Myrica rubra(MR) is rich in anthocyanins, and it has good anti-cancer, anti-aging, antioxidant, and antiviral effects. The proportion of disability and death caused by ischemic stroke gradually increased, becoming a major disease that is harmful to human health. However, research on effects of anthocyanin from MR on cerebral ischemia-reperfusion (I/R) injury is rare. In this study, we prepared eight purified anthocyanin extracts (PAEs) from different types of MR, and examined the amounts of total anthocyanin (TA) and cyanidin-3-O-glucoside (C-3-G). After one week of PAE treatment, the cerebral infarction volume, disease damage, and contents of nitric oxide and malondialdehyde were reduced, while the level of superoxide dismutase was increased in I/R mice. Altogether, our results show that Boqi1MR contained the most TA (22.07%) and C-3-G (21.28%), and that PAE isolated from Dongkui MR can protect the brain from I/R injury in mice, with the mechanism possibly related to the Toll-like receptor 4 (TLR4)/ nuclear factor-κB (NF-κB) and NOD-like receptor pyrin domain-containing 3 protein (NLRP3) pathways. Keywords:Myrica rubra;anthocyanins;cyanidin-3-O-glucoside;cerebral ischemia-reperfusion;TLR4/NF-κB;NLRP3

The Myrica rubra pomace polyphenols (MRPP) were used to study their hypoglycemic effects and mechanisms using Type 2 diabetes (T2D) (db/db) mice as the model. The results showed that the oral administration of MRPP (low-dose myricitrin, 50 mg/kg BW by gavage; high-dose myricitrin, 250 mg/kg BW by gavage; and Myrica rubra pomace, 500 mg/kg BW by gavage) for 4 weeks significantly reduced fasting blood glucose, glycated serum protein, serum insulin, and insulin resistance index in mice, as well as the dyslipidemia in mice was improved. MRPP was able to improve the structural morphology of hepatocytes and pancreatic β-cells in diabetic mice. Its hypoglycemic mechanism may involve the upregulation of GLUT-4 and IRS-1 genes expression in the PI3K and AMPK signaling pathways, downregulation of GSK-3β, AMPK, PI3K, and AKT genes expression, and enhancement of the activity of enzymes related to glycogen synthesis and glucose metabolism. Research findings on 16S rRNA suggest that MRPP has the ability to alter the composition of the gut microbiota, impede the growth of harmful bacteria, and foster the growth of beneficial bacteria. Therefore, MRPP was capable of reshaping the specific gut microbial community and supporting its application as a novel supplement in functional foods for the treatment of T2D.

The anti-obesity effects of different proanthocyanidin extracts (PE) from Chinese bayberry (Myrica rubra Sieb.et Zucc.) leaves were evaluated in a high-fat (HF) diet-induced obese rat model. The leaves of Chinese bayberry (LCB), a non-desaccharified proanthocyanidin extract (NDPE), and a desaccharified proanthocyanidin extract (DPE) were compared with orlistat, a conventional anti-obesity drug used as a positive control. The rats from all of the PE-treated groups showed obvious weight loss, with the greatest weight loss in the DPE group (P < 0.05). Compared to the HF group, almost all of the PE-treated groups showed significantly reduced levels of serum TC, TG, AST, ALT, ALP, LDL-C, ADP and a LEP/ADP ratio, and increased levels of HDL-C and LEP. Furthermore, the expression levels of SIRT1, PPAR-γ, C/EBP-α, BMP4 and UCP1 were investigated in the liver, kidney, epididymis and abdominal adipose tissue. Compared to the HF group, PPAR-γ and C/EBP-α levels were significantly reduced and SIRT1, BMP4 and UCP1 levels were significantly upregulated in the DPE-treated group. Our results suggested that PE exerted its anti-obesity effects by upregulating the expression of SIRT1, thus inducing the deacetylation of PPAR-γ and downregulating the expression of C/EBP-α, and also upregulating the expression of BMP4 to boost the levels of brown fat.

