Redox/pH/NIR-responsive degradable silica nanospheres with fluorescence for drug release and photothermal therapy

Xinli Li,Mengru Chen,Yanbao Zhao,Lei Sun

In this work, S S bond-doped porous silica nanoparticles with CuS nanocores (CuS/OS) were prepared as drug carriers for chemical-photothermal synergistic therapy and redox/pH/light triple-response drug release. The synthesized CuS/OS carriers have dendritic pore structure with particle size of about 100 nm. Tiopronin (MPG) was used as model drug to evaluate the release performance of the carrier. Chitosan (CS) and green fluorescent curcumin (Cur) were used as plugs to seal the loaded drug, which endows the CuS/OS carrier with pH-responsive release and monitoring properties. The obtained MPG-loaded CuS/OS-CS-Cur has pH/GSH/NIR triple responsive release and redox-responsive degradation. The introduction of CuS nanocores gives the CuS/OS system a good photothermal therapy function. The CuS/OS-CS-Cur and its degradation products have good biocompatibility.

Beibei Zhu,Jun Shi,Chongchong Liu,Jingguo Li,Shaokui Cao

As a novel two-dimensional material, Ti 3 C 2 MXenes has attracted lots of attention in biomedical filed for its large surface area and excellent near-infrared (NIR) responsiveness. In this paper, an in-situ growth and self-assembly approach was employed to combine gold nanorods (GNRs) with Ti 3 C 2 nanosheets to prepare intelligent sandwich-like Ti 3 C 2 @GNRs/PDA/Ti 3 C 2 nanohybrids . Compared with Ti 3 C 2 nanosheets, in-situ growth Ti 3 C 2 @GNRs possessed excellent photothermal conversion efficiency (45.89%), caused by the distinguished photothermal synergy between GNRs and Ti 3 C 2 nanosheets. Moreover, the high specific surface area of Ti 3 C 2 MXene and outstanding adhesion performance of PDA endowed Ti 3 C 2 @GNRs/PDA/Ti 3 C 2 nanohybrids with superior drug loading ability for doxorubicin hydrochloride (DOX) (95.88%). Besides, Ti 3 C 2 @GNRs/PDA/Ti 3 C 2 nanohybrids displayed distinct pH/NIR responsive drug release properties upon NIR irradiation owing to the strong π-π stacking interaction between Ti 3 C 2 @GNRs/PDA/Ti 3 C 2 and DOX, along with the excellent NIR-responsiveness of Ti 3 C 2 @GNRs/PDA/Ti 3 C 2 . This paper offers a practicable method to prepare Ti 3 C 2 MXene-based nanoplatform with synergistically enhanced NIR drug release behavior, brilliant biocompatibility and high drug loading efficiency, which is expected to be applied in remote cancer therapy.

Construction and Evaluation of DOX@MOF-199-PDA-HA Nanoparticle Drug Delivery System

Zhaoxiang Zeng,Lihua Huang,Caiyan Xu,Siqi Rao,Yin Chen,Wenjin Xu,Hao Hong

Metal-organic frameworks (MOFs) are a unique type of coordination polymer characterized by their high porosity and enormous specific surface area, which enable their use as carriers. We have constructed a novel MOFs-based nanodelivery system designed for multimodal combination therapy to enhance the delivery efficiency and efficacy of chemotherapeutic drugs. The synthesis of MOF-199, which inherently possesses a drug-release response function, was used as a nanodrug carrier. Doxorubicin (DOX) was encapsulated through physical adsorption, and the carrier surface was modified with a biomimetic material, polydopamine (PDA). Both PDA and hyaluronic acid (HA) molecules contain carboxyl groups, which can be connected through Fe3+-mediated coordination interactions. Transmission electron microscopy (TEM) and dynamic light scattering (DLS) were used to test the size, dispersity, and structural characteristics of the synthesized products, confirming their appropriate size and dispersity. BET(BET surface area) and TGA test results indicated that the nanocarrier exhibits good stability, a large specific surface area, and high porosity, making it suitable for use as a drug carrier. Ultraviolet spectrophotometry confirmed that the nanocarrier MOF-199-PDA-HA has the ability to consume GSH, with the drug loading rate and encapsulation efficiency of DOX in the system were measured as 39.58% and 73.26%, respectively. In cellular experiments, the DOX@MOF-199-PDA-HA group produced toxic hydroxyl radicals following H2O2 addition. After near-infrared light irradiation, both cellular toxicity and reactive oxygen species fluorescence intensity increased, indicating further enhanced anticancer efficacy. Using free HA as a control, it was confirmed that HA enhances tumor-specific accumulation of DOX@MOF-199-PDA-HA by binding to CD44 receptors on cancer cell surfaces.DOX@MOF-199-PDA-HA is a pH/GSH/NIR-responsive nanodelivery system with targeting functionality. This system ensures that drugs are released with higher efficiency in the tumor microenvironment through pH and glutathione (GSH) responsive drug-release mechanisms, thereby reducing toxicity to healthy tissues

