Arbuscular Mycorrhiza Reduced Nitrogen Loss via Runoff, Leaching, and Emission of N2O and NH3 from Microcosms of Paddy Fields

Zhang Shujuan,Yun Wenfei,Xia Yu,Wu Sikai,You Zhaoyang,Matthias C. Rillig

This study aims to investigate the role of arbuscular mycorrhizal (AM) symbiosis in reducing N loss from paddy fields, using two rice lines: a mycorrhiza-defective rice line (non-mycorrhizal) and its mycorrhizal progenitor. Two rice lines were grown in the presence of an AM fungal isolate. In this study, N loss of runoff, leaching, N 2 O emission, and NH 3 volatilization were measured, and in addition, N uptake of rice, soil aggregates, and plant available N concentration of soil. The results obtained suggest that N loss via runoff, leaching, NH 3 volatilization, and N 2 O emission of mycorrhizal rice was 11%, 8%, 6%, and 1%, lower than that of non-mycorrhizal rice, respectively. Meanwhile, mycorrhizal rice has higher biomass and plant N uptake. Our study shows that the AM symbiosis contributes to the sustainability of rice production by reducing N loss, enhancing soil aggregation and increasing plant N uptake.

Potential Risk of Significant N2O Emission without Changing NOx Conversion on Commercial V2O5/TiO2 Catalyst under Working Conditions

Jiaying Xing,Qitong Xue,Jianjun Chen,Jinxing Mi,Xiaoping Chen,Jianqiang Shi,Zhiming Liu,Junhua Li

Vanadium-based catalysts play a pivotal role in the emission control of industrial NOx via selective catalytic reduction (SCR) technology. However, little attention has been paid to the potential emission of greenhouse gas N2O under complex working conditions. This work reports that a commercial V2O5/TiO2 catalyst may lead to significant N2O emission without greatly changing the outlet NOx concentration after chromium (Cr) deposition. With a Cr loading of 2 wt %, N2O concentration increased from 27.8 to 199.2 ppm at 350 °C with the value of outlet N2O/(N2O+N2) from 2.5% to 19.4%. Experimental results combined with DFT+U calculations suggest that nonselective catalytic reduction (NSCR) is the main route for N2O formation in a wide temperature range of 250 ∼ 400 °C. It is stemmed from the fact that the covalent interaction between Cr and V species on the V2O5/TiO2 surface accelerates the conversion of V4+ + Cr6+ → V5+ + Cr3+, leading to a larger proportion of surface V5+. More importantly, surface V5+ is highly related to the redox property of the V2O5/TiO2 catalyst, which is beneficial to NSCR reaction rather than the standard SCR process. The work suggests that to better inhibit the emission of greenhouse gases during the NH3–SCR process, monitoring N2O emission should be included along with the NOx concentrations, especially in complex flue gases.

Suitability of Mycorrhiza-Defective Rice and Its Progenitor for Studies on the Control of Nitrogen Loss in Paddy Fields via Arbuscular Mycorrhiza

Zhang Shujuan,You Zhaoyang,Guo Xinyue,Yun Wenfei,Xia Yu,Rillig Matthias C.

Employing mycorrhiza-defective mutants and their progenitors does not require inoculation or elimination of the resident microbial community in the experimental study of mycorrhizal soil ecology. We aimed to examine the suitability of mycorrhiza-defective rice (non-mycorrhizal, Oryza sativa L., cv. Nipponbare) and its progenitor (mycorrhizal) to evaluate nitrogen (N) loss control from paddy fields via arbuscular mycorrhizal (AM) fungi. We grew the two rice lines in soils with the full community of AM fungi and investigated root AM colonization. In the absence of AM fungi, we estimated rice N content, soil N concentration and microbial community on the basis of phospholipid fatty acids; we also quantified N loss via NH3 volatilization, N2O emission, runoff and leaching. In the presence of AM fungi, we did not find any evidence of AM colonization for non-mycorrhizal rice while mycorrhizal rice was colonized and percentage of root colonization was 17–24%. In the absence of AM fungi, the two rice lines had similar N content, soil N concentration and microbial community. Importantly, there was no significant difference in N loss via all the four pathways between mycorrhizal and non-mycorrhizal systems. This mycorrhizal/non-mycorrhizal rice pair is suitable for further research on the role of AM fungi in the control of soil N loss in paddy fields.