Twig blight disease is the primary disease that affects the production ofMyrica rubrain China. It was reported that exogenous brassinolide (BL) can improve disease resistance in plants. Here, we examined the effects of exogenous BL on disease resistance, chlorophyll contents, antioxidant enzyme activity, ROS accumulation, and key gene expression ofM. rubrato analyze the mechanism of BR-induced resistance of twig blight disease inM. rubra. The results demonstrated that 2.0 mg·L−1of BL could significantly lessen the severity of twig blight disease inM. rubra. Exogenous BL increased the contents of chlorophyll a, chlorophyll b, carotenoids, and total chlorophyll. Moreover, exogenous BL also significantly enhanced the activity of antioxidant enzymes such as superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT), and decreased malondialdehyde (MDA) content and reactive oxygen species (ROS) accumulation in leaves, such as H2O2and O2·−. Additionally, exogenous BL dramatically up-regulated the expression of pathogenesis-related (PR) genes such asMrPR1,MrPR2, andMrPR10, as well as important genes such asMrBAK1,MrBRI1, andMrBZR1involved in brassinosteroid (BR) signaling pathway. The transcriptome analysis revealed that a total of 730 common differentially expressed genes (DEGs) under BL treatment were found, and these DEGs were primarily enriched in four Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways. Based on these findings, nine important candidate genes related to the resistance of twig blight disease under BL treatment were further identified. In this study, we elucidated the effects of exogenous BL on enhancing the resistance ofM. rubrato twig blight disease and preliminary analyzed the potential mechanism of resistance induction, which will provide a crucial foundation for the management and prevention of twig blight disease inM. rubra. Keywords:Myrica rubra;exogenous brassinolide;twig blight disease;activity of antioxidant enzymes

The increasing rate of hyperlipidemia has drawn great attention and natural products may play a potential role in prevention of these disorders. Chinese bayberry leaves proanthocyanidin (BLPs) with epigallocatechin-3- O -gallate (EGCG) as its terminal and major extension units is unusual in the plant kingdom. Crude, purified and ultrasound depolymerized BLPs (CBLPs, SPBLPs and UDBLPs) were prepared with the aim to investigate their hypolipidemic mechanisms. BLPs inhibited the lipase activity in vitro and reduced the accumulation of total triacylglycerols (TAG) and cholesterol induced by oleic acid in HepG2 cells. SPBLPs and UDBLPS significantly increased the phosphorylation of AMP-activated protein kinase (AMPK) and thus reduced TAG and sterols biosynthesis by inhibiting its downstream proteins, such as acetyl-CoA carboxylase, 3-hydroxy-3-methylglutaryl-CoA reductase and sterol regulatory element-binding protein. BLPs also promoted glucose consumption in oleic acid induced HepG2 cells. Overall, this study suggests the hypolipidemic property and promotion of glucose consumption of BLPs via AMPK pathway.

Myrica rubrapomace accounts for 20% of the fruit’s weight that is not utilized when it is juiced. The pomace contains bioactive phenolic substances such as anthocyanins and flavonoids. To improve the utilization value ofMyrica rubrapomace, an optimized extraction method for the residual polyphenols was developed using response surface methodology (RSM). The resulting extract was analyzed by high performance liquid chromatography (HPLC), and the in vitro hypoglycemic activity and antioxidant activity of the polyphenolic compounds obtained were also investigated. The optimum extraction conditions (yielding 24.37 mg·g−1total polyphenols content) were: extraction temperature 60 °C, ultrasonic power 270 W, ethanol concentration 53%, extraction time 57 min, and solid to liquid ratio 1:34. Four polyphenolic compounds were identified in the pomace extract by HPLC: myricitrin, cyanidin-O-glucoside, hyperoside, and quercitrin. DPPH and hydroxyl radical scavenging tests showed that theMyrica rubrapolyphenols extract had strong antioxidant abilities. It is evident that the residual polyphenols present inMyrica rubrapomace have strong hypoglycemic activity and the juiced fruits can be further exploited for medicinal purposes. Keywords:Myrica rubraresidue;diabetes management;polyphenols;α-Glucosidase inhibition;HPLC

