Neuron-specific enolase (NSE), a tumor marker of small cell lung cancer (SCLC), has high application value in the early diagnosis of SCLC. In this study, a dual signal electrochemical aptasensor for NSE was constructed based on hemin/reduced graphene oxide/multi-walled carbon nanotube (H–rGO–MWCNT) nanocomposites. Hemin played a dual role, functioning not only as an in situ electrochemical probe but also exhibiting excellent peroxidase-like properties, effectively catalyzing the electroreduction of H2O2. Reduced graphene oxide and multi-walled carbon nanotubes exhibited excellent conductivity. Through their binding with hemin, the nanocomposites achieved a larger specific surface area, providing numerous active sites for capturing the NSE aptamer. In the presence of NSE, the specific adsorption between the antigen and the aptamer formed a stable antigen–aptamer structure, which inhibited the performance of hemin, resulting in the weakening of the electrochemical signals of hemin and H2O2. Leveraging these characteristics, the sensitive and cost-effective dual-signal electrochemical aptasensor has been fabricated for the detection of NSE. One signal corresponded to differential pulse voltammetry (DPV) of hemin, while the other signal was derived from chronoamperometry, capturing the catalytic reduction of H2O2. The linear ranges for NSE were 1 pg mL−1 to 1 μg mL−1 and 100 pg mL−1 to 100 ng mL−1 with the limit of detection (LOD) of 0.21 pg mL−1 and 11.22 pg mL−1 by DPV and chronoamperometry, respectively. In addition, this aptasensor exhibited good reproducibility, stability and specificity. The recovery of NSE in human blood serum samples was from 89% to 131%. It provided a promising strategy for the detection of NSE in clinical diagnostics.

As one of the specific markers for small cell lung cancer (SCLC), the clinical value for serum neuron-specific enolase (NSE) detection in diagnosis, monitoring and therapy of SCLC is considerable. Herein, a sensitive amperometric aptasensor was developed for the detection of NSE based on nickel-manganese oxide@acetylene black/platinum nanoparticles (Ni 6 MnO 8 @AB/PtNPs) composite as signal amplifier. The combination of Ni 6 MnO 8 , AB and PtNPs not only showed synergistically catalytic activity to hydrogen peroxide (H 2 O 2 ), but also possessed lots of binding sites for loading the thiol-terminated signal probes (TSPs) through Pt-S bonds. Additionally, nitrogen-doped carbon nanotubes/poly (diallyldimethylammonium chloride)/ZnO microflowers (N-CNTs/P/ZnO MFs) exhibited outstanding electric conductivity and was used as substrate material to load more biotin-terminated capture probes (BCPs). In the presence of the NSE, the TSPs and BCPs specifically recognized the NSE to form a sandwich-type electrochemical aptasensor, which exhibited a broad linear response range from 0.00001 ∼ 100 ng mL −1 and a low detection limit of 2.86 fg mL −1 (S/N = 3). In addition, the electrochemical aptasensor obtained favorable detection results in diluted human serum, indicating that it had potential application in clinical NSE detection.

The construction of advanced systems capable of accurately detecting neuron-specific enolase (NSE) is essential for rapidly diagnosing small-cell lung cancer. In this study, an electrochemiluminescence (ECL) resonance energy transfer immunosensor was proposed for the ultra-sensitive detection of NSE. The co-reactants C2O42− and Ru(bpy)32+ were integrated to form a self-enhanced ECL luminophore (Ru-ZnMOF) as the ECL donor. The abundant carboxyl functional groups of Ru-ZnMOF supported antibody 1 via an amidation reaction. Polydopamine-modified zinc dioxide nanoflowers, as ECL acceptors, inhibited Ru-ZnMOF ECL signaling. The linear range of NSE was 10 fg/mL to 100 ng/mL with a detection limit of 3.3 fg/mL (S/N = 3), which is suitably low for determining NSE in real samples.

