Due to the deep biological penetrability and therapeutic depth, the photothermal therapy over second near-infrared region (NIR-II) is booming against deep-seated tumors. Intensive endeavors are committed to looking for suitable photothermal agents (PTAs), but the progress seems not so satisfied toward the choice of PTA dosage. Herein, a comprehensive parameter, incident photon-to-thermal conversion coefficient (IPTCE), is used to evaluate the overall conversion of PTAs at different dosage, which will benefit for determining the optimized dosage of PTAs in pursuit of complete healing together with reduced long-term damages of nanodrugs. To prove the possibility, a series of anionic solid solution MXenes are chosen as hosts due to their versatile chemical compositions and correspondingly tunable light response. By deconvoluting fundamental structure–composition–property relationships, anionic regulation with extra electron injection leads to tunable free carrier densities and enhanced NIR-II harvesting. Ti 3 C 1.23 N 0.77 with high-level nitrogen exhibits extraordinary extinction coefficient (43.5 L g −1 cm −1 ) than other MXenes. The parameter of IPTCE can guide the choice of PTA concentration for complete photothermal healing in vitro and vivo. This proof-of-principle demonstration highlights synthetically tailoring of the light harvesting over NIR-II biowindow for a given host material by anionic regulation and further optimizes tumor photothermal therapy at low dose.

Near-infrared-II (NIR-II, 900–1700 nm) fluorescence imaging as one of the emerging imaging methods has shown the advantages of excellent spatial resolution and sensitivity. In order to obtain high-quality NIR-II fluorescence images and effective treatment results, it is essential to develop theranostic fluorescent nanoprobes with good tumor targeting and treatment capabilities. Herein, we designed and synthesized novel NIR-II fluorescent theranostic nanoprobes based on the organic dye IR783. IR783 was further encapsulated into liposomes, along with the chemotherapeutic drug doxorubicin (DOX) and the cervical cancer-targeting peptide TMTP1 (NVVRQ), to construct integrated nanoprobes for imaging and treatment. The constructed IR-DT nanoprobes, with its good tumor targeting ability, exhibit excellent imaging performance and significant therapeutic effects. With its powerful imaging capabilities, the IR-DT nanoprobes can achieve high-resolution fluorescence imaging of blood vessels, tumor lesions, and sentinel lymph nodes and could dynamically monitor the dissolution of carotid artery thrombi. The treatment results demonstrate that the nanoprobes possess significant advantages in both tumor-targeting capability and therapeutic efficacy. These findings suggest that it holds great potential to be developed into a theranostic nanoprobe for cervical cancer.

Multifunctional gold (Au)-based nanomaterials with high atomic number (symbol Z) and strong absorbance in the second near-infrared window (NIR-II) property are emerging as promising candidates for tumor thermo-radiotherapy. The main limitations of applying Au-based nanomaterials to biomedical studies include the absence of active tumor-targeting ability, penetrating efficiency, and stability. In this study, we present a novel type of tumor cell-derived stellate plasmonic exosomes (TDSP-Exos) for penetrative targeted tumor NIR-II thermo-radiotherapy and photoacoustic imaging. The TDSP-Exos are abundantly and easily produced by the incubation of tumor cells with gold nanostars, based on which gold nanostars promote the exocytosis of exosomes from tumor cells. Compared with bare gold nanostars, the TDSP-Exos exhibit pronounced accumulation in deep tumor tissues and perform well in both PA imaging and NIR-II thermo-radiotherapy against the tumor. Moreover, the TDSP-Exos improve tumor hypoxia to enhanced radiotherapy by NIR-II photothermal therapy. This work indicates that the tumor cell-derived exosomes have the potential to function as a universal carrier of photothermal agents for targeted tumor NIR-II thermo-radiotherapy.

The luminescence of traditional phosphorescence-based hypoxia probes is limited to the visible and first near-infrared wavelength regions (<1000 nm), which has defects of higher light scattering and lower penetration depth in contrast with the second near-infrared wavelength window (NIR-II, 1000–1700 nm) for optical bioimaging. Herein, 5,15-bis(2,6-bis(dodecyloxy)phenyl)-porphyrin platinum(II) (PpyPt) with J-aggregation induced NIR-II phosphorescence is reported. J-aggregates of PpyPt are confirmed by the X-ray diffraction data in the crystalline state. Moreover, the emission and excitation spectra of PpyPt in the solid states reveal NIR-II luminescence feature of PpyPt in J-aggregates. More importantly, by preparation of water-soluble PpyPt nanoparticles (PpyPt NPs 4.76 ) with J-aggregates, it has NIR-II phosphorescent lifetime of microseconds and good oxygen-sensitivity in water. Moreover, the good biological hypoxia-sensing potential of PpyPt NPs 4.76 is demonstrated in cells and 4T1-tumor-bearing mice. This study provides an efficient strategy to design NIR-II phosphorescent probe for sensitive tumor-hypoxia detection through the construction of J-aggregates.

