Baicalin ameliorates CUMS-induced depression-like behaviors through activating AMPK/PGC-1α pathway and enhancing NIX-mediated mitophagy in mice

Xiaohui Jin,Leilei Zhu,Shuaifei Lu,Caiyin Li,Ming Bai,Erping Xu,Jiduo Shen,Yucheng Li

Mitochondrial dysfunction has been reported to be involved in the pathogenesis of depression, and mitophagy is a key pathway for mitochondrial quality control. This study aimed to investigate the effect of baicalin on mitophagy in the hippocampus of mice exposed to chronic unpredictable mild stress (CUMS) and explore its potential mechanism. After exposure to CUMS for 6 weeks, mice were given baicalin (20 mg/kg) or fluoxetine (20 mg/kg) by oral gavage for 4 weeks, and HT22 cells were injured by corticosterone (CORT) in vitro. Depression-like behaviors were assessed by sucrose preference test and tail suspension test . The mitochondrial structure was observed by transmission electron microscopy . Detection of mitophagy and mitophagy-related protein by mitophagy kit and Western blot . The results showed that baicalin improved depressive-like behaviors in CUMS mice, and ameliorated mitochondrial structural impairment in the hippocampus neuron. Baicalin significantly down-regulated light chain 3(LC3)II/I, protein sequestosome 1 (P62), and translocase of the outer membrane 20 (TOM20), and up-regulated Nip-like protein (NIX), Adenylate activated protein kinase (AMPK), and Peroxisome proliferator-activated receptor-gamma coactivator (PGC)-1α. Furthermore, molecular docking showed that baicalin interacts with AMPK through hydrogen bonding. Baicalin increased NIX and AMPK, and improved mitophagy level and mitochondrial function in HT22 cells. Treatment with Phorbol 12-Myristate 13-acetate demonstrated that up-regulation of NIX ameliorated CORT-induced mitochondrial dysfunction in HT22 cells. In conclusion, the present study suggested that the antidepressant effect of baicalin may be related to the enhancement of NIX-mediated mitophagy through activating the AMPK/PGC-1α pathway by directly binding to AMPK.

Dinotefuran induces BNIP3/NIX-mitophagy, necroptosis, and damage in the uterus of adolescent mice

Jiawei Lei,Yushan Li,Mingxin Bai,Haoyue Du,Xueqin Shen,Hu Fu,Zhengli Yan,Yongfei Zhu

Xenobiotics such as pesticides can easily damage the reproductive organs during adolescence. Dinotefuran, the latest neonicotinoid insecticide, is considered to have low toxicity in vertebrates. However, research on its reproductive toxicity, especially damage to the female reproductive system in mammals is scarce. In this study, in vivo uterine toxicity was induced by oral administration of dinotefuran to mice at a dose ≥1/2 ADI (0.1 mg/kg/day), while in vitro toxicity was resulted from exposing cultured uteri to dinotefuran at levels equivalent to human blood levels (30–6000 ng/L). Hematoxylin-eosin staining revealed that dinotefuran caused uterine damage both in vivo and in vitro , while transmission electron microscopy detected mitophagy in the uterine cells. Furthermore, this study confirmed that dinotefuran triggered oxidative stress (decreased SOD and GSH, and elevated MDA), reduced mtDNA, and disrupted the BAX-to-BCL ratio in the uterus. Double immunofluorescence, qPCR, and western blot analyses found that dinotefuran induced excessive PRKN-independent mitophagy regulated by BNIP/NIX in the uterus, along with activation of NF-κB/NLRP3 signaling and the necroptosis pathway. It also revealed that dinotefuran increased uterine Caspase-1 /Cleaved CASPASE-1 levels by qPCR and western blot analysis. Collectively, these findings indicate that dinotefuran at concentrations ≥1/2 ADI value or equivalent to human blood levels induces mouse uterine toxicity in vivo and in vitro , respectively, and they all trigger oxidative stress, BNIP3/NIX-mediated mitophagy, NF-κB/NLRP3 signaling, and necroptosis in the uterus.

High-efficiency electrochemical H2O2 synthesis by heteroatom-doped NiX/Ni nanocomposites with honeycomb-like porous carbon

Mengran Liu,Canyu Liu,Tianfang Yang,Shixiang Hu,Siyun Li,Shizhe Liu,Yang Liu,Ye Chen,Bingcheng Ge,Shuyan Gao

