This study investigated the conditions for polygonati rhizoma (PR) fermentation by Monascus ruber and the lipid-lowering activity of the resulting product. Monascus ruber was used for the solid-state fermentation of PR, and the response surface method was used to optimize the fermentation culture medium parameters. The optimal fermentation system yielded a maximum monacolin K concentration of 21.82 ± 0.34 mg/g. The fermentation conditions were as follows: 12.5 mmol/kg of L-glutamic acid, 0.64 mmol/kg of cyclic adenosine monophosphate , 3.0 wt% of yeast extract powder, a fermentation period of 15 days, and a fermentation temperature of 30 ℃. The binding capacities of unfermented PR, “Nine-Steam-Nine-Bask” processed Polygonati rhizoma (NPR), and Monascus ruber solid-state fermented polygonati rhizoma (MPR) to sodium cholate, sodium taurocholate , and sodium glycolate were determined. MPR showed a stronger bile acid-binding capacity than unfermented PR. In addition, MPR exhibited the highest cholesterol inhibitory capacity of 51.43±1.41 %. Thus, MPR can be used as a potential antilipemic agent with potent hypolipidemic effects.

Blue light, as an important environmental factor, greatly affects the production of Monascus pigments (MPs) and citrinin in Monascus spp.. In this study, the deletion, complementation, and overexpression mutants of MrcreD from Monascus ruber M7, which encodes an arrestin-like protein, were constructed and cultivated on PDA (Potato dextrose agar) medium to study the effects of blue light on MPs and citrinin production. The results revealed that blue light inhibited the formation of cleistothecia, conidia, and the production of MPs and citrinin in M. ruber M7. However, under blue light, in contrast to M. ruber M7, MrcreD -overexpressing strain displayed increased production of extracellular yellow pigments and intracellular orange pigments, whereas MrcreD -deleted strain showed enhanced production of intracellular yellow and orange pigments. Then, the extracellular citrinin production decreased in both mutants. The RT-qPCR results demonstrated that compared to M. ruber M7, overexpressing MrcreD increased the expression of genes involved in MPs biosynthesis, and decreased the genes involved in citrinin biosynthesis, while deleting MrcreD increased the expression of citrinin-relative genes. This is the first time that the functions of MrcreD gene in filamentous fungi have been researched under blue light, and it provides a strategy for exploring complex light-regulatory systems in filamentous fungi.

YellowMonascuspigments can be of two kinds: Natural and reduced, in which natural yellowMonascuspigments (NYMPs) attract widespread attention for their bioactivities. In this study, the antioxidative and antibreast cancer effects of the water-soluble NYMPs fermented byMonascus ruberCGMCC 10910 were evaluated. Results showed that water-soluble NYMPs had a significantly improved antioxidative activities compared to the reduced yellowMonascuspigments (RYMPs) that were chemically derived from orange or redMonascuspigments. Furthermore, NYMPs exhibited a concentration-dependent inhibition activity on MCF-7 cell growth (p< 0.001). After a 48-h incubation, a 26.52% inhibition yield was determined with 32 μg/mL of NYMPs. NYMPs also significantly inhibited the migration and invasion of MCF-7 cells. Mechanisms of the activities were associated with a down-regulation of the expression of matrix metalloproteinases and vascular endothelial growth factor. Rather than being alternatively used as natural colorants or antioxidants, this work suggested that NYMPs could be selected as potential functional additives in further test of breast cancer prevention and adjuvant therapy. Keywords:natural yellowMonascuspigments;water-soluble;antioxidation;migration;invasion;MCF-7 cells

Obesity and hyperlipidemia, chronic diseases resulting from excessive energy intake and abnormal lipid metabolism, respectively, are intricately associated with cardiovascular disease and diabetes and have become global health problems. Monascus ruber ( M. ruber ), a filamentous fungus with edible properties, produces bio-functional metabolites including a good deal of enzymes. In this study, the effect of the biological transformation of fermented food on obesity and hyperlipidemia, was systematically investigated using the mixture of medicine and food homology (MFH) hawthorn and millet as fermentation substrate. Results showed that fermentation promoted the release of bioactive compounds such as phenolic compounds. Pancreas lipase assay indicated that the inhibition rate and IC 50 of free phenolic compounds in fermented MFH food were 70.51% ± 2.59%, and 1.34 mg/mL, respectively. In vitro , the binding rates of free phenolic compounds in fermented MFH food to Sodium cholate (SC), sodium glycinate (SGC) and sodium taurocholate (STC) were 84.99% ± 6.59%, 80.29% ± 6.23%, and 79.68% ± 6.18%. Further, Monascus -fermented hawthorn and millet (MFHM) of fermented MFH food markedly suppressed lipid accumulation in oleic acid (OA)-induced HepG2, with cell triglycerides (TG) and total cholesterol (TC) clearance of 24.31% ± 1.81% and 73.88% ± 2.72%, respectively. These findings implied that the fermentation of a mixture of MFH millet and hawthorn by M. ruber holds promise for reducing obesity and lowering lipid levels, thereby serving as a valuable foundation and reference for future research on the development of multifunctional food products.

Ginseng (Panax ginseng C. A. Meyer) is a famous Traditional Chinese Medicine, which is widely used to treat cardiovascular disease. Monascus ruber (M. ruber) is a fungus used in food and medicine fermentation, and lovastatin, its metabolite, is used extensively in the treatment of dyslipidemia. In this study, ginseng has been fermented by M. ruber, and the response surface methodology (RSM) was applied to optimize fermentation parameters to obtain optimal fermentation system, with further exploring to lipid-lowering activity of P. ginseng C. A. Meyer–M. ruber fermentation products (PM). The concentration of ginseng, temperature, and rotating speed were set as variables and the lovastatin yield was optimized by a Box–Behnken design (BBD) analyzed by RSM. The binding capacity of PM for sodium taurocholate and sodium cholate was assayed by UV spectrophotometry. The highest content of lovastatin production (85.53 μg g−1) was obtained at a ginseng concentration of 1.96%, temperature of 30.11 °C, and a rotating speed of 160.47 rpm. PM exhibited bile acid binding capacity, which was stronger than unfermented ginseng. The RSM can be used to optimize the fermentation system to obtain the best fermentation process. In addition, the fermentation of ginseng by M. ruber can enhance the lipid-lowering effect.