Nanoparticles of Zinc Oxides Mitigated N2O Emissions in Tea Plantation Soil

Jing Wang,Linfang Guo,Fengmin Yang,Jian Xiang,Lizhi Long,Kang Ni

The excessive application of nitrogen in tea plantations leads to severe soil acidification and N2O emission boosting. To promote sustainable agriculture, nanoparticles (NPs) have emerged as alternative fertilizers, but their effects on soil nitrification and greenhouse gas emissions in tea plantations remain unclear. In this study, the effects of NP type (ZnO-NPs and Fe2O3-NPs) and dose (0, 1, 10, and 100 mg·kg−1) on soil N2O emissions were investigated via a lab incubation trial. Soil pH, ammonium, and nitrate changes were also monitored during the incubation period. The abundance of functional genes related to nitrification and denitrification processes was analyzed as well. The results showed that ZnO-NPs led to a decrease in N2O emissions. The reduction effect was stronger with increasing dose and resulted in a 33% reduction at an addition rate of 100 mg·kg−1. The cumulative N2O emissions had significantly positive correlations with NH4+-N and NO3−-N. ZnO-NP addition showed a significantly negative effect on Ammonia-Oxidizing Archaea (AOA) but a positive effect on Ammonia-Oxidizing Bacteria (AOB) gene abundance. In contrast, Fe2O3-NPs showed an insignificant impact on N2O emissions and soil N content, as well as nitrification–denitrification gene abundance, regardless of different doses. These results imply that the application of ZnO-NPs may inhibit nitrification through the retarding of AOA activity. This study provided us with a potential practice to reduce N2O emissions in tea plantations by applying ZnO-NPs, but the efficiency of this reduction needs further examination under ambient conditions before field application.

Biochar remediates denitrification process and N2O emission in pesticide chlorothalonil-polluted soil: Role of electron transport chain

Xiaoxuan Su,Yiyu Wang,Qiang He,Xuebin Hu,Yi Chen

Agricultural production causes large amounts of pesticide residues into soil, disturbing soil denitrification and N 2 O emission. Recently, biochar amendment has been regarded as the effective way to stimulate soil denitrification and reduce N 2 O emission. However, the underlying mechanism has not been well understood. Here, the effects of biochar on denitrification and N 2 O emission in the chlorothalonil (CHT)-polluted soil were explored. Biochar (50 g kg −1 ) was added to soil at the beginning of the experiment, and CHT (5 and 25 mg kg −1 ) was repeatedly applied to soil at the 30-day interval for 105 days. The obtained results showed that CHT remarkably declined soil respiration rates (52–77%) and denitrification rates (50–56%), but increased N 2 O emissions by 820–1030%. After biochar application, however, soil denitrification rates were increased by 13–26%, and N 2 O emissions were declined by 442–809% compared with non-biochar spiked soils. A further mechanism study indicated that biochar remarkably promoted the activities of microbial electron transfer chain, electron transporters (complex I, II, III and cytochrome c), and four denitrifying reductases (NAR, NIR, NOR, NOS) in CHT-polluted soils, suggesting that electron transport and consumption capacities during denitrification were stimulated by biochar. Soil denitrification was thus improved and N 2 O emission was subsequently reduced. Furthermore, biochar significantly upregulated soil denitrifying gene abundances and increased denitrifiers abundances (i.e., Enterobacter and Pseudarthrobacter ). Overall, this work will promote the current understandings of molecular mechanisms through which N 2 O emission in pesticide contaminated soils responds to biochar remediation.

