The antifungal activity of Nerol (NEL) was evaluated against Aspergillus niger , a known cause of grape spoilage, and possible modes of action were explored. The antifungal efficacy of NEL against A. niger has been proven in a dose-dependent manner through in vitro (mycelial growth and spore germination) and in vivo (grapes) tests. An obvious increase in the membrane permeability was detected through the determination of extracellular pH and conductivity. The damage in the cell membrane was measured by monitoring the influx of Propidium iodide (PI) using flow cytometry and was further verified through the inhibition of ergosterol synthesis in the cell membrane. NEL induced morphological changes, and the integrity of the A. niger fungal cells decreased with increasing NEL concentrations. A dose-dependent decrease in ergosterol production was also seen in A. niger cell membranes upon exposure to NEL. The mechanism-of-action study revealed that NEL activated a membrane-active mechanism that inhibits ergosterol synthesis and consequently compromised the membrane integrity of A. niger . As a result, the membrane permeability changes and causes cell death. The membrane lesion mechanism makes NEL a natural alternative to commercial fungicide for control of stored grape spoilage.

The antifungal efficacy of nerol (NEL) has been proved against Aspergillus flavus by using in vitro and in vivo tests. The mycelial growth of A. flavus was completely inhibited at concentrations of 0.8 μL/mL and 0.1 μL/mL NEL in the air at contact and vapor conditions, respectively. The NEL also had an evident inhibitory effect on spore germination in A. flavus along with NEL concentration as well as time-dependent kinetic inhibition. The NEL presented noticeable inhibition on dry mycelium weight and synthesis of aflatoxin B1 (AFB1) by A. flavus, totally restraining AFB1 production at 0.6 μL/mL. In real food system, the efficacy of the NEL on resistance to decay development in cherry tomatoes was investigated in vivo by exposing inoculated and control fruit groups to NEL vapor at different concentration. NEL vapors at 0.1 μL/mL air concentration significantly reduced artificially contaminated A. flavus and a broad spectrum of fungal microbiota. Results obtained from presented study showed that the NEL had a great antifungal activity and could be considered as a benefit and safe tool to control food spoilage.

Abstract Candida albicans invasion is one of the most serious fungal infections in clinical history. In recent years, because of the widespread use of immunosuppressive drugs, chemotherapy drugs, glucocorticoids, and broad-spectrum antibiotics, serious drug resistance has been reported; therefore, a new type of antifungal drug needs to be developed. In this study, we found that Nerol (NEL) had strong antimicrobial activity and 0.77 μL/mL NEL was the minimum inhibitory concentration (MIC) effective against C. albicans . We determined the change of the growth curve of NEL for C. albicans , to identify the trend of NEL activity against C. albicans . Through the determination of the ergosterol content and glucose-induced extracellular fluid acidification of NEL on C. albicans , we found that NEL inhibits the growth of C. albicans by destroying cell membranes. This finding was also supported by the expression of SAP (secreted aspartyl proteinase) involved in cell membrane synthesis. Finally, demonstrations of phenotype investigation, colony-forming unit (CFU) counts, and PAS (periodic acid-Schiff) staining were conducted to prove that NEL had the ability to treated mouse oral C. albicans infection and vaginal C. albicans infection. This research may help us to investigate new antimicrobial agents for treating C. albicans infections. Key Points • NEL can inhibit the growth of C. albicans. • NEL destroys the cell membrane formation and permeability of C. albicans. • NEL can treat vulvovaginal candidiasis and oropharyngeal candidiasis in mice. • NEL could be used as a possible antifungal agent.

