A Novel Copolymer-Based Functional SPECT/MR Imaging Agent for Asialoglycoprotein Receptor Targeting

Pu Zhang,Zhide Guo,Deliang Zhang,Chang Liu,Guibing Chen,Rongqiang Zhuang,Manli Song,Hua Wu,Xianzhong Zhang

The aim of this study is to develop a copolymer-based single-photon emission computed tomography/magnetic resonance (SPECT/MR) dual-modality imaging agent that can be labeled with both technetium-99m (99mTc) and gadolinium (Gd) and target asialoglycoprotein receptor (ASGPR) via galactose. Monomers of N-p-vinylbenzyl-6-(2-(4-dimethylamino)benzaldehydehydrazono) nicotinate (VNI) for labeling of 99mTc, 5,8-bis(carboxymethyl)-3-oxo-11-(2-oxo-2-((4-vinylbenzyl)amino)ethyl)-1-(4-vinylphenzyl)-2,5,8,11-tetraazatridecan-13-oic acid (V2DTPA) for labeling of Gd, and vinylbenzyl-O-β-d-galactopyranosyl-d-gluconamide (VLA) for targeting ASGPR were synthesized, respectively. Then the copolymer P(VLA-co-VNI-co-V2DTPA) (pVLND2) was synthesized and characterized by gel permeation chromatography, dynamic light scattering, and high-performance liquid chromatography analysis. After labeling with 99mTc and Gd simultaneously, the radiochemical purity, toxicity, relaxivity (r1), and in vivo SPECT/MR imaging in mice were evaluated. Single-photon emission computed tomography/magnetic resonance imaging and biodistribution results showed that pVLND2 could target ASGPR well. The significantly improved signal to noise ratio was observed in mice liver during MR imaging. All the results suggest that this novel kind of copolymer has the potential to be further developed as a functional SPECT/MR imaging agent.

Hydrothermal Synthesis of Monodispersed BaGdF5:Yb/Er Nanoparticles for CT and MR Imaging

Rongjin Sun,Ting Yin,Peng Huang,Guo Gao,Joseph George Shapter,Yulan Shen,Jingjing Zhang,Daxiang Cui

In this paper, we report the development of a facile one-pot hydrothermal method for the controlled synthesis of monodispersed BaGdF 5 :Yb/Er nanoparticles for computed tomography (CT) and magnetic resonance (MR) imaging. The as-prepared nanoparticles have uniform size with a diameter of 20–25 nm. MTT tests show that the cell viability surpasses 90% even with a concentration of nanoparticles of 500 µg/mL, suggesting that the as-prepared BaGdF 5 :Yb/Er nanoparticles possess low toxicity. For both BaGdF 5 :Yb/Er nanoparticles and iopromide, the Hounsfield unit (HU) values increase with the their concentration. The slope of BaGdF 5 :Yb/Er nanoparticles is about 26.47, which is much higher than that of iopromide (16.98), indicating a better CT imaging effect. Interestingly, we find that the synthesized BaGdF 5 :Yb/Er nanoparticles exhibit paramagnetism. Our in vitro and in vivo imaging experiments demonstrate that the synthesized monodispersed BaGdF 5 :Yb/Er nanoparticles can serve as effective contrast agents for CT and MR imaging.

Synthesis of gadolinium-based Bi2S3 nanoparticles as cancer theranostics for dual-modality computed tomography/magnetic resonance imaging-guided photothermal therapy

Hongying Guo,Xiaoshuang Zhao,Hongmei Sun,Hongda Zhu,Honghao Sun

Development of a safe, efficient and inexpensive multifunctional nanoplatform using a facile approach for multimodal imaging and therapeutic functions becomes more and more practically relevant but challenging. In this work, we demonstrated a novel nanocomposites (Bi2S3-Gd) for computed tomography (CT)/magnetic resonance (MR) imaging-guided photothermal therapy (PTT) for cancer in vitro. It was achieved by modification of hydrophobic Bi2S3 with a smart amphiphilic gadolinium-chelated ligand. The as-prepared nanocomposites composed of low-cost Bi2S3 and gadolinium complexes, showed high stability, excellent biocompatibility and good photostability. It was observed that Bi2S3-Gd nanocomposites can efficiently convert the NIR light into heat, and then suppressed the growth of tumor cells under NIR laser irradiation. Apart from serving as an effective photothermal agent, the as-prepared nanomaterials could induce an efficient contrast enhancement for both CT and MR imaging at low concentrations of Bi and Gd, rendering more accurate diagnosis. This work suggests the potential of Bi2S3-Gd nanomaterials as a novel multifunctional nanoplatform for CT/MR imaging-guided PTT for cancer.

