Proanthocyanidins (PAs) are important phenolic compounds and PA biosynthesis is regulated by a ternary MBW complex consisting of a R2R3-MYB regulator, a bHLH factor and a WDR protein. In this study, a tobacco R2R3-MYB factor NtMYB330 was characterized as the PA-specific regulator in which the PA biosynthesis was promoted in the flowers of NtMYB330-overexpressing lines while decreased in the flowers of ntmyb330 mutants. NtMYB330 can interact with flavonoid-related bHLH partner NtAn1b and WDR protein NtAn11-1, and the NtMYB330-NtAn1b complex is required to achieve strong transcriptional activation of the PA-related structural genes NtDFR1, NtANS1, NtLAR1 and NtANR1. Our data reveal that NtMYB330 regulates PA biosynthesis in seeds and affects seed germination, in which NtMYB330-overexpressing lines showed higher PA accumulations in seed coats and inhibited germination, while ntmyb330 mutants had reduced seed coat PAs and improved germination. NtMYB330 affects seed germination possibly through two mechanisms: modulating seed coat PAs to affect coat-imposed dormancy. In addition, NtMYB330 regulates the expressions of abscisic acid (ABA) and gibberellin acid (GA) signaling-related genes, affecting ABA-GA crosstalk and seed germination. This study reveals that NtMYB330 specifically regulates PA biosynthesis via formation of the MBW complex in tobacco flowers and affects germination through adjustment of PA concentrations and ABA/GA signaling in tobacco seeds.

Strawberries ( Fragaria × ananassa ) are prized for bioactive flavonoids and anthocyanins, contributing to antioxidant capacity and health benefits. However, decades of breeding for yield and shelf-life have eroded their phytochemical diversity. To explore this trade-off, we compared wild strawberries ( Fragaria nilgerrensis , HM) with three cultivated varieties (pink-FY, red-RF, white-DX) using integrated LC-MS/MS metabolomics and transcriptomics. Wild HM strawberries contained twice the vitamin C (134.2 mg/100g), 2.5× more total flavonoids (138.7 mg/100g), and higher antioxidant activity (367 μg Trolox/g) than commercial varieties. In contrast, red-fruited RF showed abundant anthocyanins (pelargonidin-3-glucoside 6.6% abundance), while the wild HM genotype accumulated higher levels of flavonoids, with neohesperidin, lonicerin, hyperin and ternatumoside II collectively representing 20.9% of total flavonoids - concentrations 3-169 times greater than those observed in the three commercial varieties (FY, RF, DX). Transcriptomic analysis (RNA-seq, Illumina HiSeq 2500) revealed distinct MYB transcription factor associations: FxaYL_611g0111200 (MYB66) showed strong positive correlation with anthocyanin biosynthesis in RF (r=0.92, **p<0.005 ), while FxaYL_242g0472670 (MYB44-like) was significantly associated with HM's unique antioxidant flavonoid profile (r=0.96, ***p<0.0001 ). These regulatory relationships were corroborated by hierarchical clustering demonstrating varietal-specific expression patterns and principal component analysis (82.6% variance explained), collectively providing molecular targets for enhancing nutritional quality in strawberry breeding programs.

Background (TF) R2R3-MYB transcription factors (TFs) play crucial roles in regulating plant secondary metabolism, which is vital for growth, development, and stress responses. Salvia miltiorrhiza , a medicinal plant, is known for its bioactive compounds such as salvianolic acids. Understanding the R2R3-MYB TF family in S. miltiorrhiza could provide insights into the regulation of secondary metabolism and contribute to quality improvement in this important medicinal plant. Methods and Results We conducted a comprehensive analysis of the R2R3-MYB TF family in S. miltiorrhiza , identifying 431 MYB-like TFs, including 149 R2R3-MYB TFs. Phylogenetic analysis classified these TFs into 27 distinct groups. Chromosomal localization analysis revealed their widespread distribution across all chromosomes. Structural analysis showed that most R2R3-SmMYB genes contain untranslated regions (UTRs) and conserved motifs, with motif5 potentially playing a key role in differentiation. Interspecific collinearity analysis demonstrated the highest collinearity between S. miltiorrhiza and Salvia baicalensis , with 12 intraspecific collinear genes exhibiting interspecific collinearity with multiple species. Promoter analysis identified abundant light- and hormone-responsive elements, particularly MeJA-responsive elements. Expression profiling highlighted SmMYB50 as highly responsive to MeJA, ABA, and YE induction, promoting salvianolic acid accumulation. Transgenic validation confirmed that SmMYB50 activates key biosynthetic genes such as SmCYP98A14 , crucial for SalB synthesis. Conclusions Our findings provide valuable insights into the functional roles of R2R3-SmMYB-TFs in regulating secondary metabolism in S. miltiorrhiza . SmMYB50 was identified as a key regulator, offering new directions for future research on metabolic pathways and quality improvement in this important medicinal plant.

