Co-assembled supramolecular hydrogels of cell adhesive peptide and alginate for rapid hemostasis and efficacious wound healing

Ziran Zhai,Keming Xu,Leixia Mei,Can Wu,Jing Liu,Ziqi Liu,Lanxin Wan,Wenying Zhong

Injectable hydrogels are promising materials for applications in non-compressive wound management. Yet difficulties remain for the fabrication of mechanically stable hydrogel materials with inherent functionalities in both hemostatic control and wound healing without additional supplements of growth factors. Herein, we reported the co-assembly of a cell adhesive peptide conjugate (Pept-1) and alginate (ALG), to confer supramolecular hydrogels with excellent mechanical properties and high efficacy in both hemostatic control and wound healing requiring no additional growth factors. The co-assembling process of Pept-1 and ALG, which was mediated by electrostatic interactions and metal chelation, afforded a composite hydrogel with denser nanofibrillar structures and better mechanical strength when comparing to the Pept-1 gel alone. As-prepared Pept-1/ALG hydrogels exhibited excellent injectability and thixotropic properties, making them ideal materials for wound dressing. The composite hydrogel induced fast hemostasis when spiked with whole blood in vitro, and reduced the amount of bleeding to ∼18% of the untreated control in a liver puncture mouse model. Meanwhile, it promoted adhesion and migration of fibroblast NIH3T3 cells in vitro, and accelerated the rate of wound healing in a full-thickness skin defect model of mice. In addition, the Pept-1/ALG hydrogel showed excellent biocompatibility with no obvious hemolytic activity. In future, the strategy of utilizing co-assembled nanostructures composed of biofunctional peptides and polysaccharides could be further exploited to construct a broad range of nanocomposite materials for a variety of biomedical applications.

Human umbilical cord mesenchymal stem cell-derived exosome ameliorate doxorubicin-induced senescence

Zhen Yang,Feng Yan,Jiangwei Yuan,Manjun Yang,Jinyu Wang,Changqiao You,Kaiqun Ren

Background Cellular senescence refers to a condition where cells permanently cease division while maintaining metabolic activity. Doxorubicin (Dox) is known as an agent of induction of cellular senescence. This study aimed to explore the potential role of human umbilical cord mesenchymal stem cell-derived exosomes (hucMSC-Exo) in mitigating Dox induced senescent. Method NIH3T3 cells were treated by various concentrations of Dox with or without hucMSC-Exo, cell morphology, viability, migration, senescence-associated SA-β-Gal staining were monitored. Cellular senescence was induced in C57BL/6J mice via administration of 5 mg/kg Doxorubicin, followed by treatment with hucMSC-Exo or metformin. Assessments included body weight, liver and kidney weight, colon length, SA-β-Gal staining of kidney and skin, molecular biomarkers of aging such as p16 INK4A , p53, and p21 Waf1/Clip1 to evaluate senescence status. Result We found that after the treatment of exosomes or metformin improved several aging-related phenotypes in both mouse and cellular models, including increases in body weight, liver and kidney weights, and the reduction of SA-β-Gal positive cells in kidney and skin tissues as well as cell models. At the molecular level, hucMSC-Exo resulted in the downregulation of inflammatory factors and senescence markers in liver and kidney tissues as well as cell models. Conclusion Our study demonstrates hucMSC-Exo may ameliorate Dox induced senescence either in NIH3T3 cells or in mice.

Synergistic effects of electroactive antibacterial material and electrical stimulation in enhancing skin tissue regeneration: A next-generation dermal wound dressing

Xi-Liang Zang,Fei Gao,Zhao Zhang,Lin-Hua Shen,Yue-Hai Pan

Objective We aimed to develop an electroactive antibacterial material for the treatment of skin wound diseases. Method To this aim, we modified chitosan (CS), a biocompatible polymer, by coupling it with graphene (rGO) and an antimicrobial polypeptide DOPA-PonG1. The material's effect on skin injury healing was studied in combination with external electrical stimulation (EEM). The structure, surface composition, and hydrophilicity of the modified CS materials were evaluated using scanning electron microscopy (SEM), Fourier-transform infrared spectroscopy (FTIR), and contact angle measurements. We studied NIH3T3 cells cultured with modified materials and subjected to EEM to assess viability, adhesion, and tissue repair-related gene expression. Results SEM data demonstrated that rGO was distributed uniformly on the surface of the CS material, increasing surface roughness, and antimicrobial peptides had minimal impact on surface morphology. FTIR confirmed the uniform distribution of rGO and antibacterial peptides on the material surface. Both rGO and DOPA-PonG1 enhanced the hydrophilicity of CS materials, with rGO also improving tensile strength. The dual modification of CS with rGO and DOPA-PonG1 synergistically increased antibacterial efficacy. Cellular events and gene expression relevant to tissue repair process were enhanced by these modifications. Furthermore, EEM accelerated epidermal regeneration more than the material alone. In a rat skin wound model, DOPA-PonG1@CS/rGO dressing combined with electrical stimulation exhibited accelerated healing of skin defect. Conclusion Overall, our results demonstrate that CS materials modified with rGO and DOPA-PonG1 have increased hydrophilicity, antibacterial characteristics, and tissue regeneration capacities. This modified material in conjunction with EEM hold promise for the clinical management for dermal wounds.

