Screening and evaluating honokiol from Magnolia officinalis against Nocardia seriolae infection in largemouth bass (Micropterus Salmoides)

Xin-Yuan Jiang,Zong-Yi Ren,Nian-Kun Zhang,Ke-Chen Yang,Gao-Xue Wang,Hai-Feng Jiang

Nocardiosis caused by Nocardia seriolae is a major threat to the aquaculture industry. Given that prolonged therapy administration can lead to a growth of antibiotic resistant strains, new antibacterial agents and alternative strategies are urgently needed. In this study, 80 medicinal plants were selected for antibacterial screening to obtain potent bioactive compounds against N. seriolae infection. The methanolic extracts of Magnolia officinalis exhibited the strongest antibacterial activity against N. seriolae with the minimal inhibitory concentration (MIC) of 12.5 μg/ml. Honokiol and magnolol as the main bioactive components of M. officinalis showed higher activity with the MIC value of 3.12 and 6.25 μg/ml, respectively. Sequentially, the evaluation of antibacterial activity of honokiol in vivo showed that honokiol had good biosafety, and could significantly reduce the bacterial load of nocardia-infected largemouth bass ( p < .001). Furthermore, the survival rate of nocardia-infected fish fed with 100 mg/kg honokiol was obviously improved ( p < .05). Collectively, these results suggest that medicinal plants represent a promising reservoir for discovering active components against Nocardia , and honokiol has great potential to be developed as therapeutic agents to control nocardiosis in aquaculture.

Nine phenylethanoid glycosides from Magnolia officinalis var. biloba fruits and their protective effects against free radical-induced oxidative damage

Ge Lanlan,Zhang Wenhui,Zhou Gao,Ma Bingxin,Mo Qigui,Chen Yuxin,Wang Youwei

To systematically study the chemical constituents in Magnolia officinalis var. biloba fruits, nine phenylethanoid glycosides were isolated by solvent extraction, silica gel, and preparative high-performance liquid chromatography (HPLC). Their structures were elucidated by 1D and 2D NMR analyses, including COSY, HMQC and HMBC correlations, and HPLC analysis of sugar residue. Nine phenylethanoid glycosides, namely, magnoloside I a ( 1 ), magnoloside I c ( 2 ), crassifolioside ( 3 ), magnoloside I b ( 4 ), magnoloside III a ( 5 ), magnoloside IV a ( 6 ), magnoloside II a ( 7 ), magnoloside II b ( 8 ) and magnoloside V a ( 9 ), were first isolated from the n -butanol fraction of Magnolia officinalis var. biloba fruits alcohol extract. Free radical scavenging activities of the nine phenylethanoid glycosides were assessed using the DPPH, ABTS, and superoxide anion radical scavenging assays. Simultaneously, protective effects of all compounds against free radical-induced oxidative damage were evaluated by two different kinds of mitochondrial damage model. The protective effects were assessed by mitochondrial swelling, the formations of malondialdehyde (MDA) and lipid hydroperoxide (LOOH), the activities of catalase (CAT), glutathione reductase (GR) and superoxide dismutase (SOD). All phenylethanoid glycosides showed significant protective effects.

Effect and mechanism of Magnolia officinalis in colorectal cancer: Multi-component-multi-target approach

He Zhang,Jun Tang,Huiliang Cao,Chenguang Wang,Chong Shen,Jun Liu

Ethnopharmacological relevance Colorectal cancer (CRC) is a prevalent malignant tumor of the digestive tract. Traditional Chinese medicine (TCM) has a long history of treating CRC, with advantages such as effectiveness, multi-target, multi-pathway, and minimal side effects. TCM Magnolia officinalis (M. officinalis) refers to the dried bark, root bark, and branch bark of either Magnolia officinalis Rehd.et Wils. or Magnolia officinalis Rehd.et Wils. var. biloba Rehd.et Wils . It is commonly utilized to alleviate the side effects of chemotherapy for CRC, owing to its anti-inflammatory and anti-tumor properties. However, current research primarily focuses on the individual components and does not take into consideration the characteristics of multi-component-multi-target action. Aim of the study Our aim is to study the new action characteristics of M. officinalis in the treatment of CRC. Materials and methods Utilizing network pharmacology to identify potential active ingredients, key targets, and main signaling pathways of M. officinalis for the treatment of CRC. The binding effect was further validated through molecular docking analysis. Furthermore, the aforementioned components were identified using liquid chromatography-mass spectrometry (LC-MS), and the cleavage pathways of the main components were analyzed. Subsequently, both in vitro and in vivo experiments were carried out to investigate the anti-CRC effect of the active ingredients of M. officinalis and its potential mechanism. Results Network pharmacology and Molecular docking identified 5 main active ingredients and 6 core targets of M. officinalis for the treatment of CRC. Then, LC-MS identified the active components of M. officinalis. At the same time, both in vitro and in vivo experiments have confirmed the ability of Eucalyptol (Euc) and Obovatol (Obo)to inhibit inflammation and tumor cell proliferation. The possible mechanism involved is that Euc and Obo counteract CRC by inhibiting the over-activation of NF-κBp65/JAK and Bcl-2/Caspase signaling pathways, respectively. They also play a role in the anti-CRC effect of M. officinalis. Conclusion Magnolol (MAG), Honokiol (HK), Euc, Obo, and Neohesperidin (NHP) in M. officinalis may be the pharmacological substance basis for its anti-cancer effect on CRC. The treatment of CRC with M. officinalis is characterized by its multi-component, multi-target, and multi-pathway approach. These findings provide a theoretical basis for further inspiring the clinical application of M. officinalis and the development of efficacy targets.

