Shucking is an indispensable step in the preparation of cooked mussel products, as it facilitates the detachment of meat from the shell. In this study, we comprehensively investigated the effects of boiling, steaming, and microwaving on taste constituents in half-cooked mussel meat. Two-dimensional correlation spectroscopy revealed the key differential taste components of the different shucking groups. Structural equation modeling (SEM) indicated the positive effects of saltiness and bitterness on umami taste, while sweetness and sourness had negative effects on umami taste in half-cooked mussel meat. Furthermore, Glu, Asp, Ala, Arg, betaine, malic acid, succinic acid, glycogen, Cl − , Na + , K + , and PO3- 4 were quantitatively determined as the main taste compounds. The steaming shelling group had the most enriched taste components, with the highest equivalent umami concentration compared to the other shelling groups. Hence, steaming shucking may be favored due to abundant tastes and nutrients.

Perfluorooctanoate (PFOA) is widely used as a surfactant and has metabolic, immunologic, developmental, and genetic toxicity on marine organisms. However, the effects of PFOA on individual defense functions in mussels in the presence of titanium dioxide nanoparticles (nano-TiO 2 ) are poorly understood. To investigate the defense strategies and regulatory mechanisms of mussels under combined stressors, the thick-shell mussels Mytilus coruscus were exposed to different PFOA concentrations (0, 2 and 200 μg/L) and nano-TiO 2 (0 and 0.1 mg /L, size: 25 nm) for 14 days. The results showed that, compared to the control group, PFOA and nano-TiO 2 significantly reduced the number of byssal threads (NBT), byssal threads length (BTL), diameter of proximal threads (DPB), diameter of middle threads (DMB), diameter of distal byssal threads (DDB), adhesive plaque area (BPA), and breaking force of byssal threads (N). Under the influence of PFOA and nano-TiO 2 , the morphological surface smoothness of the fractured byssal threads surface increased, concurrently inducing an increased surface roughness in the adhesive plaques. Additionally, under the presence of PFOA and nano-TiO 2 , the foot displayed dispersed tissue organization and damaged villi, accompanied by an increased incidence of cellular apoptosis and an upregulation of the apoptosis gene caspase-8 . Expression of the adhesion gene mfp-3 and byssal threads strength genes ( preCOL-D, preCOL-NG ) was upregulated. An interactive effect on the performance of byssal threads is observed under the combined influence of PFOA and nano-TiO 2 . Under co-exposure to PFOA and nano-TiO 2 , the performance of the byssal threads deteriorates, the foot structure is impaired, and the genes mRNA expression of byssal thread secretory proteins have compensated for the adhesion and byssal threads strength by up-regulation. Within marine ecosystems, organic and particulate contaminants exert a pronounced effect on the essential life processes of individual organisms, thereby jeopardizing their ecological niche within community assemblages and perturbing the dynamic equilibrium of the overarching ecosystem. Environmental Implication Perfluorooctanoic acid (PFOA) is prone to accumulate in marine organisms. TiO 2 nanoparticles (nano-TiO 2 ) are emerging environmental pollutants frequently found in marine environment. The effects of PFOA and nano-TiO 2 on marine mussels are not well understood, and their toxic mechanisms remain largely unknown. We investigated the impacts of PFOA and nano-TiO 2 on mussel byssus defense mechanisms. By assessing byssus performance indicators, morphological structures of the byssus, subcellular localization, and changes in byssal secretion-related genes, we revealed the combined effects and mechanisms through which these two types of pollutants may affect the functional capabilities and survival of mussels in the complex marine ecosystem.

Triphenylphosphine oxide (TPPO) as a new organophosphorus flame retardant is extensively used in the industrial and pharmaceutical fields due to its chemical stability and abundance. In marine environments, TPPO accumulates as a result of natural ecological processes. This study aimed to evaluate the accumulation process and toxicological effects of TPPO utilizing mussels ( Mytilus coruscus ), a species traditionally employed in marine monitoring and commonly cultivated in offshore aquaculture. GC–MS/MS was employed to quantify the accumulation of TPPO in the gill, gonad, muscle, and digestive gland tissues of mussels exposed to three sublethal concentrations (20.0, 100.0, and 500.0 μg/L) over durations of 3, 7, 14, and 28 days, as well as the examining of the chemical and molecular parameters. Results showed that the accumulation of TPPO (100.0 and 500.0 μg/L) increased throughout the 28 d of exposure, and the peak accumulation of 20.0 μg/L TPPO was detected on 14 d. Short-term TPPO exposure did not cause oxidative stress in mussels. Chronic exposure for 28 days resulted in significant oxidative damage and oxidative stress in mussels, as evidenced by elevated expression and activity of SOD, increased levels of glutathione and malondialdehyde, and upregulation of HSP70 and HSP90, metallothionein, and CYP4Y1, alongside the initiation of apoptotic cell death. TPPO exposure activated natural immune genes and disrupted lipid metabolism in mussels. This study demonstrated the persistent accumulation of TPPO in mussels, highlighting its ecotoxicological hazards and underscoring the necessity to predict the risks associated with widespread environmental exposure to TPPO.