A chemophotothermal and targeting multifunctional nanoprobe with a tumor-diagnosing ability

Li Niannian,Cheng Jiejun,Zhang Ying,Wang Jie,Huang Gang,Zhu Jun,He Dannong

The development of multifunctional nanoparticles for diagnosis and therapy of cancer has been a focus of research in recent years. Owing to excellent properties of chemophotothermal therapy and tumor targeting accompanied by magnetic resonance (MR) or computed tomography (CT) imaging, mesoporous silica has been widely applied to tumor diagnosing carriers. However, previous research focused more on chemotherapy combined with photothermal therapeutic strategies but ignored intuitive assessment of tumors. In this work, we report a novel nanoprobe consisting of hyaluronic-acid–modified Gold Nanorods@mSiO2@Mn@DOX (GNR@MMH@DOX) nanoparticles, which realize multifunctional integration of therapeutics and imaging. Au nanorods (GNRs) were synthesized without a seed precursor by a water bath method, then sequentially modified with mesoporous silica, doped with Mn 2+ , and loaded with doxorubicin in a solution. In vitro and in vivo assays verified their great biocompatibility, good stability, and attractive specificity of targeting to receptor CD44, which is overexpressed on cancer cells. In addition, GNR@MMH nanoparticles proved to be a more distinguishable CT/MR imaging contrast compared to commercial contrast agents. The article demonstrates promising composition for CT/MR imaging and photothermal therapy of tumors, giving new insights into the diagnosis and therapy of different kinds of malignant tumors.

Active targeted dual-modal CT/MR imaging of VX2 tumors using PEGylated BaGdF5 nanoparticles conjugated with RGD

Tao Wang,Guorong Jia,Chao Cheng,Qiuhu Wang,Xiao Li,Yanyan Liu,Chaofan He,Luguang Chen,Gaofeng Sun,Changjing Zuo

Multimodal imaging can improve the accuracy of cancer diagnosis by combining two or more imaging modalities into one system. In this study, PEGylated BaGdF5 nanoparticles (NPs) were synthesized by a hydrothermal method, and the formed PEG-BaGdF5 NPs showed good X-ray attenuation properties and positive MR contrast effects in vitro. Considering that the overexpression of integrin αvβ3 in the endothelial cells of angiogenic tumor vasculature is universal, RGD peptide, which can recognize and bind integrin αvβ3 specifically, was connected to the surface of PEG-BaGdF5 NPs. The resulting RGD-PEG-BaGdF5 NPs had good biocompatibility and a long half-life in vivo. In VX2 tumor-bearing mice, the HU values and MR signal enhancement of tumor sites in the targeted group were both higher than that in the non-targeted group after intravenous injection of RGD-PEG-BaGdF5 NPs or PEG-BaGdF5 NPs, respectively. Therefore, the RGD-PEG-BaGdF5 NPs may be used as dual-modal CT/MR contrast agents of different types of solid tumors via an active RGD-mediated vasculature targeting pathway.

Microwave–assisted synthesis of water–disperse and biocompatible NaGdF4: Yb,Ln@NaGdF4 nanocrystals for UCL/CT/MR multimodal imaging

Songxia Yu,Zhiqiang Wang,Ruijun Cao,Lingjie Meng

In this paper, we have developed a series of rare earth fluoride nanocrystals (NCs) that can be used for multimodal imaging applications. Three highly monodisperse and water dispersible NaGdF 4 :Yb,Ln@NaGdF 4 (Ln   =   Er, Tm, Ho) upconversion luminescence (UCL) NCs with an average size of 75   nm were prepared using a facile and efficient microwave–assisted solvothermal reaction in ethylene glycol (EG). The morphology, crystal structures, size distributions and zeta potentials of the resulting NCs were fully characterized by transmission electron microscopy (TEM), X–ray diffraction (XRD) and Nano–ZS90 Zetasizer. The obtained NaGdF 4 :Yb,Ln@NaGdF 4 (Ln   =   Er, Tm, Ho) NCs could emit bright blue, green and red luminescence in water upon laser excitation at 980   nm. Furthermore, these core–shell NCs exhibited low cytotoxicity for HeLa and C9H2 cells, separately. Moreover, these NCs also displayed excellent Hounsfield units (HU) values in X–ray computed tomography (CT) imaging and high relaxivity parameters (r 1 ) in magnetic resonance (MR) imaging in vitro . Thus, these small, water–dispersible and biocompatible NCs show excellent potential as contrast agents for UCL/CT/MR multimodal imaging and other biomedical applications.

