Ti3C2 MXene/MoS2@AuNPs ternary nanocomposite for highly sensitive electrochemical detection of phoxim residues in fruits

Jiani Zhang,Xiaohui Guo,Jian Zhang,Xiaobing Guo,Yuying Xu,Lijuan Chen

Phoxim, extensively utilized in agriculture as an organothiophosphate insecticide, has the potential to cause neurotoxicity and pose human health hazards. In this study, an electrochemical enzyme biosensor based on Ti 3 C 2 MXene/MoS 2 @AuNPs/AChE was constructed for the sensitive detection of phoxim. The two-dimensional multilayer structure of Ti 3 C 2 MXene provides a robust framework for MoS 2 , leading to an expansion of the specific surface area and effectively preventing re-stacking of Ti 3 C 2 MXene. Additionally, the synergistic effect of self-reduced grown AuNPs with MoS 2 further improves the electrical conductivity of the composites, while the robust framework provides a favorable microenvironment for immobilization of enzyme molecules. Ti 3 C 2 MXene/MoS 2 @AuNPs electrochemical enzyme sensor showed a significant response to phoxim in the range of 1 × 10 −13  M to 1 × 10 −7  M with a detection limit of 5.29 × 10 −15  M. Moreover, the sensor demonstrated excellent repeatability, reproducibility, and stability, thereby showing its promising potential for real sample detection.

DNA cyclic assembling control in an electrochemical strategy with MoS2@AuNPs for determination of kanamycin

Wang Lina,Zhang Li,Yu Ying,Lin Bixia,Wang Yumin,Guo Manli,Cao Yujuan

A sensitive electrochemical strategy was established for kanamycin determination. A specific aptamer was modified on the electrode as the probe, followed by a cyclic hybridization chain reaction (HCR) with methylene blue, causing an increasing signal response. In the presence of kanamycin, it can initiatively convolve the aptamer and prevent further DNA assembling, resulting in a signal distinction sensitive to the target amount. However, the signal reproducibility is low. To improve the precision, the HCR procedure was investigated. The results demonstrated that the optimal amount of assembled DNA is 12-fold to that of aptamer. This amount was then controlled in further assays. Admittedly, controlled DNA assembling commonly indicates a limited signal amplification. To further enhance the sensitivity, a nanocomposite based on MoS 2 and AuNPs was modified on the electrode. The results of the assay proved that the signal distinction sensitive to target amount increased by 50%. A linearity range is obtained from 0.01 nM to 1.0 μM of kanamycin, and the LOD is 8.4 pM. Subsequently, this strategy was employed to detect kanamycin in chicken liver and milk sample; the recovery results suggest that it possess a satisfactory application prospect in analysis of agricultural products. 3D MoS2-AuNPs carbon paper probe for ultrasensitive detection and discrimination of p53 gene

Jiaying Zhao,Danqun Huo,Xintong Geng,Jing Bao,Jingzhou Hou,Zhengfan Shui,Huisi Yang,Yanli Qi,Yian Hu,Mei Yang,Changjun Hou

p53 gene is one of the most important anticancer genes and has been recognized as a typical biomarker for the early diagnosis, classification, and prognosis evaluation of cancers. In this work, a novel biosensing strategy integrating MoS 2 -AuNPs modified carbon paper (CP) conductive substrate and enzyme signal amplification technique was developed for p53 gene detection. We constructed an immobilized DNA S1 probe and a biotinylated DNA S3 probe, which could induce the formation of an S1-p53-S3 DNA sandwich complex in the presence of target sequence p53. After conjugating avidin-HRP via biotin-avidin reaction, 3,3’,5,5’-tetramethylbenzidine would be catalyzed for signal amplification in the presence of H 2 O 2 so that the induced electrochemical response would change with proportional to the abundance of p53 gene. The proposed biosensor exhibited two linearities in the range of 1.0 × 10 −15 ∼ 1.0 × 10 -12 M and 1.0 × 10 -12 ∼ 1.0 × 10 -6 M, and the limit of detection (LOD) was 68 fM. No significant interferences were observed during detecting real serum samples. Moreover, it is worth pointing out that the biosensor together with its disposable, low-cost, and stable features can discriminate well between the mutated and wild type p53 gene extracted from tumor cells (wtTP53 in MCF-7 and mutated in SK-BR-3), demonstrating a promising applicability for clinical test.