Fritillaria hupehensis cultivated under the canopy of Magnolia officinalis demonstrated superior anti-inflammatory and expectorant effects

Xiaoyue Wang,Yuanyuan Duan,Haiyan Wang,Rui Zhao,Fanfan Wang,Tao Tang,Qingfang Wang,Jingmao You

Background Fritillaria hupehensis is a medicinal plant used for treating cough and inflammation. Its cultivation under the canopy of Magnolia officinalis (U- F. hupehensis ) increases alkaloid content compared to traditionally field-grown cultivation (T- F. hupehensis ). Purpose This study aims to evaluate the superior therapeutic efficacy of U- F. hupehensis against cough and inflammation and to elucidate the role of its rhizosphere bacterial community in enhancing the accumulatio of key bioactive compounds. Methods The anti-inflammatory and expectorant efficacy of U- F. hupehensis was evaluated in animal models (ammonia-induced cough, phenol red excretion, and LPS-induced bronchitis). Network pharmacology and cellular experiments identified the key bioactive compounds. Soil physicochemical properties and 16S rRNA sequencing were used to analyze the rhizosphere bacterial community. The relationship between the rhizosphere microbiome and bioactive compound accumulation was analyzed using structural equation modeling. Results U- F. hupehensis exhibited significantly stronger anti-inflammatory and expectorant effects than T- F. hupehensis . Hupehenine, peimine, and sipeimine were validated as the primary active components responsible for this enhanced efficacy. Understory cultivation increased rhizosphere microbial diversity. A specific keystone microbial module (Module 1) was identified and shown to directly mediate the accumulation of the bioactive compounds. Conclusion The superior anti-inflammatory and expectorant efficacy of understory-cultivated F. hupehensis is driven by a marked increase in the key bioactive alkaloids (hupehenine, peimine, and sipeimine). Our study identifies a specific rhizosphere microbial community as the pivotal mediator of this enhanced alkaloid accumulation. These results provide a pharmacologically relevant mechanism that explains the differential therapeutic potential of F. hupehensis from distinct cultivation sources. Construction of an immobilised acetylcholinesterase column and its application in screening insecticidal constituents from Magnolia officinalis

Yong-Hao Ye,Cong Li,Jun Yang,Liang Ma,Yu Xiao,Jun Hu,Nasir Ahmed Rajput,Cong-Fen Gao,Ying-Ying Zhang,Ming-Hua Wang

BACKGROUND Application of a matrix-immobilised target enzyme for screening inhibitors is widely used in drug development, but there are few studies in insecticide discovery. In this paper, an economical and effective immobilised acetylcholinesterase (AChE) column was prepared using the sol–gel embedment method, which was further combined with high-performance liquid chromatography for screening the AChE inhibitors and insecticidal compounds from complex natural products. RESULTS AChE inhibitory constituents magnolol and honokiol were isolated from the ethanol extract of Magnolia officinalis , with IC 50 values of 0.069 and 0.057 mM respectively. In an in vivo bioassay, magnolol and honokiol showed insecticidal activity against Nilaparvata lugens , with LC 50 values of 0.324 and 0.137 mM, which are comparable with that of commonly used insecticide chlorpyrifos (0.233 mM). Moreover, molecular docking was carried out against a homology model of N. lugens AChE. The complexes showed that magnolol and honokiol placed themselves nicely into the active site of the enzyme and exhibited an interaction energy that was in accordance with our activity profile data. CONCLUSION These results demonstrate that magnolol and honokiol have great applied potential to be developed as natural insecticides, and an immobilised AChE column is very useful as a rapid screening tool for target enzymes towards potent inhibitors. © 2014 Society of Chemical Industry

