Spermine, a key biogenic amine indicating food freshness, is typically detected using costly and time-consuming chromatographic methods. This study introduces a more efficient, eco-friendly alternative—a label-free colorimetric sensing platform using gold nanoparticles (AuNPs) functionalized with 11-mercaptoundecanoic acid and capped with an N-hydroxysuccinimide (NHS)-ester. Transmission electron microscopy revealed monodisperse, spherical AuNPs (13 nm), with an increase in size upon functionalization. Fourier transform infrared spectra confirmed successful functionalization. The hydrodynamic size of the AuNPs increased from 17.26 nm to 2167 nm, and the zeta potential shifted from −48.86 mV to −35.59 mV. The platform takes advantage of the selective interaction between spermine and NHS-ester-functionalized AuNPs, inducing nanoparticle aggregation, as shown by a red shift in surface plasmon resonance (SPR). UV-vis spectroscopy demonstrated a robust linear correlation (R2 = 0.958) between spermine concentration (1.0–4.0 μM) and nanoparticle aggregation index, with a limit of detection (LOD) of 0.77 μM. The sensor also exhibited high reproducibility in pork extract matrices (coefficient of variation <5%) and selectivity for spermine amid various interfering analytes. Its eco-friendly design and rapid response time position it as a viable tool for real-time spermine monitoring in food spoilage, offering comparable performance metrics to traditional chromatographic techniques while addressing sustainability concerns.

γ-PGA-NHS ester, which was prepared using poly(γ-glutamic acid) (γ-PGA) and N-hydroxysuccinimide (NHS) as the raw materials, was synthesized to be a novel cross-linker of collagen. Fourier transform infrared spectra analysis suggested that the products displayed the characteristic absorption peak of ester. Results from nuclear magnetic resonance analysis indicated that the esterification degree of γ-PGA-NHS ester was increased with the increase of NHS. Modified collagen was prepared and characterized. The results of circular dichroism analysis indicated modified collagen retained the triple helix structure of natural collagen. Sodium dodecyl sulphate polyacrylamide gel electrophoresis revealed that the molecular weight of collagen was increased after cross-linking. Peptide mapping of collagen suggested that cross-linked collagen possessed an enhanced resistance to trypsin degradation. Differential scanning calorimeter results showed that the denaturation temperature of collagen was improved from 68.1 ± 0.4 to 91.2 ± 0.5 °C (p < 0.05). Dynamic viscoelastic measurements demonstrated the improvement of thermal stability and reflected the exponential increase in η*. The cross-linked collagen retained porous structure and the pore size became larger as observed by scanning electron microscopy. The investigation results provided useful information to produce collagen with improved physicochemical properties, particularly the thermal stability via the use of γ-PGA-NHS ester as a biomacromolecule-based cross-linker.

Polymer brushes of polymethacrylic acid (PMAA) and PMAA-associated polymer blends of PMAA/PNIPAM (poly-N-isopropylacrylamide) were prepared on porous silicon for further investigation of the EDC/NHS (1-ethyl-3-(3-dimethylaminopropyl)carbodiimide/N-hydroxysuccinimide) activation mechanisms by infrared spectroscopy. When the fragmentation degree of PMAA blocks in PMAA-associated polymer blends is increased, the production of anhydride wanes from dominant to recessive, whereas a complementary product of the NHS-ester waxes from recessive to dominant. The Thorpe–Ingold effect was applied to explain the formation of anhydride: the gem-dialkyl groups of PMAA next to the carboxylic acids compress the acid side chains close to each other; thus, once the intermediate of O-acylisourea forms, it will be attacked by the intramolecular neighboring acid much faster than any other nucleophiles such as NHS and water, and therefore the six-membered ring of the anhydride will be formed. All acid side chains in PMAA standing next to each other will form an anhydride, primarily due to the Thorpe–Ingold effect, unless they are sterically hindered, whereas only isolated acid side chains form the NHS-ester. The EDC/NHS activation results for four small molecules of dicarboxylic acids in aqueous media, namely, glutaric acid and 2,2-dimethyl glutaric acid, which generate disuccinimidyl ester with high yield, and succinic acid and 2,2-dimethyl succinic acid, which remain intact, can also be explained by the Thorpe–Ingold effect. A clear understanding of the EDC/NHS activation mechanisms of PMAA will take us a step closer for resolving the mechanistic ambiguity of the carbodiimide/additive coupling reactions for amide bond formation.