Sensitive and fast analysis of terbutaline sulfate in food using a modified electrode based on a MoS2/AuNPs nanocomposite

Xiaoyun Lin,Yanfang Wang,Miaomiao Zou,Yongnian Ni

Terbutaline sulfate (TBS) is a β2-adrenoceptor agonist, overdoses of which can cause serious harm and even be life-threatening to humans. Herein, we demonstrate a simple and facile method for the preparation of a molybdenum disulfide–gold nanoparticle composite (MoS2/AuNPs). The obtained MoS2 and MoS2/AuNPs were characterized and investigated by X-ray powder diffraction, scanning electron microscopy, and transmission electron microscopy. An MoS2/AuNPs-modified glassy carbon electrode (MoS2/AuNPs/GCE) was constructed using the dropping method and used to study the electrochemical behavior of TBS. The results indicated that the sensor had excellent electrocatalytic activity for TBS, with detection sensitivity for TBS more than 100 times higher compared with the bare GCE. With the successive addition of TBS, the peak current of the differential pulse stripping voltammogram (DPSV) of TBS was proportional to its concentration within a certain range, with a linear range of 1–85 nmol L−1 and limit of detection (LOD) of up to 0.47 nmol L−1. Furthermore, the modified electrode showed good stability, reproducibility, and anti-interference ability, meaning that it could be used to detect TBS in real samples.

Sensitive amperometric immunosensor for pathogen antigen based on MoS2@AuNPs assembling dual-peptide as bioprobes with significant dual signal amplification

Zongmei Zheng,Mingyang Wang,Jinlong Yuan,Junchong Liu,Haipeng Yu,Zhonghuang Yang,Wanjian Liu,Aihua Liu

It is crucial to timely and accurately identify the causative virus for early treatment and urgent prevention. Viral antigen detection can identify those people who are most likely at risk of spreading the disease, but most based on antibodies with limited stability and sensitivity. Peptides offer several advantages over antibodies, such as low cost, smaller size and good stability. The development of electrochemical immunoassay using specific peptide probes have the merits of good sensitivity and selectivity as well as good stability. Herein we report an amperometric immunosensor using peptides as capture probe and recognition probe. The molecular docking suggests that the two peptides of Pi (sequence: NFWISPKLAFALGGGKKKSC) and FK11 (sequence: WFLNDSELISML), bioscreened from phage display, bind to N-terminal domain of SARS-CoV-2 spike protein (SP). The peptide Pi is assembled on MoS 2 @AuNPs modified electrode to capture SARS-CoV-2 SP, which is recognized by peptide FK11-displayed phage to form Pi/SARS-CoV-2 SP/FK11-phage sandwich. Then anti-M13 phage conjugated horseradish peroxidase (HRP) (anti M13-HRP) was introduced to recognize the phage capsid protein pVIII to form M13 phage/anti M13-HRP to enrich thousands of HRP, which can further electrochemically catalyze H 2 O 2 reduction at highly conductive MoS 2 @AuNPs at − 0.35 V. Then amperometric immunosensor was constructed with linear range of 0.1−5000 pg/mL SARS-CoV-2 SP and detection limit of 0.074 pg/mL. The sensor also has good selectivity, batch reproducibility and stability, capable of detecting down to 10 transducing units/mL SARS-CoV-2 pseudoviruses. This work represents the first example of dual-peptide probes based sandwich-type electrochemical immunosensor integrated with dual signal amplification, which may provide a cost-effective assay platform in detecting real SARS-CoV-2 viruses for early diagnosis. The flexible and modular strategy can be extended to develop other type biosensors for a wide range of applications.

Bifunctional reduced graphene oxide/polyelectrolytes/MoS2-AuNPs hybrids with Vis/NIR photocatalytic and photothermal activities for biosafe and efficient antibacterial applications

Meng Fu,Chang Luo,Qi Yuan,Zequn Ma,Quansheng Long,Baihui Liu,Dang Wu,Jingshui Xu,Xiangming Li

The development of bacterial resistance to antibiotics and its global spread highlights the urgent need for new alternative antimicrobial agents. Here, we have developed a biosafe antibacterial material based on reduced graphene oxide/polyelectrolytes nanolayers and gold nanoparticles functionalized molybdenum disulfide (rGP 1.5 /MoS 2 -AuNPs) via in situ synthesis and electrostatic assembly. The rGP 1.5 /MoS 2 -AuNPs has high visible and near-infrared absorption, which can efficiently provide remarkable photothermal conversion efficiency and generate reactive oxygen species (·O 2 - , ·OH, 1 O 2 ) via photocatalytic oxygen reduction and water oxidation. The rGP 1.5 /MoS 2 -AuNPs combines the photocatalysis with photothermal effect, exhibiting a biosafe and effective killing for Gram-negative and Gram-positive bacterial. The rGP 1.5 /MoS 2 -AuNPs simultaneously optimizes three key parts for antibacterial: i) polyelectrolytes component promotes bacterial adhesion, ii) reduced graphene oxide and AuNPs components provide photothermal conversion capacity and increase light absorption, iii) MoS 2 component generates photogenerated electron-hole pairs, and the electrons are rapidly conducted from the AuNPs to the reactants, which promotes the rapid separation of the electron-hole pairs, resulting in the generation of reactive oxygen species. This work provides a simple but powerful way to prepare photocatalytic and photothermal material for antibacterial applications.
