Pyrimirhodomyrtone inhibits Staphylococcus aureus by affecting the activity of NagA

Huan Zeng,Minjing Cheng,Jingyi Liu,Chunxia Hu,Shilin Lin,Ruiqin Cui,Haibo Li,Wencai Ye,Lei Wang,Wei Huang

The epidemic of methicillin-resistant Staphylococcus aureus (MRSA) infections has created a critical health threat. The drug resistance of MRSA makes the development of drugs with new modes of action particularly urgent. In this study, we found that a natural product derivative pyrimirhodomyrtone (PRM) exerted antibacterial activity against S. aureus , including MRSA, both in vitro and in vivo . Genetic and biochemical studies revealed the interaction between PRM and N -acetylglucosamine-6-phosphate deacetylase (NagA) and the inhibitory effect of PRM on its deacetylation activity. We also found that PRM causes depolarization and destroys the integrity of the cell membrane. The elucidation of the antibacterial mechanism will inspire the subsequent development of new anti-MRSA drugs based on PRM.

From NaGa(IO3)3F to NaGa(IO3)2F2 and NaGa(IO3)4: The Effects of Chemical Substitution between F– Anions and IO3– Groups on the Structures and Properties of Gallium Iodates

Shuoxing Yang,Hongping Wu,Zhanggui Hu,Jiyang Wang,Yicheng Wu,Hongwei Yu

Introducing F– anions or substituting F– anions with IO3– groups has been proven to be ideal strategies for designing novel noncentrosymmetric (NCS) and polar materials, yet systematic investigation into the effect of F– anions or the substitution of IO3– for F– anions on structures and properties remains rarely explored. Herein, two new gallium iodates, NaGa(IO3)2F2 (1) and NaGa(IO3)4 (2), were successfully designed and synthesized based on NaGa(IO3)3F by introducing more F– anions and replacing F– anions with IO3 groups, respectively. Structurally, in compound 1, the adjacent [GaF3(IO3)3]3– polyanions are connected in an antiparallel manner, resulting in a complete cancellation of local polarity. While in compound 2, all IO3 groups in 2D [Ga(IO3)4]∞– layers are aligned, leading to large macroscopic polarization. Additionally, chemical substitution also results in a qualitative improvement in the functional properties of compound 2. It possesses strong SHG response (12 × KDP @1064 nm) and broad optical transparency, coupled with large birefringence (0.21 @1064 nm), showcasing its promise as a promising nonlinear optical (NLO) crystal. The effects of chemical substitution between F– anions and IO3– groups on the structures and properties are discussed in detail.

Efficient angiogenesis-based wound healing through hydrogel dressing with extracellular vesicles release

Zhengzhe Han,Lanlan Dong,Ang Li,Zongyue Li,Landie Fu,Zhichang Zhang,Xiang Li,Xiaolin Li

Wound healing and angiogenesis remain challenges for both clinical and experimental research worldwide. Periosteum-derived extracellular vesicles (P-sEVs) delivered by hydrogel dressings provide a potential strategy for wound defects to promote fast healing. In this study, we designed a NAGA/GelMA/Laponite/glycerol hydrogel wound dressing that can release P-sEVs to accelerate angiogenesis and wound healing (named [email protected] ) ( N -acryloyl glycinamide, NAGA). The wound dressing showed multiple functions, including efficient angiogenesis, tissue adhesion and a physical barrier. P-sEVs significantly enhanced the proliferation, migration, and tube formation of endothelial cells in vitro . The results of in vivo experiments showed that P-sE [email protected] accelerates the healing of a full-thickness defect wound model by stimulating the angiogenic process. The improved cell proliferation, tissue formation, remodeling, and re-epithelialization possibly resulted in the fast healing. This study shows that multifunctional hydrogel dressing combined with bioactive molecules can achieve fast and satisfactory wound healing in full-thickness wound defects and other related wounds.

A Facile Strategy for Synergistic Integration of Dynamic Covalent Bonds and Hydrogen Bonds to Surmount the Tradeoff between Mechanical Property and Self-Healing Capacity of Hydrogels

Minglei Ma,Jumin Yang,Zhanpeng Ye,Anjie Dong,Jinwei Zhang,Jianhua Zhang

The self-healable hydrogels have attracted increasing attention due to their promising potential for ensuring the durability and reliability of hydrogels. However, they still face a serious challenge to achieve a positive balance between mechanical and healing performance, especially for the room-temperature autonomous self-healable hydrogels. Herein, a simple but efficient strategy to fabricate a kind of dynamic boronate and hydrogen bonds dual-crosslinked double network (DN) hydrogel based on a UV-initiated one-pot in situ polymerization of N -acryloyl glycinamide (NAGA) in polyvinyl alcohol-borax slime is reported. The obtained PN- x /PB hydrogels, especially with high content of PNAGA, are shown to possess high mechanical strength, high toughness, and fatigue-resistance properties as well as excellent self-healability at room temperature (nearly 88% self-healing efficiency based on the strain compression test), due to the dynamic DN structure, and the combination of the adaptable and reconfigurable dynamic boronate bonds and hydrogen bonds. Considering the easily available materials and simple preparation process, this novel strategy should offer not only a kind of dynamic DN hydrogel with robust mechanical performance and high self-healing capability, but also enrich the methodological toolbox for synergistic integration of dynamic covalent bonds and hydrogen bonds to surmount the tradeoff between mechanical properties and self-healing capacity of hydrogels.
