Despite the significant advances of imaging techniques nowadays, accurate diagnosis of bacterial infections and real-time monitoring the efficacy of antibiotic therapy in vivo still remain huge challenges. Herein, a self-assembling peptide (FFYEGK) and vancomycin ( Van ) antibiotic molecule co-modified gadolinium (Gd) MRI nanoaggregate probe (GFV) for detecting Staphylococcus aureus ( S. aureus ) infection in vivo and monitoring the treatment of S. aureus- infected myositis by using daptomycin ( Dap ) antibiotic as model are designed and fabricated. The as-prepared GFV probe bears Van molecules, making itself good bacteria-specific targeting, and the peptide in the probe can enhance the longitudinal relaxivity rate ( r 1 ) after self-assembly due to the π–π stacking. The study showed that, based on the GFV probe, bacterial infections and sterile inflammation can be discriminated, and as few as 10 5 cfu S. aureus can be detected in vivo with high specificity and accurately. Moreover, the T 1 signal of GFV probe at the S. aureus -infected site in mice correlates with the increasing time of Dap treating, indicating the possibility of monitoring the efficacy of antibacterial agents for infected mice based on the as proposed GFV probe. This study shows the potential of GFV probe for diagnosis, evaluation, and prognosis of infectious diseases in clinics.

Contrast-enhanced magnetic resonance imaging (CE-MRI) is a promising approach for the diagnosis of kidney diseases. However, safety concerns, including nephrogenic systemic fibrosis, limit the administration of gadolinium (Gd)-based contrast agents (GBCAs) in patients who suffer from renal impairment. Meanwhile, nanomaterials meet biosafety concerns because of their long-term retention in the body. Herein, we propose a small-molecule manganese-based imaging probe Mn-PhDTA as an alternative to GBCAs to assess renal insufficiency for the first time. Mn-PhDTA was synthesized via a simple three-step reaction with a total yield of up to 33.6%, and a gram-scale synthesis can be realized. Mn-PhDTA has an r1 relaxivity of 2.72 mM–1 s–1 at 3.0 T and superior kinetic inertness over Gd-DTPA and Mn-EDTA with a dissociation time of 60 min in the presence of excess Zn2+. In vivo and in vitro experiments demonstrate their good stability and biocompatibility. In the unilateral ureteral obstruction rats, Mn-PhDTA provided significant MR signal enhancement, enabled distinguishing structure changes between the normal and damaged kidneys, and evaluated the renal function at different injured stages. Mn-PhDTA could act as a potential MRI contrast agent candidate for the replacement of GBCAs in the early detection of kidney dysfunction and analysis of kidney disease progression.

Taking advantage of the superior biocompatibility, good stability in a wide pH and temperature range, as well as its strong affinity with DNA of hydroxyapatite (HAp), tetrahedral DNA nanostructures (TDNs) conjugated with AS1411 aptamer ( anti -nucleolin overexpressed on tumor cell membranes) were employed as affinity ligands to construct a novel mono-dispersed HAp based probe with Gd 3+ doping (Apt-TDNs-GdHAp) for MR imaging. The adsorption of TDNs on the nano-HAp surface facilely accomplished the construction of the Apt-TDNs-GdHAp probes. Meanwhile, the use of hydrophilic TDNs not only favored the phase-transfer from the oil phase to the aqueous phase, but also enhanced the mono-dispersion of this probe due to the well-ordered distribution of TDNs on the surface of nano-HAp. Moreover, Apt-TDNs-GdHAp probe with a better mono-dispersion and crystalinity achieved twice higher longitudinal relaxivity (r 1 value) than that of GdHAp synthesized by microwave-assisted method (Microwave-GdHAp), exhibiting much more excellent T 1 -weighted imaging performance. With the introduction of TDNs, the stability and the tumor-targeting accessibility were also greatly improved, showing its great potential for further bio-applications.

Adenosine triphosphate (ATP) is a central mediator of cancer cell metabolism and signaling, driving processes such as proliferation, invasion, metastasis, and therapeutic resistance. Current ATP imaging modalities-including fluorescence, chemiluminescence/bioluminescence, photoacoustic imaging, and magnetic resonance imaging (MRI)-face inherent trade-offs among sensitivity, spatial resolution, and tissue penetration capability. Here, a dual-mode probe is reported that synergistically couples the anatomical resolution of MRI with the high-sensitive, long-lasting signals of afterglow imaging for visualization of ATP in the tumor microenvironment. The probe is constructed via DNA hybridization between an afterglow unit and a T 1 -weighted MRI contrast agent, with an ATP-specific aptamer mediating probe disassembly upon ATP binding. This design generates simultaneous turn-on afterglow and MRI signals, enabling the imaging of intracellular and extracellular ATP fluctuations. Metabolic modulators-coenzyme Q10 (an ATP activator) and 2-deoxy-D-glucose (2-DG, a glycolysis inhibitor)-are applied to demonstrate dynamic, bidirectional tracking of ATP levels in vivo. Furthermore, by monitoring ATP depletion during synergistic metabolic therapy (cisplatin + 2-DG), a correlation between imaging readouts and therapeutic efficacy is established. The work provides anintegrated MRI/afterglow imaging platform for high-contrast, real-time ATP sensing in cancer, offering a powerful tool for elucidating tumor metabolism and evaluating metabolic interventions.
