Investigation of the antioxidative potential of Meretrix meretrix L. derived peptides from simulated gastrointestinal digestion: In vitro and in silico insights

Chao Du,Zhenzhen Hui,Na Li,Kaiyue Liu,Yong Wang,Ping Wang,Shuyang Sun

This investigation sought to identify antioxidative peptides derived from Meretrix meretrix L. (MML) throughout simulated gastrointestinal digestion (SGD) via in vitro and in silico strategies. The study demonstrated that hydrolysates from MML captured at various SGD phases showcased robust antioxidative activity. These hydrolysates also displayed a protective role against hydrogen peroxide-induced stress in Raw 264.7 cells. Notably, hydrolysates sourced from the intestinal phase of digestion (MMLHs-I) markedly inhibited the reduction of cell viability, limited the generation of reactive oxygen species under oxidative stress conditions, and promoted the activity of key antioxidant enzymes including superoxide dismutase, catalase, glutathione peroxidase, and glutathione reductase, thus amplifying the cells' antioxidative defense system. Through nLC-MS/MS analysis, 105 peptides tracing back to 18 parent proteins were pinpointed in MMLHs-I, with 18 of these peptides previously reported in the BIOPEP-UWM database. Subsequent in silico analysis, alongside empirical antioxidant activity assays, pinpointed the peptide WLV as an exceptional antioxidative agent, demonstrating an IC 50 of 0.125 mg/mL in the ABTS assay and an impressive 15.18 μmol Trolox/mg in the ORAC assay. The outcomes highlight MML's promise as a novel source of antioxidative peptides, positioning peptide WLV as a potentially valuable addition to functional foods designed to combat oxidative stress.

A green and efficient deproteination method for polysaccharide from Meretrix meretrix Linnaeus by copper ion chelating aerogel adsorption

Yong Zhu,Hailun Li,Jingrui Ma,Tingting Xu,Xinyu Zhou,Shiqi Jia,Jiaxin Zha,Dengping Xue,Weili Tao,Qingping Xiong,Jun Yuan,Jing Chen

Polysaccharides (PMM) from Meretrix meretrix Linnaeus exhibit a variety of biological activities; however, conventional purification methods are costly, time consuming and introducing toxic agents. This paper aimed to develop a green and efficient method for the deproteinization of PMM by copper ion chelating aerogel (CCA) adsorption. First, CCA was successfully prepared by copper ion chelating composite aerogel (CA). Then, CCA adsorption was applied to remove protein mixed in PMM. The optimal parameter conditions of this deproteinization method were screened, using the deproteinization rate ( D r %), polysaccharide recovery rate ( R r %) and selectivity coefficient ( K c ) as evaluation indicators , by single factor experiment. The optimal conditions were obtained as follows: CCA dose of 1.4%, pH value of 5.0 and adsorption temperature of 30 °C. Under these conditions, equivalent D r % (93.31 ± 1.33%), higher R r % (93.34 ± 1.08%) and K c (14.01 ± 0.46) were achieved compared with the latest reported deproteinization report. Finally, the effect of deproteinization between CCA adsorption and Sevag method, which is the most classical deproteinization method, were compared to verify the practicability of CCA adsorption. As a result, CCA adsorption not only showed much higher R r % as well as K c , but also would not cause any reagent contamination and environment pressure. In conclusion, CCA adsorption would be a green and highly efficient method with great potential in large-scale environmentally sustainable deproteinization of polysaccharides in the future.

