Exonuclease I–assisted electrochemical detection of ofloxacin enhanced with MXene/AuNP accelerator

Zhang Yan,Yan Yuting,Huang Yue

An innovative electrochemical biosensor is proposed for the sensitive detection of ofloxacin (OFL) by integrating exonuclease I (Exo I)–assisted recognition-transduction strategy into the MXene/AuNP-enhanced electrochemical platform. Through ingenious design, the presence of OFL triggers the competitive displacement reaction and enables Exo I to hydrolyze the released single-stranded DNA probe containing the sequence of Pb 2+ -DNAzyme, protecting the signal probes on the electrode surface from DNAzyme cleavage, which eventually brings about a remarkable electrochemical signal positively related to the concentration of OFL. MXene/AuNPs with excellent conductivity, large specific surface area, and good chemical stability are employed as the electrode substrate to efficiently accelerate charge transfer and increase the loading capacity for signal probes, leading to the enhancement of signal output and endowing the biosensor with high sensitivity, rapid response, and fine robustness. Taking advantage of OFL recognition–initiated Exo I hydrolysis to DNAzyme, the electrochemical platform with “signal-off/on” mode can detect OFL with good specificity and reduced background interference. Benefiting from the synergistic effect of the involved sensing elements, the developed electrochemical biosensor achieves a low detection limit of 0.47 pg mL −1 with a broad linear range and exhibits satisfactory detection performance in real food samples, indicating potential practical applicability in food safety monitoring. MXene–AuNCs-based electrochemical aptasensor for ultrasensitive detection of neuron-specific enolase in early lung cancer diagnosis

Xuan Weng,Guanbo Wang,Hang Zhang,Hai Jiang

This study presents   a titanium carbide MXene–gold nanocomposites (MXene–AuNCs)-based aptasensor for the sensitive detection of neuron-specific enolase (NSE). Laser-induced graphene electrodes (LIGEs) were fabricated using polyimide film as the three-electrode electrochemical sensing system. Specifically, MXene–AuNCs were immobilized on the working electrode to enhance both the electronic conduction and the immobilization capacity of the NSE aptamers (Apt). As a labeling material, MXene–AuNCs–Apt facilitated signal amplification, enabling highly sensitive detection. Under optimized conditions, this aptasensor exhibited an ultralow detection limit of 0.13 pg/mL for NSE, a wide detection range from 0.01 ng/mL to 100 ng/mL along with satisfactory reproducibility,   repeatability, specificity and stability. Furthermore, the proposed aptasensor was successfully applied to measure NSE levels in serum samples. This work provides an efficient strategy for the fabrication of high-performance electrochemical biosensors in the preliminary   estimates and diagnosis of lung cancer at early stages.

Metal-mediated Fe3O4@polydopamine-aptamer capture nanoprobe coupling multifunctional MXene@Au@Pt nanozyme for direct and portable photothermal analysis of circulating breast cancer cells

Limin Yang,Heng Guo,Ting Hou,Jingang Zhang,Feng Li

Breast cancer (BC) is the most common cancer in the world and circulating tumor cells (CTCs) are reliable biomarkers for early breast cancer diagnosis in a non-invasive manner. However, effective isolation and sensitive detection of BC-CTCs by portable devices in human blood samples are extremely challenging. Herein, we proposed a highly sensitive and portable photothermal cytosensor for direct capture and quantification of BC-CTCs. To achieve efficient isolation of BC-CTCs, aptamer functionalized Fe 3 O 4 @PDA nanoprobe was facilely prepared through Ca 2+ -mediated DNA adsorption. To further detect the captured BC-CTCs with high sensitivity, multifunctional two-dimensional Ti 3 C 2 @ [email protected] nanozyme was synthesized, which not only possessed superior photothermal effect but also exhibited high peroxidase-like activity for catalyzing 3,3′,5,5′-tetramethylbenzidine (TMB) to produce TMB oxide (oxTMB) with a strong photothermal characteristic, combining with Ti 3 C 2 @ [email protected] to synergistically amplify the temperature signal. Moreover, numerous Ti 3 C 2 @ [email protected] nanocomposites would be selectively attained on the BC-CTCs surface through multi-aptamer recognition and binding strategy, which further enhanced the specificity and facilitated signal amplification . Therefore, direct separation and highly sensitive detection of BC-CTCs was successfully achieved in human blood samples. More significantly, the controlled release of the captured BC-CTCs without affecting cell viability could be straightforwardly realized by a simple strand displacement reaction. Thus, with the distinct features of portability, high sensitivity, and easy operation, the current method holds great promise for early diagnosis of breast cancer.

