Background Despite developments in nanotechnology for use in the pharmaceutical field, there is still a need for implementation of this technology in agrochemistry. In this study, silver nanoparticles (AgNPs) were successfully prepared by a facile and an eco-friendly route using two different ligands, 2ʹ-amino-1,1ʹ:4ʹ,1″-terphenyl-3,3″,5,5″-tetracarboxylic acid (H4L) and 1,3,6,8-tetrakis (p-benzoic acid)-pyrene (TBAPy), as reducing agents. The physiochemical properties of the as-obtained AgNPs were characterized by scanning electron microscopy (SEM), energy-dispersive X-ray (EDX), X-ray diffraction (XRD) and transmission electron microscopy (TEM). The toxicity of H4L–AgNP and TBAPy–AgNP against the brown planthopper (BPH, Nilaparvata lugens) was also measured. Results SEM and TEM analyses demonstrated the formation of quasi-spherical AgNP structures in the presence of H4L and TBAPy. Insecticidal assays showed that TBAPy is less effective against N. lugens, with a median lethal concentration (LC50) of 810 mg/L, while the toxicity of H4L increased and their LC50 reached 786 mg/L 168 h posttreatment at a high concentration of 2000 mg/L. H4L–AgNPs were also highly toxic at a low concentration of 20 mg/L, with LC50 = ~ 3.9 mg/L 168 h posttreatment, while TBAPy–AgNPs exhibited less toxicity at the same concentration, with LC50 = ~ 4.6 mg/L.Conclusion sThese results suggest that the synthesized AgNPs using the two ligands may be a safe and cheaper method compared with chemical insecticides for protection of rice plants from pests and has potential as an effective insecticide in the N. lugens pest management program.

Cuticle, mainly composed of chitin and cuticular proteins (CPs), is a multifunctional structure of arthropods. CPs usually account for >1% of the total insect proteins. Why does an insect encode so many different CP genes in the genome? In this study, we use comprehensive large-scale technologies to study the full complement of CPs (i.e., the CP-ome) of the brown planthopper (BPH), Nilaparvata lugens, a major rice plant pest. Eight CP families (CPR, CPF, TWDL, CPLCP, CPG, CPAP1, CPAP3, and CPAPn) including 140 proteins in BPH, in which CPAPn is a CP family that we discovered. The CPG family that was considered to be restricted to the Lepidoptera has also been identified in BPH. As reported here, CPLCP family members are characterized by three conserved sequence motifs. In addition, we identified a testis protein family with a peritrophin A domain that we named TPAP. We authenticated the real existence of 106 proteins among the 140 CPs. RNA interference (RNAi) experiments were conducted against 135 CP genes in early- and late-instar nymphs and newly emerged female adults, demonstrating that 32 CPs were essential for BPH normal development or egg production. Combined RNAi experiments suggested redundant and complementary functions of the large number of CPs. Transcriptomic data revealed that the CP genes were expressed in a tissue-specific manner, and there were four clusters of developmental expression patterns. This study gives a comprehensive understanding of the roles of CPs in an insect cuticle.

The brown planthopper (BPH), Nilaparvata lugens , is a major insect pest of rice. Nitenpyram, a neonicotinoid insecticide, is a key compound for its control. Understanding the underlying resistance mechanisms is critical for developing effective management strategies. In this study, a nitenpyram-resistant (NR) N. lugens strain was established and characterized. Synergism tests and detoxification enzyme assays collectively demonstrate that enhanced metabolic detoxification plays a pivotal role in nitenpyram resistance in N. lugens . Quantitative RT-PCR (qRT-PCR) analysis revealed the significant overexpression of multiple candidate genes in the NR strain, including an ABC transporter gene ( ABCG1 ), four P450 genes ( CYP6ER1 , CYP3115A1 , CYP417A2 , and CYP418A1 ), four CarE genes ( CarE2 , CarE7 , CarE15 , and CarE26 ), and a UGT gene ( UGT 2B9 ). Additionally, molecular docking and molecular dynamics simulations further corroborated the high binding affinity and structural stability of these enzymes (ABCG1, CarE7, CarE15, CYP6ER1, CYP3115A1, CYP417A2, CYP418A1) and nitenpyram. Functional validation via RNA interference (RNAi) showed that silencing of ABCG1 , CarE15 , CYP3115A1 , CYP418A1 , and CYP6ER1 significantly increased nitenpyram susceptibility, confirming their functional role in resistance. Collectively, these findings elucidate a metabolic resistance mechanism mediated by multiple detoxification enzyme genes upregulation and provide a scientific basis for optimizing resistance management strategies to prolong the efficacy of nitenpyram in rice ecosystems.

