From association to intervention: Muribaculaceae driven SCFAs production enhances boar semen quality via inflammation alleviation

Liangliang Guo,Xiaoqi Pei,Jiajian Tan,Haiqing Sun,Siwen Jiang,Hongkui Wei,Jian Peng

The gut microbiota plays a vital role in host reproduction, yet its contribution to semen quality in boars remains unclear. In this study, we analyzed 556 boars from three commercial breeds and identified Muribaculaceae as a key microbial taxon positively associated with sperm quality, with the effect mediated by short-chain fatty acids (SCFAs). This association was validated in Yorkshire boars with extreme semen phenotypes. Fecal microbiota transplantation in mice confirmed that enrichment of Muribaculaceae improved semen quality, likely through enhanced SCFA production and reduced inflammation in the gut and reproductive tract. Furthermore, in vitro fermentation and mouse experiments demonstrated that a designed functional fiber selectively promoted Muribaculaceae , increased SCFA levels, and improved sperm quality. These findings suggest a functionally supported and potentially translational association between gut microbiota and boar fertility, suggesting that targeted dietary modulation of Muribaculaceae may represent a novel strategy to enhance reproductive performance in livestock.

Muribaculaceae regulate β-muricholic acid to target S1PR2 signaling and reduce neurological injury in intracerebral hemorrhage

Song Liu,Yi Lv

Background Intestinal flora imbalance after intracerebral hemorrhage (ICH) aggravates neuroinflammation and secondary brain injury through the gut-brain axis, although the specific mechanism remains unclear. This study focuses on the regulatory effects of Muribaculaceae and β-muricholic acid (β-MA, a primary bile acid) on neurological injury after ICH, aiming to reveal the molecular mechanism by which it improves the prognosis of ICH through the sphingosine-1-phosphate receptor 2 (S1PR2). Methods A mouse ICH model was constructed by collagenase induction to evaluate the changes in gut microbiota diversity and metabolites. After intervention with Muribaculum intestinale (MI), neurological function was assessed by behavioral tests, and pathological changes of brain tissue were analyzed by Hematoxylin-Eosin and Nissl staining. Subsequently, intestinal barrier function, inflammatory factors, and total bile acid (TBA) levels were examined in ICH mice. In addition, cell viability, apoptosis, oxidative stress, inflammatory factors, and β-MA levels were analyzed in the heme-induced SH-SY5Y cell model. Molecular docking and drug affinity responsiveness target stability (DARTS) were used to analyze the interaction between β-MA and S1PR2. Intervention with a S1PR2 agonist (CYM-5520) was used to further verify the mechanism. Results Altered gut microbiota composition, elevated lipopolysaccharide levels, reduced expression of tight junction proteins, inflammatory activation, and disrupted bile acid metabolism were observed in ICH mice. Supplementation with MI ameliorated neural damage, cerebral edema, and neuronal loss in an intake dose-responsive manner, enhanced intestinal barrier integrity, and increased TBA levels. As a primary bile acid, β-MA directly mitigated hemin-induced oxidative stress and inflammation in neural cells. Mechanistically, β-MA downregulated the expression of S1PR2, but overexpression of S1PR2 counteracted the protective effects of β-MA. Furthermore, the administration of CYM-5520 attenuated the neuroprotective effects conferred by MI in vivo . Conclusion Muribaculaceae alleviated neurological injury after ICH by upregulating β-MA levels, thereby inhibiting the S1PR2 signaling pathway. This research offers a novel approach to treating ICH by focusing on the gut microbiota-bile acid metabolism-neuroprotection axis.

