Cry41-Related Mutants against Myzus persicae Based on Its Interaction with Cathepsin B

Liang Jin,Xiao-Di Zhao,Jun-Ao Liao,Li-Jun Fu,Yi Lin

Cry toxins produced by Bacillus thuringiensis (Bt) are toxic to Lepidoptera, Coleoptera, and Diptera, but display very low activity against aphids. Recently, Cry41-related toxin was found to show moderate toxicity against Myzus persicae with the 50% lethal concentration (LC50) of 32.7 μg/mL, and its mode of action was proposed to be the acceleration of the apoptosis of aphid cells by enhancing Cathepsin B activity. This study focused on constructing Cry41-related mutants against M. persicae based on its interaction with Cathepsin B. First, eight key interacting residues in Cry41-related toxin were identified using alanine scanning and site-directed saturation mutagenesis. Subsequently, the positive mutant Cry41-7M protein (mutations of Gly48, Ile59, Lys364, Gln367, Gln377, Tyr378, and Ser400 to Tyr, Ala, Arg, Lys, Lys, Lys, and Ala in Cry41-related toxin, respectively) and the negative mutant Cry41-6A protein (mutations of Gly48, Lys364, Gln367, Gln377, and Tyr378 to Ala and mutation of Pro453 to Glu in Cry41-related toxin) were constructed, expressed, and purified. We then found that Cry41-7M protein performed slightly stronger than Cry41-related toxin in enhancing the enzymatic activity of Cathepsin B, whereas Cry41-6A protein did not affect Cathepsin B activity. A further bioassay showed that the mortality caused by Cry41-7M protein (LC50 = 19.144 μg/mL) was marginally higher than that of Cry41-related toxin, while Cry41-6A protein caused a decreased mortality (LC50 = 42.478 μg/mL). These results are expected to open new avenues for improving Cry aphidicidal activity.

Possible Insecticidal Mechanism of Cry41-Related Toxin against Myzus persicae by Enhancing Cathepsin B Activity

Xiao-Di Zhao,Bin-Wu Zhang,Li-Jun Fu,Qi-Lin Li,Yi Lin,Xiao-Qiang Yu

Cry toxins produced by Bacillus thuringiensis are well known for their high insecticidal activities against Lepidoptera, Diptera, and Coleoptera; however, their activities against Aphididae are very low. Recently, it has been reported that a Cry41-related toxin exhibited moderate activity against the aphid Myzus persicae, and thus, it is highly desirable to uncover its unique mechanism. In this paper, we report that Cathepsin B, calcium-transporting ATPase, and symbiotic bacterial-associated protein ATP-dependent-6-phosphofructokinase were pulled down from the homogenate of M. persicae as unique proteins that possibly bound to Cry41-related toxin. Cathepsin B has been reported to cleave and inactivate antiapoptotic proteins and plays a role in caspase-initiated apoptotic cascades. In this study, Cathepsin B was expressed in Escherichia coli and purified, and in vitro interaction between recombinant Cathepsin B and Cry41-related toxin was demonstrated. Interestingly, we found that addition of Cry41-related toxin obviously enhanced Cathepsin B activity. We propose a model for the mechanism of Cry41-related toxin as follows: Cry41-related toxin enters the aphid cells and enhances Cathepsin B activity, resulting in acceleration of apoptosis of aphid cells.

Cytochrome P450 CYP6CY3 Mediated Cross-Resistance to Isocycloseram and Fluralaner in Myzus persicae

Ziyi Chen,Chunyan Yin,Falong Wang,Xinlin Sun,Wishwajith Kandegama,Zhenyu Wang,Guangfu Yang

The green peach aphid (Myzus persicae, Hemiptera: Aphididae) is a globally important pest. Isocycloseram (a novel isooxazoline) and fluralaner (a benzamide/sulfonamide) are novel insecticides. A Hainan-resistant population (HNR), exhibited 459- and 316-fold resistance to isocycloseram and fluralaner, respectively, compared with a Yunnan susceptible population (YNS). Synergist bioassays and enzyme activity analyses indicated that enhanced cytochrome P450 monooxygenase activity contributed to resistance. Transcriptomic and qRT-PCR analyses revealed that CYP6CY3 was upregulated 16.4- and 10.8-fold in HNR compared with YNS. RNA interference of CYP6CY3 significantly restored susceptibility to both insecticides. In vitro recombinant enzyme assays confirmed CYP6CY3-mediated metabolism of isocycloseram and fluralaner, and gene copy number analysis demonstrated a 7.09-fold amplification. These results reveal CYP6CY3-mediated cross-resistance to isocycloseram and fluralaner, representing the first mechanistic evidence of isoxazoline resistance in M. persicae.

