Myocardial infarction (MI) is the primary source of death in cardiovascular diseases. Myricitrin (MYR) is a phenolic compound known for its antioxidant properties. This study aimed to investigate the impact of MYR alone or combined with exercise on a rat model of MI and its underlying mechanism. Sprague-Dawley rats were randomized into 5 groups: sham-operated (Sham), MI-sedentary (MI-Sed), MI-exercise (MI-Ex), MI-sedentary + MYR (MI-Sed-MYR) and MI-exercise + MYR (MI-Ex-MYR). MI was induced through ligation of left anterior descending coronary artery. The treatment with exercise or MYR (30 mg/kg/d) gavage began one week after surgery, either individually or in combination. After 8 weeks, the rats were assessed for cardiac function. Myocardial injuries were estimated using triphenyltetrazolium chloride, sirius red and Masson staining. Changes in reactive oxygen species (ROS) levels, mitochondrial membrane potential (ΔΨm), apoptosis and Nrf2/HO-1 pathway were analyzed by ROS kit, JC-1 kit, TUNEL assay, western blot and immunohistochemistry. Both MYR and exercise treatments improved cardiac function, reduced infarct size, suppressed collagen deposition, and decreased myocardial fibrosis. Additionally, both MYR and exercise treatments lowered ROS production induced by MI, restored ΔΨm, and attenuated oxidative stress and apoptosis in cardiomyocytes. Importantly, the combination of MYR and exercise showed greater efficacy compared to individual treatments. Mechanistically, the combined intervention activated the Nrf2/HO-1 signaling pathway. These findings suggest that the synergistic effect of MYR and exercise may offer a promising therapeutic approach for alleviating MI.

Diospyros lotus L. leaves are used as a functional tea and folk medicine in several Asian countries. This study aimed to evaluate the effects of phenolic extracts of the leaves on scavenging nitrite and inhibiting N-nitrosamines (NAs) formation and to determine the active ingredients responsible for these effects. Of the 7 fractions (Fr1-Fr7) prepared from the extract of D. lotus leaves, Fr5 contained the highest phenolic content and exhibited the strongest activity. Five active ingredients of Fr5 were discovered, and 4 of them were identified as myricitrin (Mytr), myricetin (Myt), myricetin-3-O-β-d-glucoside (Myt-Glc), and myricetin-3-O-β-d-galactoside (Myt-Gal). The content of Mytr was much higher than those of the other 3 ingredients, both in Fr5 and extracts of D. lotus leaves. Finally, Mytr and Myt were proved to have stronger activities by the 1,1-diphenyl-2-trinitrophenyl hydrazine scavenging, nitrite scavenging, and inhibition of NAs formation assays. These results indicated that Mytr was the main active ingredient of D. lotus leaves. Myt, Mytr, and Fr5 from the leaves could be used as natural agents for antioxidant, nitrite scavenging, and inhibition of NAs formation in food and the human body.

Myricitrin has many pharmacological effects, such as anti-inflammation, liver protection and anti-oxidation. However, its clinical application is limited by poor solubility and low oral bioavailability. The preparation of myricitrin-loaded proliposomes (MPs) was achieved via the combination of thin-film dispersion technique and freeze-drying method. The in vitro release of MPs compared with free myricitrin was measured in different dissolution media while the pharmacokinetic study was also conducted in rats. Moreover, the uric acid-lowering activity of MPs was investigated in the hyperuricemic rat model. The prepared myricitrin appeared to be spherical. Notably, compared with the free myricitrin, the cumulative release in vitro and in vivo oral bioavailability of MPs were markedly increased. Besides, the MPs could significantly lower the serum uric acid level as well as ameliorate liver and kidney damage in hyperuricemic rats compared with the model group. Therefore, the present work supports the fact that MPs improved the oral bioavailability of myricitrin for the prospect of clinical application.

Background: Aging involves progressive functional decline driven by oxidative stress and mitochondrial dysfunction, and dietary flavonoids are promising aging interventions due to their multi-target regulatory pathways. Methods: This study systematically evaluated the anti-aging activity of myricitrin (MYR) and its underlying mechanisms usingCaenorhabditis elegans, testing MYR at 0.25 mg/mL, employing a rotenone-induced model of mitochondrial oxidative stress, and performing reporter strain analyses, qRT-PCR, and molecular docking analysis with acetylcholinesterases (ACE-1/2). Results: MYR at 0.25 mg/mL significantly extended lifespan and improved locomotor performance without detectable toxicity regarding growth, feeding, or reproduction; reduced reactive oxygen species accumulation and restored antioxidant enzyme activities in the rotenone-induced model; was accompanied by the nuclear translocation of DAF-16 and the upregulation of associated downstream antioxidant genes (sod-3,gst-4); enhanced mitochondrial quality control by upregulatingsir-2.1and modulating genes associated with the mitochondrial unfolded protein response (atfs-1,hsp-6) and mitochondrial dynamics (fzo-1,drp-1); and indicated a favorable interaction between MYR and acetylcholinesterases (ACE-1/2). Conclusions: MYR represents a promising candidate compound for mitigating aging-related phenotypes, with its beneficial effects linked to a DAF-16-associated antioxidant response and SIR-2.1-correlated mitochondrial quality control network.