Acute response of soil denitrification and N2O emissions to chlorothalonil: A comprehensive molecular mechanism

Xuebin Hu,Yiyu Wang,Xiaoxuan Su,Yi Chen

The fungicide chlorothalonil (CHT) has been widely used in the tea orchard due to its high-efficiency and sterilization. It has been reported that repeated application of CHT inhibits soil nitrification process. However, the acute impact of CHT on soil denitrification and associated N 2 O emissions is unclear. This study evaluated nitrate (NO 3 − ) removal, denitrifying gene abundance and denitrifying enzyme activity of tea orchard soil after a 72-h-exposure to CHT. It was found that increasing CHT from 5 to 25 mg kg −1 suppressed the NO 3 − removal efficiency from 74.6% to 54.1%, but increased N 2 O emissions from 23.1% to 94.8%. Following treatment with 25 mg kg −1 of CHT, the abundances of the nir K, nir S and nos Z genes were reduced by 31.6%, 22.1%, and 50.7%, respectively. Alternatively, the declines of the electron transport system activity (ETSA) value and adenosine triphosphate (ATP) content suggested that CHT had an inhibitory effect on microbial metabolism. Enzyme activity studies further revealed that the decrease of nitrate reductase (NAR), nitrite reductase (NIR) and nitric oxide reductase (NOR) activities was the main reason for the suppression of denitrification by CHT. Furthermore, positive correlations were observed between denitrifying reductase activity and the intracellular metabolism, indicating that the decrease in microbial metabolism should also be responsible for the inhibitory effect of CHT on the denitrifying process. Overall, it was found that the acute exposure of soil to CHT could inhibit the denitrification process and significantly increase N 2 O emissions, which might result in destruction of the soil nitrogen cycle and exacerbation of global warming .

Effects of humic acid modified oyster shell addition on lignocellulose degradation and nitrogen transformation during digestate composting

Mingyi Lu,Xiaoshuang Shi,Quan Feng,Xu Li,Shujuan Lian,Mengdan Zhang,Rongbo Guo

The aim of this work was to investigate the performance of a novel humic acid modified oyster shell (MOS) bulking agent on the digestate composting. MOS was prepared by immobilizing humic acid onto oyster shell using solid phase grafting method, and then applied to the composting process. Results showed more obvious degradation of lignocellulose was observed in the MOS treatment, which was probably due to the high relative abundance of Actinobacteria . Moreover, the addition of MOS could significantly preserve NH 4 + and reduce the NO 3 − generation with the decreasing abundance of ammonia-oxidizing bacteria and archaea. Besides, adding MOS reduced the N 2 O emission by 59.63% compared with the control. After composting, excitation-emission matrix fluorescence spectra demonstrated that the humification degree as well as compost maturity was enhanced with MOS added.

Three-stage hybrid NSD/DC plasma assisted n-C5H12/O2/N2 ignition: Improved energy efficiency and reduced NOx/N2O emissions

Nan Liu,Qi Chen,Xianwu Jiang,Jie Chen,Lidong Zhang,Jiaying Pan,Haiqiao Wei,Xingqian Mao