Molecular characterization of a catalase gene from the green peach aphid (Myzus persicae)

Mao-Ye Li,Yun Wang,Xiao Lei,Chuan-Tao Xu,Dong-Dong Wang,Su Liu,Shi-Guang Li

The green peach aphid, Myzus persicae (Sulzer), is a serious agricultural pest with a worldwide distribution. Catalase (CAT), which is encoded by the catalase ( Cat ) gene, is an extremely important antioxidant enzyme that plays a pivotal role in protecting cells against the toxic effects of hydrogen peroxide. The Cat gene has not been characterized in M. persicae ; therefore, this study describes the identification of the Cat ( MpCat ) gene from M. persicae . MpCat contains an open reading frame of 1515 bp and encodes a MpCAT protein consisting of 504 amino-acid residues. MpCAT possesses features typical of other insect catalases, including 7 conserved amino acids involved in binding heme and 15 involved in binding nicotinamide adenine dinucleotide phosphate. Phylogenetic analysis showed that MpCAT was closely related to orthologs from other aphid species. MpCat consisted of nine exons and eight introns, and the number and insertion sites of introns are consistent with those of Cat genes from Acyrthosiphon pisum (Harris) and Aphis gossypii Glover. The mRNA transcripts of MpCat were detected at all tested developmental stages, with the highest mRNA level in alate adults. The expression of MpCat was significantly upregulated when M. persicae was exposed to low and high temperatures, ultraviolet radiation, Beauveria bassiana , and permethrin. The transcription of MpCat and the activity of catalase were suppressed by RNA interference, and knockdown of MpCat significantly reduced the survival rate in M. persicae under heat stress. The results provide valuable information for further study on the physiological functions of MpCat . Research Highlights A catalase gene ( MpCat ) was characterized in Myzus persicae . Transcription of MpCat was responsive to various environmental stresses. Knockdown of MpCat increased mortality in M. persicae under heat stress.

Inhibition of Nicotinamidase Impacts Growth, Development, and Lifespan of Drosophila melanogaster and Myzus persicae

Hailiang Guo,Wenjin Xia,Changyang Chen,Song Yang,Dongmei Sun,Jizu Qiao,Jianya Su

Nicotinamidase (Naam), which is essential for insect NAD synthesis but absent in mammals, represents a potential pesticide target. Virtual screening identified 1-[4-(trifluoromethyl)-3-pyridinyl]-ethanone (4-TFPE) as a potent inhibitor of Myzus persicae (IC50 = 4.156 μM) and Drosophila melanogaster (IC50 = 2.855 μM) Naam. While 4-TFPE reduced pupal weight, development, and longevity in both species, it showed no lethal toxicity. Crucially, Naam expression was upregulated by nicotinamide accumulation, revealing a feedback regulation mechanism that compromises its pesticidal potential. Furthermore, Flumetnicam’s neurotoxic effects were found to be Naam-independent. These findings highlight how target compensation mechanisms can undermine pesticide development focused on single metabolic enzymes, suggesting the need for multitarget strategies.

Delta-Class Glutathione S-Transferase Modulates the Susceptibility of Myzus persicae to λ-Cyhalothrin

Wang-Xi Shen,Jia-Min Yu,Xin-Yi Liu,Yu-Xing Zhang,Mao-Ye Li,De-Xin Chen,Su Liu

Glutathione S -transferases (GSTs) constitute a critical enzyme superfamily responsible for xenobiotic detoxification in insects. The green peach aphid, Myzus persicae , is a globally significant agricultural pest that has exhibited declining field susceptibility to λ-cyhalothrin (LCT), a widely applied pyrethroid insecticide. While elevated GST activity has been correlated with LCT tolerance in various aphid populations, the precise molecular mechanisms driving this process have not been fully elucidated. In this study, we functionally characterized a delta-class GST gene (designated MpGSTd1 ) from M. persicae . The encoded protein possesses hallmark structural motifs of the GST family, including distinct glutathione-binding (G-site) and hydrophobic substrate-binding (H-site) pockets. Transcriptional profiling revealed maximal MpGSTd1 expression during the late nymphal to adult stages, and its transcript levels were markedly induced following LCT challenge. Biochemical analysis of the recombinant MpGSTd1 protein confirmed its catalytic competency toward the model substrate CDNB, exhibiting a V max of 0.91 µmol/min/mg and a K m of 2.73 mM. Notably, LCT exposure inhibited the enzyme's activity in vitro with an IC 50 of 0.02 mM, and high-performance liquid chromatography (HPLC) assays demonstrated that MpGSTd1 can directly deplete LCT by 11.4%. Furthermore, RNA interference (RNAi)-mediated suppression of MpGSTd1 in vivo significantly elevated aphid mortality upon subsequent LCT exposure. Collectively, these findings delineate an essential role for MpGSTd1 in modulating LCT susceptibility in M. persicae and underscore its potential utility as a molecular target for future aphid management strategies. Summary A delta-class GST from Myzus persicae was characterized. MpGSTd1 transcription is induced by LCT exposure. Recombinant MpGSTd1 depletes LCT in vitro . Knockdown of MpGSTd1 increases insect susceptibility to LCT.