Gold nanorods/graphene oxide nanosheets immobilized by polydopamine for efficient remotely triggered drug delivery

Qi Zeer,Shi Jun,Zhu Beibei,Li Jingguo,Cao Shaokui

Abstract In this paper, a novel one-pot strategy was utilized to immobilize gold nanorods (GNRs) onto the surface of PEGylated graphene oxide (GO-PEG) via polydopamine (PDA) to fabricate GNRs/GO@PDA hybrid nanosheets. The GNRs/GO@PDA nanosheets possessed excellent photostability and photothermal conversion efficiency (36.10%), due to the robust interfacial immobilization of PDA between GNRs and GO-PEG. Besides, GNRs/GO@PDA also could be utilized to be an efficient drug nano-vehicle that possessed an excellent loading ability for anticancer drug doxorubicin hydrochloride (DOX) (86.16%). Furthermore, the drug release of hybrid nanosheets could be triggered by pH value and near-infrared (NIR) light irradiation, which could be mainly ascribed to the strong π – π stacking interaction between GNRs/GO@PDA and DOX, as well as the brilliant NIR-responsive property of GNRs/GO@PDA. It was found that GNRs/GO@PDA possessed extremely low cytotoxicity to MCF-7 cells even at 250 μg mL −1 , while GNRs/GO@PDA(DOX) showed obvious cytotoxicity at 100 μg mL −1 . Thus, this paper presents a feasible strategy to fabricate GNRs/GO hybrid nanosheets with high drug loading capacity, excellent photothermal conversion property and pH/NIR-stimuli-responsive drug delivery performance, which possesses brilliant potentials to be an efficient platform for remotely triggered drug delivery. Gold nanorods/polydopamine-capped hollow hydroxyapatite microcapsules as remotely controllable multifunctional drug delivery platform

Yuchan Cao,Jun Shi,Zheng Wu,Jingguo Li,Shaokui Cao

In the present paper, hollow hydroxyapatite/polydopamine/gold nanorods (H-HAP/PDA/AuNRs) hybrid microcapsules was prepared for multi-responsive drug delivery via self-assembly method. PDA/AuNRs hybrid shell was employed to obstruct the initial burst drug release and endow the hybrid vehicle with superior photothermal conversion capacity, robust photothermal stability and brilliant near-infrared (NIR)-responsive delivery property. Moreover, due to the mesoporous structure of H-HAP as well as the strong mutual adsorption between PDA and drug, H-HAP/PDA/AuNRs exhibited excellent drug loading efficiency (95.6%). The drug release results demonstrated that the hybrid vehicle possessed distinct pH-/NIR- dual responsiveness, which due to the disintegration of HAP/PDA matrix in acid condition and the synergistically enhanced NIR responsiveness of PDA/AuNRs. Furthermore, cell viability results demonstrated that H-HAP/PDA/AuNRs exhibited outstanding biocompatibility and low bio-toxicity. Thus, the as-prepared H-HAP/PDA/AuNRs hybrid microcapsules hold great promise as novel smart drug carriers for remotely controllable drug delivery.

A multifunctional nanoplatform based on MoS2-nanosheets for targeted drug delivery and chemo-photothermal therapy

Yanbo Yang,Jianrong Wu,David H. Bremner,Shiwei Niu,Yu Li,Xuejing Zhang,Xiaotian Xie,Li-Min Zhu

Synergistic tumor treatment has recently attracted more and more attention due to its remarkable therapeutic effect. Herein, a multifunctional drug delivery system based on hyaluronic acid (HA) targeted dual stimulation responsive MoS 2 nanosheets (HA-PEI-LA-MoS 2 -PEG, HPMP) for active interaction with CD44 receptor positive MCF-7 cells is reported. Melanin (Mel), a new type of photothermal agent and doxorubicin (DOX) are both loaded onto the HPMP nanocomposite and can be released by mild acid or hyperthermia. The prepared HPMP nanocomposite has a uniform hydrodynamic diameter (104 nm), a high drug loading (944.3 mg.g −1 HPMP), a remarkable photothermal effect (photothermal conversion efficiency: 55.3%) and excellent biocompatibility. The DOX release from HPMP@(DOX/Mel) can be precisely controlled by the dual stimuli of utilizing the acidic environment in the tumor cells and external laser irradiation. Meanwhile, loading of Mel onto the surface can enhance the photothermal effect of the MoS 2 nanosheets. In vitro experiments showed that the HPMP@(DOX/Mel) nanoplatform could efficiently deliver DOX into MCF-7 cells and demonstrated enhanced cytotoxicity compared to that of the non-targeted nanoplatform. In vivo experiments in a breast cancer model of nude mice further confirmed that the HPMP@(DOX/Mel) significantly inhibited tumor growth under near infrared (NIR) laser irradiation, which is superior to any single therapy. In summary, this flexible nanoplatform, based on multi-faceted loaded MoS 2 nanosheets, exhibits considerable potential for efficient pH/NIR-responsive targeted drug delivery and chemo-photothermal synergistic tumor therapy.