Differential toxic effects of nano-titanium dioxide on clams (Meretrix meretrix) with various individuality

Xiaoxue Li,Erzhou Yao,Jie Li,Weiqun Lu

Nano-TiO 2 is inevitably released into aquatic environment with increasing of nanotechnology industries. Study pointed that different individuality showed divergent behavioral and physiological response when facing environmental stress. However, the effects of nano-TiO 2 on tolerance of bivalves with different individualities remain unknown. In the study, clams were divided into two types of individuality - proactive and reactive by post-stress recovery method. It turned out that proactive individuals had quicker shell opening level, stronger burrowing behavior, faster feeding recovery, higher standard metabolic rate and more rapid ammonia excretion ability than reactive individuals after exposed to air. Then, the survival rate, hemocytes response and oxidase activity of classified clams were evaluated after nano-TiO 2 exposure. Results showed that after 30 d exposure, proactive individuals accelerated burrowing behavior with higher survival rate. Moreover, proactive clams had better adaptability and less hemocytes response and oxidative damage than reactive clams. The study highlights the individualities of marine shell fish determine individual capacity to adapt to environmental changes, play important roles in aquaculture and coastal ecosystem health.

Modification of gel properties of Meretrix meretrix (clam) with polysaccharides: physical characterization and interaction mechanism

Jia-Cheng Liu,Lin-Da Zhang,Yue Liu,Tian-Qi Zhou,Bin Lai,Ce Wang,Jia-Nan Yan,Hai-Tao Wu

BACKGROUND This study investigated the contribution of 11 polysaccharides (2%, w/w ), including pectin (PC), κ -carrageenan (KC), ι -carrageenan (IC), gellan gum (GG), guar gum (GM), sodium alginate (SA), konjac gum (KG), gum arabic (GA), fucoidan (FC), locust bean gum (LBG), and curdlan (CD), to the gel and microstructural properties of Meretrix meretrix clam gel (MMG). RESULTS The hardness, springiness and chewiness of MMG with KC, IC, GG, SA and FC addition increased by ~10%–250%, while PC, GM, KG and LBG groups decreased by ~0.6% to 69%. KC, IC, SA, GG and FC decreased the cooking loss rate (CLR) by 69.4% to 88.7% and correspondingly enhanced the water holding capacity (WHC) by 10.2% to 21.4%, which was accompanied by an increased bound water and immobilized water area and high hydrogen proton density. The addition of KC transformed the MMG microstructure from a loose network with large pores to a compact, dense network, reducing lacunarity by 57.9%. The primary intermolecular forces in MMG with the incorporation of KC, IC, GG, SA and FC were hydrophobic interactions and disulfide bonds, which increased by 32.8%-105.3% and 45.6%-114.5% than MMG alone, respectively. CONCLUSION Collectively, KC, IC, GG, SA and FC could improve the gel properties of MMG and the strongest synergistic combination was found in the MMG/KC system. This study suggests that the incorporation of polysaccharides is a strategy with potential for modifying the gel properties of shellfish surimi products. © 2024 Society of Chemical Industry.

Comparison of the ecotoxicological effects of biochar and activated carbon on a marine clam (Meretrix meretrix)

Hao Zheng,Xia Liu,Guocheng Liu,Xianxiang Luo,Fengmin Li,Zhenyu Wang

Biochar (BC) and activated carbon (AC) amendments have been applied to remediate contaminated sediments because of their benefits in reducing toxicity and/or the bioavailability of contaminants. However, their ecotoxicity to benthic organisms, which are not well understood, should be carefully considered before their practical application. Therefore, a wood-derived BC (WBC) or a coconut shell-derived AC (CAC) was incorporated into a marine sediment at 5% (dw%) to investigate their effects on the growth and nutritional quality of a marine clam ( Meretrix meretrix ). CAC and WBC had no effect on the pH and calcium content of the sediment, but significantly increased the electrical conductivity (EC) and total organic carbon (TOC), and decreased the dissolved organic carbon (DOC). After 28 days of cultivation, WBC reduced the shell growth of the clam by 36.4%, and CAC reduced the soft tissues of the clam by 9.05%. This was probably because WBC and CAC adsorb organic matters and the feeding algae may colonize within their pores, thus, reducing food bioavailability for the clams. Water extracts of CAC and WBC had negligible effects on soft tissue growth, whereas the CAC extract inhibited the growth of clam shells by 57.9%. WBC and CAC had little influence on the nutritional quality of clams, including moisture, crude protein , and crude lipid contents. These findings are helpful to understand the ecotoxicological risks of BC and AC amendments to benthic organisms when used for the remediation of contaminated marine sediments.

