Using polyethylene glycol to promote Nannochloropsis oceanica growth with 15 vol% CO2

Yanxia Zhu,Jun Cheng,Xiaodan Xu,Hongxiang Lu,Yangang Wang,Xi Li,Weijuan Yang

CO 2 capture with microalgae has been put forward in response to global concern on greenhouse gas emission . However, the short residence time and slow diffusion of CO 2 in water limits the growth of microalgae. In order to improve CO 2 transfer from gas phase to liquid phase and utilization by algal cells, polyethylene glycol 200 (PEG 200) was used as CO 2 absorbent to promote growth of Nannochloropsis oceanica with the bubbling of 15 vol% CO 2 . Total inorganic carbon (TIC) absorbed in culture medium remained constant at 5.6 mM when 15 vol% CO 2 was bubbled continuously. PEG 200 in the medium provided additional CO 2 absorption from 0.6 to 4.8 mM when PEG 200 concentration increased from 0.5 to 4 mM. The specific growth rate of N. oceanica reached the maximum (1.41 d −1 ) with 1 mM PEG 200 in culture medium, which was 21.5% higher than the specific growth rate without PEG 200. About 79% of the increase in biomass was attributed to the increased TIC with more CO 2 dissolution in culture medium because of PEG 200, and about 21% was attributed to PEG 200 itself utilized as an organic carbon source. In conclusion, PEG 200 as a CO 2 absorbent can effectively capture flue-gas CO 2 for algal growth.

Exogenous arginine induced metabolic modulation and biomass enhancement in Nannochloropsis oceanica under high CO2 stress

Ruixue Ma,Jun Cheng,Ying Liu,Dongwei Jia,Jiahao Wang

Given that excessive CO 2 induces reactive oxygen species (ROS) which inhibit photosynthesis and microalgal survival, this study explores how exogenous arginine enhances stress resilience and biomass yield of Nannochloropsis oceanica under high CO 2 stress. Arginine promoted NO synthesis, enhancing antioxidant enzyme activities to combat ROS damage. Transcriptomic analysis revealed that key enzymes in the Calvin cycle (PRK, GAPDH, FBPase and SBPase) and TCA cycle (CS, IDH, SCS and MDH) were significantly up-regulated, supporting carbon fixation and energy metabolism. Arginine also alleviated cell necrosis by extending the G1 phase, promoting DNA replication and protein synthesis, which increased the proportion of viable cells from 47 % to 84 %. Furthermore, the synthesis of chlorophyll a and carotenoid were enhanced, leading to 46 % increase in biomass yield. The results highlighted arginine as a promising metabolic regulator for enhancing stress tolerance and carbon fixation efficiency of N. oceanica under high CO 2 conditions.

Low-temperature and iron-doping to tailor Nannochloropsis Oceanica-derived carbon for high-performance dopamine biosensing

Zhuanzhuan Shi,Qianqian Fu,Jiatao Gu,Ruoyu Wei,Xiaoshuai Wu,Xiaokun Fu,Feng Wang,Ruizhi Zhu,Chunxian Guo,Chang Ming Li

Nannochloropsis Oceanica ( N. Oceanica ), widely distributed across global marine environments, is rich in proteins and amino acids, serving as an excellent source of nitrogen self-doped biomass carbon. Further modification and optimization of this microalga for constructing high-performance sensing platforms hold significant research and practical implications. This study strategically employs iron doping during low-temperature carbonization to modulate the structural and electronic properties of N. Oceanica -derived carbon. Results demonstrate that varying iron concentrations greatly accelerates graphitization, tailors the pore architecture, and significantly increases the specific surface area, thereby creating abundant catalytic active sites. The optimized iron-doped carbon material serves as a highly effective electrocatalyst when integrating into silk fabric-based screen-printed electrodes. These biosensors achieve exceptional dopamine (DA) detection performance, featuring a remarkably low detection limit (3 nM) and a broad linear range (0.01–2000 µM). Crucially, mechanistic analysis reveals that iron facilitates electrocatalytic activity and significantly enhance charge-transfer efficiency, thereby significantly boosting catalytic ability. This work establishes a novel, low-temperature iron-doping strategy for transforming biomass carbon into a cost-effective, eco-friendly, and high-performance platform for flexible electrochemical DA biosensors, paving a way for advanced bioanalytical applications.

Ammonium sulfate (coking byproduct) downregulates assimilatory nitrate pathway to save energy for carbon sequestration of Nannochloropsis oceanica with at stable pH using HEPES-NaOH

Ying Liu,Xiangjin Liang,Jun Lu,Yapeng Chen,Jun Liu,Baoying Wang,Ruixue Ma,Junchen Xu,Jun Cheng

How to utilize low-cost easily-acidified ammonium sulfate, a byproduct of coking industry, to farm microalgae for CO2 fixation is a difficult problem. Ammonium sulfate was mixed with sodium nitrate to cultivate Nannochloropsis oceanica at stable pH using HEPES-NaOH for solving acidification problem, and downregulate assimilatory nitrate reduction to save energy for improving carbon sequestration. Transcriptomic sequencing showed that nitrate assimilation consumed much ATP and reductivity derived from photosystem I, and competed with Calvin cycle for energy to limit CO2 fixation rate of microalgae in conventional nitrate medium. However, pure ammonium culture attenuated GS/GOGAT cycle (a main pathway for ammonium assimilation and amino acids synthesis) to limit nitrogen utilization rate of microalgal cells. In the mixed ammonium and nitrate supply modes, nitrogen metabolic pathway was down-regulated, while the Calvin cycle pathway was up-regulated to improve CO2 fixation. Nitrate reduced the extent of up-regulation of the ammonium transporter to prolong effective utilization time of ammonium. Asparagine synthetase activity in accessorial pathway of nitrogen assimilation substantially increased, while asparaginase was down-regulated to avoid harmful overaccumulation of ammonium in the cell. Down-regulation of tricarboxylic acid cycle attenuated intermediate 2-oxoglutarate shift to ammonium assimilation and improved efficiency of nitrogen utilization. The optimum condition for microalgal growth was at total nitrogen concentration of 5.28 mM and a nitrate to ammonium molar ratio of 2:3. Compared with the control (2.64 mM nitrate), the maximum photosynthetic electron transfer efficiency rETR increased by 34.9%, chlorophyll a concentration increased by 110% and average biomass growth rate increased by 33.7%.