Interaction between a Sulfated Polysaccharide from Sea Cucumber and Gut Microbiota Influences the Fat Metabolism in Rats

Yujiao Zhang,Haoran Song,Zhengqi Liu,Chunqing Ai,Chunhong Yan,Xiuping Dong,Shuang Song

Due to its significant physiological effects, a sulfated polysaccharide has been considered an important nutrient of sea cucumber, but its metabolism in vivo is still unclear. The present study investigated the metabolism of a sea cucumber sulfated polysaccharide (SCSP) in rats and its influence on the metabolite profiles. The quantification by HPLC-MS/MS revealed that the blood level of SCSP achieved a maximum of 54.0 ± 4.8 μg/mL at 2 h after gavage, almost no SCSP was excreted through urine, and 55.4 ± 29.8% of SCSP was eliminated through feces within 24 h. These results prove the utilization of SCSP by gut microbiota, and a further microbiota sequencing analysis indicated that the SCSP utilization in the gut was positively correlated with Muribaculaceae and Clostridia_UCG-014. In addition, the non-targeted metabolomic analysis demonstrated the significant effects of SCSP administration on the metabolite profiles of blood, urine, and feces. It is worth noting that the SCSP supplement decreased palmitic acid, stearic acid, and oleic acid in blood and urine while increasing stearic acid, linoleic acid, and γ-linolenic acid in feces, suggesting the inhibition of fat absorption and the enhancement of fat excretion by SCSP, respectively. The present study shed light on the metabolism in vivo and the influence on the fat metabolism of SCSP.Keywords:marine polysaccharides;intestinal flora;nontargeted metabolomics

Optimization of a gelatin/carboxymethylcellulose-based probiotic microcapsule and its application in preventing dextran sodium sulfate–induced colitis in mice

Xiao Zhu,Yiming Zhi,Xiaoyi Heng,Lihui Zhou,Chang Liu,Yiwen Zhao,Yijun Wang,Jun Liu,Jinbao Huang

Oral administration of probiotics has demonstrated substantial potential in alleviating colitis. However, most of the ingested microorganisms struggle to survive the harsh conditions of the gastrointestinal tract, leading to decreased efficacy. In the present study, using double emulsification (W 1 /O/W 2 ) and complex coacervation methods, we developed a gelatin/carboxymethyl cellulose (CMC)-based probiotic microcapsule and analyzed the efficacy of encapsulated probiotics in preventing dextran sodium sulfate (DSS)-induced colitis in mice. Our results reveal that nearly 90% of the encapsulated probiotics remained viable after 30-day storage at 4°C and approximately 38.1% of viable bacteria (4.0 × 10 8 cfu/g) survived after 4-h simulated gastrointestinal digestion. In a DSS-induced colitis model, pretreatment with probiotics exerted significant protective effects, with the bacterial microcapsule-treated group having superior outcomes to those of the bacterial suspension plus empty carrier group. Probiotic treatments, especially those administered in the encapsulated form, significantly increased fecal short-chain fatty acid contents, and altered the intestinal microbial composition. The family Muribaculaceae , dominant bacteria in the mouse gut, may be the key microorganism involved in the BM regulation process. Our study presents an alternative approach to treating colitis using probiotics. Practical Application The encapsuled probiotic showed remarkable storage stability at 4°C, maintained good vitality after simulated digestion, and gained superior outcomes in preventing colitis. Our results offer an alternative approach for the probiotic preparations aiming to prevent the intestinal inflammation.

Impact of structurally diverse polysaccharides on colonic mucin O-glycosylation and gut microbiota

Zhao Tong,Zhang Yue,Nan Linhua,Zhu Qing,Wang Shukai,Xie Yutao,Dong Xinling,Cao Cui,Lin Xiaoliang,Lu Yu,Liu Yuxia,Huang Linjuan,Gong Guiping,Wang Zhongfu

Understanding how dietary polysaccharides affect mucin O -glycosylation and gut microbiota could provide various nutrition-based treatments. Here, the O -glycan profile of the colonic mucosa and gut microbiome were investigated in C57BL/6J mice fed six structurally diverse dietary polysaccharides and a mixture of six fibers. Dietary polysaccharides increased total O -glycans, mainly by stimulating neutral glycans. Highly branched arabinogalactan promoted terminally fucosylated core 1 O -glycans; whereas linear polysaccharides, including pectin, konjac glucomannan, inulin, and the fiber mixture, favored terminally di-fucosylated O -glycans. The last three polysaccharides also lowered the level of sulfated O -glycans and sialylated mono-fucosylated O -glycans. Varied monosaccharide composition in mixed polysaccharides had a synergistic beneficial effect, boosting fucosylated neutral glycans, decreasing acidic glycans, and stimulating microbial richness and diversity. Dietary polysaccharides containing arabinose and sulfate groups enhanced the relative abundances of Akkermansia and Muribaculaceae , respectively. The present comparison reveals the relationship between dietary polysaccharide structure, mucin O -glycan composition, and intestinal microorganisms.

