NdF 3 nanoflakes and hexagonal NaNdF 4 nanorods were successfully prepared in a facile manner by the one-pot microwave-assisted hydrothermal reaction of Nd(NO 3 ) 3 ·6H 2 O and NaF within 3 h at pH = 1 and pH = 3–10, respectively. Ethylenediaminetetraacetic acid disodium salt (C 10 H 14 N 2 O 8 Na 2 ·2H 2 O, EDTA–2Na) was used as the morphology-controlling agent. The size, morphology, and crystallinity of the resulting nanocrystals can be easily tuned by adjusting the reaction temperature and time. The as-prepared NdF 3 nanoflakes and NaNdF 4 hexagonal prisms were characterized by X-ray diffraction and scanning electron microscope. All nanocrystals exhibited a typical hexagonal phase, and their crystallization improved with increasing reaction temperature and reaction time. A strong emission spectrum was observed at approximately 850–900, 1060, and 1330 nm for both NdF 3 nanoflakes and NaNdF 4 nanorods under a laser excitation of 354 nm, indicating that Nd 3+ exhibits high luminescence in these two nanocrystals.

Photothermal therapy (PTT) has been widely used for the treatment of various medical conditions due to their noninvasive and cost-effective advantages. However, the light absorption and scattering of the biosystem limit the deep tissue applications of conventional PTT probes. In this paper, we proposed the sub-10 nm NaNdF4 nanocrystals with both incident and emission wavelengths located at the optical window. Under 800 nm laser excitation, the maximum light-to-heat conversion efficiency of these ultrasmall photothermal agents is evaluated to be 85%. In addition, the conversion efficiency can be further adjusted through varying the doping concentration, changing the probe size, and coating the inert shell. Meanwhile, temperature feedback on the basis of the Nd3+ luminescence signal validates the precise temperature manipulating inside the biological tissue, evidencing the feasibility of the proposed probes for PTT treatment. Beneficial from the efficient photothermal conversion and low energy loss of excitation/emission photons in organic tissue, the effective treatment depth of the nanoprobes goes to 6 mm with the actual laser power density only ∼0.2 W/cm2. Finally, based on the rapid heating response of the formed nanoprobes, fast ablation treatment of a 1.5 cm pork tissue is achieved using pulsed laser.

This work reports the light-to-heat conversion (LHC) behavior of NaNdF4doped with Sm3+. Due to the cross-relaxation between Nd3+and Sm3+, the improved LHC is obtainable by introducing 5% Sm3+. When the laser power density is only 1.72 W/cm2, the spot temperature of NaNdF4:5%Sm3+powder reaches as high as 138.7 ± 4.04 °C. More importantly, the photoheating response to the pump laser has favorable linear characteristics within a specific power range. A simple physical model is applied to analyze the relationship between photothermal heating and pump power. Finally, the temperature-responsive luminescence anti-counterfeiting is designed by combining the LHC material with the NaYF4:Yb3+/Ho3+/Ce3+microcrystals. This novel strategy only requires two laser beams, and thus is more convenient to apply.

The narrow absorption and emission bands, long fluorescence lifetime, and excellent stability of rare earth nanoparticles (referred to as RE NPs) make them very attractive for multimodal imaging and therapy of cancer. Their narrow absorption requires the careful selection of laser wavelength to achieve the best performance, particularly for RE NPs simultaneously having photothermal and photoluminescent properties (e.g., Nd-based nanoparticles), which has not been investigated. Herein, we prepared a series of different-sized NaNdF4 nanoparticles (referred to as NNF NPs) (i.e., 4.7, 5.9, 12.8, and 15.6 nm) from ultrasmall nanoclusters and investigated their in vitro and in vivo size-dependent photothermal conversion and photoluminescence under irradiation by a 793 nm laser and an 808 nm laser, respectively. We find that all nanoparticles exhibited the better photothermal conversion performance under the irradiation of the 808 nm laser than under the 793 nm laser, of which 12.8 nm NNF NPs showed the best performance, and the temperature of their solution can be quickly increased from 30 °C to around 60 °C within 10 min under the irradiation of the 808 nm laser with a power intensity of 0.75 W/cm2. When we used the 793 nm laser to excite these NNF NPs, we found that all nanoparticles exhibited the stronger photoluminescence in the second near-infrared window (NIR-II) than under the excitation by the 808 nm laser, of which 15.6 nm NNF NPs possessed the strongest NIR-II luminescence. We then modified 12.8 nm NNF NPs with phospholipid carboxyl PEG and functionalized with RGD for actively targeted imaging of cancer. The NaNdF4@PEG@RGD nanoparticles (referred to as NNF-P-R NPs) have good biocompatibility, stability, and excellent targeting capability. The in vivo result show that 12.8 nm NNF NPs exhibited better photothermal conversion performance under the irradiation of the 808 nm laser, and stronger NIR-II fluorescence under irradiation of the 793 nm laser, which are consistent with the in vitro result. This work demonstrates the significance of selection of the proper laser wavelength for maximally taking advantage of RE nanoparticles for the diagnosis and treatment of cancer.

