, we developed a programmable and facile screening PROTAC development platform based on peptide self-assembly termed split-and-mix PROTAC (SM-PROTAC). In this study, we applied this technology for the development of lysosome-based degraders, named a split-and-mix chaperone-mediated autophagy-based degrader (SM-CMAD). We successfully demonstrated SM-CMAD as a universal platform by degrading several targets, including ERα, AR, MEK1/2, and BCR-ABL. Different from other lysosomal-based degraders, SM-CMAD was capable of facile screening with programmable ligand ratios. We believe that our work will promote the development of other multifunctional molecules and clinical translation for lysosomal-based degraders.

Elastic Deformation of Cellulose/Lignin-Based Anode for Rejuvenating Aged Mix-Cultured Electroactive Biofilms

Xue Liu,Zheng Zhang,Qingwen Zheng,Chengcheng Suo,Bailing Dong,Huiying Song,Jiayi Wang,Jia Liu,Runfeng Yuan,Sailike Milanbieke,Sha Luo,Chenhui Yang,Zhijun Chen,Ruiwen Wang,Wei Li,Shouxin Liu

Electroactive biofilms (EABs) are essential components of both natural and artificial bio-electrochemical systems (BESs). However, the inevitable decay of EABs during prolonged operation can diminish their performance. In this contribution, an effective and noninvasive strategy for rejuvenating aging biofilms by the elastic deformation of anode material is approved. The synthesized wood tracheid-like structures anode material showed excellent compressibility and fatigue resistance in a wet state. The findings indicate that after the elastic deformation of the anode, aged biofilm exhibited a 37.5% increase in metabolic activity, and multi-SIM images confirmed the removal of dead cells. Analysis of the extruded substance revealed a significant removal of loosely bound extracellular polymeric substance which doesn't contribute directly to electron transfer. Community analysis demonstrated the rejuvenation process suppressed the ecological competition from non-exoelectrogens. Overall, there is a notable 25.97% increase in power density following the elastic deformation of the anode. Additionally, ion diffusion, specific capacitance, and catalytic response current all improved. To the knowledge, this is the first report employing an anode deformation strategy to restore decayed mix-cultured electroactive biofilm, which is vital for the practical long-term application of BESs. This work also offers new insights into the mechanical influence of anode materials on microorganisms.

Robust Mix-Charged Polyzwitterionic Hydrogels for Ultra-Efficient Atmospheric Water Harvesting and Evaporative Cooling

Xuanxuan Du,Zhiheng Xie,Hanchao Zhang,Shoukun Jiang,Xing Su,Jintu Fan

Atmospheric water harvesting (AWH) presents great potential in addressing the increasing global challenges in freshwater and energy supply, especially in arid and semi-arid regions. The recent AWH materials focus primarily on maximizing water uptake, while conventional approaches prioritize hygroscopicity at the expense of mechanical integrity, which severely limits their applicability in real-world scenarios. In this study, a novel tunable hygroscopic mix-charged polyzwitterionic hydrogel (THMPH) is reported that achieves dual excellence in outstanding moisture absorbency and mechanical robustness. Owing to the broad ionic crosslink's degree enabling the rigid skeletal framework and energy-dissipative sacrificial networks, THMPH exhibits more than 200 times higher mechanical ductility (225 kPa tensile strength retention at 200% mass swelling ratio) in comparison with the commonly-used AWH zwitterionic polybetaine. The optimized topological structure coupled with improved lithium chloride binding affinity results in excellent water uptake (2.9 g g −1 at 25 °C, 70% RH). When THMPH is used for daytime photovoltaic panel cooling, it can provide a 15 °C temperature reduction of a PV panel under 1 kW m −2 solar irradiation, resulting in a 7.33% increase in solar energy conversion efficiency. This hydrogel design paradigm, synergizing superior hygroscopicity with exceptional mechanical robustness, demonstrates significant potential for advancing practical applications.

Research on the Preparation and Performance of Biomimetic Warm-Mix Regeneration for Asphalt Mixtures

Xin Jin,Haoxuan Fu,Deli Li,Ye Yang,Yanhai Yang,Yanfeng Li,Fengchi Wang,Jiupeng Zhang

To determine the formula for biomimetic warm-mix regeneration and fulfill the requirements of a “high waste asphalt mixture content, high quality, and high level” for its usage in reclaimed asphalt pavement (RAP), this paper first determined the suitable preparation process and formula for biomimetic warm-mix regeneration based on orthogonal experiments and a gray correlation analysis. Then, the optimum dosage of the warm-mix regenerant was determined by a uniaxial penetration test, low-temperature splitting test, and freeze–thaw penetration test. The rutting test was conducted to characterize the high-temperature performance of the asphalt mixture. The Immersion Marshall Test and the freeze–thaw splitting test were used to characterize the water stability of the recycled asphalt mixture. The low-temperature small beam test was employed to study the low-temperature performance of the recycled asphalt mixture. The asphalt’s short-term and long-term aging processes were simulated using the rotary thin-film oven test (RTFOT) and the pressure aging test (PAV). The action mechanism of biomimetic warm-mix regeneration was revealed by Fourier-transform infrared spectroscopy (FTIR). Finally, a comprehensive thermal performance test was conducted on the aged asphalt after biomimetic warm-mix regeneration. The results showed that the self-made biomimetic warm-mix regeneration agent exhibited an excellent regenerative effect on RAP and significantly reduced the mixing temperature of the styrene–butadiene–styrene (SBS)-modified asphalt mixture. In addition, the self-made biomimetic warm-mix regeneration agent effectively improved the high- and low-temperature performance of the recycled asphalt mixture, but had no noticeable effect on the water stability. The suggested dosage of the biomimetic warm-mix regeneration agent was 6%, and the mixing temperature was 130 °C. The microscopic chemical analysis revealed that biomimetic warm-mix regeneration restored the performance of aged asphalt by supplementing the light component. The change rules of the chemical functional groups and the comprehensive thermal properties of the recycled mixture showed a good correlation with the change rules of its high- and low-temperature performance.