Multiple Insecticide Resistance and Associated Metabolic-Based Mechanisms in a Myzus Persicae (Sulzer) Population

Jinfeng Hu,Feng Chen,Jun Wang,Wenhua Rao,Lei Lin,Guocheng Fan

The green peach aphid,Myzus persicae(Sulzer) (Hemiptera: Aphididae), is an economically devastating crop pest worldwide. TheM. persicae(SEF-R) population of a cabbage field in China was tested for susceptibilities to 13 insecticides. Compared with the susceptible population (FFJ-S), extremely high and high resistance to beta-cypermethrin (324-fold) and imidacloprid (106.9-fold) was detected in SEF-R. More importantly, this is the first report of resistance in the fieldM. persicaepopulation to sulfoxaflor (32.4-fold), flupyradifurone (9.5-fold), pymetrozine (34.8-fold), spirotetramat (8.1-fold), flonicamid (5.8-fold), and broflanilide (15.8-fold) in China when compared with FFJ-S. The resistance factor decayed to a low level to sulfoxaflor and pymetrozine after 15 generations without any selection pressure. The resistance-related mutations (R81T and kdr) detected in SEF indicated target-site resistance to neonicotinoids and pyrethroids, respectively. Biochemical assays revealed the involvement of monooxygenase, carboxylesterase, superoxide dismutase, and peroxidase in a multi-insecticide resistance mechanism. The overexpression of P450s, esterases, and a UDP-glycosyltransferase might be responsible for the multi-insecticide resistance in SEF-R. The knockdown of CYP6CY3 in SEF-R increased its susceptibility to imidacloprid, thiacloprid, and thiamethoxam, which verified that P450s play vital roles in neonicotinoid metabolism. Our findings provide guidance for the rational use of insecticides to delay resistance development in GPA.Keywords:Myzus persicae;insecticides;resistance monitoring;detoxification enzymes;protective enzymes;metabolic mechanism

Identification and Functional Analysis of Differentially Expressed Genes in Myzus persicae (Hemiptera: Aphididae) in Response to Trans-anethole

Ding Chao-Yang,Ma Yu-Meng,Li Bin,Wang Yun,Zhao Le,Peng Jiang-Nan,Li Mao-Ye,Liu Su,Li Shi-Guang

Plant essential oils, with high bioactivity and biodegradability, provide promising alternatives to synthetic pesticides for pest control. Trans-anethole is the major component of essential oil from star anise, Illicium verum Hook. The compound has a strong contact toxicity against the green peach aphid, Myzus persicae (Sulzer) (Hemiptera: Aphididae), which is a major insect pest of many vegetables and crops. However, little information is known about how M. persicae responds to trans-anethole at the molecular level. We conducted a comparative transcriptome analysis of M. persicae in response to a LD50 dose of trans-anethole. A total of 559 differentially expressed genes were detected in the treated individuals, with 318 genes up-regulated, and 241 genes down-regulated. Gene ontology (GO) analysis revealed that these genes were classified into different biological processes and pathways. We also found that genes encoding ATP-binding cassette (ABC) transporters, DnaJ, and cuticle proteins were dramatically up-regulated in response to trans-anethole. To study the function of these genes, we performed RNA interference (RNAi) analysis. Knockdown of an ABC transporter gene (ABCG4) and a DnaJ gene (DnaJC1) resulted in a significantly increased mortality rate in M. persicae following trans-anethole exposure, indicating the involvement of these two genes in the toxicity response to trans-anethole. The findings provide new insights into the mechanisms of M. persicae in coping with plant essential oils.

V101I and R81T mutations in the nicotinic acetylcholine receptor β1 subunit are associated with neonicotinoid resistance in Myzus persicae