A double-antibody sandwich SERS immunoassay was developed for the highly specific capture of NSE in clinical serum. • The interpretable 1D-CNN model achieved an AUC of 0.9704, significantly outperforming DNN and RNN architectures. • SHAP and saliency maps deciphered the “black box”, identifying key Raman shifts at 607 and 1078 cm -1 as biomarkers. • A high-performance PSS@Cu-Ag@Ag SERS substrate with core-shell structure was fabricated using magnetron sputtering. • The periodic array provided dense “hot spots” with superior reproducibility, achieving RSD values below 6.4%. To address the issues of low imaging sensitivity in the clinical diagnosis of ischemic stroke (IS) and the cumbersome procedures associated with traditional Neuron-Specific Enolase (NSE) testing methods, this study proposes a rapid diagnostic approach which integrates Surface-enhanced Raman scattering (SERS) immunonanosensing technology with deep learning. Using magnetron sputtering, a periodic core–shell structured PSS@Cu-Ag@Ag active substrate was fabricated on a periodic microsphere array (PSS). The NSE biomarker in serum was specifically captured via a double-antibody sandwich immunoassay, and the spectra were subjected to deep feature extraction using a one-dimensional convolutional neural network (1D-CNN). Results showed that the model achieved an area under the curve average AUC of 0.9704 under five-fold cross-validation, and the SHapley Additive exPlanations (SHAP) interpreter successfully identified key differential Raman shift such as 607 cm −1 and 1078 cm −1 , enhancing the interpretability of the algorithm. This approach enables robust and transparent classification of clinically heterogeneous samples, providing a highly sensitive and intelligent medical solution with great potential for practical clinical implementation in the auxiliary diagnosis of IS. In the field of biomedical detection, chemiluminescence (CL) immunosensors are valued for their high sensitivity, strong selectivity, and straightforward operation. Nonetheless, the majority of conventional CL immunosensors are flash-type, requiring large-scale instruments to capture CL signals, which restricts their use in point-of-care testing (POCT). In this work, konjac glucomannan (KGM) is firstly carboxymethylated to form CKGM, and chitosan (CS) is then connected through electrostatic interaction to obtain CKGM-CS hydrogel. Cu 2+ is subsequently loaded onto CKGM-CS hydrogel via coordination and electrostatic interactions. Luminol is finally encapsulated in CKGM-CS hydrogel by electrostatic interaction and adsorption. The obtained CKGM-CS@luminol/Cu 2+ exhibits a high density of “glow-type” CL emission. On this basis, an ultrasensitive label-free immunosensor was developed for the detection of neuron-specific enolase (NSE) by immobilizing specific antibodies, where the quantitation relies on the immunorecognition event directly modulating the CL signal. This dual-detection platform integrates a CL instrument with a smartphone. For the CL instrument, the linear range and limit of detection (LOD) of the established method are 0.1–1000 pg/mL and 0.027 pg/mL, respectively. For the smartphone, the linear range and LOD are 0.1–1000 pg/mL and 0.043 pg/mL, respectively. Ultimately, the technique can track NSE concentration in actual samples using two distinct capture approaches, offering a versatile method for developing sensitive, cost-effective, and simple dual detection platform label-free immunosensors for NSE detection. Moreover, employing a smartphone as a readout device simplifies the detection process, making it convenient for POCT and providing a novel approach for early small cell lung cancer (SCLC) diagnosis.

Purpose To explore the potential of circRNAs as biomarkers in non-invasive body fluids for monitoring chemotherapy resistance in SCLC patients. Methods CircRNAs were screened and characterized using transcriptome sequencing, Sanger sequencing, actinomycin D treatment, and Ribonuclease R assay. Our study involved 174 participants, and serum samples were collected from all chemotherapy-resistant patients (n = 54) at two time points: stable disease and progressive disease. We isolated and identified serum extracellular vesicles (EVs) from the patients using ultracentrifugation, transmission electron microscopy, nanoflow cytometry, and western blotting analysis. The expression levels of serum and serum EVs circRNAs were determined by quantitative real-time polymerase chain reaction (qRT-PCR). The impact of circRNA on the function of SCLC cells was assessed through various assays, including proliferation assay, scratch assay, transwell assay, and cisplatin resistance assay. Results Hsa_circ_0041150 was found to be upregulated in chemoresistant SCLC cells and played a role in promoting proliferation, invasion, migration, and cisplatin resistance. Furthermore, the expression levels of hsa_circ_0041150 in serum and serum EVs increased when SCLC patients developed resistance after a first-line chemotherapy regimen. When combined with NSE, the monitoring sensitivity (70.37%) and specificity (81.48%) for chemotherapy resistance significantly improved. Moreover, the expression level of hsa_circ_0041150 showed significant associations with time to progression from SD to PD, and high hsa_circ_0041150 levels after drug resistance were more likely to cause chemotherapy resistance. Additionally, hsa_circ_0041150 demonstrated valuable potential in monitoring the progression from initial diagnosis to chemotherapy resistance in SCLC patients. Conclusion Thus, EVs hsa_circ_0041150 holds promise as a biomarker for monitoring chemotherapy resistance in SCLC patients.