Magnolia officinalis Rehder & E.H.Wilson. Bark Extract and Magnolol Alleviate Allergic Rhinitis via Modulating NF-κB/MAPK Signaling

Leyuan Huang,Xu Zhou,Guanfeng He,Haixin Li,Xiaoying Chen,Jingwen Xu,Lei Zhou

Magnolia officinalisRehder & E.H.Wilson. bark is famous as a traditional herbal medicine used in prescriptions for treating gastrointestinal discomfort, respiratory and inflammatory disorders. Magnolol, one of its principal bioactive constituents, exhibits potent anti-inflammatory and immunomodulatory properties. However, its therapeutic mechanisms in allergic rhinitis (AR) remain to be elucidated. In this study, the anti-allergic effects and molecular mechanisms ofM.officinalisbark aqueous extract (MOAE) and magnolol were investigated using an ovalbumin (OVA)-induced AR mouse model. Nasal symptoms, histopathological alterations, and serum inflammatory mediators, including histamine and immunoglobulins (IgE, IgG1, IgG2a), were evaluated to assess efficacy. Both MOAE and magnolol significantly alleviated nasal rubbing and sneezing, reduced eosinophil infiltration and mucus hypersecretion, and improved tissue morphology in nasal and lung sections. Moreover, treatment markedly decreased serum levels of histamine and OVA-specific antibodies. Integrative network pharmacology, RNA sequencing, and molecular docking analyses revealed 33 co-regulated target genes mainly involved in the NF-κB and MAPK signaling pathways, suggesting that modulation of these pathways underlies the observed anti-inflammatory effects. These findings demonstrate that MOAE and magnolol exert protective effects against AR through the regulation of key inflammatory signaling cascades. This study provides modern pharmacological evidence supporting the traditional use ofM.officinalisbark and highlights its potential as a natural therapeutic candidate for AR.

Antioxidant activity of the different polar solvent extracts of Magnolia officinalis leaves and purification of main active compounds

Tan Li-Hong,Zhang Dan,Yu Bao,Zhao Sheng-Ping,Cao Wei-Guo

The leaf of Magnolia officinalis is abundant in natural resources as a novel potential antioxidant and has great commercial value. In this study, the contents of total phenolics, total flavonoids, magnolol and honokiol in different solvent extracts (acetone, methanol and water) were measured. Meanwhile, the antioxidant activity of extracts from M. officinalis leaves were evaluated using 1,1-diphenyl-2-picrylhydrazyl free radical scavenging assay, 2,2-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt free radical scavenging assay, FRAP assays and β-carotene/linoleic acid system. In addition, the correlation between the antioxidant activity and the contents of the active compounds was analyzed. Experimental results indicated that three extracts exhibited antioxidant effects in a concentration-dependent manner, and acetone extracts had the highest antioxidant activity, next methanol and water. There was an intimate relativity between the contents of magnolol and honokiol in different solvent extracts and the antioxidant capability of them. So the magnolol and honokiol of M. officinalis leaves as a representative of the active compounds were extracted and purified by using the macroporous resin column. The purity of depurated sample could reach 82.6 %, which indicates the M. officinalis leaves have the potential to substitute the production of magnolol and honokiol in M. officinalis . The results of this study can provide theoretical and technical references for the reasonable use of the M. officinalis leaves and alleviate the shortage of M. officinalis resources.

Antimicrobial activity and mechanism of Magnolia officinalis root extract against methicillin-resistant Staphylococcus aureus based on mannose transporter

Jingwen Sun,Yu Xie,Zhiyuan Chen,Yipin Fan,Yunfeng Liu,Qi Gao,Jianqiang Li,Jingwen Bai,Yu Yang

Magnolia officinalis root extract was obtained by high-pressure-assisted extraction, and the yield of honokiol (HNK) was 86.97 ± 1.05 mg/g dry power. The purified extracts and HNK inhibited the growth of methicillin-resistant Staphylococcus aureus (MRSA). Assays for alkaline phosphatases , proteins, nucleic acids leakage, and electrical conductivity, along with scanning and transmission electron microscopy measurements, confirmed the damaging effects of HNK on the cell wall and membrane of MRSA. Intake of sugar, protein production, molecular docking , and dynamic simulations revealed that HNK interfered with the transport of mannose-based carbohydrates by binding to VAL196, ILE291, ALA293, PRO294, and MET214 on the mannose transporter (ManP) in the phosphotransferase system, and thereby inhibited the carbohydrate metabolism of MRSA. These results indicated that HNK prevented MRSA from using carbohydrates by cutting off its nutritional process, and ultimately caused bacterial rupture and lysis. Therefore, ManP may be a potential target for HNK to combat MRSA infections.

