Identification and Characterization of Nematicidal Volatile Organic Compounds from Deep-Sea Virgibacillus dokdonensis MCCC 1A00493

Dian Huang,Chen Yu,Zongze Shao,Minmin Cai,Guangyu Li,Longyu Zheng,Ziniu Yu,Jibin Zhang

Root-knot nematode diseases cause severe yield and economic losses each year in global agricultural production.Virgibacillus dokdonensisMCCC 1A00493, a deep-sea bacterium, shows a significant nematicidal activity againstMeloidogyne incognitain vitro. However, information about the active substances ofV. dokdonensisMCCC 1A00493 is limited. In this study, volatile organic compounds (VOCs) fromV. dokdonensisMCCC 1A00493 were isolated and analyzed through solid-phase microextraction and gas chromatography–mass spectrometry. Four VOCs, namely, acetaldehyde, dimethyl disulfide, ethylbenzene, and 2-butanone, were identified, and their nematicidal activities were evaluated. The four VOCs had a variety of active modes onM. incognitajuveniles. Acetaldehyde had direct contact killing, fumigation, and attraction activities; dimethyl disulfide had direct contact killing and attraction activities; ethylbenzene had an attraction activity; and 2-butanone had a repellent activity. Only acetaldehyde had a fumigant activity to inhibit egg hatching. Combining this fumigant activity against eggs and juveniles could be an effective strategy to control the different developmental stages ofM. incognita. The combination of direct contact and attraction activities could also establish trapping and killing strategies against root-knot nematodes. Considering all nematicidal modes or strategies, we could useV. dokdonensisMCCC 1A00493 to set up an integrated strategy to control root-knot nematodes. Keywords:virgibacillus dokdonensis;Meloidogyne incognita;volatile organic compound;fumigant;attraction;repellent;integrated strategy

Gen-Qiang Chen,Yan-Fei Xia,Jin-Ming Yang,Zhi-Ping Che,Di Sun,Shen Li,Yue-E Tian,Sheng-Ming Liu,Jia Jiang,Xiao-Min Lin

Gramine can be intelligently and efficiently supplied with N, N-dimethylamino group and then reacted with the corresponding sulfonyl chlorides to synthesize N, N-dimethylarylsulfonamides. We herein designed and controlled synthesis of N, N-dimethylarylsulfonamide derivatives, and first reported the results of the nematicidal activity of 15 title compounds 3a-o against Meloidogyne incongnita in vitro, respectively. Among all of the title derivatives, compounds 3a, 3c, 3k, and 3o exhibited potent nematicidal activity with median lethal concentration (LC50) values ranging from 0.22 to 0.26 mg/L. Most noteworthy, N, N-dimethyl-4-methoxyphenylsulfonamide (3c) and N, N-dimethyl-8-quinolinesulfonamide (3o) showed the best promising and pronounced nematicidal activity, with LC50 values of 0.2381 and 0.2259 mg/L, respectively.

Sulfonamide modified chitosan oligosaccharide with high nematicidal activity against Meloidogyne incognita

Zhaoqian Fan,Kun Gao,Linsong Wang,Yukun Qin,Song Liu,Ronge Xing,Huahua Yu,Kecheng Li,Pengcheng Li

Chitosan oligosaccharide (COS) modification is a feasible way to develop novel green nematicides. This study involved the synthesis of various COS sulfonamide derivatives via hydroxylated protection and deprotection, which were then characterized using NMR, FTIR, MS, elemental analysis, XRD, and TG/DTG. In vitro experiments found that COS-alkyl sulfonamide derivatives ( S6 and S11 – S13 ) exhibited high mortality (>98 % at 1 mg/mL) against Meloidogyne incognita second-instar larvaes (J2s) among the derivatives. S6 can cause vacuole-like structures in the middle and tail regions of the nematode body and effectively inhibit egg hatching. In vivo tests have found that S6 has well control effects and low plant toxicity. Additionally, the structure-activity studies revealed that S6 with a high degree of substitution, a low molecular weight, and a sulfonyl bond on the amino group of the COS backbone exhibited increased nematicidal activity. The sulfonamide group is a potential active group for developing COS-based nematicides.

