Naringenin alleviates heat stress-induced liver injury in Ningdu yellow chickens by decreasing RIPK3 and PDC binding

Zhenni Liu,Ruoyun Tang,Qiurong Qi,Siting Lin,Ping Liu,Gaofeng Cai,Zhanhong Zheng,Xiaoquan Guo,Xiaona Gao

Naringenin, a flavonoid extract, possesses anti-inflammatory, antioxidant, hepatoprotective, antitumor, and antineurotoxic properties. This study investigated the anti-heat stress effects in broilers by adding 200mg/kg naringenin to the diet of Ningdu yellow chicken under heat stress conditions. Heat stress conditions was controlled at 37 ± 2°C (7:00 a.m. to 7:00 p.m.) and 24 ± 2°C (7:00 p.m. to 7:00 a.m.) at humidity maintained at 60-65%. The results suggest that naringenin elevated the body weight and the ratio of liver mass to weight of Ningdu yellow chicken significantly. Additionally, naringenin significantly reduces heat stress level, improves liver function and antioxidant capacity. Meanwhile, the levels of necroptosis indexes (CYLD, RIPK1, RIPK3 and MLKL) and oxidative stress indexes (PDC, PYGL, GLUL and GLUD1) are downregulated by naringenin. Naringenin mitigated liver damage by decreasing inflammatory indexes caused by heat stress, including NF-κB, IL-1β, IL-18 and HMGB1. This anti-inflammatory effect arose through the downlink binding of the necroptosis index (RIPK3) and the oxidative stress index (PDC) as shown in results of fluorescence co-localization and co-immunoprecipitation. The use of naringenin in poultry may be a possible feed additive to address clinical heat stress.

Flavonoids‑targeted metabolomic analysis following rice yellowing

Yuqian Liu,Jinguang Liu,Caiyun Tang,Victoria Anthony Uyanga,Longhua Xu,Fengjiao Zhang,Jingyu Sun,Yilun Chen

Flavonoids are the main metabolites responsible for yellowing of rice. However, the accumulation pattern of flavonoids and the metabolic basis of flavonoid biosynthesis during rice yellowing remain unclear. Thus, flavonoid-targeted metabolomics was used to investigate the composition and concentration of flavonoids in rice during yellowing. The results indicated the differential flavonoids at Month 3 and Month 5 of storage were more in composition and concentration with higher antioxidant capacity. Accumulated flavonoids were mainly flavones, flavonols, isoflavones, and anthocyanidins, of which rutin, farrerol, naringenin , cyanidin 3-rutinoside, and diosmetin were the indicators of rice yellowing. Metabolic association among flavonoids demonstrated the formation of yellow pigments was jointly induced by flavones, flavonols, isoflavones, and anthocyanidins metabolism. Examination of flavonoid metabolism presented in this study enhanced current understanding of the relationship between flavonoid metabolites and development of rice yellowing. It also offers a theoretical basis for targeted prediction of rice yellowing in the future.

Pharmaceutical cocrystals of naringenin with improved dissolution performance

Chun Luo,Wendong Liang,Xin Chen,Jianming Wang,Zongwu Deng,Hailu Zhang

Naringenin (NAR), a natural flavanone compound, has received increasing attention in recent years due to its diverse pharmacological activities. However, this bioflavonoid shows poor solubility and therefore is difficult to absorb on oral ingestion. To improve the solubility of NAR, co-crystallization trials were performed, and four NAR cocrystals were obtained for the first time. All products were comprehensively characterized by a range of analytical methods, including single crystal and powder X-ray diffraction (XRD), liquid and solid state nuclear magnetic resonance (NMR), Fourier transform infrared spectroscopy (FTIR), and differential scanning calorimetry (DSC). The dissolution studies revealed that all the four cocrystals, namely, NAR–isonicotinamide (NAR–INM), NAR–picolinic acid (NAR–PCA), and two modifications of NAR–betaine (NAR–BTN), exhibit improved apparent solubilities and intrinsic dissolution rates (IDRs) relative to the parent NAR. Two NAR–BTN forms are monotropically related. Additionally, BTN was used for the first time to form cocrystals with flavonoid compounds, which may provide crystallographic inspiration for the cocrystal design and screening of these kinds of bioactive molecules.

