Plant-based essential oils have been considered to be used as fish anesthetics and as suitable alternatives to synthetic anesthetics. In this study, we evaluated the effect of Cinnamomum camphora var. linaloofera Fujita essential oil (CFEO), MS-222 and linalool on anesthetic induction and recovery of spotted seabass, Lateolabrax maculatus , and verified their sedative effects during simulated transportation. A total of 198 live spotted seabass samples were subjected to various concentrations of MS-222, CFEO, and linalool: 10, 20, 30, 50, 100, 150, and 200 mg L −1 , individually, to evaluate the anesthetic induction time. MS-222, CFEO, and linalool all showed a deep sedation in spotted seabass at 20 mg L −1 . The minimum concentration required to achieve profound anesthesia in spotted seabass was found to be 50 mg L −1 MS-222, 100 mg L −1 CFEO, and 50 mg L −1 linalool, respectively. In addition, 120 healthy spotted seabass (400 g ± 3.5 g) were randomly divided into four groups, which were tranquilized by 20 mg L −1 CFEO, linalool, and MS-222, and the control fish were kept in clean seawater without anesthetics and essential oils. Then, the spotted seabass was simulated to be transported in a vibrating conveyor under 100 rpm at 12 °C for 24 h. The results showed that the fish transported with MS-222 and CFEO showed lower expressions in cytokine (tumor necrosis factor-α, interleukin-6, and interleukin-1 beta) compared to fish transported with linalool. Sedation with linalool reduced oxidative stress (higher superoxide dismutase, catalase, and glutathione peroxidase activities) in transport compared to the control but did not prevent changes in the activities of heat shock protein. In general, the efficacy of CFEO in anesthetizing and sedating spotted seabass and mitigating transport stress was as good as or better than that of MS-222, demonstrating it has the potential to be used as a new and natural sedation for spotted seabass during transport.

Background/Objectives: MS-222 is a commonly used anesthetic for fish. Research on the anesthetic mechanism of MS-222 is scarce, especially in largemouth bass. Therefore, this study investigated the tissue-specific transcriptomic and metabolomic effects of MS-222 anesthesia on largemouth bass (Micropterus salmoides). Methods: Experimental groups exposed to 40 mg/L MS-222 for 12 h were compared with untreated controls, and then transcriptomic and metabolomic analyses were performed on gill and liver samples. Results: Gill tissues exhibited 3252 differentially expressed genes (DEGs; 2309 upregulated and 943 downregulated) enriched in cardiac muscle contraction, cytoskeletal regulation, glycolysis, and toll-like receptor pathways for anesthetic adaptation. In contrast, liver tissues showed fewer DEGs (1140; 654 upregulated and 486 downregulated) primarily linked to metabolic network reorganization such as endoplasmic reticulum protein processing, PPAR signaling, and ribosome biogenesis. Metabolomic profiling demonstrated inverse patterns, with 173 differential metabolites in gills versus 297 in liver samples. Methyl nicotinate and N-acetyl-L-phenylalanine were the most significantly upregulated in the gill and liver samples. Metabolic pathway enrichment analysis revealed that MS-222-induced differential metabolites in the gill and liver of largemouth bass were predominantly associated with pathways involved in amino acid, fatty acid, phenylalanine, and nucleotide metabolism. Conclusions: These findings reveal that MS-222 anesthesia triggers organ-specific physiological adaptations through the differential regulation of metabolic and immune pathways, which provide multi-omics insights into the mechanistic basis of anesthetic responses in fish, highlighting distinct tissue strategies for managing chemical stress.

Anaesthetics are widely used in aquaculture as a method of stress relief. However, few studies have reported the influence of anaesthetics on the immune system of fish. Therefore, in the present study, we selected two different anaesthetics, MS-222 (100 mg/L) and eugenol (20 mg/L), to anaesthetize crucian carp and analyse the transcriptome. It was found that over 137 million high-quality reads were generated and de novo assembled into a final set of 97,556 unigenes. A total of eight differentially expressed genes (DEGs) related to the immune response were co-expressed in the two anaesthetic groups. GO and the KEGG revealed that these genes functioned primarily to enrich antigen processing and presentation pathways, including MHCI , MHCIα , p-MHCIIα , P-MHCIIα , CD74 , MHCII antigen and MHCII antigenα . Moreover, LRRFIP2 was found be associated with the immune response. Consistent with the transcriptome findings, qPCR verified the changes in the relative level of expression of these genes. Furthermore, long-term exposure to low concentrations of MS-222 (30 mg/L) and eugenol (8 mg/L) showed an impact on these immune genes. In conclusion, anaesthetics used in high or low concentrations for treatment and transport could affect the immune system in fish species. While eugenol was associated with an earlier activation of immune gene expression, MS-222 exhibited more significant effects on the immune response. These findings improve the understanding of the mechanisms of anaesthetics on immune damage and will be of great value for future studies involving anaesthetic selection and treatment for fish.