Xiao Xu,Qian Ding,Xiu Wang,Ruijie Wang,Farman Ullah,Xiwu Gao,Dunlun Song

BACKGROUND The peach-potato aphid, Myzus persicae (Sulzer) (Hemiptera: Aphididae), is a major pest worldwide. The intensive use of insecticides has led to the development of resistance against neonicotinoid insecticides. The R81T mutation in the nicotinic acetylcholine receptor (nAChR) beta1 subunit is considered a crucial mechanism adaptation to neonicotinoid resistance in M. persicae and Aphis gossypii . RESULTS Resistance-related mutations (R81T and V101I) were detected in the imidacloprid-resistant M. persicae AH19 population. The V101I mutation is reported for the first time. The V101I and R81T mutations existed separately, indicating that the two mutations evolved independently. Imidacloprid resistance in the AH19 population was stable without insecticide exposure. Four mutant strains were selected from the population with stable resistance. The resistance of the AH19-T, AH19-I, and AH19-TI strains to imidacloprid, thiamethoxam, and dinotefuran was significantly increased compared with the AH19-W strain. Synergism bioassays showed that the inhibition of three detoxification enzymes did not affect imidacloprid resistance in the AH19-T and AH19-I strains. Expression of nAChR β1 subunits in the AH19-W, AH19-T, and AH19-I strains remained unchanged. CONCLUSION The V101I mutation is associated with neonicotinoid resistance in M. persicae . The resistance of the AH19-T and AH19-I strains to neonicotinoids appears to be mainly due to the R81T and V101I mutations, whereas these mutations, together with changes in the cytochrome P450 monooxygenases and nAChR expression may be responsible for the development of neonicotinoid resistance in the AH19-TI strain. © 2021 Society of Chemical Industry.

A heat shock protein protects against oxidative stress induced by lambda-cyhalothrin in the green peach aphid Myzus persicae

Bao Dong,Xi-Ya Liu,Bin Li,Mao-Ye Li,Shi-Guang Li,Su Liu

Lambda-cyhalothrin (LCT) is a pyrethroid insecticide widely used to control insect pests . Insect exposure to LCT may cause abnormal accumulation of reactive oxygen species (ROS) and result in oxidative damage. Heat shock proteins (HSPs) may help protect against oxidative stress . However, little is known about the role of HSPs in response to LCT in the green peach aphid , Myzus persicae . This insect is an important agricultural pest causing severe yield losses in crops. In this study, we characterized a cDNA sequence ( MpHsp70 ) encoding a member of the HSP70 family in M. persicae . MpHsp70 encoded a 623 amino acid protein putatively localized in the cytosol. The highest expression level of MpHsp70 occurred in fourth-instar nymphs. Treatment of M. persicae with LCT resulted in oxidative stress and significantly increased H 2 O 2 and malondialdehyde levels. This led to an elevated transcription level of MpHsp70 . Injection of H 2 O 2 into M. persicae also upregulated the MpHsp70 expression level, suggesting that MpHsp70 is responsive to ROS, particularly H 2 O 2 , induced by LCT. Recombinant MpHSP70 protein was expressed in Escherichia coli . E. coli cells overexpressing MpHSP70 exhibited significant tolerance to H 2 O 2 and the ROS generators, cumene hydroperoxide and paraquat . This indicated that MpHSP70 protects against oxidative stress. Furthermore, knockdown of MpHsp70 by RNA interference resulted in increased susceptibility in apterous adults of M. persicae to LCT. These findings indicate that MpHsp70 plays an important role in defense against LCT-induced oxidative stress and insecticide susceptibility in M. persicae .

Mechanisms of first- to fourth-generation neonicotinoid resistance in Myzus persicae from Beijing peach orchards: Roles of target-site mutations and detoxification metabolism

Ren Li,Tao Su,Zehua Wang,Guanghang Qiao,Fan Yang,Yunlan Jiang,Shanning Wang

Myzus persicae is a globally destructive pest that has evolved severe resistance to neonicotinoids. However, the resistance status and underlying mechanisms in peach orchards of Beijing, China remain largely unknown. In this study, the resistance of six field populations collected from Beijing peach orchards against five neonicotinoids (representing first- to fourth-generation compounds) was investigated, and the molecular mechanisms were elucidated. Bioassays revealed resistance to imidacloprid (15.99–30.97-fold), acetamiprid (9.16–50.79-fold), sulfoxaflor (1.28–25.26-fold), thiamethoxam (6.12–16.95-fold), and dinotefuran (6.63–12.27-fold) across these populations. Mutation screening identified the known R81T and V101I mutations, along with a novel R81S substitution in the nAChR β1 subunit. Genotyping indicated that V101I single mutations were predominant (69.75% of individuals), while R81T/S + V101I combination mutations were detected in 22.56% of individuals. The results revealed that the R81T/S + V101I combination mutations confer substantially higher resistance (73.10–>1146.79-fold) than V101I alone (13.73–394.21-fold) using monoclonal strains. Enzyme activity assays demonstrated that elevated P450 (1.18–1.56-fold), GST (1.32–2.18-fold), and CarE (1.64–2.59-fold) activities are associated with the neonicotinoid resistance across all genotype backgrounds, with PBO showing the strongest synergistic effects (synergism ratios: 1.39–7.90). TPP and DEM exhibited synergism ratios ranging from 0.69 to 3.10. Molecular dynamics simulations showed that all mutations altered nAChR binding-pocket geometry, with combination mutations causing more pronounced structural perturbations and reduced imidacloprid binding affinity than V101I alone. These findings show that target-site mutations and detoxification enzymes act concertedly to drive neonicotinoid resistance in M. persicae , highlighting the need for insecticide rotation for sustainable management.