Dual drug delivery and sequential release by amphiphilic Janus nanoparticles for liver cancer theranostics

Lingyu Zhang,Manjie Zhang,Li Zhou,Qinghe Han,Xiangjun Chen,Shengnan Li,Lu Li,Zhongmin Su,Chungang Wang

Co-delivery of two drugs with diverse physicochemical properties and specific administration order for cancer theranostics are vitally important for drug resistance conquering and side effects reducing. Consequently, we explored a unique amphiphilic PCL-AuNC/Fe(OH) 3 -PAA Janus nanoparticle (JNP) to simultaneously preserve the hydrophilic drug (doxorubicin) and hydrophobic drug (docetaxel) in their distinct domains. Owing to their extraordinary heterostructure and independent pH and NIR sensitive properties, the optional sequential drug release by a single inorganic JNP was realized for the first time, and the results presented the synchronous release of two drugs had 5% better therapeutic effect. In addition, the excellent computed X-ray tomography/magnetic resonance (CT/MR) imaging capabilities from AuNC and Fe(OH) 3 suggested our JNPs could effectively guide the cancer therapy. Furthermore, the mice treated with dual drug loaded PCL-AuNC/Fe(OH) 3 -PAA JNPs under near infrared (NIR) laser irradiation showed better tumor inhibition than solo drug, cocktail and dual drug treated groups, indicating the effectivity and significance of combined cancer therapy.

Metal-organic nanostructure based on TixOy/Ruthenium reaction Units: For CT/MR Imaging-Guided X-ray induced dynamic therapy

Dongsheng Li,Qixuan Dai,Hu Chen,Huirong Lin,Zhixiang Lu,Huili Zheng,Peng Lv,Wei Li,Gang Liu,Chengchao Chu

We report a computed tomography/magnetic resonance (CT/MR) imaging-guided X-ray induced dynamic therapy (XDT) nanotheranostic platform based on Ti x O y /Ru reaction units. A new metal–organic nanostructure (TiZrRu-MON) was prepared via a solvothermal reaction of [Ti 8 Zr 2 O 12 (COO) 16 ] clusters and bipyridine-(COOH) 2 /Ru(II)-bipyridine-(COOH) 2 co-organic ligands. We demonstrate that the cluster can absorb X-ray and transform the energy to Ru(II)-bipyridine-(COOH) 2 with effective reactive oxygen species (ROS) production, which is five times more than TiZr-MON. In addition, Gd(III) ions were anchored to the bipyridine of MON for concurrent CT/MR imaging. Notably, the TiZrRuGd-MON possesses ultrahigh longitudinal relaxivity ( r 1 value up to 53.09 mM −1 ·s −1 at 1.5 T, 7-fold higher than commercial Gd-DTPA). With the guidance of CT/MR dual-mode imaging that provides comprehensive and complementary contrast information, the TiZrRuGd-MON is monitored to accumulate in tumor sites and eliminate tumor with high efficiency under X-ray irradiation. The TiZrRu-MON-based XDT strategy opens a pathway for clinical translational research of cancer theranostics.

Core–shell BaYbF5:Tm@BaGdF5:Yb,Tm nanocrystals for in vivo trimodal UCL/CT/MR imaging

Peng Zhang,Yangyang He,Jianhua Liu,Jing Feng,Zhiqiang Sun,Pengpeng Lei,Qinghai Yuan,Hongjie Zhang

Lanthanide-doped nanocrystals have been researched extensively and used for bioimaging because of their optical properties, magnetic properties and X-ray absorption. Core–shell-structured lanthanide-doped nanocrystals have been developed and characterized by TEM and XRD analysis. The nanocrystals are composed of BaYbF5:0.5% Tm as the core and BaGdF5:20% Yb, 0.5% Tm as the shell. Apart from characterization of the nanocrystals, evaluation of both their cytotoxicity via MTT assays and their long-term toxicity via histological analysis showed that their cytotoxicity was low, indicating the possibility of further in vivo imaging. This work combined the functions of trimodal imaging into one nanoplatform and then UCL, CT, and MR imaging with core–shell-structured nanocrystals were investigated both in vitro and in vivo. Taking into consideration its structural characteristics and trimodal imaging abilities, it is expected that the developed multifunctional nanoplatform may be potentially useful for diagnosing diseases at an early stage.