The development of cheap, efficient and stable hydrogen-producing catalysts is the current research focus, especially the catalysts that can work stably in the whole pH range. In this work, a myrica rubra-like structure of P–Co 1-x S/C was synthesized by hydrothermal reaction, sulfurization and phosphorization using spherical flower-like cobalt glycerate (CoG) as precursor. The design of this multilevel structure of P–Co 1-x S/C effectively increases the specific surface area. The doping phosphorus and the protection of carbon layer greatly improve catalytic performance and enhance the stability of Co 1-x S in the hydrogen evolution reaction (HER) at full pH range. When the current density is 10 mA cm −2 , the HER overpotentials of P–Co 1-x S/C in acidic, alkaline and neutral solutions are 86, 93 and 167 mV, and respectively. The Tafel slopes of P–Co 1-x S/C in acidic, alkaline and neutral solutions are 82, 100 and 103 mV dec −1 , respectively. In addition, the electrocatalyst also displays excellent electrochemical long-term stability. After 2000 cycles or 20 h long-term testing, its catalytic activity remains stable without obvious decrease at full pH range.

Tree age influences plant secondary metabolism, but its effect on antioxidant components in Chinese bayberry leaves remains unclear. Combining chemical assays and widely targeted metabolomics, we analyzed bayberry leaves of different ages, including young (<100 years) and mature (∼300, ∼400, and ∼500 years old) groups. We found that the contents of polyphenols and flavonoids significantly increased with tree age, which in turn enhanced their antioxidant activity. The 500-year-old leaves exhibited the highest activity. Metabolomic analysis identified a total of 3152 metabolites, with terpenoids, flavonoids, and phenolic acids being the predominant classes, collectively representing 50.44% of the total identified metabolites (20.43%, 20.11%, and 9.90%, respectively). Subsequent multivariate analysis revealed that the “Subclass 1” metabolites-comprising phenolic acids, flavonoids, and terpenoids-were positively correlated with antioxidant activity. Furthermore, two compounds, picfeltarraenin X and isothankunic acid, were identified in Chinese bayberry leaves for the first time. These results demonstrate that tree age enhances antioxidant capacity by regulating the accumulation of key secondary metabolites, predominantly terpenoids, flavonoids, and phenolic acids, thereby providing a scientific basis for the high-value utilization of bayberry leaves.

Protein glycation and formation of advanced glycation end-products (AGEs) impose threats to the human health. This study firstly investigated the inhibition of Chinese bayberry ( Myrica rubra ) phenolics on AGEs formation through mechanistic analysis. Four common Chinese bayberry cultivars were selected to prepare phenolic-rich extracts (CBEs) and characterized for phenolic composition, and their anti-AGE properties were evaluated in multiple in vitro systems. Total sixteen phenolics were quantified in CBEs by UPLC-ESI-MS/MS. CBEs reduced total and specific fluorescent AGEs formation in various simulating models, and protected the protein from structural modification, oxidation, and cross-linking. Mechanistic analysis unveiled that scavenging of free radicals, inactivation of transition metals, interaction with protein to form complexes, and trapping of reactive α-dicarbonyls to form adducts underlain the mechanisms of the anti-glycative actions of CBEs. Chinese bayberry fruits, especially the cultivars Biqi and Wuzi, may be a promising dietary strategy to mitigate AGEs load in the human body.