Dental erosion is a common problem in dentistry, and it refers to the chronic pathological loss of dental hard tissues due to nonbacterially produced acids, primarily caused by the exposure of teeth to exogenous acids. Dietary factors play a pivotal part in the pathogenesis of dental erosion, with a high intake of acidic beverages leading to an increased prevalence of dental erosion in adolescents. Fluoride is mainly used in clinical practice to prevent dental erosion. However, long-term fluoride intake may lead to chronic fluorosis symptoms caused by fluoride overdose. Nano-coatings on dental surfaces have become a popular area of research in dental materials in recent years. The objective of this study was to develop a novel nerol-segmented waterborne polyurethane nano-coating to protect teeth from direct contact with an acidic environment and to provide a safe, effective method for preventing dental acid erosion.

Nerol, a linear monoterpenoid, is naturally found in essential oils of various plants and is widely used in the fragrance, food, and cosmetic industries. Nerol synthase, essential for nerol biosynthesis, has previously been identified only in plants that use NPP as the precursor. In this study, a novel fungal nerol synthase, named PgfB, was cloned and characterized from Penicillium griseofulvum. In vitro enzymatic assays showed that PgfB could directly convert the substrate GPP into nerol. Furthermore, the successful expression of PgfB and its homologous protein in Saccharomyces cerevisiae resulted in the heterologous production of nerol. Finally, crucial amino acid residues for PgfB’s catalytic activity were identified through site-directed mutagenesis. This research broadens our understanding of fungal monoterpene synthases and presents precious gene resources for the industrial production of nerol.

Monilinia fructicola , an extremely destructive pathogen, induces the postharvest decay of stone fruits. Nerol completely inhibited the growth of M. fructicola at a minimum inhibitory concentration of 0.2 mL/L. It disrupted mycelial membranes and released intracellular substances, such as nucleic acid, protein, and K + . Excessive reactive oxygen species (ROS) resulted in lipid peroxidation, which is responsible for membrane disruption. For the scavenging of excessive ROS, the antioxidant enzyme activities of catalase, superoxide dismutase, and peroxidase increased. However, glutathione (GSH) and GSH peroxidase activities decreased. Lasting oxidative stress disrupted mitochondria, which exhibited cristae loss and matrix transparency. The activities of key enzymes in tricarboxylic acid (TCA) cycle and ATPase were reduced, thereby causing ATP depletion. RNA sequencing revealed that nerol down-regulated the genes for L-glutamate and GSH synthesis while up-regulating those for GSH metabolism, resulting in a net GSH depletion. Metabolic disruptions occurred in membrane lipids and cell wall integrity, primarily involving steroids, sphingolipids, glycerophospholipids, and trehalose. Nerol disrupted the expressions of most genes involved in the TCA cycle. In vivo, 0.4 mL/L nerol completely inhibited the growth of M. fructicola on peaches. These results highlight nerol’s multiple antifungal mechanisms, supporting its development as a bio-preservative against M. fructicola .

The antifungal activity of Nerol (NEL) against Candida albicans , a pathogenic fungus, has a minimum inhibitory concentration (MIC) of 4.4 mM that causes noteworthy candidacidal activity through an apoptosis-like mechanism. Calcium (Ca 2+ ) levels and reactive oxygen species (ROS) production, which are the major causes of apoptosis, were determined in C. albicans cells treated with NEL and were found to increase, which related to mitochondrial dysfunction and disruption. A series of characteristic changes of apoptosis caused by NEL, including mitochondrial membrane depolarization, cytochrome c (cyt c) release, and metacaspase activation were examined using a flow cytometer and Western blot . The results showed that an increase in intracellular Ca 2+ and ROS led to dramatically decreased mitochondrial membrane potential (MMP); cyt c was also released from the mitochondria to the cytosol. Other early apoptotic features were also observed with the metacaspase activation. Finally, the morphological changes of the cells were observed, including phosphatidylserine (PS) externalization, nuclear condensation, and DNA fragmentation through Annexin V-FITC and PI double staining, TUNEL assay , and DAPI staining. The results supported the hypothesis that NEL was involved in the apoptosis of C. albicans cells not only at the early stages, but also at the late stages. In summary, NEL can trigger mitochondrial dysfunction and disruption via elevation of Ca 2+ and ROS leading to apoptosis in C. albicans . This research on the mechanism of cell death triggered by NEL against C. albicans has important significance for providing a novel treatment of C. albicans infections.