Benzothiazole kills Meloidogyne incognita quickly by inhibiting GST to trigger ROS bursting

Qiyi Zhu,Xiaofeng Zhu,Ning Yang,Yuxi Duan,Yuanyuan Wang,Xiaoyu Liu,Di Zhao,Haiyan Fan,Lijie Chen

Root-knot nematodes ( Meloidogyne incognita ) present a significant threat to global agriculture, and the development of multi-drug resistance in these nematodes exacerbates this problem. Benzothiazole, a heterocyclic compound has been reported as a potential nematicide, however, its mode of action is not fully understood. This study aims to elucidate the nematicidal mechanism of benzothiazole against M. incognita . In the toxicity assays, benzothiazole exhibited rapid and effective nematocidal activity, significantly compromising egg masses and inhibiting egg hatching while killing newly hatched second-stage juveniles (J2s) of M. incognita . Microscopic observations revealed that after 48 h of incubation, a marked reduction in protein and carbohydrate levels within the J2s was observed. Notably, benzothiazole at a concentration of 14 mmol/L significantly inhibits glutathione S-transferase (GST) enzyme activity, leading to the accumulation of reactive oxygen species (ROS), ultimately resulting in rapid nematode death. Molecular docking and dynamics simulations demonstrated that benzothiazole forms a stable complex with GST, thereby disrupting its antioxidant function. Furthermore, in pot experiments, benzothiazole effectively reduced the gall formations of M. incognita on tomato roots. Overall, this novel inhibitory mechanism of glutathione S-transferase (GST) differs from that of the neurotoxicant abamectin, which targets glutamate-gated chloride channels (GluCl). This mechanism holds significant promise for the development of environmentally friendly nematicides. It offers a potential solution to the growing problem of multidrug resistance in root-knot nematodes and could help mitigate the substantial economic losses caused by these pests in global agriculture.

Fluoroalkenyl-Grafted Chitosan Oligosaccharide Derivative: An Exploration for Control Nematode Meloidogyne Incognita

Zhaoqian Fan,Yukun Qin,Song Liu,Ronge Xing,Huahua Yu,Kecheng Li,Pengcheng Li

The exploration of novel, environmentally friendly, and efficient nematicides is essential, and modifying natural biomacromolecules is one feasible approach. In this study, 6-O-(trifluorobutenyl-oxadiazol)-chitosan oligosaccharide derivative was synthesized and characterized by FTIR, NMR, and TG/DTG. Its bioactivity and action mode against root-knot nematodeM. incognitawere estimated. The results show that the derivative shows high nematicidal activity against J2s, and egg hatching inhibitory activity at 1 mg/mL. The derivative may affect nematode ROS metabolism and further damage intestinal tissue to kill nematode. Meanwhile, by synergism with improving crop resistance, the derivative performed a high control effect on the nematode with low phytotoxicity. These findings suggested that chitosan oligosaccharide derivatives bearing fluoroalkenyl groups are promising green nematicides. Keywords:chitosan oligosaccharide;nematicide;Meloidogyne incognita;trifluorobutenyl group

Transcriptomic and physiological analysis of the effect of octanoic acid on Meloidogyne incognita

Jian-Yu Wang,Qiu-Yue Li,Li Ren,Cheng Guo,Jian-Ping Qu,Zheng Gao,Hui-Fang Wang,Qian Zhang,Bo Zhou

Root knot nematodes are the most devastating root pathogens, causing severe damage and serious economic losses to agriculture worldwide. Octanoic acid has been reported as one of the nematicides , and its mode of action is not fully understood. The main objective of this study was to elucidate the effect of octanoic acid on Meloidogyne incognita by transcriptomic analysis combined with physiological and biochemical assays. In the toxicity assays with octanoic acid, the threshold concentration with nematicidal activity and the maximum concentration to which nematodes could respond were 0.03 μL/mL and 0.08 μL/mL respectively. Microscopic observation combined with protein and carbohydrates assays confirmed that the structure of the second-stage juveniles (J2s) was severely disrupted after 72 h of immersion in octanoic acid. Transcriptome analysis has shown that octanoic acid can interfere with the nematode energy metabolism, lifespan and signaling. Although the effects are multifaceted, the findings strongly point to the cuticle, lysosomes , and extracellular regions and spaces as the primary targets for octanoic acid. In addition, nematodes can withstand the negative effects of low concentration of octanoic acid to some extent by up-regulating the defense enzyme system and heterologous metabolic pathways. These findings will help us to explore the nematicidal mechanism of octanoic acid and provide important target genes for the development of new nematicides in the future.

Biocontrol efficacy of Bacillus velezensis and its major bioactive metabolites against Meloidogyne incognita

Yongpan Xu,Ruiyan Li,Chaoyue Hu,Jian Zhang,Zihan Li,Meiying Liu,Gangyong Zhao,Zhaopeng Wei,Jianqing Ma,Yungen Ran,Dandan Cao