Electrochemiluminescence biosensor for determination of lead(II) ions using signal amplification by Au@SiO2 and tripropylamine-endonuclease assisted cycling process

Zhai Hongguo,Wang Yue,Yin Jiaqi,Zhang Yuhao,Guo Qi,Sun Xia,Guo Yemin,Yang Qingqing,Li Falan,Zhang Yanyan

MXene@Au as the base and Au@SiO 2 as signal amplification factor were used for constructing an ultrasensitive “on–off” electrochemiluminescence (ECL) biosensor for the detection of Pb 2+ in water. The use of MXene@Au composite provided a good interface environment for the loading of tris(2,2-bipyridyl)ruthenium(II) (Ru(bpy) 3 2+ ) on the electrode. Based on resonance energy transfer, the Au (core) SiO 2 (shell) (Au@SiO 2 ) nanoparticles stimulate electron transport and promote tripropylamine (TPrA) oxidation. The luminescence effect of Au@SiO 2 was five times that of AuNPs and SiO 2 nanomaterials alone, and the ECL intensity was greatly improved. In addition, Pb 2+ activated the aptamer to exert its endonuclease activity, which realized the signal cycle amplification in the process of Pb 2+ detection. When Pb 2+ was added, the ECL signal weakened, and the Pb 2+ concentration was detected according to the decreased ECL intensity. Under optimized experimental conditions, this aptamer sensor for Pb 2+ has a wide detection range (0.1 to 1 × 10 6  ng L −1 ) and a low detection limit (0.059 ng L −1 ). The relative standard deviation (RSD) of the sensor is 0.39–0.99%, and the recovery of spiked standard is between 90.00 and 125.70%. The sensor shows good selectivity and high sensitivity in actual water sample analysis. This signal amplification strategy possibly provides a new method for the detection of other heavy metal ions and small molecules. Graphical abstract

Dual-model photoelectrochemical biosensor via DNAzyme walker integrated nanoprobe for ultrasensitive ratiometric detection of microRNA-155

Liang Tian,Jiyuan Zhang,Yanci Zhang,Huiling Fan,Chun Liu,Yihong Wang,Jiansheng Cui

Ultrasensitive detection for low concentration and abnormal expression of microRNA-155 (miR-155) in serum (fM level) at the early stage of cancer still exist great challenges. Therefore, a dual-model photoelectrochemical biosensor was developed based on label-free trimetallic nanoprobe and Mg 2+ dependent DNAzyme driven DNA walker to generate ratiometric dual-signal output for sensitive biosensing of miR-155 from breast cancer cells (MCF-7) and cervical cancer cells (HeLa). In signal amplification strategy, PtCo@Prussian Blue nanozyme was selected as a signal nanoprobe with high catalytic activity and large specific surface area, while two dimensional MXene/AuNPs as surface modification material was applied to improve the active sites of the electrode. With the presence of the target, the DNAzyme assisted DNA walker process was initiated and abundant signal nanoprobe was introduced to the electrode surface , producing high current response and weak ultraviolet-visible spectrophotometry (UV–vis) response. Hence, the biosensor for miR-155 shows a low limit of detection of 1.2 fM from the linear range of 5 fM to 5 nM, exhibiting excellent stability and reproducibility. Moreover, the proposed biosensor demonstrates superior performance for miR-155 in complex human serum samples and tumor cell lysates, presenting great application potential for early-stage clinical diagnosis.

