New strategy for improving the rainfastness and control effect on Monochamus alternatus of clothianidin via a castor oil-based polyurethane nanoemulsion

Rui Zhao,Zheng Zhang,Meng Yu,Rui Gao,Zhe Sun,Shou-he Pan,Ying-jian Ma,Yong Xu,Xue-min Wu

Pine wilt disease (PWD) is one of the most destructive diseases affecting global forest ecosystems, and Monochamus alternatus (MA) is the main insect vector of pine wood nematodes. Effective control methods that cover the entire emergence period of vector insects are urgently required. Improving the rainfastness of agrochemicals on leaves can extend the holding period. Thus, a new environmentally friendly castor oil-based polyurethane nanoemulsion (CWPU-NE) was developed to enhance the rainfastness of clothianidin. CWPU adsorbs on the surface of clothianidin particles through electrostatic and intermolecular hydrogen bonding and attaches to the surface of leaves through hydrogen bonding and film formation. The results showed that clothianidin was still effective against MA 60 d after application. The developed CWPU-NE functioned as an efficient rainfastness adjuvant that enhanced the rainfastness, retention, protection, and sustained release behavior of clothianidin. Thus, it has great potential for PWD control and wider agricultural applications.

Preparation of sustainable release mesoporous silica nano-pesticide for control of Monochamus alternatus

Gehui Wang,Xiaoyu Xu,Qingqing Cheng,Jiamin Hu,Xueying Xu,Yiwu Zhang,Sen Guo,Yingchao Ji,Chenggang Zhou,Feng Gao,Long Yang,Yanxue Liu,Shuyan Yin,Chenyu Su

Monochamus alternatus is an important stem-borer that endangers economic and ecological forest, and also the main vector of pine wilt disease, a significant forest quarantine disease, which caused seriously losses for economic and ecological services. At present, M. alternatus mainly focuses on chemical control, among which Avermectin (AVM) is widely used as a high-performance spectrum insecticide. However, there are some problems, such as short duration, and environmental pressure caused by excessive use, which limit its application in pest control. In this paper, a drug delivery system based on dendritic mesoporous silica (DMSNs) was constructed to deliver biological pesticide AVM, so as to develop a new type of nano-pesticide, which could effectively control the stem borer M. alternatus and control Bursaphelenchus xylophilus . The [email protected] NPs composite was prepared by simple physical adsorption, and the loading rate was 85.73%, which significantly improved the utilization rate of AVM. The local application of AVM optimized by DMSNs showed that the contact toxicity (LC 50  = 14.27 μg/mL) and stomach toxicity (LC 50  = 10.90 μg/mL) of M. alternatus larvae were higher than those of AVM/EC and AVM treatment, which was based on the good permeability and adhesion of DMSNs. At the same time, DMSNs had better degradation ability in the natural environment: in PBS buffer solutions with different pH values, DMSNs completely degraded in 28 days. In addition, [email protected] NPs has a good absorption and transmission effect in trees, which indicates that nanoparticle-mediated pesticide delivery system will be a significant progress in the control of forest and fruit pests.

Identification of pine wood nematode (Bursaphelenchus xylophilus) loading response genes in Japanese pine sawyer (Monochamus alternatus) through comparative genomics and transcriptomics

Xiangying Zou,Jiayu Jia,Tengfei Zhu,Shouping Cai,Yiman He,Shunde Su,Yu Fang,Jian Li,Guifang Lin,Jun Su