Comparative peptidomic profile and bioactivities of Pacific oyster, Mytilus edulis, Argopecten irradians, Meretrix meretrix L. after simulated gastrointestinal digestion

Liu Kaiyue,Li Na,Fan Jiayi,He Siliang,Tang Weixiang,Cao Yaxin,Shen Liang,Sun Shuyang,Du Chengcheng,Du Chao

Purpose This study investigated the basic composition, in vitro digestibility, biological activities, and potential bioactive peptides of Pacific oyster (PO), Mytilus edulis (ME), Argopecten irradians (AIS), and Meretrix meretri x L. (MML) after simulated gastrointestinal digestion (SGD). Methods SGD products were prepared using the INFOGEST 2.0 model, and their nutritional properties and biological activities were analyzed. A comprehensive peptidomic profiling of the digested samples was conducted using nano-liquid chromatography-tandem mass spectrometry (nano-LC MS/MS) analysis. Results All four shellfish exhibited a nutritional profile characterized by high protein and low-fat content. The digestibility of shellfish increased progressively, while the average molecular weight (MW) of the hydrolysates gradually decreased during SGD, with the intestinal hydrolysates showing the highest overall digestibility. Biological activity analysis demonstrated that intestinal digestion products from all four species exhibited notable ABTS radical scavenging activity and α-glucosidase inhibition activity, with PO and ME exhibiting the strongest effects. A total of 349, 99, 314 and 105peptides were identified from the digestion products of PO, ME, AIS, and MML, respectively. Among these, PO and AIS showed the highest sequence similarity, sharing 69 identical peptides. By contrast, MML displayed distinct peptidomic characteristics, including a higher proportion of large MW peptides. Conclusion This study provides a comprehensive peptide catalog of shellfish digestion products and offers new insights into the biological activities of shellfish-derived peptides, supporting their potential as functional food resources.

Identification of novel α-glucosidase inhibitory peptides in Meretrix meretrix Linnaeus and their inhibitory kinetics using in silico and in vitro analyses

Na Li,Kaiyue Liu,Yue Zhang,Zhenzhen Hui,Ping Wang,Shuyang Sun,Chao Du

This study aimed to identify novel peptides with α-glucosidase inhibitory activity from Meretrix meretrix Linnaeus hydrolysates (MMLHs) following simulated gastrointestinal digestion (SGD) and to elucidate their inhibitory mechanisms. The molecular weight distribution of MMLHs progressively decreased during SGD, with hydrolysates from the intestinal digestion phase (MMLHs-I) exhibiting the strongest α-glucosidase inhibition (IC 50 : 0.14 ± 0.02 mg/mL). A total of 127 peptides were identified in MMLHs-I, among which 58 were unique compared to those from the gastric digestion phase. In silico screening and molecular docking identified six novel peptides—FAGDDAPR, VPLF, YRRL, WDH, PPLA, and WSG—with significant α-glucosidase inhibitory activity, displaying IC 50 values of 0.90, 1.30, 1.90, 1.20, 1.00, and 1.80 mg/mL, respectively. Among them, FAGDDAPR and PPLA exhibited the highest bioactivity. Kinetic analysis and fluorescence quenching assays demonstrated that both peptides acted as mixed-type inhibitors, forming stable complexes with α-glucosidase. Their inhibitory effects were primarily driven by hydrogen bonding and hydrophobic interactions, which hindered substrate binding and induced conformational changes in the enzyme. These findings suggest that peptides derived from MMLHs through gastrointestinal digestion possess potential antidiabetic properties, highlighting their promise as functional food ingredients for mitigating hyperglycemic damage.