Objective: To explore the effect and underlying mechanism of Myricitrin (Myr) in regulating fibroblast-like synoviocyte (FLS)-mediated synovitis and joint destruction in RA. Methods: FLSs were isolated from synovial tissues from patients with RA. Gene expression was measured using quantitative RT-qPCR. Protein expression was detected by immunohistochemistry or Western blot. Cell apoptosis was performed by an Annexin-PI staining assay. EdU incorporation was used to assess the proliferation of RA FLS. Transwell assay was used to characterize the cell migration and invasion ability of RA FLS. The potential target of Myr was identified by RNA sequencing analysis. The in vivo effect of Myr was assessed in a collagen-induced arthritis (CIA) model. Results: Myr treatment inhibited the lamellipodia formation, migration, and invasion, but not the apoptosis and proliferation, of RA FLSs. Myr also reduced the expression of CCL2, IL-6, IL-8, MMP-1, MMP-3, and MMP-13 induced by TNF-α. The RNA-seq results indicated that AIM2 may be a target gene of Myr in RA FLSs. Furthermore, compared to healthy controls, AIM2 expression showed higher levels in synovial tissues and FLSs from RA patients. AIM2 knockdown also inhibited RA FLS migration, invasion, cytokine, and MMP expression. In addition, either Myr treatment or AIM2 knockdown reduced the phosphorylation of AKT induced by TNF-α stimulation. Importantly, Myr administration relieved arthritis symptoms and inhibited AIM2 expression in the synovium of CIA mice. Conclusion: Our results indicate that Myr exerts an anti-inflammatory and anti-invasion effect in RA FLSs and provide evidence of the therapeutic potential of Myr for RA.

Polyphenols have been widely recognized for their ability to modulate the host inflammatory response and orchestrate the intestinal microenvironment. In this study, epigallocatechin gallate (EGCG), dihydromyricetin-3′-O-sulfate (DMS), and myricitrin were identified as the major bioactive polyphenols in bayberry leaves and subsequently isolated with high purity (≥98%) by preparative liquid chromatography. Among them, myricitrin exhibited strong anti-inflammatory effects without cytotoxicity in Caco-2 cells by suppressing NF-κB activation and downstream inflammatory cytokines. In a DSS-induced colitis model, myricitrin effectively alleviated disease symptoms, preserved colonic tissue integrity, promoted goblet cell activity, and reduced inflammation-related cytokines. It also reshaped gut microbiota composition by enriching beneficial genera such as Akkermansia and Ligilactobacillus . Molecular dynamics simulations further revealed stable hydrogen bonding between myricitrin and NF-κB at key residues, providing mechanistic support for its anti-inflammatory activity. Together, these findings highlight high-purity myricitrin as a promising functional food ingredient for intestinal inflammation intervention and microbiota-targeted modulation.

Myricitrin, a naturally occurring polyphenol hydroxy flavonoid, has been reported to possess anti-inflammatory properties. However, the precise molecular mechanism of myricitrin’s effects on LPS-induced inflammation is unclear. In the present study, myricitrin significantly alleviated acute lung injury in mice. Myricitrin also markedly suppressed the production of NO, TNF-α, IL-6, and MCP-1 in RAW264.7 macrophage cells. The inhibition of NO was concomitant with a decrease in the protein and mRNA levels of iNOS. The phosphorylation of JAKs and STAT-1 was abrogated by myricitrin. Furthermore, myricitrin inhibited the nuclear transfer and DNA binding activity of STAT1. The JAK-specific inhibitor ruxolitinib simulated the anti-inflammatory effect of myricitrin. However, myricitrin had no impact on the MAPK signalling pathway. Myricitrin attenuated the generation of intracellular ROS by inhibiting the assembly of components of the gp91phox and p47phox. Suppression of ROS generation using NAC or apocynin or by silencing gp91phox and p47phox all demonstrated that decreasing the level of ROS inhibited the LPS-induced inflammatory response. Collectively, these results confirmed that myricitrin exhibited anti-inflammatory activity by blocking the activation of JAKs and the downstream transcription factor STAT1, which may result from the downregulation of NOX2-dependent ROS production mediated by myricitrin.

Inflammatory bowel disease (IBD) is closely associated with gut microbiota imbalance and intestinal barrier dysfunction. Bayberries improve intestinal health, with myricitrin (MYRR) as their main active component. However, the mechanism by which MYRR modulates the “microbiota–metabolite–host” axis to alleviate ulcerative colitis remains unclear. This study employs a dextran sodium sulfate (DSS)-induced colitis mouse model, integrating metagenomics, metabolomics, and colonic organoid cultures to explore MYRR's effects. The results demonstrate that MYRR alleviates colitis symptoms and restores the expression of key intestinal barrier proteins, MUC2 and occludin. Multi-omics analysis reveals that MYRR reshapes the gut microbiota by enriching beneficial bacteria and modulating bile acid metabolism, particularly allocholic acid (ACA), in both serum and feces. In vitro intestinal organoid experiments further confirm that ACA enhances MUC2 expression. In conclusion, MYRR protects against colitis by regulating the “microbiota–metabolite–intestinal barrier” axis, offering a theoretical foundation for microbiota-based therapeutic strategies for IBD.