Triple-enhanced Raman scattering sensors from flexible MXene/Au nanocubes platform via attenuating the coffee ring effect

Xin Liu,Alei Dang,Tiehu Li,Tung-Chun Lee,Yiting Sun,Yuhui Liu,Fei Ye,Shuze Ma,Yong Yang,Weibin Deng

Developing substrates that combine sensitivity and signal stability is a major challenge in surface enhanced Raman scattering (SERS) research. Herein, we present a flexible triple-enhanced Raman Scattering MXene/Au nanocubes (AuNCs) sensor fabricated by selective filtration of Ti 3 C 2 T x MXene/AuNCs hybrid on the Ti 3 C 2 T x MXene membrane and subsequent treatment with 1H,1H,2H,2H-perfluoro-octyltriethoxysilane (FOTS). The resultant superhydrophobic MXene/AuNCs-FOTS membrane not only provides the SERS substrate with environmental stability, but also imparts analyte enrichment to enhance the sensitivity (LOD = 1 × 10 −14  M) and reliability (RSD = 6.41%) for Rhodamine 6G (R6G) molecules owing to the attenuation of the coffee ring effect. Moreover, the triple enhancement mechanism of combining plasmonic coupling enhancement from plasmonic coupling (EM) of nearby AuNCs at lateral and longitudinal direction of MXene/AuNCs-FOTS membrane, charge transfer (CT) from Ti 3 C 2 T x MXene and target molecules and analyte enrichment function provides the substrate with excellent SERS performance (EF = 3.19 × 10 9 ), and allows efficient quantification of biomarkers in urine. This work could provide new insights into MXenes as building blocks for high-performance substrates and fill existing gaps in SERS techniques.

PEC-SERS Dual-Mode Detection of Foodborne Pathogens Based on Binding-Induced DNA Walker and C3N4/MXene-Au NPs Accelerator

Fanglei Liu,Jiayi Zhao,Xinyu Liu,Xi Zhen,Qiumei Feng,Yingqiu Gu,Guohai Yang,Lulu Qu,Jun-Jie Zhu

In this paper, a photoelectrochemical (PEC)-surface-enhanced Raman scattering (SERS) dual-mode biosensor is constructed coupled with a dual-recognition binding-induced DNA walker with a carbon nitride nanosheet (C3N4)/MXene-gold nanoparticles (C/M-Au NPs) accelerator, which is reliable and capable for sensitive and accurate detection of Staphylococcus aureus (S. aureus). Initially, a photoactive heterostructure is formed by combining C3N4 and MXene via a simple electrostatic self-assembly as they possess well-matched band-edge energy levels. Subsequently, in situ growth of gold nanoparticles on the formed surface results in better PEC performance and SERS activity, because of the synergistic effects of surface plasmon resonance and Schottky barrier. Furthermore, a three-dimensional, bipedal, and dual-recognition binding-induced DNA walker is introduced with the formation of Pb2+-dependent DNAzyme. In the presence of S. aureus, a significant quantity of intermediate DNA (I-DNA) is generated, which can open the hairpin structure of Methylene Blue-tagged hairpin DNA (H-MB) on the electrode surface, thereby enabling the switch of signals for the quantitative determination of S. aureus. The constructed PEC-SERS dual-mode biosensor that can be mutually verified under one reaction effectively addresses the problem of the low detection accuracy of traditional sensors. Experimental results revealed that the effective combination of PEC and SERS is achieved for amplification detection of S. aureus with a detection range of 5–108 CFU/mL (PEC) and 10–108 CFU/mL (SERS), and a detection of limit of 0.70 CFU/mL (PEC) and 1.35 CFU/mL (SERS), respectively. Therefore, this study offers a novel and effective dual-mode sensing strategy, which has important implications for bioanalysis and health monitoring.