Bioadhesive hydrogels show great promise in wound closure due to their minimally invasive nature and ease of use. However, they typically exhibit poor wet adhesion and mechanical properties on wet tissues. Herein, a ready-to-use bioadhesive hydrogel (denoted as PAA-NHS/C-CS) with rapidly robust adhesion and high mechanical strength is developed via a simple one-pot UV crosslinking polymerization of acrylic acid (AA), catechol-functionalized chitosan (C-CS), and acrylic acid N-hydroxysuccinimide ester (AA-NHS ester). Benefitting from the hydrogen bonds and electrostatic attractions formed between PAA-NHS and C-CS, the as-prepared hydrogel exhibits high tensile strength (∼630 kPa), fracture strain (∼1950%), and toughness (∼4250 kJ m−3) in the fully swollen state. Besides, the noncovalent interactions and covalent crosslinking formed between the dual adhesive moieties (the NHS ester and catechol groups) and the tissue surface endow the hydrogel with high shear strength (∼160 kPa), interfacial toughness (∼630 J m−2), and burst pressure (∼447 mmHg) on wet porcine skin. By integrating the high mechanical properties, rapid robust adhesion, and operational convenience, the as-prepared PAA-NHS/C-CS hydrogel shows great promise in wound closure.

The repair of abdominal wall defects is currently a clinical challenge. A naturally derived extracellular matrix (ECM) such as small intestine submucosa (SIS) has received great attention in abdominal wall defect repair because of its remarkable bioactivity, biodegradability and tissue regeneration. The match between material degradation and tissue remodeling is very important for the realization of ideal repair effectiveness. In this study, a near-infrared (NIR) fluorescent dye Cy5.5 NHS ester was used to label ECM-based (ECMB) composites consisting of SIS and chitosan/elastin electrospun nanofibers for monitoring material degradation. The tissue remodeling in the ECMB composites for a full-thickness abdominal wall defect repair was systematically investigated by a series of tests including wall thickness measurement, muscle regeneration analysis and angiogenesis assessment. The main findings were: (1) real-time and noninvasive degradation monitoring of the ECMB composites until complete degradation could be realized by chemical conjugation with a Cy5.5 NHS ester. (2) In a full-thickness abdominal wall defect model, the explant thickness could be used as an intuitional indicator for evaluating the tissue remodeling efficiency in the ECMB composites, and the accuracy of this indicator was verified by various examinations including collagen deposition, angiogenesis, and muscle regeneration. The present study could provide new insight into evaluating tissue repair effectiveness of the ECMB composites.

Reproducing the microstructure of the natural cornea remains a significant challenge in achieving the mechanical and biological functionality of artificial corneas. Therefore, the development of cascade structures that mimic the natural extracellular matrix (ECM), achieving both macro-stability and micro-structure, is of critical importance. This study proposes a novel, efficient, and general photo-functionalization strategy for modifying natural biomaterials. Collagen microfibers obtained through electrospinning are functionalized with an active N-Hydroxysuccinimide (NHS) ester, to impart light-curing ability. This approach expands the construction of photo-controllable hydrogel systems beyond conventional single methacrylate (MA) modifications or di-tyrosine bonding, enabling integration with other biomaterials for comprehensive ECM remodeling. Subsequently, the collagen microfibers are then photo-embedded into gelatin methacryloyl (GelMA) via covalent crosslinking to form a fibrous hydrogel, which supports both structural and functional requirements. In terms of biological functionality, the hydrogel promotes significant inward migration and retention of human corneal fibroblasts (hCFs), replicating ECM-like environments. Furthermore, its excellent burst resistance suggests potential as a bioadhesive. In a rabbit model, the hydrogel achieved effective repair of large-sized (6 mm) corneal defects, facilitates epithelial migration, and maintained long-term stability. This work provides valuable guidance for designing efficient and simplified bioactive materials for corneal repair and broader tissue engineering applications.