Comprehensive exploration of a traditional Chinese medicinal plant of Magnolia officinalis based on high-coverage mass spectrometry and multidimensional chemical-biological analysis

Wen-Yu Wang,Ya-Mei Song,Jia-Nuo Zhang,Ming-Yue Zhao,Wen-Han Pei,Hui Zhang,Hai-Bo Yin,Zhi-Li Xu,Gui-Zhong Xin,Ming Xie,Ting-Guo Kang,Yue-Hua Chen,Hui-Peng Song

Magnolia bark is a traditional Chinese medicine used for hypoglycaemia. With the widespread use of Magnolia bark, its resources are facing a serious shortage. To address this issue, a strategy based on high-coverage mass spectrometry (HCMS) and multidimensional chemical-biological analysis (MCBA) was proposed for the comprehensive exploration of Magnolia officinalis which is the main source of Magnolia bark. The strategy is divided into three main steps. In the first step, the stem bark, stem xylem, root bark, root xylem, leaf and rootlet of Magnolia officinalis were comprehensively analyzed using high-coverage mass spectrometry. In the second step, multivariate statistical analysis was used to explore the heterogeneity of the six parts and detect differential chemical components. In the third step, a combination of experimental screening and molecular docking was used to explore α-glucosidase inhibitors from Magnolia officinalis . Multidimensional chemical-biological analysis (MCBA) of Magnolia officinalis was achieved by combining the last two steps. Finally, a total of 103 compounds were identified from the whole plant of Magnolia officinalis . Differential components of stem bark, stem xylem, leaf, root bark, root xylem and rootlet were systematically revealed. A pair of positional isomers, namely magnolol and honokiol, were found to be α-glucosidase inhibitors. The activity of their combination is superior to that of each single compound, indicating that magnolol and honokiol are in a synergistic relationship. This strategy contributes to comprehensive exploitation of functional plants and effective alleviation of resource shortage. This study also provides a research paradigm for other similar traditional Chinese medicinal plants.

Deciphering the multi-target mechanism of Magnolia officinalis in irritable bowel syndrome with diarrhea: A combined network pharmacology, machine learning, and in vivo validation study

Xiran Wang,Yuli Wei,Mengze Wang,Yu Zhang,Longfei Liu,Xiaokai Ma,Li Gao,Haixia Ji

Background The bark of Magnolia officinalis Rehder & E. Wilson, used in TCM for abdominal distension, pain, and diarrhea, aligns with IBS-D symptoms. It is now supported by evidence for its multi-component, multi-target action. This study aims to elucidate the core bioactive components and their mechanisms against IBS-D. To achieve this, we employed an analytical strategy that uses machine learning to refine network pharmacology predictions, rather than relying on network pharmacology alone. Materials and methods This study integrates network pharmacology and machine learning to identify core therapeutic targets. The affinity of the components for these targets is validated through molecular docking studies. An IBS-D rat model was used to evaluate the effects of the core components (magnolol, honokiol, obovatol) on behavior (sucrose preference, open field), gastrointestinal motility, intestinal permeability, serum inflammatory cytokines (ELISA), and core target gene expression (qRT-PCR). Results Computational analyses identified five potential active components, including magnolol (Mag), honokiol (Hnk), and obovatol (Obo), and eight core targets (e.g., EGFR, MET) involved in PI3K-Akt and FoxO pathways. Molecular docking confirmed stable binding. In vivo, Hnk, Mag, and Obo ameliorated IBS-D-related anxiety-like behavior, intestinal hypermotility, and barrier dysfunction. They significantly downregulated EGFR and MET expression and reduced serum levels of IL-8, TNF-α, and MyD88. Notably, all treatments increased fecal short-chain fatty acids (SCFAs). Conclusion The core components of M. officinalis , represented by Hnk, Mag, and Obo, alleviate IBS-D likely through a multi-target mechanism involving suppression of EGFR/MET and subsequent PI3K-Akt signaling, leading to reduced inflammation, improved barrier function, and modulated gut-brain axis. This provides a mechanistic foundation for the traditional use of M. officinalis in treating gastrointestinal disorders.