Artificial naringinase system for cooperative enzymatic synthesis of naringenin

Chi Wang,Pei-Xu Chen,Qiong Xiao,Jun Chen,Fu-Quan Chen,Qiu-Ming Yang,Hui-Fen Weng,Bai-Shan Fang,Yong-Hui Zhang,An-Feng Xiao

Naringinase is a kind of glycosidase that catalyzes the biotransformation of naringin into naringenin . It is a multi-compound enzyme that provides the activity of α- L -rhamnosidase and β-glucosidase. In this study, an artificial naringinase system was constructed by co-immobilizing α- L -rhamnosidase ( Aspergillus oryzae FJ0123, Ao-rha) and β-glucosidase ( Thermotoga maritima MSB8, Tm-glu) on magnetic silica-based chitosan microspheres (MSC). The molar ratio of enzymes and the preparation conditions were optimized for improving synergistic effect and immobilization efficiency. Under optimized conditions (the molar ratio of Ao-rha and Tm-glu, 3:1; temperature, 25 °C; glutaraldehyde concentration, 2.0%; pH, 3.0; time, 9 h), the immobilization yield and activity recovery were 61.4% and 37.3% for Ao-rha and 90.1% and 56.3% for Tm-glu, respectively. Biochemical characterization indicated that MSC-naringinase had better adaptability and stability than free enzymes. The activity of MSC-naringinase was maintained at 58.7% after 10 times of recycling. The catalytic conditions were also investigated. The cascade reactions of naringin to prunin and prunin to naringenin by using MSC-naringinase resulted in a yield of 0.62 mg/mL and a conversion rate of 96.9%, respectively, without prunin accumulation. These results indicated the great potential of MSC-naringinase as an efficient artificial naringinase system for cooperative enzymatic synthesis of naringenin.

Preparation of Naringenin Nanosuspension and Its Antitussive and Expectorant Effects

Zhengqi Dong,Rui Wang,Mingyue Wang,Zheng Meng,Xiaotong Wang,Meihua Han,Yifei Guo,Xiangtao Wang

Naringenin (NRG) is a natural flavonoid compound abundantly present in citrus fruits and has the potential to treat respiratory disorders. However, the clinical therapeutic effect of NRG is limited by its low bioavailability due to poor solubility. To enhance the solubility, naringenin nanosuspensions (NRG-NSps) were prepared by applying tocopherol polyethylene glycol succinate (TPGS) as the nanocarrier via the media-milling method. The particle size, morphology, and drug-loading content of NRG-NSps were examined, and the stability was evaluated by detecting particle size changes in different physiological media. NRG-NSps exhibited a flaky appearance with a mean diameter of 216.9 nm, and the drug-loading content was 66.7%. NRG-NSps exhibited good storage stability and media stability. NRG-NSps presented a sustainable release profile, and the cumulative drug-release rate approached approximately 95% within 7 d. NRG-NSps improved the antitussive effect significantly compared with the original NRG, the cough frequency was decreased from 22 to 15 times, and the cough incubation period was prolonged from 85.3 to 121.6 s. Besides, NRG-NSps also enhanced expectorant effects significantly, and phenol red secretion was increased from 1.02 to 1.45 μg/mL. These results indicate that NRG-NSps could enhance the bioavailability of NRG significantly and possess a potential clinical application. Keywords:naringenin nanosuspension;media-milling method;bioavailability;antitussive effect;expectorant effect