Transition metal Ni anchored in carbon material represents outstanding 2e − oxygen reduction reaction (ORR) catalytic selectivity, but enhancing the adsorption strength of intermediate *OOH to promote its selectivity remains a major challenge. Herein, the NiX/Ni@NCHS composite catalyst with heteroatom doping (O,S) is modulated by controlling partial pyrolysis strategies on honeycomb-like porous carbon to manipulate the electronic structure of the metal Ni. With the synergistic effect of honeycomb structure and O atom, NiO/Ni@NCHS-700 exhibits an exceptional H 2 O 2 selectivity of above 89.1% across a wide potential range from 0.1 to 0.6 V in an alkaline electrolyte, and an unexpected H 2 O 2 production rate up to 1.47 mol g cat ‒1 h ‒1 @0.2 V, which outperforms most of the state-of-the-art catalyst. Meanwhile, NiS/Ni@NCHS exhibits excellent electrocatalytic performance, with 2e − ORR selectivity of 91.3%, H 2 O 2 yield of 1.85 @0.3 V. Density functional theory simulations and experiments results reveal that the heteroatom doping (O,S) method has been employed to regulate the adsorption strength of Ni atoms with *OOH, and combined with the self-sacrificing template-assisted pyrolysis approach to improve the microstructure of catalysts and optimize the active site. The heteroatom doping method in this work provides significant guidance for promoting 2e − ORR electrocatalysis to produce H 2 O 2 .

Tunable product selectivity in the hydrodeoxygenation of guaiacol over Nix-Fey/Al2O3 catalysts under mild conditions

Hao Zhao,Chongbo Cheng,Pengfei Li,Fengjie Bian,Dekui Shen,Xiaoxiang Jiang

The selective hydrodeoxygenation (HDO) of lignin-derived phenolics such as guaiacol into high-value chemicals under mild conditions presents a persistent challenge in biomass valorization. In this work, a series of Ni x -Fe y /Al 2 O 3 catalysts were synthesized by a sol-gel method and evaluated for the HDO of guaiacol. Characterization (XRD, XPS, H 2 -TPR, NH 3 -TPD, etc.) revealed that the Ni x -Fe y /Al 2 O 3 catalysts possess enhanced textural properties, improved metal dispersion, and tailored surface acidity, collectively contributing to their high activity and selectivity. The introduction of Fe promotes the formation of a Ni 3 Fe alloy and defect-rich FeO x species. Under mild conditions (180 °C, 2 MPa H 2 ), Ni 5 /Al 2 O 3 achieved full guaiacol conversion with > 85% selectivity to 1,2-cyclohexanediol. By introducing Fe and increasing the temperature to 260 °C (2 MPa H 2 ), the Ni 5 -Fe 1 /Al 2 O 3 catalyst shifted the reaction pathway toward deeper deoxygenation, yielding cyclohexane with over 78% selectivity. This tunable selectivity originates from a synergistic mechanism in which metallic Ni/Ni 3 Fe sites facilitate hydrogenation and hydrogenolysis, while acid sites and FeO x -derived oxygen vacancies promote the dehydration of diol intermediates, especially at higher temperatures. This study provides a design-oriented bimetallic strategy for the targeted valorization of lignin into distinct high-value chemicals.

Air-stable and reusable porous silica-supported ultrafine Nix-NiO nanoparticles for semi-hydrogenation of alkynes

Yaxin Xing,Yatao Su,Luyao Li,Zengli Jia,Gang Feng,Hongchao Wang,Yuangong Zhang,Xin Wen

The practical application of non-noble metal-based hydrogenation catalysts is limited because of their low activity and easy oxidation in air. Here, porous silica-supported ultrafine nickel oxide (NiO) was prepared by calcining an organic–inorganic hybrid 3D nanomaterial that was formed via the Michael addition/Schiff’s base reaction, and hydrolyzation of triethoxysilane. NiO nanoparticles with ca. 2.6 nm diameter was uniformly dispersed and confined in the cogenerated mesoporous SiO 2 skeleton, thanks to the coordination between pyrogallol/amino units and Ni 2+ ions, and the isolation effect of Si-O-Si network. The as-prepared catalyst featured mesoporous structure and high specific surface area (715 m 2 /g), and exhibited excellent activity, selectivity and reusability in the semi-hydrogenation of alkynes. Small amount of metallic nickel (Ni x -NiO) that formed on the surface of NiO nanoparticles during the semi-hydrogenation acted as the active species, and the moderate adsorption/desorption energies on Ni x -NiO facilitated the activity enhancement. Most importantly, it showed constant catalytic activity and selectivity after a long-term storage in air. This work provides a green and feasible strategy, as an alternative to the traditional impregnation method, for the preparation of ultrafine metal-supported catalysts with air-stability and reusability.