α-Glucosidase is a key enzyme related to starch digestion and type-2 diabetes. In the present study, the inhibition effect and the underlying mechanism of Chinese bayberry leaves proanthocyanidins (BLPs) on α-glucosidase were investigated by enzyme kinetic analysis, multi-spectroscopy and molecular docking simulation. The results revealed that BLPs was a high potential noncompetitive-type inhibitor of α-glucosidase with the half maximal inhibitory concertation (IC50) value of 0.037 ± 0.001 mg mL −1 and acarbose equivalent (AE) of 517.01 mmol AE g −1 . What’s more, BLPs may interact with some amino acids surrounded in the allosteric site of α-glucosidase to form a complex driven by hydrogen bonding and hydrophobic interaction, and thus change the structure and microenvironment of α-glucosidase, leading to the decrease of activity of α-glucosidase. The present study suggested that BLPs as epigallocatechin-3-O-gallate (EGCG) polymers could be a novel α-glucosidase inhibitor and had potential to be further used in functional food or anti-diabetic drug.

Background: The hypoglycemic effects of Chinese bayberry leaves proanthocyanidins (BLPs) have been demonstrated. It is unclear, nevertheless, whether BLPs reduced postprandial blood glucose levels by regulating glucose uptake and glucose transport.
Method: This study investigated the effect of BLPs (25, 50 and 100 μg/mL) on glucose uptake and glucose transport in human intestinal epithelial cells (Caco-2 cells). The uptake of 2-Deoxy-2-[(7-nitro-2,1,3-benzoxadiazol-4-yl) amino]-D-glucose (2-NBDG) and disaccharidases activity in Caco-2 cells were measured. The glucose transport ability across the cell membrane was determined using the established Caco-2 monolayer model. The transcript and protein levels of key glucose transporters were analyzed using real-time quantitative polymerase chain reaction (RT-qPCR) and western blotting, respectively.
Results: The results showed that BLPs significantly decreased glucose uptake and disaccharidases activity (p<0.05). Otherwise, BLPs treatment obviously inhibited glucose transport across the Caco-2 monolayer in both simulated-fast (5 mM glucose) and simulated-fed (25 mM glucose) conditions. It was attributed to the suppression of glucose transporter2 (GLUT2) and sodium-dependent glucose cotransporter 1 (SGLT1) by BLPs. BLPs were found to significantly downregulated the transcript level and protein expression of glucose transporters (p<0.05). Meanwhile, the mRNA expression of phospholipase C (PLC) and protein kinase C (PKC) involved in the signaling pathway associated with glucose transport were decreased by BLPs.
Conclusion: These results suggested that BLPs inhibited intestinal glucose transport via inhibiting the expression of glucose transporters. It indicated that BLPs could be potentially used as a functional food in the diet to modulate postprandial hyperglycemia.

To enable more sustainable utilization of waxberry pomace, this study developed an ultrasound-assisted deep eutectic solvent (UAE-DES) method for efficiently recovering total phenolic content (TPC) and total anthocyanin content (TAC). Based on physicochemical properties and extraction efficiency, choline chloride–lactic acid (ChCl–LA, 1:2) was identified as the best system. Low-field nuclear magnetic resonance (LF-NMR) and Fourier-transform infrared spectroscopy (FTIR) revealed the DES's hydrogen-bonding architecture restricted molecular mobility, explaining its superior solvation capacity. Optimization by single-factor and response surface methodology (RSM) gave the optimization conditions, including ultrasonication for 58 min at 360 W (16W/L), a solid-liquid ratio of 1:35 g/mL, and a water volume fraction of 36% (v/v). Under these settings, TPC and TAC reached 44.26 mg GAE/g DW and 6.92 mg C3GE/g DW, respectively. Furthermore, comprehensive UPLC-QTOF-MS profiling identified 11 major bioactive constituents in the extract. In comparative antioxidant assays, the DES extract markedly outperformed the ethanol extract, with IC 50 values of 0.025 mg/mL for DPPH•, 0.038 mg/mL for ABTS• + , 0.114 mg/mL for •OH, and 0.675 mg/mL for total antioxidant capacity (T-AOC). These results indicated ultrasound-assisted ChCl-LA extraction offered a sustainable and tunable strategy that achieved both high phytochemical recovery and preserved antioxidant functionality from waxberry pomace.