Protective Effect of Naringenin on Cadmium-Induced Kidney Injury in Rats

Hao Ling,Junbing Mao,Bing Xu,Yaning Shi,Jicang Wang

Cadmium (Cd) exposure in occupational settings poses significant nephrotoxic risks, yet effective preventive strategies remain limited. This study investigated the protective effects of Naringenin (Nar) against Cd-induced nephrotoxicity. Twenty-four male SD rats (4 weeks old) were randomly assigned to control group, Cd group, Nar group, and Cd+ group for 14 days. Histopathological damage characterized by tubular necrosis and inflammatory infiltration. Cd exposure significantly impaired renal function, as evidenced by elevated serum uric acid and creatinine levels, increased oxidative stress markers (renal glutathione and malondialdehyde accumulation). Molecular analysis confirmed Cd-induced apoptosis through dysregulation of key apoptotic markers: downregulation of anti-apoptotic Bcl-2 and upregulation of pro-apoptotic Bax at both mRNA and protein levels, accompanied by increased cytochrome c release and activation of Caspase-9 and Caspase-3. These findings were further supported by TUNEL staining, which showed increased apoptosis in renal tubular cells. This study supports the potential application of naringin in alleviating cadmium-induced nephrotoxicity.

Naringenin protects against septic cardiomyopathy in mice by targeting HIF-1α

Jiajia Pan,Lijun Meng,Rujun Li,Zicheng Wang,Wenjie Yuan,Yucheng Li,Lin Chen,Qinhao Shen,Weili Liu,Li Zhu

Myocardial dysfunction is a prevalent complication of sepsis (septic cardiomyopathy) with a high mortality rate and limited therapeutic options. Naringenin, a natural flavonoid compound with anti-inflammatory and antioxidant properties, holds promise as a potential treatment for sepsis-induced myocardial dysfunction. This study investigated the pharmacological effects of naringenin on septic cardiomyopathy. In vivo and in vitro experiments demonstrated that naringenin improved cardiomyocyte damage. Network pharmacology and database analysis revealed that HIF-1α is a key target protein of naringenin. Elevated expression of HIF-1α was observed in damaged cardiomyocytes, and the HIF-1α inhibitor effectively protected against LPS-induced cardiomyocyte damage. Molecular docking studies confirmed the direct binding between naringenin and HIF-1α protein. Importantly, our findings demonstrated that naringenin did not provide additional attenuation of cardiomyocyte injury on the biases of HIF-1α inhibitor treatment. In conclusion, this study proves that naringenin protects against septic cardiomyopathy through HIF-1α signaling. Naringenin is a promising therapeutic candidate for treating septic cardiomyopathy.

Synergistic action of naringenin and amikacin in the treatment of avian colibacillosis

Yi Lankun,Chen Xu,Cao Mingze,Liu Qing,Huang Yang,Pang Jianing,Wang Zeyu,Guan Ding,Ma Tenghe,Zhu Zhen,Wan Yongbiao

Background Avian colibacillosis is an infectious disease caused by pathogenic Escherichia coli (E. coli) and is one of the significant causes of economic losses in the global poultry industry. The overuse of antibiotics has led to the emergence of resistant strains, and the presence of antibiotic residues in chickens and eggs poses a threat to human health. Therefore, exploring safe adjuvants to enhance the efficacy of existing antibiotics is one of the critical issues in poultry farming and public health research. Naringenin (NG), a naturally occurring flavonoid, has been extensively studied as a therapeutic agent for various diseases, but its efficacy in combination with antibiotics for treating avian colibacillosis has not yet been explored. Results In this study, we established a model of avian colibacillosis and treated it with NG and amikacin (AMK) alone or in combination. Compared with monotherapy, the combination of NG and AMK significantly decreased bacterial loads in the heart, liver, spleen, lung, and kidney by 5.0–6.0 log 10 CFU/g ( p  < 0.01). Histopathological analysis revealed that the combination treatment markedly alleviated pathological damage in the lung and duodenum compared to the AMK alone group. Furthermore, the combination therapy significantly exerted anti-inflammatory and antioxidant effects, as evidenced by reduced serum levels of tumor necrosis factor-alpha (TNF-α), interleukin-1 beta (IL-1β), and malondialdehyde (MDA), alongside increased superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities ( p  < 0.05). 16 S rRNA sequencing demonstrated that the combination treatment significantly modulated the gut microbiota composition, decreasing the relative abundance of Proteobacteria from 44% in the infection group to 1.2% ( p  < 0.01), while increasing Firmicutes and Bacteroidetes to levels comparable to healthy controls. Conclusions The research results provides evidence for the therapeutic potential of NG as an effective antibiotic synergist when combined with AMK against avian colibacillosis.