Ecdysteroids , insect steroid hormones , play key roles in regulating insect development and reproduction. Hemipteran insects require ecdysteroids for egg production; however, ecdysteroid synthesis (ecdysteroidogenesis) details have not been elucidated. We identified all known genes encoding ecdysteroidogenic enzymes in Nilaparvata lugens and clarified their necessity during nymphal and ovarian development . We confirmed that N. lugens utilized 20-hydroxyecdysone as an active hormone. Assays using heterologous expression of enzymes in Drosophila S2 cells showed conserved functions of enzymes Neverland, CYP306A2, CYP314A1 and CYP315A1, but not CYP302A1. RNA interference and rescue analysis using 20-hydroxyecdysone demonstrated that most of the genes were necessary for nymphal development. The identified N. lugens enzymes showed conserved functions and pathways for ecdysteroidogenesis . Knockdown of ecdysteroidogenic enzyme genes in newly molted females caused failure of egg production: less vitellogenic and mature eggs in ovaries , fewer laid eggs and embryonic development deficiency of laid eggs. Considering the high expressions of ecdysteroidogenic enzyme genes in adults and ovaries, ecdysteroidogenesis in ovaries was critical for N. lugens ovarian development. Our study presents initial evidence that hemipteran insects require ecdysteroidogenesis for ovarian development.

Sometimes, crop breeding varieties demonstrate high resistance to target insects under laboratory conditions but exhibit significantly low resistance in the field. This research aimed to explain this phenomenon based on inter-species interactions among insects, as herbivory by one insect species can trigger physiological changes in plants that enhance their attraction to other insect species. The striped stem borer (SSB),Chilo suppressalis(Walker), and the brown planthopper (BPH),Nilaparvata lugens(Stål), are pests of rice (Oryza sativaL.) that cause major losses in grain production. In this study, we investigated BPH performance and behavior on the planthopper-resistant rice variety “Mudgo” with pre-feeding of SSB. BPHs showed better growth and development, as well as feeding behavior, on SSB-damaged plants compared to undamaged plants. Then, gene expression and phytohormone analysis revealed that jasmonic acid (JA) biosynthesis was induced by SSB feeding. The JA pathway is a central defense signaling hub in rice responding to chewing herbivores like SSB; however, our findings reveal that its induction can have contrasting ecological consequences, inadvertently reducing resistance to a subsequent piercing-sucking pest (BPH). Finally, we discovered that volatile emissions induced by SSB damage attracted BPH and benefited its development. In summary, we found that JA biosynthesis triggered by SSB herbivory played a vital role in rice defense against BPH. This provides insight into the molecular and biochemical mechanisms underlying BPH preferences for SSB-damaged rice plants. Our study emphasizes the crucial role of inter-species interactions in enhancing host plant resistance to insect pests and evaluating germplasm resistance. These findings can serve as a basis for controlling BPH.

The brown planthopper (BPH), Nilaparvata lugens is a typical pesticide-induced resurgent rice pest. A previous study showed that a fungicide, jinggangmycin (JGM)-treated rice led to markedly increased sugar content and (Insulin-like Peptide 2) ILP2 in response to sugar-mediated TOR signaling and stimulated fecundity in BPH. However, the role of the other ILPs in response to types of carbohydrate compounds remained poorly understood. The present studies showed that JGM treatment led to strongly increased glucose content and slightly increased sucrose and trehalose contents in rice plants. The 5th instar nymphs feeding with JGM-treated rice or exogenous glucose-supplied artificial diets markedly upregulated transcription of ILP2 and ILP3 , triggered ILP/Foxo signaling transduction, and further induced ILP3 secretion in the female brain. Moreover, silencing of ILP3 in the nymphal stage through RNAi led to a significant decrease of ILP3 secretion in the brain compared with dsGFP-treated females and inhibited ILP/Foxo signaling transduction. Suppression of ILP3 decreased soluble protein content in the ovary and fat body, inhibited Vg expression and protein synthesis, and further blocked ovarian development in female adults. Finally, depletion of ILP3 significantly influenced reproductive parameters, including the number of laid eggs, oviposition period, number of offspring, hatching rate, and population growth indexes of BPH. The current study revealed the mechanism of diet glucose regulating BPH reproduction by inducing ILP3 expression, which complements the theory of pesticide-induced resurgence of rice pests and also provides an important gene target for green control of BPH.