Botrytis cinerea , the second prevalent fungal pathogen worldwide, threatens a wide range of plants through its aggressive gray mold infection. This study investigates the antifungal effect of nerol against B. cinerea . Its minimum inhibitory concentration is 0.25 mL/L. Colony diameter reduced by 88.86 % at 48 h. Nerol exposure induced ROS accumulation, increasing malondialdehyde levels, which disrupted membrane integrity. It further induced mitochondrial dysfunction by suppressing the enzyme activities of succinic dehydrogenase and citrate synthase, reducing membrane potential, and promoting cytochrome C release and caspase-3 activation. Combined with phosphatidylserine externalization, these results suggested that nerol inhibited the growth of B. cinerea by a mitochondrial apoptosis pathway. Nerol regulated autophagy-related genes expression and activated autophagy. In postharvest cherry tomatoes, nerol controlled gray mold on postharvest fruits by inhibiting the growth of B. cinerea , but also inducing fruit resistance by elevating phenolic content by 50.13 %. The study elucidates nerol's multifaceted antifungal and fruit-protective effects against B. cinerea , supporting its development for the postharvest preservation of cherry tomatoes.

The contamination of food with Aspergillus flavus and subsequent aflatoxins is one of the most serious safety problems in the world. In this study of nerol (NEL)’s antifungal mechanism of action, we observed morphological and physiological changes in Aspergillus flavus. We found that NEL resulted in elevated levels of reactive oxygen species (ROS) and calcium ions (Ca 2+ ). On ROS assays, compared with the controls, the proportion of fluorescent cells treated with concentrations of 0.25, 0.5, 1, and 2 μL/mL NEL increased to 8.4 ± 1.07%, 10.2 ± 1.72%, 13.4 ± 0.50%, and 26.2 ± 4.21%, respectively. Increased mitochondrial dysfunction and oxidative stress induced by the interactions between Ca 2+ and ROS subsequently activate the release of cytochrome c and caspase activity. Characteristic changes of apoptosis were also observed via various detection methods, including phosphatidylserine externalization, nuclear condensation, and DNA fragmentation. Meanwhile, we found that the expression of CaMKs increased significantly in NEL-treated cells. In conclusion, our findings indicate that NEL has great potential as an eco-friendly antifungal agent for food preservation.

Ceratocystis fimbriata is the most devastating phytopathogen causing significant losses in post-harvest sweet potato. In this study, monoterpene nerol (NEL), the active compound in neroli essential oil, was found to dose-dependently inhibit the mycelial growth and spore germination of C. fimbriata at a minimum inhibitory concentration (MIC) of 0.25 mL L -1 . NEL vapor treatments significantly reduced the incidence and lesion diameter of black rot in sweet potato infected by the fungus and regulated the defense-related enzyme activity of phenylalanine ammonia lyase (PAL). Using RNA sequencing (RNA-seq) and biochemical assays, it was demonstrated that NEL treatment impaired cell membrane integrity via down-regulating the expression of ergosterol synthesis genes and reduced the ergosterol content. Moreover, an analysis of a series of apoptotic events revealed that NEL treatment caused mitochondrial membrane damage by reducing the mitochondrial membrane potential (MMP, Δψm), which led to down-regulation of genes involved in ATP production, then induced accumulation of intracellular reactive oxygen species (ROS) generation. Simultaneously, NEL caused nuclear chromatin condensation and concomitant DNA cleavage, which led to the up-regulation of DNA repair genes expression, and the cell-cycle arrest principally occurred at the G2/M phase in C. fimbriata . Altogether, these findings provide information about the underlying antifungal mechanism of NEL against C. fimbriata and suggest that NEL could be a useful alternative for controlling C. fimbriata in post-harvest spoilage of sweet potato.