Osteosarcoma as an aggressive malignant neoplasm is commonly treated by surgical resection, chemotherapy, and radiotherapy, but surgery induces bone defects and chemo/radiotherapy causes severe systemic toxicity. We have designed a novel low-toxic bifunctional 0D/2D heterojunction (HJ) that can be used to generate effective photothermal effects in the second NIR window (NIR-II, 1000–1350 nm) for simultaneous osteosarcoma treatment and bone regeneration. To construct this 0D/2D nanostructure, biocompatible, positively charged NIR-II-responsive carbon dots (CDs) were synthesized by an ultrafast microwave-assisted hydrothermal method, followed by electrostatically driven assembly on both sides of negatively charged WS 2 nanosheets. We found that the HJs exhibited enhanced NIR-II absorption and photothermal conversion efficiency. Due to the deep-tissue penetration ability of 1064 nm laser, complete osteosarcoma ablation was realized at a low power density even when being covered by a 10-mm-thick additional tissue. Furthermore, osteogenic differentiation was remarkably enhanced by mild photothermal effects of CD/WS 2 HJs, which significantly up-regulated bone-related gene expressions, particularly, heat shock protein expression. Designed CD/WS 2 HJs can be used in osteosarcoma treatment and bone-tissue engineering.

Multiplexed fluorescence imaging is mainly applied to cellular imaging. However, the application of an analogous technique to mammals remains challenging due to the presence of substantial scattering and autofluorescence in biological tissues in the visible (350–700 nm) and near-infrared I region (NIR-I, 700–900 nm). Herein, we report multiplexing excitation and single-channel detection in the near-infrared II region (NIR-II) with two fluorescent probes for dual-color imaging in vivo. The first one, FNIR-1090 was constructed on Pr-1099 derivative by attaching with 1-Ethylpiperazine through the SRN1 displacement reaction. The second one, IR-765 was synthesized by a one-step reaction between the Vilsmeier–Haack reagent with 2-(3,5,5-trimethylcyclohex-2-enylidene) malononitrile (TEMO). It is noteworthy that the absorption spectrum of FNIR-1090 is almost completely separated from that of IR-765, which facilitates dual-color in vivo imaging. The two dyes can be used for high-contrast dual-channel vivo imaging of different tissues. All these prominent performances show the great promise of FNIR-1090 and IR-765 in non-invasive, real-time, and dual-color NIR-II imaging (>900 nm) in vivo.

Here, we report the first PbS quantum dot (QD)-based micromotors with NIR-I light-driven photocatalytic propulsion and NIR-II fluorescence. Under the irradiation of NIR-I light (808 nm), PbS QD-doped cuprous oxide (Cu2O@PbS) micromotors can display efficient propulsion in a variety of biocompatible fuels such as malic acid, glucose, and urea. Among them, the Cu2O@PbS micromotors exhibit the best propulsion performance in a very low concentration of malic acid, with an average speed as high as 11.86 μm/s. The enhanced NIR-I photocatalytic activity of Cu2O@PbS micromotors benefits from the doping of NIR-I PbS QDs that can be excited by NIR-I light and exhibit high electron transport efficiency. The doped PbS QDs can effectively increase the absorption efficiency of the micromotors in the NIR-I region while also inhibiting the recombination of photogenerated electron–hole pairs. Interestingly, due to the presence of NIR PbS QDs, the Cu2O@PbS micromotors demonstrate prominent and stable NIR-II fluorescence (emission wavelength: 1100 nm), which offer promising potential for visualization of their position in vivo. In comparison to other photocatalytic micromotors, the simple fabrication strategy, excellent NIR-II fluorescence, together with the NIR-I light-dependent propulsion behavior of the current Cu2O@PbS micromotors, thus pave the way for further development of advanced smart “robots” for intelligent biomedical applications.

Breast-conserving surgery (BCS) has become the standard care for early-stage breast cancer. The accurate assessment of tumor margins is urgently required for BCS because positive resection margins often lead to local recurrence. To address this clinical dilemma, a Hf(IV)-coordinated NIR-II fluorescence “turn-on” nanoprobe based on the clinically approved NIR-II fluorescent dye indocyanine green (ICG) for intraoperative tumor visualization is developed. Notably, the fluorescence of ICG can be efficiently quenched by Hf(IV) and subsequently recovered in vivo, showing a remarkable fluorescence “quenching-recovery-amplification” capacity. This nanoprobe can effectively accumulate in tumor sites, accurately identifying submillimeter-sized primary and residual tumors with high sensitivity. In addition, subcutaneous, muscle-infiltrating, and orthotopic breast cancer models are built to repeatedly prove that this ultrasensitive nanoprobe is feasible for precise imaging-guided surgery in breast cancer. Overall, this study constructs an activatable fluorescent nanoprobe for real-time intraoperative tumor margin visualization, holding promise for complete surgical resection and reduction of local recurrence.