Cancer cell membrane-coated magnetic nanoparticles for MR/NIR fluorescence dual-modal imaging and photodynamic therapy

Jiong Li,Xuandong Wang,Dongye Zheng,Xinyi Lin,Zuwu Wei,Da Zhang,Zhuanfang Li,Yun Zhang,Ming Wu,Xiaolong Liu

Theranostic nanoprobes integrated with dual-modal imaging and therapeutic functions, such as photodynamic therapy (PDT), have exhibited significant potency in cancer treatments due to their high imaging accuracy and non-invasive advantages for cancer elimination. However, biocompatibility and highly efficient accumulation of these nanoprobes in tumor are still unsatisfactory for clinical application. In this study, a photosensitizer -loaded magnetic nanobead with surface further coated with a layer of cancer cell membrane (SSAP-Ce6@CCM) was designed to improve the biocompatibility and cellular uptake and ultimately achieve enhanced MR/NIR fluorescence imaging and PDT efficacy. Compared with similar nanobeads without CCM coating, SSAP-Ce6@CCM showed significantly enhanced cellular uptake, as evidenced by Prussian blue staining, confocal laser scanning microscopy (CLSM) and flow cytometric analysis. Consequently, SSAP-Ce6@CCM displayed a more distinct MR/NIR imaging ability and more obvious photo-cytotoxicity towards cancer cells under 670 nm laser irradiation. Furthermore, the enhanced PDT effect benefited from the surface coating of cancer cell membrane was demonstrated in SMMC-7721 tumor-bearing mice through tumor growth observation and tumor tissue pathological examination. Therefore, this CCM-disguised nanobead that integrated the abilities of MR/NIR fluorescence dual-modal imaging and photodynamic therapy might be a promising theranostic platform for tumor treatment.

X-ray activated near-infrared persistent luminescence nanoparticles for trimodality in vivo imaging

Jinyuan Wang,Xia Sun,Jixuan Xu,Lin Liu,Peng Lin,Xiaofang Luo,Yan Gao,Junpeng Shi,Yun Zhang

As promising luminescence nanoparticles, near-infrared (NIR) persistent luminescence nanoparticles (PLNPs) have received extensive attention in the field of high-sensitivity bioimaging in recent years. However, NIR PLNPs face problems such as short excitation wavelengths and single imaging modes, which limit their applications in vivo reactivated imaging and multimodal imaging. Here, we report for the first time novel Gd2GaTaO7:Cr3+, Yb3+ (GGTO) NIR PLNPs that integrate X-ray activated NIR persistent luminescence (PersL), high X-ray attenuation and excellent magnetic properties into a single nanoparticle (NP). In this case, Cr3+ is used as the luminescence center. The co-doped Yb3+ and coating effectively enhance the X-ray activated NIR PersL. At the same time, the presence of the high-Z element Ta also makes the GGTO NPs exhibit high X-ray attenuation performance, which can be used as a CT contrast agent to achieve in vivo CT imaging. In addition, since the matrix contain a large amount of Gd, the GGTO NPs show remarkable magnetic properties, which can realize in vivo MR imaging. GGTO NPs combine the trimodal benefits of X-ray reactivated PersL, CT and MR imaging and are suitable for single or combined applications that require high sensitivity and spatial resolution imaging

CT/MR detectable magnetic microspheres for self-regulating temperature hyperthermia and transcatheter arterial chemoembolization

Chengxiong Wei,Chengwei Wu,Xin Jin,Peinan Yin,Xiaogang Yu,Chao Wang,Wei Zhang

The embolic microspheres containing magnetic nanoparticles and anti-tumor drugs have been proposed for transcatheter arterial chemoembolization (TACE). However, this technique still suffers the poor control of hyperthermia temperature and drug release behavior. Herein, the magnetic microspheres based on low Curie temperature superparamagnetic iron oxide nanoparticles are developed by emulsification cross-linking of gelatin, genipin, and sodium alginate. The magnetic microspheres can self-regulate the hyperthermia temperature at around 50°C, un-necessitating any temperature control facilities. The magnetic microspheres can load doxorubicin hydrochloride and the loaded drug can be released in a controllable way by using an alternating magnetic field. Cytocompatibility and hemolysis evaluations confirm the non-cytotoxicity and negligible hemolysis of magnetic microspheres. The embolization model on rabbit auricular artery demonstrates that the magnetic microspheres can occlude the targeted blood vessel and are visualized under CT/MR imaging. All these findings suggest that the prepared magnetic microspheres could be used as the embolic agent in TACE. Statement of significance The existing magnetic embolic microspheres suffer the poor control of hyperthermia temperature and drug release behavior in TACE. In this work, we developed the magnetic embolic microspheres based on superparamagnetic iron oxide nanoparticles with a low Curie temperature. Upon the application of alternating magnetic field, the embolic microspheres can self-regulate the hyperthermia temperature at around 50°C and the drug loaded in the microspheres can be released in a somewhat controllable manner. The embolic microspheres are also detectable to both CT and MR. These characteristics enable the developed microspheres to simultaneously realize self-regulating temperature hyperthermia, on-demand drug release, embolization, and CT/MR imaging.