BACKGROUND Pine wood nematode (PWN; Bursaphelenchus xylophilus ) is the causative agent of pine wilt disease (PWD), which is considered the most dangerous biohazard to conifer trees globally. The transmission of PWN relies on insect vectors, particularly the Japanese pine sawyer (JPS; Monochamus alternatus ). However, the molecular mechanism underlying PWN-JPS assembly remains largely unknown. RESULTS Here, we found that both geographical and gender could significantly affect the PCA (PWN carrying amount) of JPS; thus, JPS transcriptomes from diverse locations and genders were explored regard to PWN loading. Due to the shortage of genomes, we developed a full-length reference transcriptome for analyzing next-generation sequencing data. A comparative genomic study was performed, and 11 248 potential PWN-carrying associate genes (β) were nominated in JPS by using the reported genomes of PWN and non-PWN carrier insect species. Then, 151 differentially expressed transcripts (DETs), 28 of them overlapped with β, correlated with the PCA of JPS were nominated by RNA-Seq, and found that fatty acid β-oxidation might be the key factor that affected the PCA of JPS. Furthermore, JPS fatty acid β-oxidation rates were experimentally decreased using the inhibitor Etomoxir, leading to an increased PCA of JPS. Meanwhile, silencing MaCPT1 in JPS by RNA interference led to a decreased fatty acid β-oxidation rate and increased PCA of JPS. CONCLUSIONS In conclusion, MaCPT1 was able to decrease the PWN-JPS assembly formation through the fatty acid β-oxidation of JPS. These results provide new insights for exploring the impact of PWN invasion on JPS. © 2024 Society of Chemical Industry.

Fluorescence-Labeled Abamectin Nanopesticide for Comprehensive Control of Pinewood Nematode and Monochamus alternatus Hope

Chenyu Su,Yingchao Ji,Shanshan Liu,Shangkun Gao,Shenghan Cao,Xiaoyu Xu,Chenggang Zhou,Yanxue Liu

Pine wilt disease is a major forestry quarantine disease caused by pinewood nematode and spread by the vector insect Monochamus alternatus (MA), which has brought destructive harm to pine trees. Avermectin (AVM) is a broad-spectrum fungicide, insecticide, and nematicide. However, AVM easily degrades under ultraviolet light and is almost insoluble in water, which seriously affect its control effect on the pine wilt disease. In this study, an original nanopesticide was designed to improve the bioactivity and utilization of AVMactive ingredients, with antiphotolysis and slow release properties especially. Bovine serum albumin (BSA) was dissolved in water, taking advantage of the oleophobic properties of the protein to wrap AVM in an oil phase by self-assembly. The obtained AVM BSA nanoparticles (AVM@BSA·NPs) had preferable sustained-release performance, the cumulative release reached 90.2%, the nanoparticles could be released continuously for about 20 days, the particle size of the AVM@BSA·NPs was about 65 nm, and the loading efficiency reached 78%. The nanodelivery system could effectively protect AVM from photolysis, and the retention of AVM@BSA·NPs was close to 80 and 75% after 48 and 72 h under ultraviolet light, respectively. In addition, the nanopesticides were prepared by labeling AVM@BSA·NPs with fluorescein isothiocyanate (FITC) to research the mechanism of contact and stomach toxicity of AVM using the probe function of FITC. Compared with AVM original, the stomach toxicity and contact toxicity of AVM@BSA·NPs increased by 35.3 and 19.6%, respectively. There are broad application prospects of AVM@BSA·NPs for the prevention and control of forest diseases and other pests.

Transcriptome profiling and RNA interference reveals relevant detoxification genes in Monochamus alternatus response to (+)-α-pinene

Xuanyu He,Zhaodong Hao,Hui Li,Tian Xu,Ruixu Chen,Xiaohong Xia,Shouyin Li,Dejun Hao

