Peach fruit is highly susceptible to Monilinia fructicola ( M. fructicola ), a pathogen that causes significant postharvest losses. Enhancing the efficacy and reducing the dosage of chemical fungicides are critical for controlling brown rot caused by M. fructicola . Natural volatile organic compounds (VOCs) have been proposed as a selective method for managing postharvest pathogen infection. Among these, 2-decanone, a natural volatile produced by fungi and insects, has demonstrated potential in inhibiting pathogen growth. In this study, 60 μL/L 2-decanone effectively inhibited the growth of M. fructicola mycelia. It also reduced spore germination and appressorium formation by downregulating the expression of MfBmp1 and MfPls1 , respectively. In addition, 2-decanone induced spore apoptosis by increasing reactive oxygen species (ROS) accumulation and exacerbating mitochondrial damage. When applied to peach fruit, 2-decanone fumigation significantly reduced disease incidence and lesion diameter compared to the control group, without adversely affecting fruit quality during storage. Furthermore, we observed that 2-decanone exhibited synergistic effects when combined with boscalid, effectively inhibiting the growth of M. fructicola . Applying boscalid 15 days before harvest and fumigating with 2-decanone postharvest provided similar brown rot suppression as boscalid alone but used only one-tenth the amount of fungicide. Overall, 2-decanone fumigation presents an effective strategy for controlling postharvest brown rot disease in peaches, and its combined use with boscalid introduces a promising approach for enhancing postharvest peach fruit preservation.

Brown rot caused by Monilinia fructicola ( M. fructicola ) severely affects Shengzhou nane ( Prunus salicina var. taoxingli ). Natamycin (NAT), a natural antimicrobial preservative, has demonstrated good efficacy in mitigating post-harvest decay and maintaining fruit quality. This study investigated antifungal efficacy and underlying mechanism of NAT against M. fructicola in vitro and in vivo settings. In vitro assay demonstrated that NAT exhibited potent antifungal activity against mycelial growth and spore germination of M. fructicola , with efficacy increasing in correlation with higher concentrations of NAT. Specifically, 20.0 mg L −1 NAT induced notable mycelial and spore deformation, characterized by twisting and breakage, while 25.0 mg L −1 NAT completely inhibited mycelial growth and spore germination. NAT exposure suppressed antioxidative enzyme activity, for example, 20 mg L −1 NAT decreased SOD activity by 61.49% of the control at 24 h, and downregulated related gene expression, leading to intracellular accumulation of ROS and oxidative stress. Furthermore, NAT disrupted the integrity of cellular membrane, resulting in leakage of intracellular contents. For instance, DNA leakage increased by 3.2-fold after 24 h. NAT exposure damaged mitochondrial structural and impaired its function, as indicated by the decreased adenosine triphosphate content, by 58.54% of the control, lower energy charge levels, and the reduced activities of malate dehydrogenase, succinate dehydrogenase, Ca 2+ /Mg 2+ -ATPase, and Na + /K + -ATPase by 79.67%, 77.57%, 81.34%, and 89.79% of the control, respectively, following a 24 h exposure. In vivo experiments indicated that NAT treatment resulted in 93.30% reduction in disease incidence at concentration of 200 mg L −1 , as well as a decrease in lesion diameter in Shenzhou nane fruit. Additionally, NAT enhanced the activities of antioxidative enzymes, including superoxide dismutase, catalase, and peroxidase a significant reduction in malondiadehyde levels. NAT also promoted the activity of phenylalanine ammonia-lyase, chitinase, β-1, 3-glucanase, and polyphenol oxidase. The alleviation of oxidative stress and enhancement of defense enzyme activity together increased fruit resistance to M. fructicola . Consequently, higher fruit quality was achieved during storage, thus, 200 mg L −1 NAT increased total soluble solids by 2.42 folds of the control. In conclusion, our findings demonstrated that NAT exhibited a dual-action effect against antibacterial M. fructicola , acting both as a direct antifungal agent and an inducer of host fruit's innate defense mechanism. These findings highlight the potential of NAT treatment as an effective strategy for controlling brown rot caused by M. fructicola in Shengzhou nane fruit

Monilinia fructicola , an extremely destructive pathogen, induces the postharvest decay of stone fruits. Nerol completely inhibited the growth of M. fructicola at a minimum inhibitory concentration of 0.2 mL/L. It disrupted mycelial membranes and released intracellular substances, such as nucleic acid, protein, and K + . Excessive reactive oxygen species (ROS) resulted in lipid peroxidation, which is responsible for membrane disruption. For the scavenging of excessive ROS, the antioxidant enzyme activities of catalase, superoxide dismutase, and peroxidase increased. However, glutathione (GSH) and GSH peroxidase activities decreased. Lasting oxidative stress disrupted mitochondria, which exhibited cristae loss and matrix transparency. The activities of key enzymes in tricarboxylic acid (TCA) cycle and ATPase were reduced, thereby causing ATP depletion. RNA sequencing revealed that nerol down-regulated the genes for L-glutamate and GSH synthesis while up-regulating those for GSH metabolism, resulting in a net GSH depletion. Metabolic disruptions occurred in membrane lipids and cell wall integrity, primarily involving steroids, sphingolipids, glycerophospholipids, and trehalose. Nerol disrupted the expressions of most genes involved in the TCA cycle. In vivo, 0.4 mL/L nerol completely inhibited the growth of M. fructicola on peaches. These results highlight nerol’s multiple antifungal mechanisms, supporting its development as a bio-preservative against M. fructicola .

This work fabricated natamycin-loaded zein-casein nanoparticles (N-Z/C NPs) by a pH-driven approach and applied to control postharvest peach brown rot caused by Monilinia fructicola . When casein and phosphoric acid were used as a stabilizer and neutralizing acid, respectively, NPs with mean particle sizes < 100 nm and zeta-potentials < −30 mV could be obtained. The NPs could increase the aqueous dispersibility of natamycin and showed high stability against environmental changes, which could be attributed to both hydrophobic stacking and hydrogen bonds between natamycin and zein. Besides, the effects of N-Z/C NPs on the storage of peach were assessed in vitro and in vivo . Nanoencapsulation did not affect the antifungal activities of natamycin. The NPs with 20 and 80 µg/mL of natamycin could basically inhibit the spore germination and mycelial growth of M. fructicola , respectively. The N-Z/C NPs coatings exhibited better results than natamycin in controlling of peach brown rot.