The carotenoid profiling of Mytilus coruscus generally depends on the dietary microalgae, and the ingested carotenoids are modified and processed depending on the properties of different tissues. Although much research effort has focused on analyzing carotenoid profiling between microalgae and shellfish, the detailed metabolic pathways of dietary carotenoids in shellfish are poorly understood. In the present study, we carried out feeding experiments with different unialgal cultures ( Chaetoceros mulleri and Dicrateria inornata ) during the growth stages and examined the carotenoid profiles between the dietary microalgae and the corresponding tissues in order to study the resorption and metabolic transformations of dietary carotenoids in M. coruscus . The results showed that 1) carotenoids mainly occurred in the pre-spawning phase of M. coruscus; the contents of most carotenoids showed the same trends as in dietary microalgae. 2) An association analysis of the total content of carotenoids and relevant gene expression demonstrated that the main tissue for carotenoid synthesis in M. coruscus was the hepatopancreas . The synthetic reactions of the allenic to acetylenic carotenoids pathway and the 4-keto oxidative pathway in M. coruscus coexisted. 3) Quantitative analysis revealed that the carotenoids mytiloxanthin, alloxanthin, and pectenolone containing acetylene groups were selectively assimilated in gonad (69.3–74.1%), mantle (83.2–102%), and muscle (86.9–100%), forming a large percentage of the carotenoids present in those tissues. These results provide new insights into the biosynthetic and metabolic transformation pathways of carotenoids in M. coruscus .

Peroxiredoxins (Prxs) are crucial antioxidant enzymes that protect against oxidative damage and participate in immune responses. However, the immunological function of Prx5 in bivalves remains unclear. In this study, the peroxiredoxin gene McPrx5 was cloned from Mytilus coruscus , and its molecular characteristics and biological functions were analyzed. A 1491 bp cDNA sequence was obtained, containing a 573 bp open reading frame (ORF) that encodes a 190-amino-acid protein (20.29 kDa) with features of an atypical 2-Cys Prx. Tissue distribution analysis revealed that McPrx5 was highly expressed in the foot, gill, and digestive gland. Immune challenges with LPS, poly I:C, and Perkinsus olseni induced dynamic changes in McPrx5 expression. Subcellular localization analysis of the McPrx5-GFP fusion protein showed that McPrx5 was exclusively localized in the cytoplasm. Recombinant McPrx5 enhanced the tolerance of Escherichia coli BL21 to H₂O₂, confirming its antioxidant activity. Importantly, in vivo experiments demonstrated that McPrx5 negatively regulates the expression of key molecules in the TLR4 / MyD88 / NF-κB signaling pathway, as well as FOXO transcription factors, suggesting its important role in linking redox homeostasis with immune regulation. These findings clarify the biological functions of McPrx5 in M. coruscus and provide a new basis for understanding the coordinated regulatory mechanisms linking antioxidant defense to innate immunity in bivalves.