The brown planthopper (BPH), Nilaparvata lugens (Stål), is a major insect pest of rice in Asia and is mainly controlled with insecticides. Dinotefuran has been commercialized for control of N. lugens in China since 2013; however, the characteristics and mechanisms of resistance to dinotefuran are still unclear. Here, we conducted a long-term (from 2013 to 2021) and large geographical scale (11 provinces) resistance monitoring program for dinotefuran in the BPH in China. Before 2020, field populations of N. lugens have developed moderate resistance to dinotefuran, while since 2020, most field populations of N. lugens have developed high resistance. To develop a better resistance management strategy, a dinotefuran-resistant (Din-R) strain of N. lugens was selected in the laboratory to investigate the inheritance, possible mechanisms of resistance and potential fitness costs. This Din-R strain developed resistance to imidacloprid, nitenpyram and thiamethoxam. The dinotefuran resistance was controlled by multiple genes. RNA interference (RNAi)-mediated suppression of NlCYP6ER1 , NlCYP6CS1 and NlCYP314A1 expression increased the susceptibility of N. lugens to dinotefuran. Transgenic lines of Drosophila melanogaster expressing NlCYP6ER1 or NlCYP314A1 were less susceptible to all four neonicotinoids. Furthermore, the Din-R strain had a relative fitness of 0.64 with a substantially decreased survival rate, emergence rate and fecundity. Collectively, our results showed that dinotefuran resistance was mediated by multiple CYP genes. The high expression of these genes was related to the cross-resistance of N. lugens to other neonicotinoids. This study provided valuable information for facilitating the development of neonicotinoids resistance management strategies.

The brown planthopper (BPH, Nilaparvata lugens ) is a notorious sap-sucking insect pest that damages rice ( Oryza sativa ) plants throughout Asia. During BPH feeding, saliva enters rice plant tissues, whereas during oviposition egg-associated secretions (EAS) are deposited in damaged plant tissue. Dynamic changes in rice to planthopper salivary effectors have been widely reported. However, the effects of EAS from planthopper on rice immunity remains largely unexplored. In this study, we found that both infestation of rice by gravid BPH female adults and treatment with the EAS elicited a strong and rapid accumulation of jasmonic acid (JA), JA-isoleucine, and hydrogen peroxide in rice. EAS enhanced plant defenses not only in rice but also in tobacco, and these impaired the performance of BPH on rice, as well as the performance of aphids and whiteflies on tobacco. High-throughput proteome sequencing of EAS led to 110 proteins being identified and 53 proteins with 2 or more unique peptides being detected. Some proteins from BPH EAS were also found in the salivary proteome from herbivores, suggesting potential evolutionary conservation of effector functions across feeding and oviposition; however, others were only identified in EAS, and these are likely specifically related to oviposition. These findings point to novel proteins affecting interactions between planthoppers and rice during oviposition, providing an additional source of information for effector studies.

Insects have evolved highly sensitive olfactory sensory systems to detect plant hosts and mates, with plant volatiles playing an important role in informing insect behavior. Chemosensory proteins (CSPs) are thought to play a key role in this process, but in this respect, there is limited information on brown planthopper Nilaparvata lugens , one of the most destructive pests of rice. To expand our understanding of CSP function in N. lugens we explored expression profiles and binding characteristics of NlugCSP3. The ligands with higher binding affinity were also validated by molecular docking and behavioral assays. NlugCSP3 mRNA was expressed at relatively higher levels in antennae and abdomen of 3-day-old unmated macropterous males as well as in antennae of 3-day mated macropterous and brachypterous females. Fluorescence competitive binding assays revealed that 5 out of 25 candidate volatiles are strong binders (Ki < 10 μM). Behavioral assays revealed that nonadecane and 2-tridecanone, which have high binding affinities in fluorescence competition-binding assays, displayed strong attractiveness to N. lugens . Pursuing this further, molecular docking analysis identified key amino acid residues involved in binding volatile compounds. Overall, our data provide a base for further investigation of the potential physiological functions of CSP3 in Nilaparvata lugens , and extend the function of NlugCSP3 in chemoreception of N. lugens.

