Bio-inspired enzyme–inorganic hybrid nanomaterials are extensively employed for biological applications owing to their capability of enhancing enzyme activity and stability compared with free counterpart. They play a vital role in signal output and amplification during bioassay, but usually suffer from deactivation and costing problems, which motivates us to seek a more robust signal generation pattern. The coordination interaction between metal ions and amide groups in enzyme drives the evolution of various morphology and structure. Inspired by this, C-reactive protein (CRP) aptamer, with amide groups in their nucleobases, is introduced as an organic part serving as a linker and biorecognition unit to prepare aptamer-inorganic hybrid nanosheets, and be applied for CRP detection. Unlike using organic enzymes as a signal enhancer, the inorganic part, Mn 3 (PO 4 ) 2 , is exploited for generating and transducing firm current signals, which stem from electrochemical active molybdophosphate precipitates reacted by phosphate and external molybdate. This enzyme-free electrochemical aptasensor based on biomineralized Mn 3 (PO 4 ) 2 /aptamer exhibits an excellent detection performance for CRP. And the reliability is also acceptable compared to the traditional immunoturbidimetric method at the hospital, suggesting our method good feasibility for clinical diagnosis.

The excellent selectivity and anti-interference ability of enzyme-based biosensors have attracted considerable attention by researchers. However, the short lifetime, difficult electron transfer, and easily leak from the supporting materials restrict the practical applications of enzyme. In this study, an enzyme–inorganic hybrid biosensor constructed from slack Hb–Mn3(PO4)2·3H2O nanosheets was prepared by a simple and effective in situ immobilization method. Designed as an H2O2 sensor, it exhibits a fast electron transfer rate constant (ks = 4.16 s−1), an ultra-wide linear range for H2O2 detection (20 to 56 100 μM), and long life-time (85% of initial response after 29 days), which may be attributed to the effective immobilization of the enzyme and the good biocompatibility of Mn3(PO4)2·3H2O. For the detection of a real sample, the range of recovery of our designed biosensor was around 98.2–102.6%, confirming that our proposed method presents a promising immobilization enzyme-based biosensor for use in practical applications.

It is critical for fabricating flexible biosensors with both high sensitivity and good selectivity to realize real-time monitoring superoxide anion (O 2 •− ), a specific reactive oxygen species that plays critical roles in various biological processes. This work delicately designs a Mn 3 (PO 4 ) 2 /MXene heterostructured biomimetic enzyme by assembling two-dimensional (2-D) Mn 3 (PO 4 ) 2 nanosheets with biomimetic activity and 2-D MXene nanosheets with high conductivity and abundant functional groups. The 2-D nature of the two components with strong interfacial interaction synergistically enables the heterostructure an excellent flexibility with retained 100% of the response when to reach a bending angle up to 180°, and 96% of the response after 100 bending/relaxing cycles. It is found that the surface charge state of the heterostructure promotes the adsorption of O 2 •− , while the high-energy active site improves electrochemical oxidation of O 2 •− . The Mn 3 (PO 4 ) 2 /MXene as a sensing platform towards O 2 •− achieves a high sensitivity of 64.93 µA·µM −1 ·cm −2 , a wide detection range of 5.75 nM to 25.93 µM, and a low detection limit of 1.63 nM. Finally, the flexible heterostructured sensing platform realizes real-time monitoring of O 2 •− in live cell assays, offering a promising flexible biosensor towards exploring various biological processes.

The authors have synthesized nanostructured manganese(II) phosphate hollow spheres [Mn 3 (PO 4 ) 2 HS] with tunable pore structure by using a micro-emulsion method. These, if deposited on a glassy carbon electrode (GCE), are shown to be a viable material for electrochemical sensing of superoxide (radical) anion (O 2 ˙ − ) at a typical working voltage of 0.7 V (vs. SCE). Hence, they act as biomimetic enzymes that allow for the determination of O 2 ˙ − with a very low detection limit (1.35 nM), wide linear range (5 nM to 0.4 mM), and good long-term stability. The modified GCE was applied in-situ to the electrochemical determination of O 2
− that is released from human malignant melanoma cells and normal keratinocyte, and it showed excellent real time analytical capability. In our perception, the use of this material offers exciting opportunities in terms of implementing nanoscale transition metal phosphates as biomimetic enzymes in enzyme-free diagnostic sensors. Conceivably, these will offer higher sensitivity and longer durability than assays based on the use of natural enzymes.

A visual colorimetric microfluidic paper-based analytical device (μPAD) was constructed following the direct synthesis of enzyme-inorganic hybrid nanomaterials on the paper matrix. An inorganic solution of MnSO 4 and KH 2 PO 4 containing a diluted enzyme (glucose oxidase, GOx) was subsequently pipetted onto cellulose paper for the in situ growth of GOx@Mn 3 (PO 4 ) 2 hybrid functional materials. The characterization of the morphology and chemical composition validated the presence of hybrid materials roots in the paper fiber, while the Mn 3 (PO 4 ) 2 of the hybrid provided both a surface for enzyme anchoring and a higher peroxidase-like catalytic activity as compared to the Mn 3 (PO 4 ) 2 crystal that was synthesized without enzyme modulation. This new approach for the in situ growth of an enzyme-inorganic hybrid on a paper matrix eliminates centrifugation and the dry process by casting the solution on paper. The sensing material loading was highly reproducible because of the accuracy and stability of pipetting, which eventually contributed to the reliability of the μPAD. The self-assembled natural and artificial enzyme hybrid on the μPADs specifically detected glucose from a group of interferences, which shows great specificity using this method. Moreover, the colorimetric signal exhibited detection limitation for glucose is 0.01 mM, which lies in the physiological range of glucose in biological samples.