This work presents a three-stage hybrid nanosecond discharge (NSD) and direct current (DC) discharge assisted n -C 5 H 12 /O 2 /N 2 ignition with lower discharge energy and higher energy efficiency. The reaction rate coefficients between O 2 (a 1 Δ g ) and n -C 5 H 12 are updated by mass/Master equation simulation. The promotive ignition enhancement via reaction channel O 2 (a 1 Δ g ) + n -C 5 H 12 → C 5 H 10 + H 2 O 2 in the hybrid NSD/DC discharge is demonstrated. The results show that the ignition delay time of the three-stage hybrid plasma assisted ignition is 1–2 orders of magnitude shorter than that of the two-stage hybrid discharge when the reduced electric field strength in the DC discharge stage is over 10 Td. Meanwhile, the plasma-enhanced energy efficiency of three-stage hybrid discharge increases. This improved efficiency on ignition enhancement is due to the kinetic enhancement by the electronically excited species and radicals (O 2 (a 1 Δ g ), O( 1 D), N 2 (B) and O). The kinetic and thermal effects by plasma are effectively intergated to enhance ignition. However, more discharge energy deposited in the vibrationally excited states N 2 ( v ) in the two-stage hybrid discharge is less efficient on ignition enhancement due to the thermal contribution via vibrational-translational relaxation. In the aspect of NO x /N 2 O emissions, reduction efficiency of the three-stage hybrid discharge is two times higher than that of the two-stage hybrid discharge. By controlling the N/N( 2 D) production pathway e + N 2 → e + N + N( 2 D), the three-stage hybrid discharge reduces NO x and N 2 O production by 61.1 % and 90 %, respectively. The results show that the ignition delay time is non-monotonically dependent on the discharge pulse number. There exist optimal pulse number, reduced electric field and effective threshold power density of DC discharge to achieve maximum ignition enhancement and low NO x /N 2 O emissions. This study provides valuable insights into the use of NSD/DC hybrid discharge technology in advanced engines to achieve energy-efficient ignition enhancement and reduce NO x /N 2 O emissions.

Tanning wastewater restructured nitrogen-transforming bacteria communities and promoted N2O emissions in receiving river riparian sediments

Xiangpeng Zeng,Yanyan Liu,Qingqing Wang,Haitao Ma,Xinlei Li,Qiang Wang,Qingxiang Yang

Physicochemical and toxicological characterization of leather tanning wastewater has been widely documented. However, few reports have examined the response of denitrification N 2 and N 2 O emissions in riparian sediments of tannery wastewater-receiving rivers. In this study, 15 N-nitrate labeling was used to reveal the effects of tanning wastewater on denitrification N 2 and N 2 O emission in a wastewater-receiving river (the old Mang River, OMR). OMR riparian sediments were highly polluted with total organic carbon (93.39 mg/kg), total nitrogen (5.00 g/kg) and heavy metals; specifically, Cr, Zn, Cd, and Pb were found at concentrations 47.3, 5.8, 1.6, 4.3, and 2.8 times that in a nearby parallel river without tanning wastewater input (the new Mang River, NMR), respectively. The denitrification N 2 emission rates (0.0015 nmol N · g −1 · h −1 ) of OMR riparian sediments were significantly reduced by 2.5 times compared with those from the NMR ( p < 0.05), but the N 2 O emission rates (0.31 nmol N · g −1 · h −1 ) were significantly increased (4.1 times, p < 0.05). Although the dominant nitrogen-transforming bacteria phylum was Proteobacteria in the riparian sediments of both rivers, 11 nitrogen-transforming bacteria genera in the OMR were found to be significantly enriched; five of these were related to pollutant degradation based on linear discriminant analysis (LDA > 3). The average activity of the electron transport system in the OMR was 6.3 times lower than that of the NMR ( p < 0.05). Among pollution factors, heavy metal complex pollution was the dominant factor driving variations in N 2 O emissions, microbial community structure, and electron transport system activity. These results provide a new understanding and reference for the treatment of tanning wastewater-receiving rivers.

Unraveling the Impact of Perfluorooctanoic Acid on Sulfur-Based Autotrophic Denitrification Process

Hong-Xu Bao,Zhou-Yang Li,Chen Chen,Min Li,Xue-Ning Zhang,Kang Song,Yi-Lu Sun,Ai-Jie Wang