As the most abundant renewable energy source , wood comprises the secondary cell wall (SCW). SCW biosynthesis involves lignin and cellulose deposition. Increasing studies have illustrated that R2R3-MYB transcription factors (TFs) play pivotal roles in affecting lignin accumulation and SCW formation. Nevertheless, the regulatory roles of R2R3-MYBs are still unresolved in Cryptomeria fortunei Hooibrenk cambium and wood formation. To dissect the potentials of CfMYBs , we successfully cloned and intensively studied the functions of CfMYB4 and CfMYB5 in SCW formation and abiotic stress response. They both contained the conserved MYB domain capable of forming a special structure that could bind to the core motifs of downstream genes. The phylogenetic tree implied that two CfMYBs clustered into different evolutionary branches. They were predominantly expressed in the stem and were localized to the nucleus. Furthermore, CfMYB4 functioned as an activator to enhance lignin and cellulose accumulation, and increase the SCW thickness by elevating the expression levels of SCW-related genes. By contrast, CfMYB5 negatively regulated lignin and cellulose biosynthesis, and decreased SCW formation by reducing the expression of SCW biosynthetic genes. Our data not only highlight the regulatory functions of CfMYBs in lignin deposition but also provide critical insights into the development of strategies for the genetic improvement of Cryptomeria fortunei wood biomass.

MYB proteins play a crucial role in plant growth and development and stress responses. In this study, 160 members of the MYB gene family from the pepper genome database were used to analyze gene structures, chromosome localization, collinearity, genetic affinity and expression in response to heavy metals . The results identified R2R3-MYB members and further phylogenetically classified them into 35 subgroups based on highly conserved gene structures and motifs. Collinearity analysis showed that segmental duplication events played a crucial role in the functional expansion of the CaMYB gene family by intraspecific collinearity, and at least 12 pairs of CaMYB genes existed between species prior to the differentiation between monocots and dicots . Moreover, the upstream CaMYB genes were mainly localized to the phytohormone elements ABRE and transcription factor elements MYB and MYC. Further analysis revealed that MYB transcription factors were closely associated with a variety of abiotic stress-related proteins (e.g., MAC-complex and SKIP). Under the stress of five metal ions, Cd 2+ , Cu 2+ , Pb 2+ , Zn 2+ , and Fe 3+ , the expression levels of some CaMYB family genes were upregulated. Of these genes, pairing homologous 1 ( PH-1 ), PH-13 , and PH-15 in the roots of Capsicum annuum were upregulated to the greatest extent, indicating that these three MYB family members are particularly sensitive to these five metals. This study provides a theoretical reference for the analysis of the molecular regulatory mechanism of MYB family genes in mediating the response to heavy metals in plants. This study reveals the mode of interaction between MYB and a variety of abiotic stress proteins and clarifies the biological functions of CaMYB family members in the regulation of heavy metal stress.

Phenolic compounds are vital bioactive constituents in fruits, yet modern strawberry breeding has often reduced their diversity. Here, we employed a multi-omics approach integrating UPLC-MS/MS-based metabolomics and RNA-seq transcriptomics to investigate the divergence in phenolic profiles and their transcriptional regulation between a wild strawberry ( Fragaria nilgerrensis , HM) and three cultivated varieties (white ‘Danxue’ (DX), pink ‘Fenyu’ (FY), and red ‘Red Face 99’ (RF)). The wild HM genotype exhibited higher antioxidant activity and a significantly more complex phenolic profile, dominated by high-abundance galloylated and benzoylated glucosides (e.g., digallic acid methyl ester, salicylic acid-2- O -glucoside) that were largely absent or depleted in cultivated fruits. In contrast, the cultivated varieties displayed specialized yet simplified profiles: DX accumulated hydroxycinnamoyl galactonic acids, FY was enriched in feruloylated glucosides, and RF was characterized by coumaroyl-glucose derivatives. Transcriptomic analysis identified a set of MYB transcription factors (e.g., FxaYL_531g0581170 , FxaYL_642g0175720 ) significantly upregulated in wild HM, with strong correlations to key bioactive phenolics such as 4-hydroxybenzoate and salicylic acid derivatives. These findings illustrate how selective breeding has reshaped phenolic composition through alterations in MYB regulatory networks. The wild strawberry germplasm thus represents a valuable natural reservoir for biofortification strategies aimed at restoring the nutritional and functional quality of modern strawberry cultivars.