Dynamically Responsive Scaffolds from Microfluidic 3D Printing for Skin Flap Regeneration

Xiaocheng Wang,Yunru Yu,Chaoyu Yang,Luoran Shang,Yuanjin Zhao,Xian Shen

Biological scaffolds hold promising perspectives for random skin flap regeneration, while the practical application is greatly limited by their insufficient vascularization ability and the lack of responsiveness during the dynamical healing process. Herein, a novel MXene-incorporated hollow fibrous (MX-HF) scaffold with dynamically responsive channels is presented for promoting vascularization and skin flap regeneration by using a microfluidic-assisted 3D printing strategy. Benefiting from the photothermal conversion capacity of the MXene nanosheets and temperature-responsive ability of poly(NIPAM) hydrogels in the MX-HF scaffolds, they display a near-infrared (NIR)-responsive shrinkage/swelling behavior, which facilitates the cell penetration into the scaffold channels from the surrounding environment. Moreover, by incorporating vascular endothelial growth factor (VEGF) into the hydrogel matrix for controllable delivery, the MX-HF scaffolds can achieve promoted proliferation, migration, and proangiogenic effects of endothelial cells under NIR irradiation. It is further demonstrated in vivo that the NIR-responsive VEGF@MX-HF scaffolds can effectively improve skin flap survival by promoting angiogenesis, decreasing inflammation, and attenuating apoptosis in skin flaps. Thus, it is believed that such responsive MX-HF scaffolds are promising candidates for clinical random skin flap regeneration as well as other diverse tissue engineering applications.

A chitosan derivative-crosslinked hydrogel with controllable release of polydeoxyribonucleotides for wound treatment

Yanzhen Sun,Xiaodong Jing,Yang Liu,Bing Yu,Hao Hu,Hailin Cong,Youqing Shen

Nucleic acid-based agents have advantages in therapeutic efficacy and biological safety. However, due to its facile degradability, it lacks an effective route of administration in wound treatment. Designing smart hydrogels for the spatiotemporally controllable delivery of nucleic acids is of great significance for clinical applications. Here, a near-infrared (NIR)-responsive nanocomposite hydrogel was prepared using methyl methacrylate (GMA)-modified chitosan as the macromolecular cross-linker, N-isopropylacrylamide (NIPAAm) as the backbone, and molybdenum disulfide nanosheets (MoS 2 NSs) as the nanocomponents. The polydeoxyribonucleotide (PDRN), a nucleic acid-based agent that promotes tissue regeneration , was loaded and delivered. The photothermal conversion capability of MoS 2 NSs enables customized care of PDRNs and antibacterial enhancement. In a full-thickness skin defect model, high-quality wound healing effects were demonstrated under the action of nanocomposite hydrogels. The proposed nanocomposite hydrogel provides a new reference for local delivery of nucleic acid-based agents.

Chen Zhengrong,Mo Quansheng,Mo Dandan,Pei Xiaomin,Liang Anru,Cai Jinhong,Zhou Bo,Zheng Li,Li Hongmian,Yin Feiying,Zhao Jinmin

Injury caused by excess reactive oxygen species (ROS) may lead to susceptibility to bacterial infection and sustained inflammatory response, which are the major factors impeding diabetic wound healing. By utilizing optimal anti-inflammatory, antioxidant, and antibacterial biomaterials for multifunctional wound dressings is critical in clinical applications. In this study, a novel electrospun PLGA/MoS2@Pd nanofiber membrane was synthesized by encapsulating antioxidant and near-infrared (NIR) responsive MOS2@Pd nanozymes in PLGA nanofibers to form a multifunctional dressing for diabetic wound repair. With excellent biocompatibility and hemostatic ability, this novel PLGA/MoS2@Pd nanofiber membrane can effectively reduce oxidative stress damage and intracellular inflammatory factors expression in fibroblasts by scavenging ROS. Additionally, the PLGA/MoS2@Pd nanofiber membrane exhibited favourable NIR-mediated photothermal antibacterial activity in vitro, with inhibition rates of 97.14% and 97.07% against Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli), respectively. In a diabetic rat wound infection model, NIR-assisted PLGA/MoS2@Pd nanofiber membrane effectively inhibited bacterial growth in the wound, reduced infection-induced inflammatory response, and promoted tissue epithelialization and collagen deposition, resulting in a wound healing rate of up to 98.5% on day 14. This study highlighted the construction of a multifunctional nanofiber membrane platform and demonstrated its promising potential as a clinical dressing for diabetic wounds.

