Fluorine-19 (19F) MRI (19F-MRI) is a promising method for quantifying biomedical research and clinical applications without background interference. Nevertheless, dependency on high-field MRI systems limits the applicability of 19F-MRI. Low-field MRI systems are more common than high-field MRI systems. Hence, developing 19F-MRI at low-field MRI devices can promote the 19F-MRI translation in medical diagnosis. The detection sensitivity of fluorine agents is critical in 19F-MRI. Reduction of the 19F spin-lattice relaxation time (T1) enables an improved detection sensitivity while requiring ultrashort echo time (UTE) imaging methods to reduce the negative spin–spin relaxation (T2) decay effect. However, conventional UTE sequences require hardware with high performance. Herein, we introduce the k-space scaling imaging (KSSI) MRI sequence that accomplishes sampling k-space with variable scales to implement hardware-friendly UTE 19F-MRI compatible with low-field MRI systems. We implemented experiments with swine bone, a perfluorooctyl bromide (PFOB) phantom, and one tumor-bearing mouse on two self-customized low-field MRI systems. The swine bone imaging validated the ultrashort TE of KSSI. Under high concentrations of manganese ferrite, a high signal-to-noise ratio was shown in the imaging of a fluorine atom concentration of 658 mM, which indicated high-sensitivity detection of KSSI. Moreover, the KSSI sequence exhibited a 7.1 times signal-to-noise ratio of spin echo sequence on the PFOB phantom imaging with a fluorine atom concentration of 3.29 M. Additionally, the various concentrations of the PFOB phantom imaging revealed quantifiable capacity. Finally, the 1H/19F imaging was implemented with KSSI on one tumor-bearing mouse. This method provides the potential for clinical translation of fluorine probes at low-field MRI systems.

Gadolinium (III)-based T 1 contrast agents have been widely used in clinical MRI. In this study, salicylic acid-gadolinium chelate was prepared via directly coordination reaction of gadolinium ion and salicylic acid. Then, three polyethylene glycols with different molecular weight were modified on the surface of salicylic acid-gadolinium by simple chemical coupling to construct three core-shell structural Gd based composites. The scanning electron microscopy (SEM) and transmission electron microscopy (TEM) characterization results show that the composite is a spherical particle with a diameter of about 100–200 nm. The longitudinal relaxation rate r 1 of the Gal-PEG-2000 is 11.097 (mmol/L) −1 /s, and the ratio of r 2 / r 1 is as low as 2.53. The composite shows good liver and intestines MRI performances after being used in in vivo imaging, showing a good prospect of biological application.

Personalized radiotherapy strategies enabled by the construction of hypoxia-guided biological target volumes (BTVs) can overcome hypoxia-induced radioresistance by delivering high-dose radiotherapy to targeted hypoxic areas of the tumor. However, the construction of hypoxia-guided BTVs is difficult owing to lack of precise visualization of hypoxic areas. This study synthesizes a hypoxia-responsive T 1 , T 2 , T 2 mapping tri-modal MRI molecular nanoprobe (SPION@ND) and provides precise imaging of hypoxic tumor areas by utilizing the advantageous features of tri-modal magnetic resonance imaging (MRI). SPION@ND exhibits hypoxia-triggered dispersion-aggregation structural transformation. Dispersed SPION@ND can be used for routine clinical BTV construction using T 1 -contrast MRI. Conversely, aggregated SPION@ND can be used for tumor hypoxia imaging assessment using T 2 -contrast MRI. Moreover, by introducing T 2 mapping, this work designs a novel method (adjustable threshold-based hypoxia assessment) for the precise assessment of tumor hypoxia confidence area and hypoxia level. Eventually this work successfully obtains hypoxia tumor target and accurates hypoxia tumor target, and achieves a one-stop hypoxia-guided BTV construction. Compared to the positron emission tomography-based hypoxia assessment, SPION@ND provides a new method that allows safe and convenient imaging of hypoxic tumor areas in clinical settings.