Near-infrared (NIR) dyes can overcome the weak absorption of lanthanide nanoparticles (NPs) by antenna sensitization, offering new avenues to develop efficient and versatile lanthanide nanomaterials. However, current research on dye-sensitized lanthanide NPs for photothermal conversion is still preliminary, and the involved excited-state dynamics and interfacial interactions remain elusive. Herein, steady-state/transient absorption spectroscopy and theoretical calculation are used to investigate the coordination and aggregation states of cypate dyes on NaNdF 4 NPs, revealing the influence of interfacial interactions on resonant energy transfer. Synergetic heat-generation mechanism of lanthanide cross-relaxation and dye intermolecular collisions is further proposed. The photothermal conversion efficiency of cypate-NaNdF 4 nanocomposites reaches 50.4%, outperforming those of typical photothermal materials with high NIR absorption. Moreover, the intersystem crossing of cypate can be inhibited due to the depopulation of the S 1 exciton via ET, thereby improving anti-photobleaching ability. These dye-sensitized NaNdF 4 nanocomposites exhibit superior photothermal effect, stability and NIR-II luminescence, showing great potential in theranostic applications.

Manipulation of autophagic processes has emerged as a promising strategy for synergizing nanoagent-mediated photothermal therapy (PTT). Most of the current studies focus on improving PTT efficacy by inhibiting pro-survival autophagy induced by the heat generated from the photothermal process. However, autophagy induced by the nanoagents is usually ignored, which may weaken the effect of autophagy-mediated efficacy improvement in PTT if induced autophagy is pro-death. Therefore, this work aims at developing a nanoagent that is able to induce heat-synergetic pro-survival autophagy to optimize the efficacy of PTT. An approach is developed to coat carbon layer, polyethylenimine (PEI), and folic acid (FA) on NaYF 4 :Er,Yb,Nd@NaNdF 4 (DCNPs@C@PEI@FA, DCPF) nanoparticles successively, giving access to the nanoagent to induce pro-survival autophagy. The synthetic imaging-guided photothermal nanoagent displays outstanding targeting ability and biocompatibility based on the surface modification of PEI and FA. By using an autophagy inhibitor chloroquine, a conspicuously synergistic effect on DCPF-mediated PTT in vitro and in vivo tumor models (HeLa) is achieved. A promising strategy is presented here to enhance the efficacy of imaging-guided PTT by modulating the autophagy induced by the nanoagent.

Rare-earth nanoparticles (RE NPs) with narrow long wavelength emissions have been recently investigated for their potential application for fluorescence imaging in the second near-infrared window (NIR-II). Previously these RE NPs have a very limited application in the diagnosis and treatment of deep-seated tumors such as brain tumors, due to their weak fluorescence in the range of 1300–1700 nm. Herein, we report a significant enhancement of more than 10 times regular emission of NaNdF 4 nanoparticles at 1340 nm wavelength by coating them with an inert layer of NaLuF 4 , followed by sensitizing with a near-infrared dye (IR-808). We deliver these highly bright nanoparticles into the brain by using focused ultrasound to temporarily open the blood-brain barrier (BBB), and then detect the orthotopic glioblastoma by fluorescence imaging at 1340 nm. The images obtained from long wavelength fluorescence ( i.e. 1340 nm) exhibited better resolution and contrast compared to the short wavelength fluorescence ( i.e. 1060 nm). Our study not only provides insights for enhancing often overlooked emissions of rare-earth nanoparticles for NIR-II fluorescence imaging of deep-seated tumors, but also demonstrates great potential of focused ultrasound based technology in delivering nanotheranostic agents.

A noninvasive strategy for in situ diagnosis and precise treatment of bacterial biofilm infections is highly anticipated but still a great challenge. Currently, no in vivo biofilm-targeted theranostic agent is available. Herein, we fabricated intelligent theranostic alginate lyase (Aly)-NaNdF4 nanohybrids with a 220 nm sunflower-like structure (NaNdF4@DMS-Aly) through an enrichment-encapsulating strategy, which exhibited excellent photothermal conversion efficiency and the second near-infrared (NIR-II) luminescence. Benefiting from the site-specific targeting and biofilm-responsive Aly release from NaNdF4@DMS-Aly, we not only enabled noninvasive diagnosis but also realized Aly-photothermal synergistic therapy and real-time evaluation of therapeutic effect in mice models with Pseudomonas aeruginosa biofilm-induced pulmonary infection. Furthermore, such nanobiohybrids with a sheddable siliceous shell are capable of delaying the NaNdF4 dissolution and biodegradation upon accomplishing the therapy, which is highly beneficial for the biosafety of theranostic agents.