Influence of pH on gel properties of Meretrix meretrix clam protein with κ-carrageenan: Phase behaviors, nonlinear rheological properties and gelation mechanism

Jia-Cheng Liu,Tian-Qi Zhou,Jia-Ni Zhou,Bin Lai,Ce Wang,Jia-Nan Yan,Hai-Tao Wu

In the present study, the complex coacervation of Meretrix meretrix clam protein (MMCP) and κ -carrageenan (KC) was investigated based on turbidimetry at various pH (12–1) and blending ratio (9:1–1:9). The pH c and pH φ1 showed a ratio-independent behavior around pH 9.5 and pH 6.0, respectively. The MMCP/KC complex were divided into three phases by crucial pH: mixed polymers, soluble complexes and insoluble coacervates. The storage modulus ( G′ ) of MMCP-KC binary hydrogels at pH 4.5 was 165 % and 71.4 % higher than that at pH 10.5 and 7.5, respectively, exhibiting the best viscoelasticity. According to nonlinear rheological analysis, the MMCP/KC binary hydrogels had weak strain overshoot properties, as well as intracycle shear thinning and strain-stiffening properties, which exhibited the best structural stability at pH 4.5. Correspondingly, the MMCP/KC binary hydrogels at pH 4.5 exhibited a well-organized and dense microstructure with the lowest lacunarity about 0.17 accompanied by microphase separation and protein self-aggregation, which allowed more water to be entangled in the gel network and enhanced the thermal stability of the binary hydrogels. Furthermore, the electrostatic and hydrophobic interactions maintained the main intermolecular forces of MMCP/KC binary hydrogels under acidic conditions, with the total proportions of 42.8 % and 34.7 %. The blue shift of amide I bands, decreased free -SH groups and increased surface hydrophobicity of MMCP/KC binary hydrogels further confirmed the main contribution of electrostatic and hydrophobic interactions in the gel network. These results provided theoretical and methodological basis for designing novel marine protein-polysaccharide binary hydrogels with excellent gel properties.

Development of Meretrix meretrix clam protein/ι-carrageenan hydrogels with soft-textured and safe-swallowing: Insights into nonlinear rheological behavior, microstructure and gelation mechanisms

Jia-Cheng Liu,Jia-Ni Gao,Zhu-Jun Zhang,Bin Lai,Ce Wang,Jia-Nan Yan,Hai-Tao Wu

The complex coacervation and gelation behavior of Meretrix meretrix clam protein/ ι -carrageenan (MMCP/IC) influenced by various blending ratios (9:1-1:9) and pH (12-1) were investigated. Compared with other blending ratios, the MMCP/IC composite hydrogels at blending ratio of 4:6 exhibited the greatest rheological properties ( G' and G'' ) and textural properties (hardness, cohesiveness, springiness and chewiness), which were increased by 0.2-3.6 folds and 31%-151%, respectively. The MMCP/IC composite hydrogels showed weak strain overshoot behaviors and strain-softening behavior , which showed the best shear resistance and stability at blending ratio of 4:6 under large deformation. The P 21 and P 22 of MMCP/IC composite hydrogels at blending ratio of 4:6 enhanced by 3.3%-73.1% than that under other blending ratios compared with that of single MMCP. Correspondingly, MMCP/IC composite hydrogels at blending ratio of 4:6 exhibited a compact network structure with the lowest lacunarity and largest aggregation, supporting highest gel viscoelasticity. The strengthened electrostatic interactions and hydrogen bonds between MMCP and IC were reflected by blue shift of Amide A and Ⅰ bands, and the former occurred earlier than the latter. Furthermore, electrostatic interactions and hydrophobic interactions were the primary forces for maintaining MMCP/IC composite hydrogels, accounting for 25.6%-32.3% and 29.7%-40.6% of the total forces. The MMCP/IC composite hydrogels at various blending ratios (6:4, 4:6 and 2:8) could be classified into level 5 dysphagia food (minced and moist) by International Dysphagia Eating Standardization Initiative (IDDSI). These findings provide insights into the development of novel soft-gel types of dysphagia foods based on marine-derived proteins and polysaccharides.