Melanoma is one of the most aggressive forms of skin cancer. Accurate and early diagnosis of melanoma is crucial for improving patient outcomes. This study develops two TYR-activatable molecular probes, Mn-TyrEDTA and Al-18F-TyrEDTA, for the selective detection of melanoma in vivo. In vitro studies reveal that Mn-TyrEDTA exhibits TYR activity-dependent relaxivity enhancement, undergoing TYR-mediated oxidative polymerization, resulting in the formation of paramagnetic oligomers. UV–vis analysis supports this mechanism, showing time- and TYR concentration-dependent increases in broad band absorbance in the UV–vis region, specifically around 475 nm, due to the formation of o-quinone intermediates and melanin-like oligomers. HPLC analysis further confirmed the presence of polar oligomeric products in Mn-TyrEDTA solutions incubated with TYR/O2. MRI studies demonstrate Mn-TyrEDTA’s selective retention and signal enhancement in TYR-expressing melanoma tissues. Furthermore, PET imaging with Al-18F-TyrEDTA conducted using a dual-xenograft mouse model reveals significantly higher uptake and retention of Al-18F-TyrEDTA in TYR-expressing melanoma compared to TYR-negative tumors. This selective retention could be attributed to a TYR-mediated proximity labeling mechanism, where highly reactive quinones form covalent bonds with nearby tumor proteins. In summary, our findings establish Mn-TyrEDTA and Al-18F-TyrEDTA as promising TYR-activatable imaging probes, offering a novel strategy for the early diagnosis and prognosis of melanoma.

Graphical In this article, a reconfigurable and mobile magnetic microswarm as MRI contrast enhancer was investigated experimentally and numerically. By changing motion mode, it could adaptively navigate multiple terrains. Furthermore, we demonstrated this microswarm performed great enhancement of MRI imaging. This microswarm held great promise for highly sensitive and accurate intestinal MRI in the clinic. Highly effective contrast enhancer that processes targeting ability and maneuverability is in great demand in clinics for accurate diagnosis. Here a new strategy using deformable and manipulatable magnetic microswarm as MRI contrast enhancer is developed. Magnetic microswarm aggregated from nanoparticles is inherently deformable and they can be controlled with multiple programmable deform abilities. It is demonstrated that spatiotemporal programming magnetic field enables the magnetic microswarm not only to exhibit both ribbon-like and round-like behaviours but also to adaptively navigate multiple terrains. Intestinal model is conducted to explore the effect of magnetic microswarm as MRI enhancer, indicating the obvious enhancement of T 2 -weighted MRI sequences. This magnetic microswarm holds great promise for highly sensitive and accurate intestinal MRI in the clinic.

Within brain tissue engineering, stem cell implantation assisted by biodegradable injectable scaffolds is a crucial method. However, implanted scaffolds, especially microcarriers, may translocate due to the fluidity of the materials. Therefore, the development of an MRI contrast enhancement microcarrier, which could noninvasively trace the location of the implant in vivo, is necessary to guide the evaluation of treatment prognosis. In this study, GdPO 4 ·H 2 O nanoparticles were utilized as the dispersed phase to endow PLGA microcarriers with T1-weighted MRI contrast ability. By adjusting the dispersed phase of Gd compound nanoparticle in PLGA Matrix, a cross-comparison experiment between MRI contrast enhancement imaging and biocompatibility was conducted, and 0.8% Gd was found to be the most suitable rate for preparing nano-composites. Moreover, 0.8% Gd/PLGA microcarriers were suspension-cultured with stem cells in vitro and implanted into traumatic brain injury rats in vivo. Excellent cytocompatibility and enhancement of the T1 phase of MRI were confirmed. This work created a T1-weighted MRI contrast-enhanced microcarrier, which provides a clinical noninvasive tracing cell scaffold for brain tissue engineering.

To noninvasively monitor the effect of gene therapy and achieve an optimal therapeutic effect, liposomes encapsulated polyethylenimine (PEI)-coated superparamagnetic iron oxide nanoparticles (SPION) with dual functions of MRI diagnosis and gene therapy were prepared. SPION was synthesized via co-precipitation, and then modified with PEI via thiourea reaction. The liposomes encapsulating PEI-SPION (LP-PEI-SPION) were prepared by ethanol injection. Fourier transform infrared spectra confirmed that PEI was successfully modified onto SPION, and thermogravimetric analysis indicated that the PEI content was about 17.1%. The LP-PEI-SPION/DNA had a small particle size of 253.07 ± 0.90 nm. LP-PEI-SPION/DNA had low cytotoxicity with more than 80% of the cell survival rates and high transfection efficiency compared with Lipofectamine® 2000/DNA. Additionally, it also showed good MRI effect on three cell lines. The liposomes encapsulating PEI-SPION (lipopolyplexes) have been successfully prepared as MRI contrast agents and gene delivery vectors, which may have great theoretical research significance and clinical potentials.Abbreviations: PEI, polyethylenimine; SPION, superparamagnetic iron oxide nanoparticles; LP-PEI-SPION, liposomes encapsulating PEI-SPION; MTT, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide; ICP-MS, inductively coupled plasma mass spectrometry; XRD, X-ray diffraction; TEM, transmission electron microscope; TGA, thermogravimetric analysis; DOTAP, 1,2-dioleoyl-3-trimethylammonium-propane; DOPE, 1,2-dioleoyl-sn-glycero-3-phosphoethanolamine; Chol, cholesterol.