Background Lavender (genus Lavandula, family Lamiaceae) is an aromatic plant widely grown as an ornamental plant. The chemical composition of lavender is characterized by monoterpenoids, sesquiterpenoids, and other compounds, which are primarily synthesized and stored in epidermal secretory structures called glandular trichomes (GTs). Volatile organic compounds (VOCs) are responsible for the aroma characteristics of plant oil that drive consumer preference. Aroma is usually regarded as a characteristic trait for the classification of aromatic plants. Interestingly, VOCs are synthesized and stored in GTs. Lamiaceae species such as purple perilla, peppermint, basil, thyme, and oregano usually possess two types of GTs: peltate glandular trichomes (PGTs) and capitate glandular trichomes (CGTs). But the development process of PGTs in lavender has been reported in only a few studies to date. Results In this study, we identified and quantified the VOCs in four lavender cultivars by headspace-solid phase micro extraction-gas chromatography mass spectrometry (HS–SPME–GC–MS). A total of 66 VOCs were identified in these four cultivars, the most prominent of which were linalyl acetate and linalool, and flowers were the main site of accumulation of these VOCs. Here, we examined the developmental process of PGTs, including the formation of their base, body, and apex. The apex cells contained secretory cavities, which produced VOCs. Based on the reference genome sequence of the lavender cultivar ‘Jingxun 2’, several R2R3-MYB subfamily genes related to GT formation were identified. These results will guide the engineering of GTs and molecular breeding of lavender for improving the VOC content. Conclusion sIn this study, we identified the VOCs in four lavender cultivars. We analyzed the formation of GTs, and compared the number and diameter size of PGTs among four lavender cultivars. Additionally, we identified four candidate genes belonging to the R2R3-MYB family.

Seed oil content and fatty acid (FA) composition collectively determine the quality and economic value ofBrassica napus. Little is known about the role of R2R3-MYB transcription factors (TFs) in regulating FA biosynthesis inB. napus. Here,BnaC04.MYB89was found to be expressed primarily in developing seeds. Overexpression ofBnaC04.MYB89consistently decreased FA levels, as evidenced by its effect in both theArabidopsis thaliana myb89-1mutant andB. napusseeds. RNA-seq of developing seeds at 30 DAP (days after pollination) revealed marked suppression of FA biosynthetic genes inBnaC04.MYB89-overexpressing plants compared to the K407 control. ChIP (Chromatin immunoprecipitation) analysis revealed that BnaC04.MYB89 directly inhibited the expression ofBnaA03.BCCP1andBnaC03.HDwhile indirectly regulating that ofBnaA09.BADC1,BnaA03.BADC3,BnaA03.MOD1, andBnaA08.FAT8, thereby reducing seed FA accumulation. Collectively, these results elucidate the role for BnaC04.MYB89 and provide new insights into the transcriptional regulatory network controlling seed oil accumulation inB. napus.

The aim of the study is to explore the protective effect of new gelatin (NG, Xin’ejiao in China) on hematopoietic injury caused by chemotherapy. Zebrafish, at 48 hours post fertilization (hpf), was treated with different chemotherapeutic drugs to establish the zebrafish hematopoietic damage model with reduced thrombocytes and erythrocytes. The protecting effects of NG on the thrombocytes and erythrocytes were observed, respectively, on zebrafish models. Then, the RT-PCR method was used to detect the change of mRNA level of the hematopoiesis-related cytokines scl1, c-myb, pu.1, GATA1, and runx1 genes. The results showed that 50 μg·mL−1 and 100 μg·mL−1 NG rescued and increased the thrombocytes numbers induced by vinorelbine (NVB) and chloramphenicol (CHL) and the erythrocytes numbers induced by methotrexate (MTX), doxorubicin (ADM), and mechlorethamine hydrochloride (MH) in zebrafish models. Meanwhile, the mRNA expression of scl1, c-myb, and GATA1 genes in the NG treatment group was raised compared with the MTX treatment group. Also, the mRNA expression of pu.1 and Runx1 in the NG treatment group was reduced compared with the MTX treatment group. In consequence, traditional Chinese medicine NG showed a certain degree protective effect on hematopoiesis injury induced by chemotherapy in this study, which may depend on the promotion of erythrocytes proliferation and the regulation of the hematopoietic genes level.

