Light quality affects plant leaf growth and its metabolism of amino acids and polyphenols. Anoectochilus roxburghii has been used for food , medicine, and ornamental purposes. To obtain the optimal light quality for improved accumulation of metabolites in A. roxburghii in vitro . The seedings were cultured under the white light (W), blue light (B), red light (R), red: blue light = 1:2 (R1B2), red: blue light = 1:1 (R1B1), red: blue light = 2:1 (R2B1). Twenty compounds in raw leaves were in situ rapidly identified within 20 s by neutral desorption-extractive electrospray ionization mass spectrometry (ND-EESI-MS) without any sample pretreatment, including eight amino acids , eleven polyphenols, and kinsenoside. Meanwhile, leaf area and the activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) were evaluated. Our results showed that leaf area and antioxidant enzyme activities of the R2B1 were significantly higher than B or R group. The red-blue light increased the content of isoleucine , histidine , serine , arginine, and promoted the accumulation of caffeic acid , ferulic acid , quercetin , myricetin , kaempferol , and rutin in phenylpropane metabolism, especially in the R2B1, which could be used as the optimal light quality. ND-EESI-MS could enhance our understanding of the effect of light quality on the accumulation of amino acids and polyphenols in A. roxburghii .

Herein, we constructed a platform of neutral desorption-extractive electrospray ionization mass spectrometry (ND-EESI-MS) for direct and rapid detection of chloramphenicol (CAP) in honey samples diluted with methanol. Under the optimized working conditions, the quantitative information of CAP residues was acquired effectively by EESI-Ion Trap MS n . Using heated methanol-N 2 as spray reagent, we reduced the limit of determination (LOD) from 73.3 ng/mL to 0.3 ng/mL, and the CAP detection is linear in the range of 1–5000 ng/mL ( R = 0.9947). For the honey samples with CAP of 10, 100, and 1000 ng/mL, the recoveries were 133.0, 80.6, and 101.1 %, and the relative standard deviations were 5.96, 8.82, and 8.71 %, respectively. The reproducibility assays showed the stability of this method. Therefore, this ND-EESI-MS method is powerful for direct, rapid, and quantitative CAP analysis in honey samples with high sensitivity, precision, and specificity.
In the current neutral desorption-extractive electrospray ionization mass spectrometry (ND-EESI-MS) method, N 2 is inlet into samples to desorb chloramphenicol (CAP). We tried to use some organic solvents as the spray reagent to dissolve CAP, and then the best neutral desorption efficiency was obtained when using methanol. We applied this modified ND-EESI-MS method to detect CAP in honey samples only with sample dilution. The limit of CAP detection was then reduced from 73.3 to 0.3 ng/mL, reaching the current EU standard. Therefore, this is a powerful method for direct, rapid, and quantitative CAP analysis in honey samples.

High-viscosity rice soup is not only a popular food item but also a traditional Chinese medicinal remedy. Herein, we developed a high-throughput characterization platform for high-viscosity rice soup analysis using Neutral Desorption Extractive Electrospray Ionization Mass Spectrometry (ND-EESI-MS) with untargeted metabolomics. The chemical fingerprint of high-viscosity rice soup was directly analyzed by ND-EESI-MS, with exogenous (e.g., heavy metals and pesticides) and endogenous pollutants detected via qualitative and quantitative tandem mass spectrometry. Multivariate statistical analysis was then applied to distinguish the quality of the high-viscosity rice soup effectively. The concentrations of heavy metals and pesticide residues in high-viscosity rice soup were quantified, demonstrating a strong linear relationship (R2 > 0.99) across the 0.05–2000.00 ng mL−1 concentration range. The limits of detection (LOD) ranged from 0.27 ng mL−1 to 2.18 ng mL−1, while the limits of quantification (LOQ) ranged from 0.82 ng mL−1 to 6.61 ng mL−1. Furthermore, three specific compounds—3-methyl-2-butenoic acid, salicylic acid, and rhamnose—were quantified. These compounds demonstrated excellent linearity (R2 > 0.99) within the linear range of 1.10–1500.00 ng mL−1. The LOD ranged from 0.56 to 1.30 ng mL−1, and the LOQ ranged from 1.71 to 3.95 ng mL−1. Owing to its capability for simultaneous detection of exogenous and endogenous contaminants in complex viscous matrices, this ND-EESI-MS platform demonstrated significant potential to enhance quality control in food safety surveillance and pharmaceutical manufacturing.

The threat of drug-resistant bacteria is challenging, and it is urgent to explore new antibiotics. Gold nanoparticles (AuNPs) are known to be a group of promising antibacterial agents for replacing conventional antibiotics. Nevertheless, their antibacterial mechanism remains to be elucidated. Herein, we directly observed the interaction between antibacterial AuNPs and bacteria at the molecular level using neutral desorption extractive electrospray ionization mass spectrometry (ND-EESI-MS). We monitored and analyzed the dynamic changes of bacterial metabolites in real time after AuNP treatment. Ten substances representing 3 major metabolic pathways, including protein and nucleic acid synthesis, energy metabolism, and quorum sensing, were identified, indicating that AuNPs may exert antibacterial effects through multiple mechanisms influencing bacterial metabolism and communication. This study deepens the understanding of the antibacterial mechanism of AuNPs and is insightful for designing and screening new antibacterial agents.