This project aims to construct cRGD functionalized mesoporous silica nanoparticles and cRGD modified mesoporous silica nanoparticles for the diagnosis and treatment of tumors, providing new ideas for targeted therapy of tumors. The mesoporous silica nanoparticles were doped with gadolinium in situ to provide excellent imaging; cRGD was coupled on the particle surface to confer particle targeting; and hyaluronic acid was loaded onto the particles by electrostatic adsorption, thereby improving the biocompatibility of the particles and prolonging their in vivo circulation time.Taking pancreatic cancer as a model, we studied its targeting ability to pancreatic cancer and its phagocytosis to cancer cells; Using methods such as cell growth experiments and flow cytometry, the anti-cancer effect and pro apoptotic effect of the system were studied. In vivo distribution, tumor targeting and therapeutic efficacy of nanoparticles evaluated in a mouse model of pancreatic cancer with loaded tumors.Evaluate the bioavailability and enrichment of nanoparticles in tumor tissue using MRI technology. Evaluate the therapeutic effect and safety through changes in tumor volume, histopathological examination, and prognosis. Characterization of the synthesis results proved that cRGD-HA-DOX-Gd 2 O 3 @MSN (cHDG@MSN) was successfully synthesized with a particle size of 230.83 ± 12.36 nm.In vitro drug release experiments of DOX were carried out at different pH values (5.5 and 7.4), where the release was only up to 22.65% at pH 7.4, whereas DOX release was increased up to 78.75% at pH = 5.5.The results confirm the pH responsiveness of this nanocarrier platform. The results of cytotoxicity studies showed that cHDG@MSN itself is not cytotoxic. Biosafety evaluation and hemolysis test results confirmed that the probe is highly biocompatible.Notably, Gd 3+ significantly enhanced the T 1 contrast of the system according to MR imaging results. The apoptosis rates of SW1990 cells treated with PBS, DOX and cHDG@MSN in flow cytometry were 13.97%, 18.38% and 29.02%, respectively, demonstrating the effectiveness of the nanoprobes at the cellular level. Animal experiments demonstrated the effectiveness of nanoprobes at the pathological level and imaging level.Cells and animals demonstrated that cHDG@MSN effectively inhibited the proliferation of pancreatic cancer cells. This research further verified the pH sensitivity of the constructed compound drug delivery system to achieve accurate diagnosis and treatment of pancreatic cancer tumor cells.

Diabetes mellitus (DM) causes skeletal muscle damage. Energy metabolism alterations in diabetic muscles remain to be elucidated. Chemical exchange saturation transfer (CEST) MRI is increasingly adopted in imaging muscle energy metabolism. This study aimed to investigate energy metabolism alterations in diabetic muscles using CEST MRI. Six adult New Zealand rabbits with alloxan-induced DM were scanned on a 5 T MR imager before and 2, 4, 6, and 8 weeks after the DM model induction, with six normal rabbits as controls. T 2 -weighted imaging, B 1 field mapping, and CEST imaging under four B 1 amplitudes of 0.75, 1.0, 1.25, and 1.5 μT were performed, respectively. Blood glucose level was measured before each MRI experiment. Inverse magnetization transfer ratio analysis was adopted to quantify CEST effects of two energy metabolites of creatine (Cr) and phosphocreatine (PCr), from which their ratio was calculated. B 1 inhomogeneity was corrected using a smoothing spline interpolation algorithm. pH-sensitive CrCEST ratiometric was measured by dividing CrCEST under B 1 of 1.25 μT with that under 0.75 μT. Paired student's t -test and one-way ANOVA were conducted with p < 0.05 considered statistically significant. Blood glucose level measurements confirmed the successfully induced DM model. The controls showed no significant changes in Cr- and PCr-weighted CEST signals throughout the study. In contrast, a significant increase of Cr-weighted CEST signals started in 2 weeks ( p = 0.049) and became more pronounced in Week 6 ( p = 0.045) and Week 8 ( p = 0.002) after the DM model induction, whereas PCr-weighted CEST signals remained constant among multiple time points, leading to significantly reduced PCr/Cr in Week 6 ( p = 0.047) and Week 8 ( p = 0.011). Meanwhile, CrCEST ratio did not show significant alterations ( p > 0.05). The significantly increased Cr-weighted CEST and reduced PCr/Cr with time implied disrupted energy metabolism in diabetic muscles, providing supplementary information for better understanding of DM-related muscle impairments from the metabolic level.