Tibetan highland barley fiber improves obesity and regulates gut microbiota in high-fat diet-fed mice

Linyao Gan,Jing Han,Chenyao Li,Jing Tang,Xuebing Wang,Yue Ma,Yefu Chen,Dongguang Xiao,Xuewu Guo

Tibetan highland barley fiber (T-Fiber) contains high dietary fiber (DF) and beta-glucose. Highland barley has been shown to reduce the risk of diseases associated with high-fat diets (HFD). In the present research, we studied whether T-Fiber could ameliorate HFD-induced obesity in mice and explored its regulatory effects on intestinal microbes . Our findings revealed that T-Fiber feeding significantly reduced weight gain and dyslipidemia , improved glucose tolerance and insulin resistance, alleviated chronic systemic inflammation, and increased short-chain fatty acids (SCFA) in feces in mice. Histological observations confirmed that T-Fiber alleviated Epididymal fat (EF) and lipid accumulation in the liver. In addition, 16s rRNA sequencing analysis of mice cecum contents showed that T-Fiber restored the HFD-induced gut microbiota. T-Fiber supplementation increased the abundance of potentially beneficial bacteria Muribaculaceae , Akkermansiaceae , and Lachnospiraceae_NK4A136_groups , while significantly reducing the abundance of Bacteroidaceae , Prevotellaceae , Rikenellaceae , and Alloprevotella . Furthermore, the increase in the production of SCFA may be related to the abundance of Lachnospiraceae_NK4A136_group . In conclusion, T-Fiber has some anti-obesity abilities under high-fat feeding and can be developed as a relevant anti-obesity product.

Benzyl Isothiocyanate and Resveratrol Synergistically Alleviate Dextran Sulfate Sodium-Induced Colitis in Mice

Jianan Liu,Qian Zhang,Hongshun Hao,Jingran Bi,Hongman Hou,Gongliang Zhang

The aim of our study was to investigate whether the combination of benzyl isothiocyanate (BITC) and resveratrol (RES) has a synergistic effect on the inhibition of inflammation in colitis. The results revealed that the BITC and RES combination (BITC_RES) was more effective than either substance alone at significantly alleviating the symptoms of dextran sodium sulfate (DSS)-induced colitis in mice, including the prevention of colon shortening and loss of body weight, a reduction in the disease activity index, and prevention of colon damage. Similarly, compared with the DSS group, BITC_RES reduced myeloperoxidase (MPO) and inducible nitric oxide synthase (iNOS) levels in the mouse colon by 1.4–3.0-fold and 1.4-fold, respectively. In addition, the combination of BITC and RES upregulated the inflammatory factor IL-10 by 1.3- and 107.4-fold, respectively, compared to the individual BITC and RES groups, whereas the proinflammatory factors, including TNF-α, IL-1β, and IL-6, were downregulated by 1.1–7.4-, 0.7–3.6-, and 0.6–2.6-fold, respectively, in the BITC_RES group compared with the individual groups. Gut microbiome analysis indicated that BITC_RES remodeled the structure of gut bacteria at the phylum, family, and genus levels, upregulating the abundance of the phylum Bacteroidetes and the familyMuribaculaceaeand the genus norank_f_Muribaculaceaeand downregulating the abundance of the phylum Firmicutes. Significant correlations between the relative levels of these proinflammatory cytokines and changes in the gut microbiota were found using Pearson’s correlation analysis. BITC and RES exhibited synergistic effects by reshaping the gut microbiota and modulating the level of serum cellular inflammatory factors, thus exerting a protective effect against colitis.