Xanthine oxidase inhibitory and anti-hyperuricemic properties of sulfonate derivatives of naringenin

Yuan Luo,Jinyan Xie,Hao Yang,Yu Huang,Xinbin Yang

Xanthine oxidase (XO) facilitates oxidation of hypoxanthine to xanthine and then to uric acid, potentially leading to hyperuricemia when production of excessive uric acid. In the present study, naringenin–3′–sulfonate sodium and naringenin–3′,6–disulfonate sodium were synthesized by chemical modification of naringenin, and evaluated for XO inhibitory in vitro. The results indicated that naringenin–3′–sulfonate sodium exhibited significantly inhibitory activities compared to naringenin and naringenin–3′,6–disulfonate sodium. Further inhibition kinetics, fluorescence spectra, circular dichroism spectra and molecular docking analysis revealed that as a reversible competitive inhibitor, naringenin–3′–sulfonate sodium could bind to the active site of XO and induce conformational changes through hydrogen bond, hydrophobic forces, π-π stacking, and electrostatic interaction. Furthermore, naringenin–3′–sulfonate sodium was effective in reducing serum uric acid levels in hyperuricemic mice, which were found to be associated with the inhibition of serum XO activity. Additionally, naringenin–3′–sulfonate sodium exhibited potential in ameliorating renal damage as evidenced by serum creatinine analysis and histopathological examination of renal tissues. These findings provide compelling evidence for the promising role of naringenin–3′–sulfonate sodium as a preventive or therapeutic agent against hyperuricemia.

A Polymethionine Nanoparticle Fluorescent Probe for Sensitive Detection of Naringin and Naringenin

Yuhong Jiao,Lu Li,Jinlong Ge,Yanfang Tai,Hui Han

In this work, we demonstrated a novel, sensitive and effective fluorescent naringin (NRG) and naringenin (NRGe) detection method using polymethionine nanoparticles (PMNPs) as a fluorescent nanoprobe. The PMNPs were first synthesized by autopolymerization of methionine at 90 °C when trace copper ions existed. The as-prepared PMNPs were thoroughly characterized by transmission electron microscopy (TEM), Fourier-transform infrared spectroscopy (FT-IR), gel permeation chromatograph (GPC), nuclear magnetic resonance spectroscopy (NMR), transient and steady-state fluorescence and UV–Vis absorption spectroscopy. The quenching mechanism was attributed to the inner filter effect (IFE). Moreover, the developed assay was used successfully to detect NRG and NRGe in real samples of citrus fruits, illustrating that this detection method has great potential application in the field of citrus fruits analysis.

Naringenin as a Novel Stabilizer for Improving the Stability of 5-Methyltetrahydrofolate

Siqi Zhou,Jing Xu,Kun Hou,Mengmeng Zhang,Ting Huang,Liping Guan,Guangbin Zhou,Jun Yin,Dan Song

This study discovered a natural flavonoid stabilizer for stabilizing 5-methyltetrahydrofolate (5-MTHF) since the instability of 5-MTHF affects the accurate determination of 5-MTHF. The optimal stabilizer was selected from six flavonoid compounds, comparing the differences between natural stabilizers and traditional stabilizers and studying the parameters of the stabilizing 5-MTHF solution. In addition, the stabilizing effect of the stabilizer on 5-MTHF was evaluated under optimal parameters under various environmental conditions. Differences in the methods with and without naringenin were compared. The concentrations of 5-MTHF were analyzed using validated liquid chromatography-tandem mass spectrometry, and percentage deviation characterized the stability. The results indicated that naringenin had a better stabilizing effect on 5-MTHF compared to dithiothreitol, similar to l-ascorbic acid, maintaining the stability of 5-MTHF under different conditions. Naringenin had no adverse effect on the stability of other metabolites. In conclusion, naringenin improved the stability of 5-MTHF, ensuring its stability during the storage and detection processes.