A novel theranostic nanoplatform for imaging-guided chemo-photothermal therapy in oral squamous cell carcinoma

Mengqin Gu,Li Jiang,Liying Hao,Junzhuo Lu,Zhenqi Liu,Zixue Lei,Yijun Li,Chengge Hua,Wei Li,Xiyu Li

Oral squamous cell carcinoma (OSCC) is highly malignant and invasive, and current treatments are limited due to serious side effects and unsatisfactory outcomes. Here, we reported the terbium ion-doped hydroxyapatite (HATb) nanoparticle as a luminescent probe to encapsulate both the near-infrared (NIR) photothermal agent polydopamine (PDA) and anticancer doxorubicin (DOX) for imaging-guided chemo-photothermal therapy. The morphology, crystal structure, fluorescence, and composition of HATb–PDA–DOX were characterized. HATb–PDA showed a high DOX loading capacity. A theranostic nanoplatform showed pH/NIR responsive release properties and better antitumor outcomes in OSCC cells than monomodal chemotherapy or photothermal therapy, while keeping side effects at a minimum. Also, the luminescence signal was confirmed to be tracked and the increase of the red/green (R/G) ratio caused by the DOX release could be used to monitor the DOX release content. Furthermore, HATb–PDA–DOX plus NIR treatment synergistically promoted in vitro cell death through the overproduction of reactive oxygen species (ROS), cell cycle arrest, and increased cell apoptosis. Overall, this work presents an innovative strategy in designing a multifunctional nano-system for imaging-guided cancer treatment.

Fabrication of a smart drug delivery system based on hollow Ag2S@mSiO2 nanoparticles for fluorescence-guided synergistic photothermal chemotherapy

Gao Minjie,Han Zehua,Wang Zhihua,Zou Xueyan,Peng Lichao,Zhao Yanbao,Sun Lei

A novel near-infrared (NIR) light-triggered smart nanoplatform has been developed for cancer targeting and imaging-guided combined photothermal-chemo treatment. Notably, Ag 2 S has a dual function of photothermal therapy and fluorescence imaging, which greatly simplifies the structure of the system. It can emit fluorescence at 820 nm under an excitation wavelength of 560 nm. The phase-change molecule of 1-tetradecanol (TD) is introduced as a temperature-sensitive gatekeeper to provide the nanocarrier with controlled release capability of doxorubicin (DOX). The nanocarrier (HAg 2 S@mSiO 2 -TD/DOX) shows a high drug loading capacity of 26.3% and exhibits an apparent NIR-responsive DOX release property. Under NIR irradiation, the photothermal effect of HAg 2 S nanocores facilitated the release of DOX through the melting of TD. The cytotoxicity test shows that the nanocarriers have good biocompatibility. As the same time, the synergistic combination leads to a better cancer inhibition effect than individual therapy alone in vitro. Cell uptake tests indicate that the carriers have excellent fluorescence imaging ability and high cellular uptake for HepG2 cells. This work provides a new strategy for the fabrication of smart nanocarriers with simple structures for fluorescence-mediated combination cancer therapy. Multimodal theranostics augmented by transmembrane polymer-sealed nano-enzymatic porous MoS2 nanoflowers

Huiling Jiang,Yilin Du,Leilei Chen,Min Qian,Yafeng Yang,Taotao Huo,Xueying Yan,Tao Ye,Bing Han,Yi Wang,Rongqin Huang

Developing an all-in-one multimodal theranostic platform that can synergistically integrate sensitive photoacoustic (PA) imaging, enhanced photothermal therapy (PTT) and photodynamic therapy (PDT) as well as the nano-enzyme activated chemodynamic therapy (CDT) presents a great challenge for the current nanomedicine design. Herein, a simple hydrothermal method was used to prepare porous molybdenum disulfide (MoS 2 ) nanoflowers. These nanoflowers were assembled by three dimensional (3D)-stacked MoS 2 nanosheets with plentiful pores and large surfaces, which thus exhibited enhanced photothermal conversion via light trapping and peroxidase (POD)-like activity via active defects exposure. Consequently, this 3D-MoS 2 nanostructure could be well-sealed by polyethylene glycol-polyethylenimine polymer modified with nucleolar translocation signal sequence of the LIM Kinase 2 protein (LNP) via strong electrostatic interaction, which not only benefited to stably deliver anticancer drug doxorubicin (DOX) into the tumor cells for pH/NIR-responsive chemotherapy, but also provided strong photoacoustic, photothermal performances and stimulated generation of reactive oxygen species (ROS) for imaging-guided PTT/PDT/CDT combined therapy. This work promised a simple all-in-one multimodal theranostic platform to augment the potential antitumoral therapeutic outcomes.