Electrophoretic deposition of hyaluronic acid-g-dopamine/bovine hemoglobin micelles for functional coatings

Sun Jiadi,Chen Weidong,Liu Xuefeng

A novel functional coating was prepared from self-assembled micelles through direct electrophoretic deposition. The micelles were obtained via self-assembly of amphiphilic hyaluronic acid- g -dopamine (HA- g -DA) in aqueous solution with encapsulation of bovine hemoglobin (BHb). The size, morphology, and zeta potential of the resulting micelles were characterized. Then, the micelles were immobilized onto conductive substrate using electrophoretic deposition technique to fabricate nanostructured coating. The structure and morphology of the resulting coating were studied by Fourier transform infrared spectroscopy (FTIR) and scanning electron microscopy (SEM). The release behavior of BHb from the coating was evaluated in a simulated body fluid (SBF), and the BHb was released up to 20 days. And in vitro assay demonstrated that the nanostructured coating is biocompatible to NIH3T3 and Hela cells, respectively. The contribution of our work lies in the creation of a novel strategy to fabricate a functional coating in viewing of the versatility of self-assembled micelles and the controllability of electrophoretic deposition process. It is believed that our work provides new ideas and reliable data to design functional coatings for biomedical applications. ᅟ

Synthesis of lanthanum doped carbon dots for detection of mercury ion, multi-color imaging of cells and tissue, and bacteriostasis

Ming Zhang,Wentao Wang,Ping Yuan,Cheng Chi,Jun Zhang,Ninglin Zhou

This work describes a hydrothermal method for the preparation of multi-functional carbon quantum dots of the type La-CQDs. The La-CQDs are 4.3   ±   0.3   nm in diameter and have a good dispersion and exhibit excellent emission property and high stability, as well as excitation-dependent emission behavior with typical excitation/emission peaks at 350/460   nm. They can also be used as an effective fluorescent probe for the quenchometric determination of mercury ion in aqueous solution with a detection limit of 0.1   µM. The particles are readily internalized by liver epithelial HepG2 cells and enable multi-color fluorescence imaging by displaying green and red emission if excited with 488 and 543   nm light, respectively. Intravenous injection of the quantum dots in nude mice, in turn, enabled tissue imaging. The La-CQDs possess abundant phosphate, hydroxyl, pyridinic, pyridine-like and pyrrole-like nitrogen groups. Therefore, they can exhibit strong fluorescence, improve the accuracy as an optical monitoring code both in vitro and in vivo , and enhance blood compatibility. In vitro investigations show that La-CQDs do not exhibit significant apoptotic effects on NIH3T3 cells even at a high concentration (2.5   mg/mL) while they exhibit improved antibactericidal capacity of the La-CQDs against gram-positive S. aureus and gram-negative E. coli bacteria was also observed. No apparent pathological damages are observed in all organs during the 30-day period, which suggests that La-CQDs do not produce detectable long-term toxicological responses.

Natural antioxidants inhibit oxidative stress-induced changes in the morphology and motility of cells

Xiong Xiang,Amakye William Kwame,Yinglu Qing,Shan Li,Min Wang,Jiaoyan Ren

Effect of antioxidants in oxidative stress-induced cell damage has been extensively studied, but its effects on cell macroscopic phenomena have rarely been explored. Cell morphology and motility are two interrelated cellular processes reflecting cellular function phenotype, and the real-time imaging technology is key for the study of these processes. In this study, we used two known antioxidants, pyrroloquinoline quinone (PQQ) and rutin , to explore the antioxidant effects on cell morphology and motility in rotenone-damaged NIH3T3 cells, as well as roles in their antioxidant function based on Cytation 7 Cell Imaging Multi-Mode Reader. First, NIH-3T3 cells were pretreated with PQQ and rutin for 4 h and then rotenone was used to induce oxidative damage. It was found that PQQ (2 μM) and rutin (300 μM) had similar protective effects on the proliferation activity of rotenone-damaged cells. Subsequently, the morphology and motility of the treated cells were quantified. Results showed that both PQQ and rutin could significantly inhibit morphological changes including cell shape index and cell surface of the damaged cells, however PQQ had a stronger effect on cell motility under the specific experimental concentrations. Using principal component analysis (PCA), we further demonstrated that the quantified cell morphology and motility were consistent with the cell proliferation activity. Consequently, quantification of cell morphology and motility as evaluation indices may serve as potential reference values for the evaluation of biological activity of antioxidants.