A type of novel macromolecular colloid was prepared from gadolinium-based poly( l -succinimide) and gold nanoparticles (GNPs) with Au S covalent bonds. The colloid displayed improved stability in aqueous media and regular arrays in partial region. Moreover, these materials enhanced the contrast 9 times more than that of the corresponding uncoated Au compound when the Au mass content was only 0.15%. Therefore, these materials might have potential as dual-modality contrast agents for magnetic resonance imaging (MRI) and computed tomography (CT).

Development of imaging methods that can detect target molecules will be of great significance for non-invasive molecular diagnosis. Vascular endothelial growth factor receptor-2 (VEGFR-2) is a broadly expressed target, and VEGFR-2-specific agents have been used in biological therapy.
In this study, amine-reactive coupling was used to label the polypeptide VEGF125–136 with a Gd(DOTA) complex to create a magnetic resonance contrast agent, VEGF125–136-Gd, which binds to VEGFR-2. Using T1-weighted magnetic resonance imaging, we explored the tumor cell-targeting
ability of VEGF125–136-Gd and its enhancement of T1-weighted image intensity using human hepatoma (HepG2) cells and found that the signal-to-noise ratio (SNR) increased commensurately with the concentration of VEGF125–136-Gd. At a concentration of 2 mM, the SNR produced by VEGF125–136-Gd
was 5.4-fold higher than that produced by a Gd-labelled non-targeting polypeptide (NTPP-Gd) control. We also evaluated the tumor-targeting efficiency of VEGF125–136-Gd in nude mice injected with human hepatoma (HepG2) cells. Preliminary in vivo imaging of VEGFR-2 in tumor tissue
revealed a targeting peak 60 min post administration of VEGF125–136-Gd, the intensity of which was 23% higher than the SNR of NTPP-Gd at the same time point. Our findings may help lay the foundation for clinical applications of VEGF125–136-Gd in targeted imaging.

Objectives: Immune checkpoint inhibitors (ICIs) have demonstrated potential in inhibiting the growth of malignant pleural mesothelioma (MPM), and their efficacy is associated with the expression of programmed death-ligand 1(PD-L1). This study evaluated a PD-L1-targeted nanoprobe for detecting PD-L1 expression in a nude mouse model of malignant pleural mesothelioma (MPM).Methods: A PD-L1-binding peptide (WL-12) was conjugated with superparamagnetic iron oxide nanoparticles (SPIONs) to create the nanoprobe WL-12@Fe₃O₄. The nanoprobe’s stability, biotoxicity, targeting ability, and in vivo magnetic resonance (MR) imaging effects were assessed and compared to non-targeted Fe₃O₄ nanoparticles. ΔT2 values and PD-L1 expression were measured in H226 and MSTO-211H tumor tissues over 4 weeks to analyze correlations. Results: The WL-12@Fe₃O₄ nanoprobe demonstrated uniform distribution and a spherical shape, with a larger size (43.82 nm) and lower surface potential (−9.34 ± 0.54 mV) compared to Fe₃O₄ (32.67 nm, −20.20 ± 0.88 mV, P < 0.05). The XPS and FT-IR analysis results indicate the successful coupling of WL-12 with Fe3O4. It was well dispersed in serum and saline and showed no cytotoxicity or organ damage in vivo. The probe selectively accumulated in PD-L1-expressing MPM cells, especially MSTO-211H, and exhibited significantly higher uptake in high PD-L1-expressing H460 cells (930.22 ± 11.75 ng/mL) compared to low PD-L1-expressing A549 cells (254.89 ± 17.33 ng/mL, P < 0.05). Tumor iron levels in the WL-12@Fe₃O₄ group were significantly elevated (141.02 ± 17.33 μg/g) compared to controls (36.43 ± 3.56 μg/g, P < 0.05), with no significant differences in other organs (P > 0.05). The T2 values of H226 and MSTO-211H tumors decreased after probe injection, with ΔT2 values significantly higher in the targeted group than the nontargeted group (P < 0.05). ΔT2 values increased over 4 weeks, correlating strongly with PD-L1 expression (P < 0.05).Conclusion: The PD-L1-targeted nanoprobe with MRI is a promising tool for noninvasive, real-time assessment of PD-L1 expression in MPM.