PFOA has garnered heightened scrutiny for its impact on denitrification, especially given its frequent detection in secondary effluent discharged from wastewater treatment plants. However, it is still unclear what potential risk PFOA release poses to a typical advanced treatment process, especially the sulfur-based autotrophic denitrification (SAD) process. In this study, different PFOA concentration were tested to explore their impact on denitrification kinetics and microbial dynamic responses of the SAD process. The results showed that an increase PFOA concentration from 0 to 1000   μg/L resulted in a decrease in nitrate removal rate from 9.52 to 7.73 mg-N/L·h. At the same time, it increased nitrite accumulation and N 2 O emission by 6.11 and 2.03 times, respectively. The inhibitory effect of PFOA on nitrate and nitrite reductase activity in the SAD process was linked to the observed fluctuations in nitrate and nitrite levels. It is noteworthy that nitrite reductase was more vulnerable to the influence of PFOA than nitrate reductase. Furthermore, PFOA showed a significant impact on gene expression and microbial community. Metabolic function prediction revealed a notable decrease in nitrogen metabolism and an increase in sulfur metabolism under PFOA exposure. This study highlights that PFOA has a considerable inhibitory effect on SAD performance.

Complementary Biotransformation of Antimicrobial Triclocarban Obviously Mitigates Nitrous Oxide Emission toward Sustainable Microbial Denitrification

Liying Zhang,Xiaodan Ma,Qian Li,Hanlin Cui,Ke Shi,Hao Wang,Yanqing Zhang,Shuhong Gao,Zhiling Li,Ai-Jie Wang,Bin Liang

Sustainable nitrogen cycle is an essential biogeochemical process that ensures ecosystem safety and byproduct greenhouse gas nitrous oxide reduction. Antimicrobials are always co-occurring with anthropogenic reactive nitrogen sources. However, their impacts on the ecological safety of microbial nitrogen cycle remain poorly understood. Here, a denitrifying bacterial strain Paracoccus denitrificans PD1222 was exposed to a widespread broad-spectrum antimicrobial triclocarban (TCC) at environmental concentrations. The denitrification was hindered by TCC at 25 μg L–1 and was completely inhibited once the TCC concentration exceeded 50 μg L–1. Importantly, the accumulation of N2O at 25 μg L–1 of TCC was 813 times as much as the control group without TCC, which attributed to the significantly downregulated expression of nitrous oxide reductase and the genes related to electron transfer, iron, and sulfur metabolism under TCC stress. Interestingly, combining TCC-degrading denitrifying Ochrobactrum sp. TCC-2 with strain PD1222 promoted the denitrification process and mitigated N2O emission by 2 orders of magnitude. We further consolidated the importance of complementary detoxification by introducing a TCC-hydrolyzing amidase gene tccA from strain TCC-2 into strain PD1222, which successfully protected strain PD1222 against the TCC stress. This study highlights an important link between TCC detoxification and sustainable denitrification and suggests a necessity to assess the ecological risks of antimicrobials in the context of climate change and ecosystem safety.

Toward N2O emission reduction in a single-stage CANON coupled with denitrification: Investigation on nitrite simultaneous production and consumption and nitrogen transformation

Peng Yan,Kai Li,Jin-Song Guo,Si-Xi Zhu,Zhi-Kang Wang,Fang Fang

A dynamic analysis approach for determining nitrite production and consumption rates was established to systematically investigate the characteristics of nitrogen transformation and N 2 O emission of the completely autotrophic nitrogen removal over nitrite (CANON) process coupled with denitrification using a sequencing batch biofilm reactor (SBBR). The results indicate that anaerobic ammonium-oxidizing bacteria was not inhibited significantly by low C/N ratios. There were no obvious differences in the nitrite production rate, nitrite consumption rate or nitrogen removal among reactors operated with C/N ratios of 0, 0.67 and 1.00, which suggested that the certain carbon source did not significantly affect the nitrite conversion and nitrogen removal in the process. More than 60% of total N 2 O emission is generated during the initial phase of each period in the SBBR. More than 94.5% of N 2 O was generated by NO 2 − -N consumption via denitrification in the process. Interestingly, total N 2 O production drops by 16.7%, when the C/N ratio increases from 0 to 1. This phenomenon may be caused by the inhibition of N 2 O production via AOB denitrification. Therefore, an appropriate carbon source (C/N = 1.00) has the beneficial effect of reducing N 2 O emission by CANON coupled with denitrification. The results of this study provide an important empirical foundation for the mitigation of N 2 O emission in the CANON process coupled with denitrification.