Slowpoke (Slo) channels are vital components of the nervous system, making them promising targets for developing neuromodulatory pesticides. However, there is limited research on the physiological roles and inhibitors of Slo channels in agricultural pests like the brown planthopper ( Nilaparvata lugens ). This study investigates the composition and essential functions of Slo channels in N. lugens through gene sequence analysis, patch-clamp electrophysiology, and inhibitor bioassays, offering valuable information for pest control approaches targeting these channels. The full coding sequence (3513 base pairs) of the N. lugens Slo gene was cloned, and five alternative splicing regions labeled A, C, E, G, and I were identified. Whole-cell patch-clamp experiments showed that the NlSlo channel exhibits typical features of calcium-activated large-conductance potassium channels, with a half-activation voltage (V 1/2 ) of 18.04 ± 0.94 mV. Removing the E region notably enhanced the channel's response to depolarization, while deleting the I region significantly reduced channel current. Among the tested inhibitors, iberiotoxin produced the strongest inhibition of the NlSlo channel at 66.21 % when applied at 10 μM, whereas plant-derived compounds inhibited the channel by 24.2 % to 48.95 % at the same concentration. In comparison, five pesticides—transfluthrin, flonicamid, tebufenozide, acynonapyr, and cypermethrin—showed inhibition rates ranging from 30.11 % to 42.74 % at 100 μM. Notably, injections of the specific inhibitor iberiotoxin significantly decreased both survival and movement abilities in N. lugens . This research highlights the critical role of Slo channel inhibition in affecting the survival and locomotion of N. lugens , providing new perspectives for developing pest management strategies targeting this species.

A previous study demonstrated that a β- N -acetyl- d -hexosaminidase ( NlHex4 ) from the brown planthopper could serve as a new potential target for biopesticides due to its requirement for molting, and lethal phenotypes were observed via RNA interference . However, the enzymatic properties of NlHex4 remain unclear. In the present study, NlHex4 was expressed, purified and characterized. Recombinant NlHex4 was functionally expressed in baculovirus-infected insect cells and purified with a molecular weight of approximately 70 kDa. Recombinant NlHex4 had an optimal pH and temperature of 6.0 and 60 °C, respectively. Moreover, recombinant NlHex4 exhibited thermal stability at 50 °C for at least 1 h. Furthermore, the activity of recombinant NlHex4 was inhibited by the metal ions Fe 2 + and Fe 3 + and the serine protease inhibitor phenylmethanesulfonyl fluoride. These results reveal the enzymatic properties of NlHex4 and provide a basis for future investigation of the interaction between HEX and chitin oligosaccharide .

Background Buprofezin, an insect growth regulator, has been widely used to control brown planthopper (BPH), Nilaparvata lugens , one of the most destructive pests of rice crops in Asia. The intensive use of this compound has resulted in very high levels of resistance to buprofezin in the field, however, the underpinning mechanisms of resistance have not been fully resolved. Results Insecticide bioassays using the P450 inhibitor piperonyl butoxide significantly synergized the toxicity of buprofezin in two resistant strains of BPH (BPR and YC2017) compared to a susceptible strain (Sus), suggesting P450s play a role in resistance to this compound. Whole transcriptome profiling identified 1110 genes that were upregulated in the BPR strain compared to the Sus strain, including 13 cytochrome P450 genes, eight esterases and one glutathione S -transferase. Subsequently, qPCR validation revealed that four of the P450 genes, CYP6ER1vA , CYP6CW1 , CYP4C77 , and CYP439A1 were significantly overexpressed in both the BRP and YC2017 strains compared with the Sus strain. Further functional analyses showed that only suppression of CYP6ER1vA , CYP6CW1 , and CYP439A1 gene expression by RNA interference significantly increased the toxicity of buprofezin against BPH. However, only transgenic Drosophila melanogaster expressing CYP6ER1vA and CYP439A1 exhibited significant resistance to buprofezin. Finally, the BPR strain was found to exhibit modest but significant levels of resistance to acetamiprid, dinotefuran and pymetrozine. Conclusions Our findings provide strong evidence that the overexpression of CYP6ER1vA and CYP439A1 contribute to buprofezin resistance in BPH, and that resistance to this compound is associated with low-level resistance to acetamiprid, dinotefuran and pymetrozine. These results advance understanding of the molecular basis of BPH resistance to buprofezin and will inform the development of management strategies for the control of this highly damaging pest. © 2022 Society of Chemical Industry.