Supported metal clusters: Nix/CuZnAl catalysts effectively improve the performance of hydrogen evolution from methylcyclohexane dehydrogenation

Junchi Meng,Xingzhou Yuan,Yang Yan,Feng Zhou,Huixia Ma,Wenyu Long,Lei Zhang,Jian Zhang

Background Recently, with the emphasis on hydrogen as alternative clean energy, it is necessary to identify a safe and efficient technique. As a hydrogen storage technique, the liquid organic hydrogen carrier (LOHC) technique has the advantages of high hydrogen storage density, high safety, and low technical cost. Methods A series of Ni x /CuZnAl catalysts were synthesized using co-precipitation and ultrasonic-assisted incipient wetness impregnation methods. Combining XRF, XRD, Uv-vis, H 2 -TPR, XPS, TEM, and SEM techniques, we show the morphology of the active site during the reaction, and further explore the effects of the formation of the metal cluster NiCuZn. Significant findings The multi-metal oxide phase in the sample disappeared after reduction and was transformed into the metal cluster NiCuAl. During the formation of the metal cluster NiCuAl on the catalyst surface, electron transfer towards Ni occurred and the reduction temperature of Ni was lowered, which contributed to the electronic effect and synergy. In this work, The Ni 10 /CuZnAl catalyst shows the best dehydrogenation activity and selectivity, the catalytic effect was 15–20 times that of the CuZnAl catalyst, and the hydrogen evolution rate reached 77.31 ± 1.25 mmol/g cat /h. Moreover, based on the characterization analysis, the work speculates that the active atom H simultaneously dissociates and desorbs on the Cu and Ni surfaces of the metal cluster. In addition, as a possible active center, with a strong metal-support interaction, the metal cluster NiCuZn promotes the dehydrogenation reaction.

Baicalin attenuates corticosterone-induced hippocampal neuronal injury by activating mitophagy in an AMPK-dependent manner

Yuan Liu,Xiaohui Jin,Caiyin Li,Jingjing Bu,Baoying Wang,Ming Bai,Pan Su,Erping Xu,Yucheng Li

Defective mitophagy is closely related to the neuronal dysfunction and major depressive disorder (MDD). Our previous study found that baicalin could enhance nip-like protein (NIX)-mediated mitophagy and exhibit antidepressant effects, and predicted that AMPK may be the pharmacological target of baicalin. However, validated experiments are lacking. In the present study, we first demonstrated the effect of baicalin on hippocampal NIX-mediated mitophagy in CORT-induced depressive mice. Secondly, we transfected siRNA to knockdown AMPK, PGC-1α, and NIX respectively in HT22 cells, and detected the effects of baicalin on mitochondrial function, AMPK/PGC-1α/NIX pathway protein expression and mitophagy levels. Finally, AAV-shAMPKα was injected into hippocampal CA3 to knockdown AMPK in mice to validate the antidepressant effects of baicalin in vivo. The results showed that CORT induced depressive-like behaviors, accompanied with neuronal damage, mitochondrial injury, and inhibited mitophagy in the hippocampus, which were prevented by baicalin (20 mg/kg) treatment. In HT22 cells, baicalin remarkably ameliorated mitochondrial dysfunction and mitophagy disturbance induced by CORT, and these protective effects of baicalin were blocked by knockdown of AMPK, PGC-1α and NIX. Moreover, the beneficial effects of baicalin on depressive-like behaviors and NIX-mediated mitophagy were suppressed by knockdown of AMPKα in mice. Our present results further demonstrated that baicalin promotes NIX-mediated mitophagy and improves depression in an AMPK-dependent manner.

Camelliasaponin B1, a Saponin from Camellia oleifera Seed, Protects Against Oxidative Stress and Is Associated with Reduced BNIP3/NIX-LC3B Expression in PC12 Cells

Xiaoqing Feng,Xiao Zhou,Shushan Jia,Jingzhen Chen,Peiwang Li,Yan Yang,Wei Wu,Lijuan Jiang,Wenbin Zeng,Changzhu Li,Qiang Liu,Yunzhu Chen

Camelliasaponins, bioactive constituents abundant in the by-products ofCamellia oleiferaoil production, exhibit diverse biological activities. However, their potential in regulating neuroprotective mitophagy remains largely unexplored. This study identifies camelliasaponin B1 (CSB1) as an abundant component inC. oleiferaseeds and investigates its cytoprotective mechanisms against oxidative stress. Using an in vitro model of H2O2-induced oxidative damage in PC12 cells, we found that CSB1 pretreatment significantly alleviated oxidative stress, as evidenced by reduced reactive oxygen species (ROS) accumulation and enhanced antioxidant enzyme activities (SOD, CAT, GSH-Px). CSB1 also preserved mitochondrial function, restoring membrane potential (ΔΨm), ultrastructure, and respiratory capacity. Mechanistically, CSB1 reduces the expression of BNIP3/NIX-LC3B pathway-related proteins, suggesting a modulatory effect on mitophagy, as supported by transcriptomic analysis, Western blotting, and immunofluorescence. Molecular docking computationally predicted potential interactions between CSB1 and BNIP3/NIX proteins, which require experimental validation. Collectively, these findings suggest that CSB1 acts as a cytoprotective agent that enhances antioxidant defenses, safeguards mitochondrial integrity, and is associated with reduced BNIP3/NIX-LC3B expression and co-localization, offering a potential molecular basis for its development as a neuroprotective agent targeting oxidative stress-related mitochondrial dysfunction.
