Microporous organic networks (MONs) have shown great promise in separation science recently. Exploring novel, simple and convenient strategy to fabricate MONs coated capillary columns for gas chromatography (GC) still remains challenging but desirable for the development of MONs in chromatographic separation. To extend the potential application of MONs in separation science and to further develop novel method for the fabrication of MONs-based capillary columns, here we demonstrate a novel covalent coupling strategy to fabricate uniform MONs bonded capillary columns for GC separation of position isomers and hydrocarbons. The bare capillary column was firstly modified with (3-bromopropyl)trimethoxysilane to provide bromine sites for coupling with alkynyl monomers . The amino- and hydroxyl-functionalized MONs (MON-NH 2 and MON-OH) were then directly grown onto the inner wall of the brominated capillary columns via the covalent coupling between bromine and alkynyl groups. The uniform MON-NH 2 and MON-OH bonded capillary columns were obtained and showed good resolution for GC separation of dichlorobenzene , chlorotoluene, bromotoluene, and propylbenzene position isomers and many other hydrocarbons including linear alkanes, alkylbenzenes , pinene isomers, cyclohexane and benzene, ketones and aldehydes. The MONs bonded capillary columns also owned good lifetime and precision for dichlorobenzene isomers with the relative standard deviations (RSDs) of 0.2–0.3%, 1.2–2.1%, and 1.7–2.5% for retention time, peak height and peak area, respectively. In addition, the fabricated MON-NH 2 and MON-OH bonded capillary columns offered better resolution than commercial InertCap-1, InertCap-5, InertCap-1701 and InertCap-WAX capillary columns for the separation of chlorotoluene and bromotoluene position isomers. These results revealed the feasibility of covalent coupling strategy to fabricate MONs-based stationary phases in GC, highlighting the potential of MONs in separation science.

Cellular defense system represented by glutathione (GSH) greatly weakens the outcomes of cancer therapy by antioxidation and detoxification. GSH depletion has been proved to be an effective way to enhance the efficacy of reactive oxygen species (ROS)-based therapies and chemotherapy. However, the existing strategies of GSH depletion still face the problems of unclear biosafety and high complexity of multicomponent co-delivery. In this study, we developed a GSH-depleting carrier platform based on disulfide-bridged mesoporous organosilica nanoparticles (MONs) to destroy the cellular defense system for cancer therapy. Responding to the high level of GSH in cancer cells, the disulfide bonds in the framework of MONs could be broken and consumed substantial GSH at the same time. Moreover, this process also promoted the degradation of MONs. In order to evaluate the effect of this platform in cancer therapy, chemotherapeutic drug cisplatin was loaded into MONs ( [email protected] ) to treat drug-resistant non-small cell lung cancer. In vitro and in vivo results indicated that [email protected] efficiently triggered GSH depletion, promoted platinum-DNA adduct formation, and induced cell apoptosis, resulting in significant tumor growth inhibition without marked toxicity. Taken together, the cellular defense system-destroying nanoparticles provide a promising platform for enhanced cancer therapy.
Microporous organic networks (MONs) are a potential class of microporous materials in analytical chemistry . As great efforts have been made in sample pretreatment and gas chromatography, the application of MONs in high performance liquid chromatography (HPLC) has largely lagged and has never been investigated. In addition, directly packing of MONs into columns for HPLC is impractical due to the high column pressure and low column efficiency resulted from the sub-micron sized and irregular MONs. To solve these issues and uncover the possibility of MONs in HPLC, here we report the fabrication of spherical silica amino-functionalized MON (SiO 2 @MON-NH 2 ) composite as a novel stationary phase for HPLC. Neutral, acidic and alkaline probes were evaluated on SiO 2 @MON-NH 2 packed column. A column efficiency of 21,246 plates per meter was obtained for n -propylbenzene. The results revealed the hydrophobic interaction was the main separation mechanism on SiO 2 @MON-NH 2 packed column. Baseline separation of benzenediol , dichlorophenol , chlorophenol and phenylenediamine position isomers, deoxynucleosides and nucleosides was also achieved on SiO 2 @MON-NH 2 packed column. The relative standard deviations of retention time, peak height and peak area for the studied position isomers, deoxynucleosides and nucleosides were 0.2–0.7%, 0.3–3.7% and 1.7–3.5%, respectively. The SiO 2 @MON-NH 2 packed column also offered better resolution for dichlorophenol isomers, deoxynucleosides and nucleosides than SiO 2 –NH 2 and commercial C18 columns. These results revealed the potential of MONs in HPLC and also provided a new way for the use of nano-sized or irregular MONs in HPLC.