Current methods for the early diagnosis of cancer can be invasive and costly. In recent years, exosomes have been recognized as potential biomarkers for cancer diagnostics. The common methods for quantitative detection of exosomes, such as nanoparticle tracking analysis (NTA) and flow cytometry, rely on large-scale instruments and complex operation, with results not specific for cancer. Herein, we present a tri-channel electrochemical immunobiosensor for enzyme-free and label-free detecting carcino-embryonic antigen (CEA), neuron-specific enolase (NSE), and cytokeratin 19 fragments (Cyfra21-1) from exosomes for specific early diagnosis of lung cancer. The electrochemical immunobiosensor showed good selectivity and stability. Under optimum experimental conditions, the linear ranges were from 10−3to 10 ng/mL for CEA, 10−4to 102ng/mL for NSE, and 10−3to 102ng/mL for Cyfra21-1, and a detection limit down to 10−4ng/mL was achieved. Furthermore, we performed exosome analysis in three kinds of lung cancer. The results showed a distinct expression level of exosomal markers in different types. These works provide insight into a promising alternative for the quantification of exosomal markers in specific diseases in the following clinical bioassays. Keywords:electrochemical immunobiosensor;exosome detection;multiple biomarkers;lung cancer

Cell replacement is a promising approach for neurodegenerative disease treatment. Somatic cells such as fibroblasts can be induced to differentiate into neurons by specific transcription factors; however, the potential of viral vectors used for reprogramming to integrate into the genome raises concerns about the potential clinical applications of this approach. Here, we directly reprogrammed rat embryonic skin fibroblasts into induced neurons (iNs) via six small-molecule compounds (SMs) (VPA, CHIR99021, forskolin, Y-27632, Repsox, and P7C3-A20). iNs exhibit typical neuronal morphology, and immunofluorescence showed that more than 96% of the iNs expressed the early neuronal marker class III beta-tubulin (TUJ1) and that more than 91% of iNs expressed the mature neuronal marker neuron-specific enolase (NSE) after 10 days of reprogramming. Quantitative real-time polymerase chain reaction also showed that most iNs expressed the dopaminergic neuron marker tyrosine hydroxylase, the neural marker Nur correlation factor 1, the (γ-aminobutyric acid, GABA) GABAergic neuronal marker GABA, and the cholinergic neuron marker choline acetyltransferase. In addition, we found that cell proliferation decreased during reprogramming and that protein synthesis increased initially and then decreased. SMs were mixed with hydrogels, and the hydrogels were implanted subcutaneously into the backs of rats. After 7 days, the TUJ1 and NSE proteins were expressed in surrounding tissues, indicating that SMs caused reprogramming in vivo. In summary, rat skin fibroblasts can be efficiently reprogrammed into iNs by SMs in vitro and in vivo.

Lung cancer is the main cancer that endangers human life worldwide, with the highest mortality rate. The detection of lung tumor markers is of great significance for the early diagnosis and subsequent treatment of lung cancer. In this study, a vertical graphene field effect transistor (VGFET) immunosensor based on graphene/C 60 heterojunction was created to offer quantitative detections for the lung tumor markers carcinoembryonic antigen (CEA), cytokeratin 19 fragment (Cyfra21-1), and neuron-specific enolase (NSE). The experimental results showed that the sensitive range for standard antigen is between 1 pg/ml to 100 ng/ml, with a limit of detection (LOD) of 5.6 amol/ml for CEA, 33.3 amol/ml for Cyfra 21–1 and 12.8 amol/ml for NSE (1 pg/ml for all). The detection accuracy for these tumor markers was compared with the clinically used method for clinical patients on serum samples. Results are highly consistent with clinically used immunoassay in its efficient diagnosis concentration range. Subsequently, the mesoporous silica nanospheres (MSNs) with an average size of 90 nm were surface modified with glutaraldehyde, and a second antibody was assembled on MSNs, which fixes nanospheres on the antigen and amplified the field effect. The LODs for three markers are 100 fg/ml (0.56 amol/ml for CEA) under optimal circumstances of detection. This result indicates that specific binding to MSNs enhances local field effects and can achieve higher sensing efficiency for tumor marker detection at extremely low concentrations, providing effective assistance for the early diagnosis of lung cancer.