Salvia miltiorrhiza Bunge is an herb rich in bioactive tanshinone and salvianolic acid compounds. It is primarily used as an effective medicine for treating cardiovascular and cerebrovascular diseases. Liposoluble tanshinones and water-soluble phenolic acids are a series of terpenoids and phenolic compounds, respectively. However, the regulation mechanism for the simultaneous promotion of tanshinone and salvianolic acid biosynthesis remains unclear. This study identified a R2R3-MYB subgroup 20 transcription factor (TF), SmMYB98 , which was predominantly expressed in S. miltiorrhiza lateral roots. The accumulation of major bioactive metabolites, tanshinones, and salvianolic acids, was improved in SmMYB98 overexpression (OE) hairy root lines, but reduced in SmMYB98 knockout (KO) lines. The qRT-PCR analysis revealed that the transcriptional expression levels of tanshinone and salvianolic acid biosynthesis genes were upregulated by SmMYB98-OE and downregulated by SmMYB98-KO . Dual-Luciferase (Dual-LUC) assays demonstrated that SmMYB98 significantly activated the transcription of SmGGPPS1 , SmPAL1 , and SmRAS1 . These results suggest that SmMYB98-OE can promote tanshinone and salvianolic acid production. The present findings illustrate the exploitation of R2R3-MYB in terpenoid and phenolic biosynthesis, as well as provide a feasible strategy for improving tanshinone and salvianolic acid contents by MYB proteins in S. miltiorrhiza .

Cold stress is a critical environmental factor adversely affecting plant growth and development. As a tropical-origin crop constituting the primary global source of sucrose, sugarcane ( Saccharum spp. hybrid) exhibits particular vulnerability to suboptimal temperature conditions, with chilling injury substantially compromising its yield potential. Despite its agricultural significance, the molecular mechanisms underlying cold acclimation in sugarcane remain poorly characterized. Here, we report a cold-repressed 1R-MYB gene, ScMYB7 , from sugarcane, whose promoter ( pro-ScMYB7 ) contains multiple cis -acting elements, including two cytosine—phosphate diester—guanine (CpG) islands. Bisulfite sequencing PCR (BSP) and qPCR results showed that low-temperature treatment increased the methylation level of the CpG islands in the promoter to reduce the transcription of the ScMYB7 gene. The outcomes of GUS enzyme activity measurement of the promoter also indicated that low-temperature treatment inhibits the promoter's transcriptional activity, and methylation inhibitors could alleviate this inhibition. By generating transgenic Arabidopsis lines overexpressing ScMYB7 , ScMYB7's roles in regulating cold tolerance were investigated. We observed that the transgenic plants reduced cold tolerance, featured by a decreased survival rate after recovery, fluctuated physiological traits, and significantly lower expression levels of the C-repeat binding factor (CBF)-dependent pathway genes ( AtCBFs , AtCOR15 , and AtRD29A ). Yeast one-hybrid assays demonstrated direct binding of ScMYB7 to the AtCBF1 promoter, while repression of sugarcane ScDREB1A occurred indirectly. Furthermore, the dual-luciferase reporter assay indicated that ScMYB7 was able to inhibit the expression of the AtCBF1 or ScDREB1A . Taken together, we propose a model in which ScMYB7 acts as a repressor of cold tolerance via the CBF-dependent pathway. Under low-temperature stress, increased methylation of the pro-ScMYB7 promoter reduces ScMYB7 expression, thereby alleviating its repression of sugarcane DREB/CBF-type transcription factors and enhancing cold adaptation.

Scutellaria baicalensis , a traditional medicinal plant originating in China, is widely cultivated for its therapeutic properties. The main bioactive substances in S. baicalensis are flavonoids, which exhibit extensive antibacterial and antiviral activities. However, the contents of these valuable natural product ingredients are relatively low in the plant. MYB transcription factors play crucial roles in regulating plant secondary metabolism, including flavonoid biosynthesis. While the regulation of MYB transcription factors has been extensively studied in various species, research on their role in S. baicalensis remains relatively scarce. In this study, we identified SbMYB111, belonging to the S7 subgroup of R2R3-MYB transcription factors, which functions as a transcriptional activator and is localized in the nucleus. Through heterologous overexpression of SbMYB111 in Arabidopsis thaliana and suppression expression in S. baicalensis , we demonstrated that SbMYB111 acts as a positive regulator in the biosynthesis of flavonoids. Furthermore, the yeast one-hybrid and dual-luciferase reporter gene assays validated that SbMYB111 activates the expression of SbC4H2 , a key enzyme gene in the flavonoid biosynthesis pathway. This study provides a theoretical basis for understanding the transcriptional regulation mechanism of flavonoid synthesis and further developing medicinal resources of S. baicalensis .