The development of high fluorescence quantum yield molecular fluorophores for second-order near-infrared (NIR-II) imaging is highly desirable but remains challenging due to the lack of reliable design guidelines. We report a method to construct “J-aggregates” by combining the inhibition of twisted intramolecular charge transfer (TICT) and electron-withdrawing strategies. Introducing electron-withdrawing substituents at the middle position of the classic boron dipyrromethene (BODIPY) backbone promotes J-type slip stacking, enhancing NIR-II fluorescence through increased electrostatic repulsion and intermolecular hydrogen bonding. The 3,5-aryl position is modified with sulfone and quaternary piperazine groups. The negative inductive effect (–I effect) from strong electron-withdrawing action increases the ionization potential (IP) of the amino group, inhibiting the TICT process and improving the quantum yield to 9.03 % and 16.9 %, respectively. Notably, the fluorescence quantum yield of NBDP-SO 2 increased to 18.2 % after self-assembly with F127, successfully applied in bioimaging. This study provides a new method for constructing high fluorescence quantum yield NIR-II emission by manipulating molecular stacking and establishing the relationship between molecular structure, intermolecular interactions, and fluorescence properties.

The second near infrared window is considered to be the optimal optical window for medical imaging and therapy as its capability of deep tissue penetration. The preparation of the gold nanorods with long wavelength absorption and low cytotoxicity is still a challenge. A series gold nanorods with large aspect ratio have been synthesized. Strong plasma absorption in the second near infrared window from 1000 to 1300 nm could be observed. The biocompatibility of the synthesized gold nanorods is dramatically improved via coating by bovine serum albumin (BSA), while the optical properties of which remains. The breast cancer tumor-bearing mouse could be well treated by the prepared gold nanorods with the NIR-II light intensity as low as 0.75 W/cm2. In summary, these results demonstrate the feasibility of using low illumination dose to treat tumor in the NIR-II region via the large aspect ratio gould nanoparticles.

The second near-infrared window (NIR-II, 1000–1700 nm) emitting phosphor is a key component for the phosphor-converted light-emitting diode (pc-LED), which has garnered significant attention. However, it remains a major challenge to discover efficient NIR-II broadband phosphors that can be excited by blue LEDs. Herein, Cr3+ and Ni2+ ions were co-doped into the Li2ZnSn3O8 (LZSO) host. LZSO:Cr3+,Ni2+ phosphor achieves a broadband NIR-II emission peak at 1465 nm with a large full width at half maximum (FWHM) of 300 nm and internal quantum efficiency (IQE) of 43.31% under excitation at 426 nm because of the efficient energy transfer (ET) from Cr3+ to Ni2+. In addition, a dual-emissive doped phosphor across the NIR-I (700–1000 nm) and NIR-II regions with a higher sensitivity at physiological temperatures was obtained. The relative sensitivity value (Sr) reaches 1.44% K−1 at 293 K, indicating that the LZSO:Cr3+,Ni2+ phosphor is a potential candidate for NIR optical thermometers in biotechnological applications. The manufactured NIR-II pc LED provides a radiative flux of 8.26 mW with a photoelectric conversion efficiency of 2.71% at a drive current of 100 mA and has promising potential in night vision, medical imaging, and spectral analysis applications.

Light-propelled nanomotors, which can convert external light into mechanical motion, have shown considerable potential in the construction of a new generation of drug delivery systems. However, the therapeutic efficacy of light-driven nanomotors is always unsatisfactory due to the limited penetration depth of near-infrared-I (NIR-I) light and the inherent biocompatibility of the motor itself. Herein, an asymmetric nanomotor (Pd@ZIF-8/R848@M JNMs) with efficient motion capability is successfully constructed for enhanced photoimmunotherapy toward hepatocellular carcinoma. Under near-infrared-II (NIR-II) irradiation, Pd@ZIF-8/R848@M JNMs convert light energy into heat energy, exhibiting self-thermophoretic locomotion to penetrate deeper into tumor tissues to achieve photothermal therapy. At the same time, functionalized with an immune-activated agent Resiquimod (R848), our nanomotors could convert a “cold tumor” into a “hot tumor”, transforming the immunosuppressive microenvironment into an immune-activated state, thus achieving immunotherapy. Dual photoimmunotherapy of the as-developed NIR-II light-driven Pd@ZIF-8/R848@M JNMs demonstrates considerable tumor inhibition effects, offering a promising therapeutic approach in the field of anticancer therapy.