Understanding the mechanism of insect resistance to toxic effects of phytochemicals can provide an insight into plant–herbivore interactions. Monochamus alternatus Hope, a main vector of pine wood nematode ( Bursaphelenchus xylophilus ), prefers to infest masson pine ( Pinus massoniana Lamb) and causes huge economic and environmental losses. α-Pinene is the primary component of resin produced by masson pine and plays a crucial role in defending against herbivorous insects, but its defensive effects on M . alternatus larvae are barely known. Here, we explored the physiological metabolism in M . alternatus larvae that were fumigated with (+)-α-pinene, (-)-α-pinene, β-pinene, (R)-(+)-limonene, myrcene and (-)-α-phellandrene. (+)-α-Pinene showed the highest fumigant toxicity to M . alternatus larvae among these monoterpenes, and the fourth instar larvae possessed the highest resistance to this compound at the larval stage. Comparative transcriptome analysis detected 423 up-regulated and 311 down-regulated genes in the larvae fumigated with (+)-α-pinene, which were mainly related to detoxification, energy and protein metabolism. Among the differentially expressed genes encoding for detoxification enzymes, 14 P450s, 13 UGTs, 1GSTs and 3ABCs were dramatically up-regulated in the larvae fumigated with (+)-α-pinene. Silencing of P450s via RNA interference led to an increased mortality in the larval fumigated with (+)-α-pinene. Piperonyl butoxide exposure significantly increased the mortality and inhibited the expressions of P450s, UGTs and ABCs in the larvae fumigated with (+)-α-pinene. These results suggested an important role of P450s in the resistance of M . alternatus larvae to (+)-α-pinene fumigation, which may facilitate the understanding of terpenoid detoxification in M . alternatus larvae.

Functional Characterization of Terpene Synthases from Masson Pine (Pinus massoniana) under Feeding of Monochamus alternatus Adults

Quanmin Wen,Ruixu Chen,Tian Xu,Dejun Hao

Conifers have evolved sophisticated terpenoid defenses for protection against herbivores and pathogens.Pinus massonianaLamb. is the most widely distributed pioneer afforestation and resin tree species in China, but is seriously harmed by pine wilt disease.Monochamus alternatusis the main vector of pine wilt disease in China. Monoterpenes, sesquiterpenes and diterpenes, the main secondary defensive compounds ofP. massoniana, are catalyzed by different terpene synthases (TPSs), which participate in the important defense pathways against external biotic and abiotic stresses. Here, we aimed to identify the terpene synthases (TPSs) inP. massoniana, responding to the feeding ofM. alternatus, and to characterize the functions and products of the mono-TPSs. We identified six differentially expressed TPS genes in theP. massonianafed upon byM. alternatus, including four mono-TPS and two sesqui-TPS genes. The functions of the four mono-TPSs were verified by analysis of the main product and by-products of these mono-TPSs. (+)-α-Pinene, (−)-α-pinene, and limonene were the major products of TPS (+)-α-pinene, TPS (−)-α-pinene, and TPS limonene, respectively, but TPS (−)-β-pinene only catalyzed a trace amount of (−)-β-pinene in the products. Our findings shed light on the potential relationships between the structure of terpene synthases and their corresponding products.

Identification and Functional Analysis of an Epsilon Class Glutathione S-Transferase Gene Associated with α-Pinene Adaptation in Monochamus alternatus

Mingyu Xue,Xiaohong Xia,Yadi Deng,Fei Teng,Shiyue Zhao,Hui Li,Dejun Hao,Wei-Yi Chen

Alpha-pinene is one of the main defensive components in conifers.Monochamus alternatus(Coleoptera: Cerambycidae), a wood borer feeding on Pinaceae plants, relies on its detoxifying enzymes to resist the defensive terpenoids. Here, we assayed the peroxide level and GST activity ofM. alternatuslarvae treated with different concentrations of α-pinene. Meanwhile, agstgene (MaGSTe3) was isolated and analyzed. We determined its expression level and verified its function. The results showed that α-pinene treatment led to membrane lipid peroxidation and thus increased the GST activity. Expression ofMaGSTe3was significantly upregulated in guts following exposure to α-pinene, which has a similar pattern with the malonaldehyde level. In vitro expression and disk diffusion assay showed that the MaGSTe3 protein had high antioxidant capacity. However, RNAi treatment of MaGSTe3 did not reduce the hydrogen peroxide and malonaldehyde levels, while GST activity was significantly reduced. These results suggested MaGSTe3 takes part in α-pinene adaptation, but it does not play a great role in the resistance ofM. alternatuslarvae to α-pinene.Keywords:glutathione S-transferase;detoxification enzymes;monoterpene