The southern root-knot nematode ( Meloidogyne incognita ) is one of the predominant pathogenic nematode species that severely threatens economically important crops. The application of biocontrol microorganisms or their metabolites represents an effective and environmentally friendly strategy for the management of root-knot nematodes. This study focused on the nematicidal activity of two Bacillus velezensis strains, RKN1111 and RKN2222, against M. incognita second-stage juveniles (J2s), and the identification of bioactive metabolites associated with their differential nematicidal effects. In vitro assays showed that, under comparable viable biomass conditions, the corrected mortality of J2s caused by RKN1111 (95.6%) was significantly higher than that caused by RKN2222 (46.2%). The LC 50 values of RKN1111 and RKN2222 were 0.58 × 10 8 and 5.86 × 10 8  CFU/mL, respectively. Metabolomic analysis revealed that, compared with RKN2222, RKN1111 exhibited 150 significantly upregulated differential metabolites. Among them, glycylvaline and N-cyclohexylformamide exhibited strong nematicidal activity against J2s, with corrected mortality rates of 88.7% and 81.7%, respectively, after 24 h treatment at 100-fold dilution of their saturated solutions. Moreover, the concentrations of glycylvaline (154 μg/mL) and N-cyclohexylformamide (4.5 μg/mL) in the original fermentation supernatant of RKN1111 were significantly higher than those in RKN2222 (43 μg/mL and 2.8 μg/mL, respectively). Further concentration-gradient bioassays combined with quantitative analysis showed that the concentration of N-cyclohexylformamide in the RKN1111 supernatant was far below the concentration threshold required for effective nematicidal activity, indicating that its direct contribution to the nematicidal activity of the supernatant was limited. In contrast, glycylvaline still exhibited marked nematicidal activity at concentrations close to those measured in the RKN1111 supernatant by HPLC. However, glycylvaline alone could not fully explain the high nematicidal activity of the RKN1111 supernatant, suggesting that it may be one of the major candidate contributors to the nematicidal activity of RKN1111 and that synergistic effects among multiple metabolites may also be involved. Overall, glycylvaline was identified as the strongest candidate bioactive metabolite screened in this study. These results provide a theoretical basis for the further development of RKN1111 as a potential biocontrol strain, as well as for further investigation of glycylvaline and the high-concentration nematicidal activity of N-cyclohexylformamide.

Diterpenoids From Erythroxylum sinense: Bidirectional Behavioral Regulation of Root-Knot Nematode Meloidogyne incognita

Han Zhang,Yu-Lian Lv,Jing Yang,Jian-Ping Huang,Dale Guo,Yong-Jiang Wang,Sheng-Xiong Huang

Root-knot nematodes (RKNs) (e.g., Meloidogyne incognita ) pose a major threat to global agriculture, driving the need for sustainable control of nematodes. Current strategies mainly rely on synthetic nematicides, but their applications were limited by concerns over environmental toxicity and public health risks. In this study, two new diterpenoids, sinensins A‒B ( 1 ‒ 2 ), together with eight analogues ( 3 ‒ 10 ), were isolated from the endemic Chinese plant Erythroxylum sinense Y. C. Wu. Four selected compounds ( 4 , 6 , 7 , and 10 ) were evaluated for their chemotaxis of M. incognita second-stage larvae (J2s) and their nematicidal activity. Among them, compound 10 with a cyclopropyl moiety exhibited a bidirectional regulation of J2s parasitic behavior: it attracted J2s at lower doses (10‒100 µg/mL), while repelled J2s at higher doses (160 and 200 µg/mL). These results highlighted that diterpenoid 10 from E. sinense represents a promising candidate for an eco-friendly “attract-kill” microsphere system to control nematodes. Graphical A special diterpenoid (compound 10 ) with a unique cyclopropane ring was isolated from Erythroxylum sinense . It exhibits bidirectional chemotactic activity toward Meloidogyne incognita J2s, acting as an attractant at low doses (10–100 µg/mL) and a repellent at higher doses (160 and 200 µg/mL). This discovery highlights its potential application in “attract-kill” strategies for sustainable nematode management.

Identification of Nematicidal Constituents of Notopterygium incisum Rhizomes against Bursaphelenchus xylophilus and Meloidogyne incognita

Gai Liu,Daowan Lai,Qi Zhi Liu,Ligang Zhou,Zhi Long Liu

During a screening program for new agrochemicals from Chinese medicinal herbs, the ethanol extract ofNotopterygium incisumrhizomes was found to possess strong nematicidal activity against the two species of nematodes,Bursaphelenchus xylophilusandMeloidogyne incognita. Based on bioactivity-guided fractionation, the four constituents were isolated from the ethanol extract and identified as columbianetin, falcarindiol, falcarinol, and isoimperatorin. Among the four isolated constituents, two acetylenic compounds, falcarindiol and falcarinol (2.20–12.60 μg/mL and 1.06–4.96 μg/mL, respectively) exhibited stronger nematicidal activity than two furanocoumarins, columbianetin, and isoimperatorin (21.83–103.44 μg/mL and 17.21–30.91 μg/mL, respectively) against the two species of nematodes,B. xylophilusandM. incognita. The four isolated constituents also displayed phototoxic activity against the nematodes. The results indicate that the ethanol extract ofN.incisumand its four isolated constituents have potential for development into natural nematicides for control of plant-parasitic nematodes.