C-type lectins (CTLs) play crucial roles in immune defense and feeding regulation, serving as key mediators of pathogen recognition and nutrient uptake in invertebrates. Although previous studies have demonstrated the involvement of CTLs in the immune response of Mytilus coruscus , their role in feeding regulation remains unclear. In this study, we identified a C-type lectin, designated MytiCTL17A , from M. coruscus and investigated its dual function in immune defense and feeding behavior. The purified recombinant MytiCTL17A binds to LPS and PGN. Additionally, it binds to both Gram-positive and Gram-negative bacteria, promoting bacterial agglutination. Notably, this agglutination can be inhibited by PAMPs. These findings confirm that rMytiCTL17A functions as a pattern recognition receptor (PRR), recognizing PAMPs, binding to bacteria, and promoting their agglutination, thereby playing a role in immune defense. Additionally, rMytiCTL17A also exhibits direct antibacterial activity. In feeding experiments, rMytiCTL17A can bind to microalgae and reduce the feeding rate of M. coruscus , indicating its involvement in the feeding process. Moreover, rMytiCTL17A can recognize carbohydrates (D-Man, D-Gal, Glu, Mal, and Suc) on the surface of microalgae and induce microalgal agglutination. Notably, this agglutination can be inhibited by D-Man, demonstrating its dependence on carbohydrate recognition. Thus, rMytiCTL17A also participates in feeding by recognizing microalgae through specific carbohydrates. This study highlights the multifunctionality of rMytiCTL17A and provides new insights into the roles of C-type lectins in bivalves, with potential implications for aquaculture in terms of enhancing disease resistance and feeding efficiency.

The increasing deposition of microplastics (MPs) in aquatic ecosystems is a worldwide concern. MPs can interact with other environmental pollutants, such as heavy metals, and change their toxicity. In this study, we focused on the effects of MPs and lead (Pb), as a toxic heavy metal, on marine mussel Mytilus coruscus under separate and co-exposure situations at environmentally relevant concentrations: MPs (1 mg/L) and Pb (50 μg/L). We found that MPs alone or in combination with Pb significantly decreased the respiration and filtration rates of the mussels ( p < 0.05). Histological observations revealed varying extents of damage to the gill and digestive gland caused by a single exposure to MPs, which was aggravated by co-exposure to Pb. In addition, co-exposure induced a higher level of oxidative stress, which was reflected by an increase in hydrogen peroxide and malondialdehyde content, and a decrease in antioxidant enzyme activity. Meanwhile, co-exposure poses a significant threat to the immune function of the mussels, as evidenced by induction of hemocytes to produce excess reactive oxygen species (ROS), significantly reducing lysosome activity ( p < 0.05), inhibiting the expression of autophagy-related genes, and inducing the expression of apoptosis-related genes, resulting in hemocyte apoptosis. Furthermore, the TLR/MyD88/NFκB signaling pathway is involved in the immune response of mussels to environmental stress. This study provides novel perspectives on the toxicity of MPs combined with Pb in marine animals, as well as the molecular mechanisms underlying their ecotoxicological effects.

In the marine environment, nanoparticles play a role in adsorbing and catalytically degrading organic pollutants, thereby mitigating their toxic effects on aquatic organisms. This study aimed to investigate the impact of nano titanium dioxide (nTiO 2 ) and tris (2-chloropropyl) phosphate (TCPP) on the hemolymph and digestive function of the thick-shell mussel Mytilus coruscus . Mussels were divided into a control group, a group exposed to TCPP alone, a group exposed to a combination of TCPP and 0.5 mg/L nTiO 2 , and a group exposed to a combination of TCPP and 1 mg/L nTiO 2 . After 14 days of exposure, oxidative stress responses, including superoxide dismutase (SOD) activity and malondialdehyde (MDA) content, immune defense responses, including acid phosphatase (ACP) and alkaline phosphatase (AKP) activities, and gene expression, including HSP70 expression, were measured in the hemolymph and digestive glands of the mussels. Compared to the control group, mussels solely exposed to 100 μg/L TCPP exhibited a significant reduction in SOD activity in the hemolymph. When TCPP was co-exposed with 0.5 mg/L nTiO 2 , there were significant increases in MDA content and AKP activity in both the digestive gland and hemolymph compared to the control group. Upon co-exposure of TCPP with 1 mg/L nTiO 2 , MDA content and AKP activity in the digestive gland significantly decreased, while SOD, ACP, and AKP activity in the hemolymph significantly increased and MDA content significantly decreased, returning to the control group levels. Furthermore, in the combined exposure, HSP70 gene expression significantly decreased as the nTiO 2 concentration increased from 0.5 mg/L to 1 mg/L. In summary, TCPP impacted the hemolymph and digestive function of mussels, whereas a concentration of 1 mg/L nTiO 2 effectively alleviated the toxic effects of TCPP. This study is crucial for assessing the ecological risks of nanoparticles and emerging organic pollutants in marine environments, and provides new insights into the interaction between nTiO 2 and TCPP, as well as the influence of nTiO 2 concentration on mitigating TCPP toxicity.