Acceleration of Healing in Full-Thickness Wound by Chitosan-Binding bFGF and Antimicrobial Peptide Modification Chitosan Membrane

Hou Lin,Wang Wei,Wang Mei-Kun,Song Xue-Song

Skin wound healing is an important clinical challenge, and the main treatment points are accelerating epidermal regeneration and preventing infection. Therefore, it is necessary to develop a wound dressing that can simultaneously cure bacterial infections and accelerate wound healing. Here, we report a multifunctional composite wound dressing loaded with chitosan (CS)-binding bFGF (CSBD-bFGF) and antimicrobial peptides (P5S9K). First, CS was used as the dressing matrix material, and P5S9K was encapsulated in CS. Then, CSBD-bFGF was designed by combining recombinant DNA technology and tyrosinase treatment and modified on the dressing material surface. The results show that the binding ability of CSBD-bFGF and CS was significantly improved compared with that of commercial bFGF, and CSBD-bFGF could be controllably released from the CS dressing. More importantly, the prepared dressing material showed excellent antibacterial activity in vivo and in vitro and could effectively inhibit the growth of E. coli and S. aureus. Using NIH3T3 cells as cellular models, the results showed that the CSBD-bFGF@CS/P5S9K composite dressing was a friendly material for cell growth. After cells were seeded on the composite dressing surface, collagen-1 (COL-1) and vascular endothelial growth factor (VEGF) genes expression in cells were significantly upregulated. Finally, the full-thickness wound of the rat dorsal model was applied to analyse the tissue repair ability of the composite dressing. The results showed that the composite dressing containing CSBD-bFGF and P5S9K had the strongest ability to repair skin wounds. Therefore, the CSBD-bFGF@CS/P5S9K composite dressing has good antibacterial and accelerated wound healing abilities and has good application prospects in the treatment of skin wounds.

Integrating supercapacitor with sodium hyaluronate based hydrogel as a novel All-In-One wound Dressing: Self-Powered electronic stimulation

Xiangya Wang,Weijie Zhang,Qi Zhou,Fen Ran

Constructing the electron transport pathway in a conductive gel membrane wound dressing could promote wound healing, but it always needs an external power supply that bright much inconvenience in clinical practice. In this paper, we design a novel all-in-one self-powered electronic stimulated wound dressing that integrating supercapacitor with sodium hyaluronate based hydrogel. The supercapacitor layer provides the required continuously electrical energy for the wound-dressing layer, where sodium hyaluronate is used as substrate material in both layers for forming the advanced all-in-one structure. During the course of clinical treatment, the electrical stimulation self-powered by supercapacitor would enable patients to perform electrical stimulation wound treatment anytime and anywhere no need on the back ward of a hospital. At the same time, introducing sodium hyaluronate as the active substance and polyelectrolyte would promote cell proliferation and accelerate wound healing, and improve electrochemical performance of supercapacitors. The all-in-one wound dressing has good mechanical properties, high water absorption capacity, and excellent biocompatibility to meet the practical requirements of clinical wound dressings. The supercapacitor layer in the all-in-one wound dressing has good electrochemical performance for charging and discharging before and after usage for wound treatment, and the wound dressing layer enhances the proliferative activity of NIH3T3 cells and accelerate the wound healing process in mice under the action of electrical stimulation provided by the supercapacitor. This study demonstrates that the all-in-one wound dressing would be an attractive candidate for portable wound healing, and broaden the scope of flexible electronics for biomedical applications.

Sheng Huang,Hua-Jiao Chen,Yan-Ping Deng,Xiu-hua You,Quan-hui Fang,Mei Lin

This work aimed to prepare a hydrogel film with antibacterial activity and excellent stability in enzymes as potential wound dressing. Using hyaluronic acid, carboxylated chitosan and gentamicin, antibacterial film HA-CC-GS was prepared through intermolecular covalent bonding. Surface morphology of film was evaluated by scanning electron microscope images. The film exhibited good water absorption capacity and appropriate water vapor permeation rate (WVPR), outstanding mechanical properties and remarkable resistance to enzymatic hydrolysis. The biocompatibility of the film was tested by CCK-8 assay, and the results showed that the viability of NIH3T3 cell was not affected by coculture with hydrogel film. In vitro antibacterial test indicated that the synthesized hydrogel film had obvious antibacterial properties. In addition, in vivo studies showed that antibacterial hydrogel film could protect wound from infection and promote wound healing. These results indicated that the prepared antimicrobial hydrogel films by cross-linking had great potential application in skin/tissue repair, suggesting as a hopeful wound dressing.