Efficacy of 2-undecanol produced by Paenibacillus polymyxa KM2501-1 in controlling Meloidogyne incognita

Fan Yang,Wei Dai,Hua Xue,Wen Chen,Chen Liu,Yaru Tian,Wanli Cheng,Jibin Zhang

Root-knot nematodes (RKNs) are notoriously challenging to control because of their broad host spectrum and swift proliferation. Consequently, there is an urgent need to develop safe and effective nematicides. Microorganisms and their metabolites have the potential to control RKNs safely and effectively. Paenibacillus polymyxa KM2501-1 can produce a range of volatile organic compounds (VOCs) with nematicidal activities, among which 2-undecanol is a potential biocontrol agent against Meloidogyne incognita. Nonetheless, the mechanism by which 2-undecanol prevents RKN infection in plant roots remains unclear. 2-Undecanol has many activities against M. incognita in vitro, including contact (LC50 = 34.5 mg/L), fumigation (LC50 = 191.6 mg/L), attraction, locomotion inhibition (frequency of head thrash and body bend reduced by 84.0% and 97.0% at concentration of 40 mg/L) and egg hatching inhibition (by 98.5% at concentration of 80 mg/L). Upon treatment with 2-undecanol, a noticeable reduction in the number of galls was observed, and the control efficacy reached 60.8% at the dose of 5 mg/pot. Metabolomics analysis revealed that the application of 2-undecanol significantly influenced the composition of root exudates of tomato. Among the 17 differential metabolites, the up-regulated 10-undecenal could attract and kill M. incognita J2 larvae at a concentration of 100 mg/L, and cyclohexylamine had nematicidal activity at the concentration of 1,000 mg/L. This study revealed the mechanism of 2-undecanol inducing plant roots to secrete nematicidal substances with attractant activity, effectively controlling RKNs, which could pave the way for the development of environmentally friendly and cost-effective nematicides.

The bioactivity of new chitin oligosaccharide dithiocarbamate derivatives evaluated against nematode disease (Meloidogyne incognita)

Zhaoqian Fan,Yukun Qin,Song Liu,Ronge Xing,Huahua Yu,Xiaolin Chen,Kecheng Li,Rongfeng Li,Xueqin Wang,Pengcheng Li

Plant-parasitic nematodes cause substantial crop losses annually; however, current nematicides are environmentally unfriendly and highly toxic to nontarget organisms. The development of green efficient nematicides from multifunctional natural bioactive substances such as chitin oligosaccharide (COS) is promising. In this paper, COS dithiocarbamate derivatives (COSDTC, COSDTA, COSDTB) were synthesized to increase nematicidal activity (against Meloidogyne incognita ), and their structures were characterized by FTIR, NMR, TGA/DTG and elemental analysis. Furthermore, the nematicidal activities, egg hatching inhibitory activities, plant growth adjustment abilities, cytotoxicity and phytotoxicity of the derivatives were evaluated. The primary mechanism was assessed by heavy metal ion absorption and GSH-binding assays. The results showed COS dithiocarbamate derivatives could possess multiple efficacies, including high nematicidal activities and egg hatching inhibitory activities, plant growth regulating effects, low cell toxicities and phytotoxicities. Additionally, it was inferred that nematicidal activity may be correlated with GSH-binding activity but not heavy metal ion complexation. COS modification has immense potential for controlling plant-parasitic nematodes.

Chemosensory Pathways Involved in the Perception of Tomato-Derived Flavonoids and Alkaloids in Meloidogyne incognita

Qian Wang,Liping Peng,Youjing Wang,Jiajia Hu,Liangying Kong,Yajun Liu

Plant-parasitic nematodes rely on the perception of host-derived chemical cues released from plant roots to locate suitable hosts, representing a critical prerequisite for successful plant infection. In this study, we selected four representative tomato (Solanum lycopersicumL.) root-derived chemical signals and demonstrated their distinct effects on the host-seeking behavior of the southern root-knot nematode (Meloidogyne incognita). Among these compounds, the flavonoids quercetin and luteolin exhibited significant attractant effects, whereas the alkaloids dihydrocapsaicin and solasodine showed repellent effects. RNA interference (RNAi) experiments further indicated that multiple chemosensory neurons are involved in the recognition of plant-derived chemical cues. Silencing of key chemosensory genes significantly impaired the directed chemotactic response ofM. incognitatoward tomato roots and reduced its infectivity, demonstrating that the chemosensory system represents a crucial molecular basis regulating host localization and parasitic establishment. This study provides new insights into host recognition mechanisms in root-knot nematodes and offers a theoretical foundation for developing environmentally sustainable strategies for nematode management by targeting host perception processes.
